Background/aim: The objective of this study was to investigate and analyze the behavioral and neurochemical effects of monosodium glutamate (MSG) injections at various and subsequent dosages on male Wistar rats during the neonatal period., Materials and methods: In order to determine the behavioral and neurochemical effects of MSG, the experiment was implemented on neonatal male Wistar rats and the test was repeated for various MSG dosages. After completing the experiment, additionally, levels of dopamine, GABA, catecholamine (dopamine, noradrenaline, and adrenaline) and glutamate in the brain cells of the decapitated rats were also measured using the ELISA method., Results: Considering the results of the behavioral test, when we compared the test values of the control group with the values of the MSG-injected groups we noted that there were significant differences in the statistical figures obtained. Additionally, we found that the statistical figures of some neurochemical parameters were also significantly different when we compared the values of the MSG group with the control values., Conclusion: MSG injection has a clear effect on the neurochemical parameters, learning memory, and locomotor activities of rats.

Arginine vasopressin (AVP), which is synthesized and secreted in the hypothalamic paraventricular nucleus (PVN), is the most important bioactive substance in the pain modulation. Our pervious study had shown that AVP plays an important role in pain modulation in caudate nucleus (CdN). The experiment was designed to investigate the source of AVP in CdN by the nucleus push-pull perfusion and radioimmunoassay. The results showed that: (1) pain stimulation increased the AVP concentration in the CdN perfusion liquid, (2) PVN decreased the effect of pain stimulation which was stronger in both sides than in one side of PVN cauterization; and (3) L-glutamate sodium would excited the PVN neurons by the PVN microinjection that could increase the AVP concentration in the CdN perfusion liquid. The data suggested that AVP in the CdN might come from the PVN in the pain process, i.e., AVP in the PVN might be transferred to the CdN to participate in the pain modulation. © 2010 Elsevier Inc. BACKGROUND: Taste receptors are expressed not only in taste buds but also in the gastrointestinal tract. It has been hypothesized that these receptors may play a role in satiety and food intake. OBJECTIVE: This study investigated the effect of intraduodenal tastant infusions (bitter, sweet, and umami) on food intake, hunger and fullness, gastrointestinal symptoms, and gastrointestinal peptide release. DESIGN: Fifteen healthy volunteers [6 male; mean +/- SEM age: 23.9 +/- 2.0 y; mean +/- SEM body mass index (in kg/m2): 22.4 +/- 0.3] received 5 treatments in a double-blind, randomized, placebo-controlled crossover design. Test days started with the insertion of a nasoduodenal catheter followed by a standardized liquid breakfast. Participants received an intraduodenal infusion 150 min after breakfast, containing quinine (bitter), rebaudioside A (sweet), monosodium glutamate (umami), a combination of the 3 tastants, or placebo (tap water) over a period of 60 min. Food intake was measured during an ad libitum meal, and visual analog scales were used to monitor gastrointestinal complaints and hunger and fullness scores. Blood samples were drawn at regular intervals for cholecystokinin, glucagon-like peptide 1 (GLP-1), and peptide YY (PYY) analysis. RESULTS: Infusion of the combination of tastants substantially decreased food intake (422 +/- 97 kcal vs. 486 +/- 104 kcal for placebo, P < 0.05), whereas both a combination of tastants and umami decreased hunger scores compared with placebo. No change in cholecystokinin, GLP-1, or PYY concentrations was observed during the infusions. Intraduodenal infusions of the tastants did not result in gastrointestinal symptoms. CONCLUSIONS: Intraduodenal infusion of umami and a combination of tastants inhibits feelings of hunger, but only the latter also reduces food intake. However, these alterations were not accompanied by changes in the plasma concentrations of the gut-derived peptides cholecystokinin, GLP-1, or PYY. Stable suspensions of nanogold (NG) and nanosilver (NS) with mean particle diameter 50 and 49 nm, respectively, were prepared by laser ablation of metals in water. To assess rat's pulmonary phagocytosis response to a single intratracheal instillation of these suspensions, we used optical, transmission electron, and semi-contact atomic force microscopy. NG and NS were also repeatedly injected intraperitoneally into rats at a dose of 10 mg/kg (0.5 mg per mL of deionized water) three times a week, up to 20 injections. A group of rats was thus injected with NS after oral administration of a "bioprotective complex" (BPC) comprised of pectin, multivitamins, some amino acids, calcium, selenium, and omega-3 PUFA. After the termination of the injections, many functional and biochemical indices and histopathological features of the spleen, kidneys and liver were evaluated for signs of toxicity, and accumulation of NG or NS in these organs was measured. From the same rats, we obtained cell suspensions of different tissues for performing the RAPD test. It was demonstrated that, although both nanometals were adversely bioactive in all respects considered in this study, NS was more noxious as compared with NG, and that the BPC tested by us attenuated both the toxicity and genotoxicity of NS. © 2013 by the authors; licensee MDPI, Basel, Switzerland.

