Objective: The aim of the present study is to investigate the cytotoxic and apoptotic effects of oregano oil from Origanum minutiflorum, which is an endemic medicinal plant in Turkey, on NB2a neuroblastoma cells.Material and Method: Cell proliferation, apoptosis, and expression of inducible and endothelial nitric oxide synthase have been determined by MTT, TUNEL, and immunohistochemistry, respectively.Result and Discussion: The cell viability was significantly decreased gradually in NB2a cells when the concentrations of both oregano oil and doxorubicin were increased. IC50 doses were found to be10.75 µl/ml for oregano oil and 5µM for doxorubicin. Furthermore, expression of inducible and endothelial nitric oxide synthase, also the number of apoptotic cells was significantly enhanced in NB2a cells after treatment with oregano oil and doxorubicin. There was a remarkable increase in inducible and endothelial nitric oxide synthase expression levels and in the number of apoptotic cells by the dual application of these agents. In addition, oregano oil enhanced the apoptotic effect of doxorubicin. In conclusion, our results indicated that oregano oil of O. minutiflorum has a cytotoxic and apoptotic effect on NB2a cells and these effects may be related to its enhancing effects on inducible and endothelial nitric oxide synthase levels., Amaç: Bu çalışmanın amacı, Türkiye'de endemik bir tıbbi bitki olan Origanum minutiflorum'dan elde edien Oregano yağının NB2a nöroblastoma hücrelerine sitotoksik ve apoptotik etkilerini incelemektir.Gereç ve Yöntem: Hücre proliferasyonu, apoptoz, İndüklenebilir ve endotelyal nitrik oksit sentazın ekspresyonu sırasıyla MTT, TUNEL ve immünohistokimya ile belirlendi.Sonuç ve Tartışma: Hem Oregano yağı hem de doksorubisin derişimleri artırıldığında NB2a hücrelerinin canlılığı kademeli olarak azaldı. IC50 dozu Oregano yağı için 10.75 ul / ml doksorubisin için 5µM olarak belirlendi. Ayrıca, Oregano yağı ve doksorubisin ile muamele edildikten sonra NB2a hücrelerinde indüklenebilir ve endotelyal nitrik oksit sentazın ekspresyonu ve apoptotik hücre sayısı anlamlı olarak yükseldi. Bu ajanların ikili uygulaması ile indüklenebilir ve endotelyal nitrik oksit sentaz ekspresyon seviyelerinde ve apoptotik hücre sayısında dikkate değer bir artış olmuştur. Ek olarak, Oregano yağı, doksorubisinin apoptotik etkisini arttırmıştır. Sonuç olarak, bulgularımız O. minutiflorum’a ait Oregano yağının NB2a hücreleri üzerinde sitotoksik ve apoptotik bir etkiye sahip olduğuna ve bu etkilerin Oregano yağının indüklenebilir ve endotelyal nitrik oksit sentaz düzeylerini arttırıcı etkileri ile ilişkili olabileceğine işaret etmektedir.

Origanum minutiflorum’dan elde edilmiş esansiyel yağa maruz bırakılmış tüm hücre dizilerinde MTT testi aracılığıyla IC50 dozu hesaplandı. Her bir hücre dizisinin IC50 dozunda eNOS, p53, PCNA ve TUNEL için immunositokimya boyaması yapıldı. Sonuçlar Graphpad yazılımı kullanılarak one-way-ANOVA kullanılarak değerlendirildi. Bulgular: Meme kanseri hücrelerinin proliferasyonu oragano yağı tarafından inhibe edildi. Oregano yağı mezenkimal kök hücrelerinde herhangi bir sitotoksik etki göstermedi. eNOS ve p53 ile apoptozisin göstergesi TUNEL’de artış yoluyla oreagono yağının kanser hücrelerinde sitotoksik olduğu ortaya çıktı ve PCNA boyamada proliferasyonun azaldığı tespit edildi. Sonuç: Bizim verilerimize göre oregano yağı MDA-MB-231 ve MCF-7 meme kanseri hücrelerinde sitotoksik bir etkiye sahiptir ve diğer biyolojik aktiviteleri için aktif komponentlerini tanımlayacak testlere ihtiyaç vardır.

The breast cancer has become a threat to women's lives at an increasing rate. The inadequate treatments of surgical procedures and chemotherapy affect negatively quality of patient life. Therefore, there has been an increase in demand for alternative and supportive agents, especially for the use of herbal products. The purpose of this study is to evaluate the in vitro anti-tumor activity of Lysimachia savranii in breast cancer cell lines compared to normal adipose tissue derived mesenchymal stem cells. The adipose tissue derived mesenchymal stem cells as normal cells with MCF-7 and, MDA-MB-231 breast cancer cells were cultured. IC50 doses of Lysimachia savranii extract for each cells were determined via MTT assay. The immunocytochemical staining was performed for the oxidative stress by eNOS, whereas apoptosis were analyzed by TUNEL method. The data exhibited with H-score and apoptotic index. The plant extract inhibited the proliferation and survival in the breast cancer cells compared to the stem cells. Lysimachia savranii extract caused the increase of eNOS staining for oxidative stress and the TUNEL labeling for apoptosis. The further molecular techniques should be used to determine the biologic activity of Lysimachia savranii extract and its content should be investigated.

