This research was carried out to examine the effects of different doses of bacterial formulation comprised of a mixture of Bacillus megaterium TV-91C, Pantoea agglomerans RK-92 and Kluyvera cryocrescens TV-113C strains on the plant development and bract quality of poinsettias (Euphorbia pulcherrima Willd. ex Klotzsch), which are one of the most important potted plants grown for their fleshy bracts of the horticultural sector. The study was carried out in a climate-controlled research greenhouse from 8 August 2018 to 15 January 2019 in Erzurum (Turkey). Rooted cuttings of poinsettia [E. pulcherrima Willd. ex Klotzsch cv. Christmas Feelings (CvF)] were used as plant materials in the study. Each of the solutions containing bacterial suspensions 52.5 (T1), 105 (T2), 210 (T3), 420 (T4) and 840 (T5) mL · L−1 of water was diluted 5 times with water and 200 mL · pot−1 was applied to the plant rhizosphere. This study revealed that positive changes incurred in plant height, main flower stem length, plant crown width, bract diameter, green leaf and bract leaf area, leaf total nitrogen content and nitrate reductase enzyme activity parameters in poinsettia plants with increasing doses of the bacterial formulation. This effect has reached the maximum level in the number of bracts, main flower stem length, bract diameter, nitrate reductase enzyme activity, plant fresh weight and maximal root length parameters with the T5 application which has the highest bacterial formulation concentration. The most efficient application of glutamine synthetase enzyme activity was determined as T4.

Despite the growing demand for organic products, research on organic farming (OF) such as genotype screening, fertilizer application and nutrition uptake remains limited. This study focused on comparisons of the apparent recovery efficiency of nitrogen fertilizer (REN) in rice grown under OF and conventional farming (CF). Thirty-two representative conventional Japonica rice varieties were field grown under five different treatments: control check (CK); organic farming with low, medium and high levels of organic fertilizer (LO, MO and HO, respectively); and CF. Comparisons of REN between OF and CF classified the 32 genotypes into four types: high REN under both OF and CF (type-A); high REN under OF and low REN under CF (type-B); low REN under OF and high REN under CF (type-C); and low REN under both OF and CF (type-D). Though the yield and REN of all the rice varieties were higher with CF than with OF, organic N efficient type-A and B were able to maintain relatively high grain yield under OF. Physiological activities in flag leaves of the four types from booting to maturity were subsequently investigated under OF and CF. Under OF, high values of soil and plant analyzer development (SPAD) and N were observed in type-B varieties, while in contrast, both indexes slowly decreased in type-C varieties under CF. Moreover, the decline in N content in type-C and D varieties was greater under OF than CF. The decrease in glutamine synthetase (GS), glutamic-pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT) activity in flag leaves was smaller under OF than CF in type-A and B varieties, while in contrast, type-C and D varieties showed an opposite trend. The findings suggest that OF slows the decline in key enzymes of N metabolism in organic N-efficient type rice, thus maintaining a relatively high capacity for N uptake and utilization and increasing yield during the late growth period. The present study was aimed at understanding the role of different hosts in ammonium transporter1;2 expressions and glutamine synthetase(GS) activity and their effects on the growth parameters in the sandal. Sandal plant associated with leguminous host expressed better growth parameters. GS activity of leguminous hosts alone and in host associated sandals was analyzed using GS transferase assay. Highest GS activity was expressed in Mimosa pudica-sandal association compared to other leguminous and non-leguminous host associations. The association of N fixing host with sandal enhanced C and N levels in order to maintain the C/N value. The role of ammonium transporters in N nutrition of sandal-host association was elucidated by cloning AMT1;2 from the leaves, haustoria and roots of host associated sandal and quantifying the relative expression by the $$ 2^{{ - \Delta \Delta {\text{C}}_{\text{T}} }} $$ method. SaAMT1;2 was strongly up-regulated in leaves, roots and haustoria of leguminous host associated sandal compared to non-leguminous host associations. The relative increase in SaAMT1;2 expressions and up-regulated GS activity positively affected the growth parameters in sandal when associated with leguminous hosts. [ABSTRACT FROM AUTHOR]

Guanosine (GUO) has been shown to act as a neuroprotective agent against glutamatergic excitotoxicity by increasing glutamate uptake and decreasing its release. In this study, a putative effect of GUO action on glutamate transporters activity modulation was assessed in hippocampal slices subjected to oxygen and glucose deprivation (OGD), an in vitro model of brain ischemia. Slices subjected to OGD showed increased excitatory amino acids release (measured by d-[H]aspartate release) that was prevented in the presence of GUO (100 µM). The glutamate transporter blockers, DL-TBOA (10 µM), DHK (100 µM, selective inhibitor of GLT-1), and sulfasalazine (SAS, 250 µM, Xc system inhibitor) decreased OGD-induced d-aspartate release. Interestingly, DHK or DL-TBOA blocked the decrease in glutamate release induced by GUO, whereas SAS did not modify the GUO effect. GUO protected hippocampal slices from cellular damage by modulation of glutamate transporters, however selective blockade of GLT-1 or Xc- system only did not affect this protective action of GUO. OGD decreased hippocampal glutamine synthetase (GS) activity and GUO recovered GS activity to control levels without altering the kinetic parameters of GS activity, thus suggesting GUO does not directly interact with GS. Additionally, the pharmacological inhibition of GS activity with methionine sulfoximine abolished the effect of GUO in reducing d-aspartate release and cellular damage evoked by OGD. Altogether, results in hippocampal slices subjected to OGD show that GUO counteracts the release of excitatory amino acids, stimulates the activity of GS, and decreases the cellular damage by modulation of glutamate transporters activity. In order to determine the best growing media for Gerbera jamesonii under alkaline conditions, a factorial experiment was conducted with two factors, namely: (i) Growing medium, and (ii) Concentrations of bicarbonate (NaHCO3) in nutrient solution. Results showed that increasing the concentration of NaHCO3 from 0 to 40 mM in nutrient solution significantly decreased plant growth, maximal quantum yield of PSII photochemistry (Fv/Fm), photosynthesis Performance Index (PI), Glutamine Synthetase (GS) activity, Leaf Relative Water Content (LRWC), Chlorophylls (Chl a, b and tota Chl) and carotenoids. Under alkalinity stress, reduction in vegetative growth, Fv/Fm, PI, LRWC, GS activity and photosynthesis pigments content was the lowest in Coconut Fiber (CF) substrate compared to the other substrates. Furthermore, plants grown in CF substrate had higher soluble sugars and proline content than those in other substrates. On the other hand, plants grown on Perlite (P) substrate had the highest reduction in growth and physiological characteristic in alkaline conditions. The alleviation of alkalinityinduced growth inhibition of plants by CF substrate may be related to improvement of photosynthesis, enhancement of GS enzyme activity and osmotic regulation. It is concluded that the use of CF substrate could provide a useful tool to improve alkalinity tolerance of gerbera plants under NaHCO3 stress. [ABSTRACT FROM AUTHOR]