Saliva from parotid glands plays a role in taste perception. Parotid saliva is also stimulated by tastants. The aim of this work is to investigate the effects of different tastants on the parotid salivary response in six subjects. Five tastants were given in different concentrations in solution and held in the mouth for 10 s. The flow rate, protein concentration, and pH of secreted parotid saliva were monitored continuously for 5 min. Stimulation by tastants on flow rate response consists of an immediate rise in flow followed by a plateau and a rapid return to prestimulus flow. Response of pH results in a slower increase while protein concentration consists in a slower decrease, both followed by a return to prestimulus levels in about 4 min. From a resting flow rate of about 140∈μL/min, an increase in flow rate to 370∈μL/min was caused by stimulation for 10 s with 10 mL of solutions of 0.01 M citric acid, 0.13 M MgSO4, 0.25 M monosodium glutamate, 0.5 M NaCl, or 0.5 M sucrose. Comparisons of the different tastants showed that the pH of stimulated parotid saliva increased linearly (r∈=∈0.9), irrespective of the nature of the tastant. Protein concentration decreased (r∈=∈-0.45) and protein amount increases (r∈=∈0.58) with increase in flow rate for all tastants. Corrected for the effects of flow rate, protein amount depended on the nature of the tastant with the greatest secretion after stimulation by citric acid. Flow rate was largely responsible for pH but tastant appears to play an additional role with flow rate on protein secretion.

Neonatal administration of monosodium glutamate (MSG) in rats causes definite neuroendocrine disturbances which lead to alterations in many organ systems. The possibility that MSG could affect tooth and salivary gland physiology was examined in this paper. Male and female pups were injected subcutaneously with MSG (4 mg/g BW) once a day at the 2nd, 4th, 6th, 8th and 10th day after birth. Control animals were injected with saline, following the same schedule. Lower incisor eruption was determined between the 4th and the 10th postnatal days, and the eruption rate was measured between the 43rd and the 67th days of age. Pilocarpine-stimulated salivary flow was measured at 3 months of age; protein and amylase contents were thereby determined. The animals treated with MSG showed significant reductions in the salivary flow (males, -27%; females, -40%) and in the weight of submandibular glands (about -12%). Body weight reduction was only about 7% for males, and did not vary in females. Saliva of MSG-treated rats had increased concentrations of total proteins and amylase activity. The eruption of lower incisors occurred earlier in MSG-treated rats than in the control group, but on the other hand the eruption rate was significantly slowed down. The incisor microhardness was found to be lower than that of control rats. Our results show that neonatal MSG treatment causes well-defined oral disturbances in adulthood in rats, including salivary flow reduction, which coexisted with unaltered protein synthesis, and disturbances of dental mineralization and eruption. These data support the view that some MSG-sensitive hypothalamic nuclei have an important modulatory effect on the factors which determine caries susceptibility. A administração neonatal de glutamato monossódico (MSG) em ratos provoca distúrbios neuroendócrinos que acarretam alterações em vários sistemas orgânicos. Neste trabalho, avaliamos as repercussões desse tratamento sobre dentes e glândulas salivares. Ratos machos e fêmeas recém-nascidos foram injetados com MSG (4 mg/g peso corporal, s.c.) uma vez ao dia nos 2º, 4º, 6º, 8º e 10º dias após o nascimento; o grupo controle recebeu solução salina no mesmo esquema. O momento da erupção do incisivo inferior foi determinado entre o 4º e o 10º dia de vida, e o ritmo de erupção foi medido entre o 43º e o 67º dia. O fluxo de saliva e o conteúdo salivar de proteína e amilase foram determinados sob estimulação com pilocarpina aos 3 meses de idade. Os animais tratados com MSG mostraram reduções significativas do fluxo salivar (machos: -27%; fêmeas: -40%) e do peso das glândulas submandibulares (cerca de -12%). Apenas em machos houve discreta redução do peso corporal (7%). A saliva dos animais tratados com MSG apresentou aumento na concentração de proteínas totais e na atividade amilásica. A erupção dos incisivos inferiores ocorreu mais precocemente nos ratos tratados do que nos controles, porém a taxa de erupção apresentou-se significativamente reduzida. A microdureza também foi menor nos animais tratados. Nossos resultados mostram que o tratamento de ratos recém-nascidos com MSG causa um quadro definido de alterações buco-dentais no animal adulto, traduzidas por redução do fluxo de saliva (sem redução da síntese protéica) e distúrbios de mineralização e erupção dentárias. Esses dados apontam para o importante papel modulador que certos núcleos hipotalâmicos sensíveis ao MSG exercem sobre os fatores que regulam a suscetibilidade à cárie.