Breast cancer is one of the most repeatedly diagnosed cancers and the foremost cause of death among women worldwide. Medicinal plants can have an anticarcinogenic effect in some cancer types. Our aim is to search the cytotoxic effect of Origanum minutiflorum O. Schwarz & P.H. Davis on breast cancer cells in vitro conditions. Methods: In this study, MDA-MB-231 and MCF-7 breast cancer cell lines and adipose tissue derived mesenchymal stem cells were used. Essential oil from Origanum minutiflorum was exposed to the all cells and IC50 dose was calculated by MTT assay. We have examined immunocytochemical staining of eNOS, p53, PCNA and TUNEL at IC50 dose for each cells. The results were evaluated using one-way-ANOVA by Graphpad software. Results: The proliferation of breast cancer cells were inhibited by oregano oil. Oregano oil did not show any cytotoxic effect in mesenchymal stem cells. It was determined that cytotoxic effect of oregano oil on cancer cells was occurred through increase of eNOS and p53 staining for survival and the TUNEL labeling for apoptosis, and decreased of PCNA staining for proliferation. Conclusion: According to our data, oregano oil has cytotoxic effect on MDA-MB-231 and MCF-7 breast cancer cell lines, and it needs to be analysis to detect active components for other biological activities.

The treatment of breast cancer includes chemotherapy, radiotherapy and chemotherapy. Recently, alternative agents have been searched for breast cancer, especially medicinal plants because their anti-cancer effects. It has been reported that the genus of Lysimachia has anti-proliferative effect on cancer cells. In our study we aimed to evaluate in vitro anti-cancer activity of Lysimachia savranii on migration of breast cancer cells. Breast cancer cell lines MCF-7 and MDA-MB-231 and also adipose tissue derived mesenchymal stem cells as normal cells. IC50 doses of Lysimachia savranii extract and Doxorubicin for each cells were calculated via MTT assay. In vitro wound model with (+) plus shape was created using pipette tips for migration analysis and IC50 doses were exposed. The oxidative stress, vascularization and apoptosis were analyzed by immunocytochemical staining of eNOS, iNOS, VEGF and TUNEL. Data was evaluated by Graphpad software using one-way analysis of variance. Lysimachia savranii and Doxorubicin have anti-proliferative effect on breast cancer cells. Breast cancer cell migration was inhibited by Lysimachia savranii. There was an increase of eNOS and iNOS stainings, and apoptotic cells whereas there was a decrease of VEGF staining. Our findings indicate that Lysimachia savranii extract is a potential herbal product for breast cancer therapy.

The natural products are promising products for anti-proliferative effects in cancer. In this study our aim was to analyze the neurotoxic effect of Origanum minutiflorum O. Schwarz & PH. Davis on neuroblastoma cells in in vitro conditions. The neuroblastoma cells were treated with Oregano oil from Origanum minutiflorum and doxorubicin, and cytotoxic activities of these agents were evaluated by MTT assay for IC50 doses. The immunocytochemistry staining of eNOS, iNOS and TUNEL assay were performed. The results were analyzed using Graphpad software for one-way-ANOVA. The immunoreactivities of eNOS and iNOS were increased in neuroblastoma cells treated with oregano oil and doxorubicin. Both of Oregano oil and doxorubicin showed inhibitory activity against neuroblastoma cells via oxidative stress. Also the number of apoptotic cells was greater than the control group. The oregano oil enhanced the cytotoxic activity of doxorubicin. The compound of oregano oil should be considered with the other neuroblastoma cells for neurotoxicity.

Recently, involvement of Wnt antagonists in oncogenesis has been demonstrated in several cancers. The investigation of their contribution to lung carcinogenesis is still under investigation. We aimed to investigate whether there is a susceptibility or preventive effect of Wnt antagonist gene polymorphisms on the development and/or prognosis of LC. We investigated 110 LC patients and 160 controls. Single-nucleotide polymorphisms of Wnt antagonist genes including DKK2 (rs17037102), DKK3 (rs3206824), DKK3 intron4 G/C (rs7396187), DKK4 (rs2073664), and sFRP4 (rs1802074) were analyzed using nested polymerase chain reaction and restriction fragment length polymorphism. Results showed that patients with DKK3 AA compared with controls have a decreased risk of LC (adjusted for smoking habit, body mass index, and familial history) (P = 0.02; odds ratio [OR],0.08; 95% confidence interval [95% CI], 0.01-0.7). It was found that, for sFRP4 polymorphism, patients with GG and GA genotypes versus AA genotype controls showed a decreased risk for LC (P = 0.01; [OR, 0.19; 95% CI, 0.05-0.73 for GG genotype]; [OR = 0.18, 95% CI, 0.04-0.72 for GA genotype]). In addition, a decreased risk of LC was also found for the genotype combination of DKK3 (rs3206824) GG and sFRP4 AG + GG (P = 0.004; OR, 0.12; 95% CI, 0.02-0.58).
