Determination of nuclear and/or cytoplasmic expression of β-catenin by immunohistochemistry in patients with endometrial cancer (EC) may constitute a potential diagnostic method for identifying patients with a catenin β1 (CTNNB1) gene mutation and those at risk of disease recurrence. The present study aimed to investigate β-catenin expression patterns in hysterectomy specimens of patients with endometrioid type EC using immunohistochemistry, and to examine the prognostic impact of β-catenin. The study was a single-institutional, retrospective cohort trial enrolling consecutive patients with a postoperative histopathological diagnosis of endometrioid EC who underwent hysterectomy between January 2015 and December 2018. Histopathology slides from 75 patients were stained with a monoclonal antibody targeting the β-catenin protein. Any percentage of nuclear staining, whether focal or diffuse, was considered 'β-catenin nuclear-positive'. The cytoplasmic staining reaction of β-catenin was assessed based on the percentage of stained cells and staining intensity. Immune-reactivity score (IRS) values were determined by multiplying the scores for the percentage of staining and staining intensity. IRS values 0 to 2 were regarded as negative expression, 3 to 4 as low expression, 6 to 8 as moderate expression, and 9 to 12 as high expression. Recurrence-free survival (RFS) was used as the prognostic endpoint. Only 2 out of 75 tissue samples (2.7%) exhibited nuclear β-catenin expression, with a low staining percentage of 5%. By contrast, cytoplasmic staining was observed in all samples (100%). According to the IRS findings, 1.3% of the samples exhibited negative cytoplasmic expression, 42.7% low expression, 38.7% moderate expression and 17.3% high expression. Cox regression analysis revealed that staining with β-catenin, either nuclear or cytoplasmic, had no impact on RFS, and stage was the sole independent prognostic factor. In conclusion, based on these results, β-catenin expression in endometrioid EC was revealed to be mostly cytoplasmic, with only 2.7% of tissue samples exhibiting nuclear expression. Despite the theoretical benefits, the favorable effect of preoperative carbohydrate loading on postoperative morbidity remains controversial. Most of the outcomes reported in the literature are derived from non-gynecologic surgery data, with only one study involving a limited number of patients specifically in gynecological oncology. The present study aimed to investigate the impact of carbohydrate loading, as a single element of enhanced recovery after surgery protocols, on postoperative course and morbidity in patients undergoing debulking surgery for epithelial ovarian cancer (EOC). The present study was a non-randomized, prospective cohort trial enrolling patients with EOC who underwent surgery between June 2018 and December 2021. An oral carbohydrate supplement with a dose of 50 g was given to patients 2-3 h before anesthesia. Data on postoperative course and morbidity were collected and compared with data of a historical cohort including consecutive patients who underwent surgery without a carbohydrate loading between January 2015 and June 2018. Analyses were performed on a total of 162 patients, including 72 patients in the carbohydrate loading group and 90 patients in the control group. Median length of hospital stay (11 days vs. 11 days; P=0.555), postoperative days 1-7 serum c-reactive protein levels (P=0.213), 30-day readmission (11.6% vs. 11.5%, P=0.985), 30-day relaparotomy (2.8% vs. 3.4%, P=0.809) and 30-day morbidity (48.6% vs. 46.7%; P=0.805) were comparable between the cohorts. No significant differences in grades of morbidities were identified between the cohorts (P=0.511). Multivariate analysis revealed that the sole independent risk factor for any postoperative morbidity was operative time. Aim: The aim of our study is to examine the clinical, surgical, and pathological factors of stage 1C adult granulosa cell tumor (AGCT) patients and to investigate the effects of adjuvant therapy on recurrence and survival rates in this patient group., Methods: Out of a total of 415 AGCT patients treated by 10 tertiary oncology centers participating in the study, 63 (15.2%) patients with 2014 FIGO stage IC constituted the study group. The FIGO 2014 system was used for staging. Patient group who received adjuvant chemotherapy was compared with patient group who did not receive adjuvant chemotherapy in terms of disease-free survival (DFS), and disease-specific survival., Results: The 5-year DFS of the study cohort was 89%, and the 10-year DFS was 85%. Those who received adjuvant chemotherapy and those who did not were similar in terms of clinical, surgical and pathological factors, except for peritoneal cytology. In the univariate analysis, none of the clinical, surgical or pathological factors were significant for DFS. Adjuvant chemotherapy and type of treatment protocol had no impact on DFS., Conclusion: Adjuvant chemotherapy was not associated with improved DFS and overall survival in stage IC AGCT. Objective: The Papanicolaou (Pap) smear test is a standard screening test that detects cervical lesions and cancers. In this multicentric study, we performed a retrospective analysis of cytological results associated with atypical glandular cells, not otherwise specified (AGC-NOS)., Methods: We retrospectively