Abstract Oxygen in marine sediments regulates many key biogeochemical processes, playing a crucial role in shaping Earth’s climate and benthic ecosystems. In this context, branched glycerol dialkyl glycerol tetraethers (brGDGTs), essential biomarkers in paleoenvironmental research, exhibit an as-yet-unresolved association with sediment oxygen conditions. Here, we investigated brGDGTs in sediments from three deep-sea regions (4045 to 10,100 m water depth) dominated by three respective trench systems and integrated the results with in situ oxygen microprofile data. Our results demonstrate robust correlations between diffusive oxygen uptake (DOU) obtained from microprofiles and brGDGT methylation and isomerization degrees, indicating their primary production within sediments and their strong linkage with microbial diagenetic activity. We establish a quantitative relationship between the Isomerization and Methylation index of Branched Tetraethers (IMBT) and DOU, suggesting its potential validity across deep-sea environments. Increased brGDGT methylation and isomerization likely enhance the fitness of source organisms in deep-sea habitats. Our study positions brGDGTs as a promising tool for quantifying benthic DOU in deep-sea settings, where DOU is a key metric for assessing sedimentary organic carbon degradation and microbial activity.

Introduction The human gut microbiota is associated with gestational diabetes mellitus (GDM), which imposes a risk of developing long-term health problems for mother and child. Most studies on GDM and microbiota have been cross-sectional, which makes it difficult to make any conclusions on causality. Furthermore, it is important to assess if a dysbiotic microbiota is passed from the mother to the child, and then being at risk of developing metabolic health problems later in life. The DANish Maternal and Offspring Microbiome study aims to identify gut microbiota-related factors involved in metabolic dysfunction in women with GDM and their offspring. Importantly, the study design allows for early detection of biological changes associated with later development of metabolic disease. This could provide us with unique tools to support early diagnosis or implement preventative measures.Methods and analysis Pregnant women are included in the study after the 11–14 weeks’ prenatal ultrasound scan and followed throughout pregnancy with enrolment of the offspring at birth. 202 women and 112 children have been included from North Denmark Regional Hospital and Aalborg University Hospital in Denmark. Mother and child are followed until the children reach the age of 5 years. From the mother, we collect faeces, urine, blood, saliva, vaginal fluid and breast milk samples, in addition to faeces and a blood sample from the child. Microbiota composition in biological samples will be analysed using 16S rRNA gene sequencing and compared with demographic and clinical data from medical charts, registers and questionnaires. Sample and data collection will continue until July 2028.Ethics and dissemination The study protocol has been approved by the North Denmark Region Committee on Health Research Ethics (N20190007). Written informed consent is obtained from all participants prior to study participation. Study results will be published in international peer-reviewed journals and presented at international conferences. The results will also be presented to the funders of the study and study participants.

Pregnancies complicated by type 1 diabetes (TID) are associated with an increased risk of obstetric and neonatal adverse outcomes. Optimal glycemic control prior to and through pregnancy is crucial to reduce complications. The use of diabetes technology is rapidly increasing. The aim of the study was to investigate the use and effects of diabetes technology in pregnant women with type 1 diabetes. A retrospective cohort study was conducted; 84 women were included in the analysis and were divided into subgroups according to their glucose monitoring method and insulin delivery method. HbA1c values declined during pregnancy in all subgroups with no significant difference between the subgroups. A difference was, however, found in birth weight z-scores. Women using a sensor and an insulin pump had larger babies compared to women without these treatment modalities. The results of the study indicate that diabetes technology, including insulin pumps and/or glucose sensors are not superior to self-monitoring blood glucose measurement and multiple daily injection insulin therapy, which is comforting in the light of the unequal access to health benefits.

