Abstract

During the second third of last century, the Orinoco Crocodile (Crocodylus intermedius) underwent a hunting process driven by the demand from the North American, European, and Japanese leather industry, resulting in a sharp decline of its populations. Currently, only two known remaining populations of this Critically Endangered species persist in the Colombian Orinoquía: in the Guayabero-Duda-Lozada and the Cravo Norte-Ele-Lipa River Systems. The latter has been the only population subject of study, including recent surveys and local conservation initiatives such as egg and hatchling ranching. Despite suggestions for population recovery based on the observed increase in clutches in the area, information regarding its genetic status has been pending assessment. This research aims to provide a genetic characterization of this remaining population and to evaluate the diversity recovered during a period of the egg ranching initiative. For this purpose, we utilized variable molecular markers, specifically 17 microsatellite loci, nuclear DNA. Despite revealing intermediate levels of genetic diversity, we identified an effective population size of 11.5–17, well below the minimum values proposed for short-term subsistence. While no evidence of inbreeding was found, it is acknowledged as a potential risk based on the population's history. Additionally, we detected a historical bottleneck possibly influenced by arid periods affecting the region since the Pleistocene. While the evaluated population presents a unique opportunity for C. intermedius conservation, it also exposes a high risk of entering the extinction vortex. 19. FABP4 Enhances Lipidic and Fibrotic Cardiac Structural and Ca2+ Dynamic Changes.

Abstract

BACKGROUND: Adipocyte FABP4 (fatty acid-binding protein 4) is augmented in the epicardial stroma of patients with longstanding persistent atrial fibrillation. Because this molecule is released mainly by adipocytes, our objective was to study its role in atrial cardiomyopathy, focusing our attention on fibrosis, metabolism, and electrophysiological changes. These results might clarify the role of adiposity as a mediator of atrial cardiomyopathy. METHODS: We used several preclinical cellular models, epicardial and subcutaneous stroma primary cell cultures from patients undergoing open heart surgery, human atrial fibroblasts, atrial cardiomyocytes derived from human induced pluripotent stem cells and isolated from adult mice, and Nav1.5 transfected Chinese hamster ovary cells. Fibrosis, glucose, mitochondrial and adipogenesis activity, gene expression, and proteomics were determined by wound healing, enzymatic, colorimetric, fluorescence assays, real-time quantitative polymerase chain reaction, and TripleTOF proteomics. Molecular changes were analyzed by Raman confocal microspectroscopy, calcium dynamics by confocal microscopy, and ion currents by patch clamp. Epicardial, subcutaneous, and atrial fibroblasts and cardiomyocytes were incubated with FABP4 at 100 ng/mL. RESULTS: Our results showed that FABP4 induced fibrosis, glucose metabolism, and lipid accumulation on epicardial and subcutaneous stroma cells and atrial fibroblasts. Besides, it modified lipid content and calcium dynamics in atrial cardiomyocytes without effects on INa. CONCLUSIONS: FABP4 exerts fibrotic and metabolic changes on epicardial stroma and modifies lipid content and calcium dynamic on atrial cardiomyocytes. Abstract

The development of self-assembled dyes able to display J-aggregation-induced emission has gained a renewed interest as an appealing and distinctive approach for the design of smart photoactive materials. F-BODIPY dyes have been exploited to this aim since J-type supramolecular organization has been effectively photoinduced under specific surrounding conditions (hydrophilic or constrained media) although, up to date, it has never been observed in pure organic media. Herein, we report the first systematic study of the self-assembly properties of F-BODIPYs in pure organic solvents and analyze in depth the control exerted by the substitution pattern of its carbon skeleton into the formation of emissive J-aggregates. Spontaneous J-aggregation in F-BODIPYs is evidenced by their lasing properties and supported by the analysis of their solid-state photophysics and X-ray crystalline packing as well as by molecular dynamics in solvent cages. These studies point out the key role of the stereoelectronic properties of the substituents orthogonally grafted at the dipyrrin core (meso and C-2 and C-6 positions) as the main driving force to control the molecular stacking arrangement of F-BODIPYs in common organic solvents. The understanding of the interplay between the molecular structure and the properties of the resulting aggregates sheds light on the workability of BODIPY-based self-assembly as a tool for advanced luminescent materials.