reviewed Pap smear tests that resulted as AGC-NOS. A total of 254 women who underwent colposcopy due to a Pap smear result of AGC-NOS were included the study between 2003 and 2021. The ages, Pap smear results, HPV results if any, colposcopic biopsy results, endocervical and endometrial pathology results, and management of these patients were analysed., Results: Two hundred fifty-four patients with AGC-NOS Pap smear results were included in the study. A total of 70 (27.6%) patients had cervical and endometrial premalignant or malignant lesions. Malignancy was observed in 17 (6.7%) patients (endometrium, n = 11 [4.3%]; cervix, n = 6 [2.4%]). Isolated premalignant or malignant lesions of the cervix and endometrium were detected in 57 (22.4%) and 12 (4.7%) patients, respectively., Conclusions: Patients diagnosed with AGC-NOS should undergo a careful evaluation with all clinicopathological features. Because cancer of the cervix and endometrium is not rare in patients diagnosed with AGC-NOS, colposcopic examination with endocervical sampling should be a priority based on a cervicovaginal smear. This study aimed to investigate the effects of the 6-week small-sided games training (SSGs) vs. high-intensity interval training (HIIT) on the psychophysiological and performance responses, and technical skills of young basketball players. Thirty-two male players (age: 14.5 ± 0.5 years of age) were randomly divided into SSGs group ( n = 16) and HIIT group ( n = 16) training methods thrice per week for 6 weeks. The players in the SSGs group performed two 5-8 min of 2 vs. 2 with 2 min rest periods, while the players in HIIT performed 12-18 min of runs at intensities (90 to 95%) related to the velocity obtained in the 30-15 intermittent fitness test (IFT). Pre-testing and post-testing sessions involved assessments of Yo-Yo Intermittent Recovery Test level 1, 30-15 intermittent fitness test, 5 and 30 m sprint times, vertical jump height, repeated sprint ability, defensive and offensive agility, and technical skills. The SSGs group demonstrated significantly higher agility-based technical responses in terms of the control dribbling and shooting skills ( d = 1.71 vs. 0.20, d = 1.41 vs. 0.35, respectively) compared with the HIIT group. Conversely, the HIIT induced greater improvements in 30 m sprint times ( d = 3.15 vs. 0.68). These findings provided that SSGs in youth basketball players may allow similar positive physical adaptations to HIIT, with an extra advantage of improving technical skills while improving enjoyability.

This study aimed to compare the order effects of combined small-sided games (SSGs) and high-intensity interval training (HIIT) on the psychophysiological responses and physical and technical performances of young soccer players. Twenty-four soccer players (aged 14.63 ± 0.71 years) were randomly divided into SSGs + HIIT ( n = 12) and HIIT + SSGs ( n = 12) for 6 weeks. The SSGs consisted of two 4-16 min rounds of 2, 3, and four-a-side games with 2 min of passive resting, whereas the HIIT consisted of 6-10 min of high-intensity runs at varying intensities (from 90 to 100%). Pre-test and post-test elements included a 5-30 m sprint test, countermovement jump test, zigzag agility test with the ball and without the ball, repeated sprint ability test, speed dribbling ability test, three-corner run test, and Yo-Yo Intermittent Recovery Test level 1. Both combined training interventions produced similar improvements in physical performance and technical responses ( p ≥ 0.05, d values ranging from 0.40 to 1.10). However, the combined HIIT + SSGs training produced meaningfully lower perceived exertion ( p = 0.00, d = 2.98) and greater physical enjoyment ( p = 0.00, d = 4.28) compared with the SSGs + HIIT intervention. Furthermore, the SSGs + HIIT group showed a higher training load than those from the HIIT + SSGs group for all weeks ( p ≤ 0.05, d values ranging from 1.36 to 2.05). The present study's results might be used by coaches and practitioners to design training programmes for youth soccer players.

Insulin regulates the glucose homeostasis by inducing tyrosine phosphorylation of insulin receptor substrate (IRS) proteins. IRS1 is the best studied member of this family and insulin-induced Tyrosine phosphorylation of (YXXM) motifs provides docking site for SH2 domain-containing proteins. Recent studies have suggested that genetic and/or environmental factors may affect the expression and phosphorylation levels of IRS1, and these could be important for development of insulin resistance. To shed light to the molecular basis of type 2 diabetes we wanted to determine whether YXXM motifs are genetically modified in these patients. We have isolated mononuclear cells of eighteen type 2 diabetes patients and prepared genomic DNA and protein lysates from these cells. The genomic DNA was used to sequence IRS1 gene, and protein lysates were used to determine the expression and phosphotyrosine levels of IRS1 after insulin stimulation. Although, we did not detect any mutations at/or near the YXXM coding regions in patients' DNA, immunprecipitation analysis of IRS1 indicated decreased levels of expression and tyrosine phosphorylation of IRS1 in patient's samples compared to that of healthy controls. Our results suggest that mononuclear cells of patients can be used to test the levels of insulin responsiveness before therapy.