Abstract In the hadal zone of the ocean (6–11 km), the characteristics of sinking marine snow particles and their attached microbial communities remain elusive, despite their potential importance for benthic life thriving at extreme pressures (60–110 MPa). Here, we used simulation experiments to explore how increasing pressure levels modify the microbial degradation, organic matter composition, and microbiome of sinking diatom aggregates. Individual aggregates were incubated in rotating tanks in which pressure was incrementally increased to simulate a descent from surface to hadal depth within 20 days. Incubations at atmospheric pressure served as controls. With increasing pressure, microbial respiration and diatom degradation decreased gradually and ceased completely at 60 MPa. Dissolved organic carbon leaked substantially from the aggregates at ≥40 MPa, while diatom lipid and pigment contents decreased moderately. Bacterial abundance remained stable at >40 MPa, but bacterial community composition changed significantly at 60–100 MPa. Thus, pressure exposure reduces microbial degradation and transforms both organic matter composition and microbiomes of sinking particles, which may seed hadal sediments with relatively fresh particulate organic matter and putative pressure-tolerant microbes.

Abstract Blue carbon is carbon stored long-term in vegetated coastal ecosystems, which constitutes an important sink for atmospheric carbon dioxide (CO2). However, because methane (CH4) and nitrous oxide (N2O) have higher global warming potentials (GWP) than CO2, their production and release during organic matter diagenesis can affect the climate benefit of blue carbon. Here, we present a meta-analysis synthesizing seagrass CH4 and N2O fluxes and long-term organic carbon burial rates, and use these data to estimate the reduced climate benefit (offsets) of seagrass blue carbon using three upscaling approaches. Mean offsets for individual seagrass species (34.7% GWP20;1.0% GWP100) and globally (33.4% GWP20;7.0% GWP100) were similar, but GWP20 offsets were higher, and GWP100 offsets were lower than globally, for the Australian region (41.3% GWP20;1.1% GWP100). This study highlights the importance of using long-term organic carbon burial rates and accounting for both CH4 and N2O fluxes in future seagrass blue carbon assessments.

Background Frailty is a major geriatric syndrome that predicts increased vulnerability to minor stressor events and adverse outcomes such as falls, fractures, disability and death. The prevalence of frailty among individuals above the age of 65 varies widely with an overall weighted prevalence of 10.7%.Objectives The purpose of this study was to examine the prevalence of prefrailty and frailty in community-dwelling older adults from the regions of Lolland-Falster, which is one of the most socioeconomically disadvantaged areas of Denmark with lower income and lower life expectancy compared with the general Danish population. Moreover, the objective was to find selected individual characteristics associated with frailty.Design An observational, cross-sectional registry-based population study with data from the regions of Lolland-Falster collected between February 2016 and February 2020.Results The study included 19 000 individuals. There were 10 154 above the age of 50 included for analysis. Prevalence of frailty in the age group of 50–64 years was 4.7% and 8.7% in the age group of 65 years and above.The study demonstrates associations between frailty and high age, female gender, low education level, low income, smoking, living alone, frequency of seeing one’s children and getting help when needed. These associations are comparable with findings from other studies.Conclusion The syndrome of frailty consists of not only physiological and medical issues but also education, life conditions such as living alone and living in poverty and how you evaluate your own health.

Abstract Background Sporadic late onset nemaline myopathy is a rare, progressive muscle disease, presenting in adulthood, mainly affecting proximal limb and bulbar muscles. Muscle biopsies show characteristic nemaline rods. The putative mechanism is considered immune-related. Other manifestations aside from neuromuscular symptoms have not been described previously. Case presentation We present a case with atypical sporadic late onset nemaline myopathy (SLONM) of a non-HIV, non-MGUS subtype, where skin manifestations preceded neuromuscular symptoms, and a residual thymus with the histology of thymic follicular hyperplasia was detected during the diagnostic workup. Thorough dermatological investigations could not explain the skin presentations. Muscle biopsy revealed variation in fiber diameter, ragged-red and COX-negative fibers associated with discrete fibrosis. Electron microscopy detected atrophic muscle fibres with disorganization of the myofibrils, nemaline rods and abnormal mitochondria. Single-fiber EMG suggested signs of a neuromuscular transmission defect, EMG showed signs of myopathy. Analyses of antibodies associated with myasthenia gravis were negative. The patient showed improvement after intravenous immunoglobulin treatment regarding both the skin and the muscle symptoms. Conclusions Our case highlights the heterogeneity of SLONM with its varied spectrum of presentation. A unique combination of dermatological symptoms and SLONM could be seen with skin lesions as primary presenting symptoms. An association can be considered between the different manifestations, presumably based on immune etiology, where immunosuppressive therapy has been beneficial.