Abstract

The VElocity foR Direct particle Identification spectrometer (VERDI) is a 2E-2v fission spectrometer that allows the measurement of the total mass distribution of secondary fission fragments with a resolving power of 1-2 u. It consists of two time-of-flight (ToF) arms, with one Micro Channel Plate (MCP) detector and up to 32 Silicon PIPS (Passive Implanted Planar Silicon) detectors per arm. The MCPs provide the start timing signals and the PIPS detectors provide both the energy and the stopping ToF signals. In real conditions, the PIPS signals are affected by the formation of plasma from the interaction between the heavy ions and the detector material. The plasma contributes to a reduction in signal amplitude, resulting in a Pulse Height Defect (PHD), and introduces a signal delay, known as Plasma Delay Time (PDT). An experiment to characterize the PDT and PHD was performed at the LOHENGRIN recoil separator of the Institut Laue Langevin (ILL). Characteristic fission fragments from the 239Pu(n,f) reaction were separated based on their A/Q and E/Q ratios, allowing the measurement of a wide range of energies from 21 to 110 MeV and masses between 80 and 149 u. Six PIPS detectors were characterized to study their individual responses to the PDT and PHD effects. The signals were recorded in a digital acquisition system to completely exploit the offline analysis capabilities. Achieved combined timing and energy resolutions for fission fragments varied between 72(2) ps and 100(4) ps and 1.4% - 2% (FWHM), respectively. Preliminary PHD and PDT data are presented from the masses A=85, 95, 130 and 143. The PHD trends are strongly correlated with both the ion energy and mass. The PDT, on the other hand, shows a strong variation as a function of the ion kinetic energy but a smaller dependence on the ion mass.

Abstract

The presence of F or CN substituents at boron in BODIPYs causes a dramatic effect on their reactivity, which allows their chemoselective postfunctionalization. Thus, whereas 1,3,5,7-tetramethyl B(CN)2-BODIPYs displayed enhanced reactivity in Knoevenagel condensations with aldehydes, the corresponding BF2-BODIPYs can experience selective aromatic electrophilic substitution (SEAr) reactions in the presence of the former. These (selective) reactions have been employed in the preparation of BODIPY dimers and tetramers, with balanced fluorescence and singlet oxygen formation, and all-BODIPY trimers and heptamers, with potential application as light-harvesting systems.

Abstract

therosclerosis is a complex disease that can lead to life-threatening events, such as myocardial infarction and ischemic stroke. Despite the severity of this disease, diagnosing plaque vulnerability remains challenging due to the lack of effective diagnostic tools. Conventional diagnostic protocols lack specificity and fail to predict the type of atherosclerotic lesion and the risk of plaque rupture. To address this issue, technologies are emerging, such as noninvasive medical imaging of atherosclerotic plaque with customized nanotechnological solutions. Modulating the biological interactions and contrast of nanoparticles in various imaging techniques, including magnetic resonance imaging, is possible through the careful design of their physicochemical properties. However, few examples of comparative studies between nanoparticles targeting different hallmarks of atherosclerosis exist to provide information about the plaque development stage. Our work demonstrates that Gd (III)-doped amorphous calcium carbonate nanoparticles are an effective tool for these comparative studies due to their high magnetic resonance contrast and physicochemical properties. In an animal model of atherosclerosis, we compare the imaging performance of three types of nanoparticles: bare amorphous calcium carbonate and those functionalized with the ligands alendronate (for micro- calcification targeting) and trimannose (for inflammation targeting). Our study provides useful insights into ligand-mediated targeted imaging of atherosclerosis through a combination of in vivo imaging, ex vivo tissue analysis, and in vitro targeting experiments.

