Acne vulgaris is one of the most common dermatological diseases and is strongly connected with the pathological growth of the Cutibacterium acnes. More than half of the cultures of this bacterium are resistant to antibiotics, resulting in the proposal of the use of antibacterial peptides as an alternative to traditional antibiotics. Ascorbic acid (AA) and its antioxidant properties may ally in acne therapy. The aim of this study was to determine the influence of the selected antibacterial peptides in the presence of ascorbic acid and 3-O-ethyl-ascorbic acid (EAA) on the properties of the monolayer formed by phosphatidylinositol. Studies of the properties of the phosphatidylinositol monolayer were carried out using the Langmuir–Wilhelmy balance. The recorded compression isotherms, hysteresis loops, and surface pressure values recorded at specific time intervals were evaluated to assess the influence of ascorbic acid and its derivatives in the presence of antimicrobial peptides on the stability and organization of phosphatidylinositol monolayers. The addition of AA to the subphase caused a faster phase transition at over 60 Å2/molecule and significantly reduced the plateau surface pressure by about 20% in most of the systems tested. The studied monolayers were found to be in the expanded liquid state (40.23–49.95 [mN/m]) or in the transition between the expanded and condensed liquid phase (51.47–60.98 [mN/m]). Compression and decompression isotherms indicated the highest flexibility of the systems at 20 °C and 25 °C. The surface pressure versus time dependence indicated the stability of the phosphatidylinositol monolayer with 3-O-ethyl–ascorbic acid and antimicrobial peptides up to 35 °C. [ABSTRACT FROM AUTHOR]

The health-promoting properties of black elderberry are related to its high content of polyphenols (natural antioxidants), which eliminate free radicals and prevent the formation of oxidative stress responsible for many diseases. The aim of this work was to determine, the anti-radical effect of Sambucus nigra infusions based on the reaction with 2,2-diphenyl-1-picrylhydrazyl (DPPH) and galvinoxyl (Glv) radicals and to determine the function describing the disappearance curves of these radicals. The antioxidant properties of infusions obtained from the flowers and fruits of this plant were tested using the modified Brand-Williams method using DPPH and Glv radicals. Higher antioxidant activity towards both the DPPH and Glv radicals was found in flowers compared to fruits. In addition, it was found that the process of quenching radicals in the reaction with Sambucus nigra infusions proceeds in accordance with the assumptions of second-order reaction kinetics. The infusion obtained from flowers quenched radicals faster than fruit infusions. The applied second-order kinetics equation may enable estimation of antioxidants levels in natural sources of radicals. [ABSTRACT FROM AUTHOR]

The aim of the study was to evaluate the effect of the peptide structure (WKWK)2-KWKWK-NH2, P4 (C12)2-KKKK-NH2, P5 (KWK)2-KWWW-NH2, P6 (KK)2-KWWW-NH2 on their physicochemical properties. The thermogravimetric method (TG/DTG) was used, which made it possible to observe the course of chemical reactions and phase transformations occurring during the heating of solid samples. Based on the DSC curves, the enthalpy of the processes occurring in the peptides was determined. The influence of the chemical structure of this group of compounds on their film-forming properties was determined using the Langmuir–Wilhelmy trough method and was followed by molecular dynamics simulation. Evaluated peptides showed high thermal stability and the first significant mass loss occurred only at about 230 °C and 350 °C. The analysis of the compressibility coefficient of individual peptides indicates that all formed peptide monolayers were in the expanded liquid phase. Their maximum compressibility factor was less than 50.0 mN/m. Its highest value of 42.7 mN/m was achieved in a monolayer made of P4. The results obtained in molecular dynamic simulation indicate that non-polar side chains played an important role in the properties of the P4 monolayer, and the same applies to P5, except that a spherical effect was observed here. A slightly different behavior was observed for the P6 and P2 peptide systems, where the type of amino acids present had an influence. Purpose: To determine the effect of the temperature of water used for the preparation of paracetamol and ascorbic acid mixture on its stability, as well as to assess the influence of humidity on the stability of single components and their mixtures. Methods: The stability of the mixtures in aqueous medium was evaluated with the aid of UV-Vis spectrophotometer interfaced with a computer. Spectral analysis was adapted to monitor changes in the aqueous medium of a commercial paracetamol and ascorbic acid mixture, an extemporaneously prepared mixture of paracetamol and ascorbic acid, and the individual preparations of paracetamol and ascorbic acid. Results: The degradation rate was lower in commercial preparation (6.80 × 10-3 min-1), compared to that of the extemporaneously prepared ascorbic acid/paracetamol mixture (2.30 × 10-2 min-1). The decomposition of the commercial product in aqueous medium was 3.38 times slower than that of the extemporaneously prepared mixture. Ascorbic acid, tested under the same conditions as the commercial product, was unstable in aqueous solutions, with a degradation rate of 1.17×10-2 min-1. Ascorbic acid, dissolved in water, degraded completely within 4 h at room temperature, whereas paracetamol remained stable under the same conditions for 11 days. Conclusion: The individual drugs in their original form retained their stability for 72 h, but some of the mixtures, in particular, the extemporaneously prepared ones showed more rapid degradation. Extemporaneous preparation of paracetamol/ascorbic acid liquid mixtures should not be encouraged. [ABSTRACT FROM AUTHOR]