In previous studies we observed an increased incidence of hyperplasia in the epithelium of the urinary bladder of rats fed cereal-based stock diet supplemented with 6% monosodium glutamate (MSG) for 3 months. Hyperplasia was not enhanced, however, when 6% MSG was fed in a purified casein diet. Further studies have been conducted to identify the dietary factor that caused the different response with the two diets. Feeding MSG had a marked alkalizing effect on the urine. Rats fed purified diet produced urine of higher acidity than did those fed stock diet, a finding attributed to the greater excess of base in the stock diet. When diets with a considerable excess of cations were fed, urinary pH showed a characteristic pattern of widely differing values during a 24-hr period, with high values (pH ≥ 8) for several hours of darkness, when food intake was high, declining during the day to a minimum at the end of the light period. Hyperplasia of the bladder epithelium was induced not only by feeding MSG, but also by feeding 5% of the alkalizing salt KHCO3, both in purified diet and in stock diet. The epithelial response to an alkalizing substance was prevented by simultaneous feeding of the acidifying salt NH4Cl. These findings indicate that the bladder changes induced by MSG are attributable to its alkalizing properties rather than to MSG per se. Moderate to severe hyperplasia of the bladder epithelium was induced also by feeding 5% NH4Cl in purified diet, a procedure accompanied by a further lowering of urinary pH. These findings showed that hyperplasia of the bladder epithelium of rats can be induced both by acidifying and by alkalizing the urine through manipulation of the acid-base balance of the basal diet. There is thus a possibility that, in carcinogenicity studies, administration of compounds to rats in the form of a salt may lead to erroneous conclusions. No. of bitstreams: 0 Previous issue date: 2003-12-01 The present study was designed to evaluate the effects of aerobic exercise training on glucose tolerance and insulin secretion of obese male Wistar rats (monosodium glutamate [MSG] administration, 4mg/g-body weight, each other day, from birth to the 14th day). Fourteen weeks after the drug administration, the rats were separated into two groups: MSG-S (sedentary) and MSG-T (T = swimming, 1 h/day, 5 days/week, with an overload of 5% body weight for 10 weeks). Rats of the same age and strain injected with saline were used as control (C) and subdivided into two groups: C-S and C-T. Insulin and glucose responses during an oral glucose tolerance test (GTT) were evaluated by the estimation of the total areas under serum insulin (AI) and glucose (AG) curves. Glucose-induced insulin secretion by isolated pancreatic islets was also evaluated. MSG-S rats showed higher AI than C-rats while MSG-T rats presented lower AI than MSG-S rats. No differences in AG were observed among the 4 groups. Pancreatic islets from MSG-rats showed higher insulin secretion in response to low (2.8) and moderate (8.3 mM) concentrations of glucose than those from their control counterparts and no differences were observed between MSG-S and MSG-T rats. These results provide evidences that the hyperinsulinemia at low or moderate glucose concentrations observed in MSG-obese rats is, at least in part, a consequence of direct hypersecretion of the B cells and that chronic aerobic exercise is able to partially counteract the hyperinsulinemic state of these animals without disrupting glucose homeostasis.