Abstract Burial of persistent organic pollutants (POPs) such as polychlorinated biphenyls (PCBs) in deep-sea sediments contributes to 60% of their historical emissions. Yet, empirical data on their occurrence in the deep-ocean is scarce. Estimates of the deep-ocean POP sink are therefore uncertain. Hadal trenches, representing the deepest part of the ocean, are hotspots for organic carbon burial and decomposition. POPs favorably partition to organic carbon, making trenches likely significant sinks for contaminants. Here we show that PCBs occur in both hadal (7720–8085 m) and non-hadal (2560–4050 m) sediment in the Atacama Trench. PCB concentrations normalized to sediment dry weight were similar across sites while those normalized to sediment organic carbon increased exponentially as the inert organic carbon fraction of the sediment increased in degraded hadal sediments. We suggest that the unique deposition dynamics and elevated turnover of organic carbon in hadal trenches increase POP concentrations in the deepest places on Earth.

In this paper, we outline the need for a coordinated international effort toward the building of an open-access Global Ocean Oxygen Database and ATlas (GO2DAT) complying with the FAIR principles (Findable, Accessible, Interoperable, and Reusable). GO2DAT will combine data from the coastal and open ocean, as measured by the chemical Winkler titration method or by sensors (e.g., optodes, electrodes) from Eulerian and Lagrangian platforms (e.g., ships, moorings, profiling floats, gliders, ships of opportunities, marine mammals, cabled observatories). GO2DAT will further adopt a community-agreed, fully documented metadata format and a consistent quality control (QC) procedure and quality flagging (QF) system. GO2DAT will serve to support the development of advanced data analysis and biogeochemical models for improving our mapping, understanding and forecasting capabilities for ocean O2 changes and deoxygenation trends. It will offer the opportunity to develop quality-controlled data synthesis products with unprecedented spatial (vertical and horizontal) and temporal (sub-seasonal to multi-decadal) resolution. These products will support model assessment, improvement and evaluation as well as the development of climate and ocean health indicators. They will further support the decision-making processes associated with the emerging blue economy, the conservation of marine resources and their associated ecosystem services and the development of management tools required by a diverse community of users (e.g., environmental agencies, aquaculture, and fishing sectors). A better knowledge base of the spatial and temporal variations of marine O2 will improve our understanding of the ocean O2 budget, and allow better quantification of the Earth’s carbon and heat budgets. With the ever-increasing need to protect and sustainably manage ocean services, GO2DAT will allow scientists to fully harness the increasing volumes of O2 data already delivered by the expanding global ocean observing system and enable smooth incorporation of much higher quantities of data from autonomous platforms in the open ocean and coastal areas into comprehensive data products in the years to come. This paper aims at engaging the community (e.g., scientists, data managers, policy makers, service users) toward the development of GO2DAT within the framework of the UN Global Ocean Oxygen Decade (GOOD) program recently endorsed by IOC-UNESCO. A roadmap toward GO2DAT is proposed highlighting the efforts needed (e.g., in terms of human resources).

