Background: It is reported that single nucleotide polymorphism (SNP) of the Translocase of outer mitochondrial membrane 40 homolog (TOMM40) gene is significantly associated with late-onset Alzheimer’s disease and cognitive decline in various ethnic groups. Abstract

Background and Objectives: The personal distress associated with caring for a family member has been well documented; however, questions about the burden of caregiving for centenarians and cross-national differences in the caregiving context, remain unanswered. Research Design and Methods: This study includes reports by caregivers of 538 near-centenarians and centenarians in the U.S. and Japan: 234 from the Georgia Centenarian Study and 304 from the Tokyo Centenarian Study. Basic descriptive and multivariate regression analyses were conducted. Mean levels of caregiver burden and near-centenarian and centenarians' characteristics (as predictors) for caregiver burden were compared between the U.S. and Japan. The near-centenarian and centenarians' functional capacity and personality were assessed as predictors. Results: Differential predictive patterns in caregiver burden were found in the two groups. In the U.S., near-centenarian and centenarians' agreeableness and conscientiousness were negatively associated with caregiver burden; whereas the near-centenarian and centenarians' neuroticism and number of diseases were positively associated with caregiver burden. In Japan, the near-centenarian and centenarians' activities of daily living, openness, and agreeableness were negatively associated with caregiving burden. Interaction effects between functional capacity and personality, on caregiver burden were observed only in the U.S. In the U.S., higher levels of agreeableness and openness significantly changed the level of caregiver burden associated with vision problems and a greater number of diseases. Discussion and Implications: Cross-national comparative predictors of caregiving burden between the two countries emphasized that caring for centenarians should be understood in the caregiving context, as well as the social context. Abstract

Background: Considerable variability exists in international prevalence and incidence estimates of dementia. The accuracy of estimates of dementia in the oldest-old and the controversial question of whether dementia incidence and prevalence decline at very old age will be crucial for better understanding the dynamics between survival to extreme old age and the occurrence and risk for various types of dementia and comorbidities. International Centenarian Consortium - Dementia (ICC-Dementia) seeks to harmonise centenarian and near-centenarian studies internationally to describe the cognitive and functional profiles of exceptionally old individuals, and ascertain the trajectories of decline and thereby the age-standardised prevalence and incidence of dementia in this population. The primary goal of the ICC-Dementia is to establish a large and thorough heterogeneous sample that has the power to answer epidemiological questions that small, separate studies cannot. A secondary aim is to examine cohort-specific effects and differential survivorship into very old age. We hope to lay the foundation for further investigation into risk and protective factors for dementia and healthy exceptional brain ageing in centenarians across diverse ethnoracial and sociocultural groups. Methods: Studies focusing on individuals aged ≥95 years (approximately the oldest 1 percentile for men, oldest 5th percentile for women), with a minimum sample of 80 individuals, including assessment of cognition and functional status, are invited to participate. There are currently seventeen member or potential member studies from Asia, Europe, the Americas, and Oceania. Initial attempts at harmonising key variables are in progress. Abstract

We present primary results from the Sequencing Quality Control (SEQC) project, coordinated by the US Food and Drug Administration. Examining Illumina HiSeq, Life Technologies SOLiD and Roche 454 platforms at multiple laboratory sites using reference RNA samples with built-in controls, we assess RNA sequencing (RNA-seq) performance for junction discovery and differential expression profiling and compare it to microarray and quantitative PCR (qPCR) data using complementary metrics. At all sequencing depths, we discover unannotated exon-exon junctions, with >80% validated by qPCR. We find that measurements of relative expression are accurate and reproducible across sites and platforms if specific filters are used. In contrast, RNA-seq and microarrays do not provide accurate absolute measurements, and gene-specific biases are observed for all examined platforms, including qPCR. Measurement performance depends on the platform and data analysis pipeline, and variation is large for transcript-level profiling. The complete SEQC data sets, comprising >100 billion reads (10Tb), provide unique resources for evaluating RNA-seq analyses for clinical and regulatory settings.

Abstract

To reduce the burden of caregivers and fulfill successful aging of family caregivers self, better supporting systems of caring elderly are requested. A questionnaire was mailed to each caregiver, who lives in Tokyo metropolitan. Quesions included burden of the caregivers and ADL of centenarians. Physical and psychological burden of caregivers is storongly affected by centenarian's ADL,cognitive level, and abnormal behaviors. Family caregivers of centenarians had a lower accumulated fatigue level and rated their health as very good to good, despite being older themselves and in spite of impoverished ADL of centenarians. Family caregivers of centenarians perceive their work not just duty or responsibility but worthwhile and satisfying. The care of centenarians might be possible model for successful caring.

Abstract

We examined the medical records of Japanese centenarians to clarify whether centenarians lived free from diseases, what kinds of morbidity they had experienced, and how such morbidity affected their autonomy. Three hundreds and thirty five centenarians living in Tokyo area were enrolled in this study. Informed consent was obtained in written form from the centenarians or their proxies. Their medical records were questioned by questionnaire and/or interview. The levels of activities of daily living (ADL) and cognitive function were also assessed using Barthel Index and mini-mental state examination (MMSE), respectively. The levels of ADL and cognitive function were significantly lower in female centenarians. About one third of centenarians escaped from grave diseases. Among their present and previous diseases, cataract, coronary and valvular heart diseases, hypertension, and cerebrovascular diseases were frequently reported. The incidence of fracture was significantly high in female centenarians. Another remarkable finding was very low incidence of diabetes mellitus (DM). The levels of ADL and cognitive function of the centenarians with the history of fracture were lower, suggesting that fracture, irrespective of its location, affected their autonomy. In conclusion, one third of Japanese centenarians escape from severe diseases, DM is unfavorable for longevity, and prevention of fracture may improve autonomy in the oldest old.

Abstract

The purpose of this study is to examine whether the personality contributes to longevity and centenarians[infinity]fADL. The NEO-FFI was administered to 272 centenarians and 1812 elderly people 65 through to 85 years old in Tokyo. Comparisons of each personality trait between the centenarians and the 65-85 elderly people showed the centenarians[infinity]f characteristics: Agreeableness was lower than the elderly in men and women, Neuroticism was higher in men, Conscientiousness and Extraversion were higher in women. Relations of the profiles of 5 personality traits and the ADL scores in the centenarians showed that the ADL level differed with the type of personality profiles. The higher level of ADL observed in the higher neuroticism and lower E, O, A, C type and the lower neuroticism and higher E, O, A, C type on ADL level than other personality types. These results suggested that the personality contributed to longevity and centenarians' ADL.

Abstract

It is reported that some personality traits in centenarians are different from those in younger controls. Recently polymorphism of dopamine receptor 4(D4DR) and serotonin transporter (5HTT) genes are shown to be related with novelty seeking and neurosis, respectively. It is not reported that polymorphism of these genes is associated with longevity. One hundred and forty five female centenarians living in Tokyo metropolitan area were enrolled in this study. As control group. 173 young females(average age 18.9 years old) were recruited. Polymorphism of D4DR in centenarians was not different from that in younger control and was not associated with the level of ADL and cognitive function of the centenarians. As for the polymorphism of 5HTT, frequency of 1 allele in centenarians was significantly higher than that in control. Centenarians with 1 allele have high level of ADL and serum albumin, compared with those with s allele. These data suggests that 5HTT polymorphism seems to be associated with longevity through behavioral and biological factors.