Abstract

Owing to the relevance of fluorescently labeled carbohydrates in the study of biological processes, we have investigated several routes for the preparation of saccharides covalently linked to borondipyrromethene (BODIPY) fluorophores. We have shown that BODIPY dyes can be used as aglycons through synthetic saccharide protocols. In particular, a per-alkylated 8-(2-hydroxy-methylphenyl)-4,4¿-dicyano-BODIPY derivative, which withstands glycosylation and protection/deprotection reaction conditions without decomposition, has been used in the stepwise synthesis of two fluorescently labeled trisaccharides. These saccharides displayed high water solubility and a low tendency to (H-)aggregation, a phenomenon that causes loss of photophysical efficiency in BODIPYs. Two additional synthetic strategies toward glyco-BODIPYs have also been described. The first method relies on a Ferrier-type C-glycosylation of the BODIPY core, leading to linker-free carbohydrate¿BODIPY hybrids. Secondly, the application of the Nicholas propargylation reaction to 1,3,5,7-tetramethyl BODIPYs provides access to 2,6-dipropargylated BODIPYs that readily undergo CuAAC reactions with azido-containing sugars. From a photophysical standpoint, the BODIPY-labeled saccharides could be used as stable and fluorescent water-soluble chromophores, thereby addressing one of the current challenges in molecular imaging.

Abstract

Lysyl oxidase (LOX) and LOX-like 1-4 (LOXL 1-4) enzymes catalyze the cross-linking of elastin and collagen in the extracellular matrix, facilitating cell migration and invasion. The inhibition of these enzymes, particularly LOXL2, has been suggested as a therapeutic strategy to prevent breast cancer metastasis. In this work, new natural LOXL2 inhibitors were searched from Aeollanthus rydingianus, a medicinal plant rich in bioactive products. Five pimarane diterpenoids, two isolated from the plant and three derivatives, were tested. These compounds have been described for their bioactive properties such as anti-tumor, anti-inflammatory, analgesic, and antibacterial activities. In this regard, we intended to explore the mechanisms of these compounds by studying their effects on LOXL2 activity. Two pimarane diterpenoids showed a mild LOXL2 inhibitory activity as evaluated by an Amplex Ultra Red-based technique. The cytotoxicity of the most active compound was analyzed by the MTT assay in the MDA-MB-231 cell line, representative of triple-negative breast cancer. This compound decreased cell viability as single agent and increased the cytotoxic effect of doxorubicin. Its glycoconjugate was considerably more toxic, likely due to a higher uptake by cancer cells., A lisil oxidase (LOX) e as proteínas tipo lisil oxidase 1-4 (LOXL 1-4) catalisam a ligação cruzada de elastina e colagénio na matriz extracelular, facilitando a migração e invasão celulares. A inibição destas enzimas, particularmente LOXL2, tem sido sugerida como estratégia terapêutica para prevenir metástases do cancro da mama. Neste trabalho, novos inibidores naturais de LOXL2 foram investigados a partir de Aeollanthus rydingianus, uma planta medicinal rica em produtos bioativos. Cinco diterpenóides de pimarano, dois isolados da planta e três derivados foram testados. Estes compostos foram previamente descritos pelas suas propriedades bioativas, tais como antitumoral, anti-inflamatória, analgésica e antibacteriana. Neste sentido, pretendemos explorar os mecanismos destes compostos, estudando os seus efeitos na atividade da LOXL2. Dois diterpenóides pimaranos mostraram uma ligeira atividade inibitória da LOXL2, avaliada por uma técnica baseada no Amplex Ultra Red. A toxicidade do composto mais ativo foi analisada pelo ensaio MTT na linha celular MDA-MB-231, representativa do cancro da mama triplo negativo. Este composto diminuiu a viabilidade celular como agente isolado e aumentou o efeito citotóxico da doxorubicina. O seu glicoconjugado foi consideravelmente mais tóxico, provavelmente devido a uma maior captação pelas célula tumorais.