Introduction The primary objective of the Danish Prostate Cancer Consortium Study 1 (DPCC-1) is to provide validation for a novel urine-based microRNA biomarker, called uCaP, for a diagnosis of prostate cancer.Methods and analysis Eligible participants are biopsy naïve men aged ≥18 years with prostate-specific antigen (PSA) levels ≥3 ng/mL, who are referred to prostate MRI due to suspicion of PC at one of the following three major urology/uroradiology centers: Aarhus University Hospital, Herlev & Gentofte University Hospital, or Odense University Hospital, where MRI and targeted biopsy are implemented in clinical use. Exclusion criteria include previous diagnosis of urogenital cancer, contraindication to MRI, gender reassignment treatment or PSA level >20 ng/mL. The participants will be asked to donate a urine sample in connection with their MRI. The study is observational, uses a diagnostic accuracy testing setup and will integrate into the current diagnostic pathway.We will measure the levels of the three microRNAs in the uCaP model (miR-222–3 p, miR-24–3 p and miR-30c-5p) in extracellular vesicle-enriched cell-free urine samples, to assess if uCaP can improve specificity and retain sensitivity for International Society of Urological Pathology Grade Group ≥2 PC, when used as a reflex test to PSA ≥3 ng/mL. We hypothesise that uCaP can improve selection for prostate MRI and reduce the number of unnecessary scans and biopsies.Ethics and dissemination This study is approved by the Central Denmark Region Committee on Health Research Ethics (reference number: 1-10-72-85-22). All participants will provide written informed consent. Study results will be published in peer-reviewed journals and presented in scientific meetings.Trial registration number NCT05767307 at clinicaltrials.gov.

Introduction: Mohs Micrographic Surgery (MMS) is a treatment option for high-risk facial nonmelanoma skin cancer with high cure rates. Especially on the nasal tip, the tissue sparing properties of MMS are appealing. The nasal tip is a common location of nonmelanoma skin cancer and can be a challenging anatomical structure for reconstructive surgery due to its prominent location in the face, the shortage of spare tissue, as well as the stiffness and composition of different skin types, cartilage and bone. Objectives: The aim of the present paper is to review and demonstrate how reconstruction of the nasal tip can be done successfully to improve the care for patients undergoing MMS in this area. Methods: Using selected literature on the area and the surgeons experience, each method of repair are described including their individual advantages and challenges. Pictures and consent were selected from one patient who underwent each repair method and three photos are presented in this paper: one after tumor resection, one immediately after repair, and one minimum 6 months post-surgery. Results: We present eight surgical methods as well as pictures from previous surgeries. Conclusions: The results demonstrate obtainable results using very different surgical methods and the importance of an individualized approach to repairing cutaneous defects of the nasal tip.

Abstract Hydroxylated isoprenoid glycerol dialkyl glycerol tetraethers (OH‐GDGTs) preserved in marine sediments are thought to be controlled by sea surface temperature (SST). However, water depth may also exert a significant influence on OH‐GDGTs. Here, we investigated sedimentary OH‐GDGTs in the Kermadec and Atacama trench regions (2,560–9,560 m water depth). Sedimentary OH‐GDGTs in hadal trenches were dominated by OH‐GDGT‐0 (72 ± 8%), potentially reflecting an adaption of source organisms to ambient cold deep water. This result, combined with global data set, revealed that the predominance of OH‐GDGT‐0 is a ubiquitous phenomenon in deep‐sea sediments, leading to a considerable underestimation of RI‐OH′‐derived SSTs. By considering both SST and water depth effects, we developed more accurate OH‐GDGT‐based paleothermometers for both shallow regions and the global ocean, encompassing the full‐ocean‐depth range. Our findings highlight the importance of accounting for the effect of water depth on OH‐GDGTs and provide improved tools for reconstructing paleo‐SSTs.

Abstract The treatment of implant‐associated bacterial infections and biofilms is an urgent medical need and a grand challenge because biofilms protect bacteria from the immune system and harbor antibiotic‐tolerant persister cells. This need is addressed herein through an engineering of antibody‐drug conjugates (ADCs) that contain an anti‐neoplastic drug mitomycin C, which is also a potent antimicrobial against biofilms. The ADCs designed herein release the conjugated drug without cell entry, via a novel mechanism of drug release which likely involves an interaction of ADC with the thiols on the bacterial cell surface. ADCs targeted toward bacteria are superior by the afforded antimicrobial effects compared to the non‐specific counterpart, in suspension and within biofilms, in vitro, and in an implant‐associated murine osteomyelitis model in vivo. The results are important in developing ADC for a new area of application with a significant translational potential, and in addressing an urgent medical need of designing a treatment of bacterial biofilms.

The accumulation and storage of nitrate by diatoms can contribute up to 1% of total pelagic nitrate and as much as 95% of total benthic nitrate, which indicates a potentially important contribution of diatoms to benthic nitrogen cycling, according to a global analysis

Krill represent a major link between primary producers and higher trophic levels in polar marine food webs. Potential links to lower trophic levels, such as heterotrophic microorganisms, are less well documented. Here, we studied the kinetics of microbial degradation of sinking carcasses of two dominant krill species Thysanoessa raschii and Meganyctiphanes norvegica from Southwest Greenland. Degradation experiments under oxic conditions showed that 6.0-9.1% of carbon and 6.4-7.1% of nitrogen were lost from the carcasses after one week. Aerobic microbial respiration and the release of dissolved organic carbon were the main pathways of carbon loss from the carcasses. Ammonium release generally contributed the most to carcass nitrogen loss. Oxygen micro profiling revealed anoxic conditions inside krill carcasses/specimens, allowing anaerobic nitrogen cycling through denitrification and dissimilatory nitrate reduction to ammonium (DNRA). Denitrification rates were up to 5.3 and 127.7 nmol N carcass-1 d-1 for T. raschii and M. norvegica, respectively, making krill carcasses hotspots of nitrogen loss in the oxygenated water column of the fjord. Carcass-associated DNRA rates were up to 4-fold higher than denitrification rates, but the combined activity of these two anaerobic respiration processes did not contribute significantly to carbon loss from the carcasses. Living krill specimens did not harbor any significant denitrification and DNRA activity despite having an anoxic gut as revealed by micro profiling. The investigated krill carcasses sink fast (1500-3000 m d-1) and our data show that only a small fraction of the associated carbon is lost during descent. Based on data on krill distribution, our findings are used to discuss the potential importance of sinking krill carcasses for sustaining benthic food webs in the Arctic.

Background: Modulation of pathological neural circuit activity in the brain with a minimum of complications is an area of intense interest. Objective: The goal of the study was to alter neurons' physiological states without apparent damage of cellular integrity using stereotactic radiosurgery (SRS). Methods: We treated a 7.5 mm-diameter target on the visual cortex of Göttingen minipigs with doses of 40, 60, 80, and 100 Gy. Six months post-irradiation, the pigs were implanted with a 9 mm-wide, eight-shank multi-electrode probe, which spanned the radiation focus as well as the low-exposure neighboring areas. Results: Doses of 40 Gy led to an increase of spontaneous firing rate, six months post-irradiation, while doses of 60 Gy and greater were associated with a decrease. Subjecting the animals to visual stimuli resulted in typical visual evoked potentials (VEP). At 40 Gy, a significant reduction of the P1 peak time, indicative of higher network excitability was observed. At 80 Gy, P1 peak time was not affected, while a minor reduction at 60 Gy was seen. No distance-dependent effects on spontaneous firing rate, or on VEP were observed. Post-mortem histology revealed no evidence of necrosis at doses below 60 Gy. In an in vitro assay comprising of iPS-derived human neuron-astrocyte co-cultures, we found a higher vulnerability of inhibitory neurons than excitatory neurons with respect to radiation, which might provide the cellular mechanism of the disinhibitory effect observed in vivo. Conclusion: We provide initial evidence for a rather circuit-wide, long-lasting disinhibitory effect of low sub-ablative doses of SRS.

The expansion of transient and permanent coastal benthic anoxia is one of the most severe problems for the coastal ocean globally. We report frequent, hidden hypoxia in the bottom 5 cm of the water column of a coastal site in the central Baltic Sea by continuous high‐resolution profiling of oxygen (O2) directly above the sediment surface. This hypoxia stood in stark contrast to 30‐yr O2 monitoring records at this site that suggest apparent continuous well‐oxygenated conditions. In situ measurements showed highly dynamic conditions in the bottom 30 cm recording frequent gradual and abrupt changes between normoxic (> 63 μmol L−1) and hypoxic (< 63 μmol L−1) conditions that would remain undetectable by conventional bottom water O2 monitoring. The temporal variability of these "hidden" hypoxia is tied to the dynamic current field and to changes in O2 consumption following resuspension events. Our observations suggest that transient benthic hypoxia is much more common than routine monitoring data indicate. [ABSTRACT FROM AUTHOR]

Diffusive equilibrium in thin films (DET) probes are passive samplers that are designed to reflect in situ porewater concentrations. In this study, we show that the gel and the plastic housing of DET probes store a substantial amount of oxygen (O2) that affects the chemical composition of porewater. To ensure complete deoxygenation, DET probes need to be treated for 7 d with continuous nitrogen flow. Such fully deoxygenated probes can be handled in the air (exposure time: < 90 s) and deployed to sediments through oxic water (exposure time: < 2 s) without significant ad(b)sorption of O2. Furthermore, we deployed a set of untreated (i.e., in equilibrium with atmospheric O2) and a set of fully deoxygenated DET probes to lake sediments. The O2 present in untreated DET probes altered iron (Fe) and phosphate (P) porewater profiles significantly. This is caused by the oxidation, immobilization, and accumulation of redox‐sensitive Fe (oxyhydr)oxides in the probe over time. Since P has a high binding affinity to Fe (oxyhydr)oxides, it is not in equilibrium with the porewater and is overestimated as well. Our results highlight the importance of thorough deoxygenation of DET probes before deployment in sediments, especially when addressing redox‐sensitive porewater species. [ABSTRACT FROM AUTHOR]

Abstract Hadal trenches are depocenters for organic material, and host intensified benthic microbial activity. The enhanced deposition is presumed to be reflected in elevated meiofaunal standing-stock, but available studies are ambiguous. Here, we investigate the distribution of meiofauna along the Atacama Trench axis and adjacent abyssal and bathyal settings in order to relate the meiofauna densities to proxies for food availability. Meiofauna densities peaked at the sediment surface and attenuated steeply with increasing sediment depth. The distribution mirrored the vertical profile of the microbial-driven oxygen consumption rate demonstrating a close linkage between microbial activity and meiofauna density. Meiofaunal standing-stock along the trench axis varied by a factor of two, but were markedly higher than values from the abyssal site at the oceanic plate. Overall, meiofaunal densities poorly correlated with common proxies for food availability such as total organic carbon and phytopigments, but strongly correlated with the microbial benthic O2 consumption rate. We argue that microbial biomass likely represents an important meiofaunal food source for hadal meiofauna. Observations from three trench systems underlying surface water of highly different productivity confirmed elevated meiofaunal densities at the trench axis as compared to abyssal sites on oceanic plates. Food availability appear to drive elevated abundance and variations in meiofauna densities in hadal sediments.

Black carbon accumulation rates in hadal trenches in the deepest regions of the oceans could be seven-fold higher than the global ocean average, according to geochemical and isotopic analyses of sediments from six trenches in the Pacific Ocean.

Abstract Meiofauna (invertebrates that pass through a 1‐mm mesh sieve, but are retained on a 40‐µm mesh) represent the most abundant and diverse animal group on Earth, but empirical evidence of their role in benthic respiration, production and carbon cycling across ecosystems is not well documented. Moreover, how meiofauna respond to changing oxygen conditions is poorly understood. We further developed an incubation system, in which oxygen and temperature conditions are easily controlled and single meiofaunal nematode respiration is resolved in glass capillary tubes, using Clark‐type oxygen microsensor. We performed the respiration measurements after exposing nematodes to different ambient oxygen concentrations, which resulted in 3–60 µM O2 during hypoxic and 80–210 µM O2 during oxic incubations in close proximity to the respective nematodes. Individual nematode respiration rates ranged from 0.02 to 1.30 nmol O2 ind.−1 day−1 and were 27% lower during hypoxic than oxic incubations. Rates derived from established allometric relations were on average fourfold higher than our direct measurements. The presented method is suitable for single nematode respiration measurements and can be adapted to a wide range of experimental conditions. Therefore, it can be used to assess meiofauna contribution to ecosystem processes and investigate species‐specific responses to changing environmental conditions, for example, oxygen stress, increasing water temperature.
