Abstract

This article examines the possiblity of the selection on phosphoglucomutase locus 1 (PGM1) during intrauterine life for deviations of the maternal-neonatal joint PGM1 and that maternal and paternal PGM1 alleles could have different associations with survival. Results show a significant relation between mother-newborn PGM1 genotype and birth weight percentile class; also, paternal PGM1*2 had negative affects with macrosomia in female infants.

Abstract

The phosphoglucomutase locus 1 (PGM1) genetic polymorphism has been studied in a sample of newborns and mothers from Penne and Rome, Italy, as well as a sample of women with repeated spontaneous abortions and their husbands from the population of Rome. Results support the hypothesis of PGM1 selection during intrauterine development.

Abstract

The relationship between human placental alkaline phosphatase (PLAP) genotype and birth weight is investigated in a sample of white, black and Puerto-Rican new-born infants from New Haven, Connecticut (total 710 subjects). Black and Puerto-Rican infants show a higher incidence of growth retardation and a higher frequency of ALPp*1/*1 genotype as compared to whites. The proportion of newborns with a low birth weight (below the 10th percentile) is lower in infants with ALPp*1/*1 genotype than in those with other PLAP genotypes, especially among non-whites. It is argued that the higher frequency of ALPp*1 allele among non-whites might be, at least in part, a consequence of their adaptation in the past to environmental conditions adverse to optimal intrauterine development. Abstract

Ninety-nine pregnant women with insulin-dependent diabetes mellitus, 83 women with gestational diabetes, and a control sample of 315 nondiabetic consecutive puerperae have been studied along with their newborn infants. Neonatal macrosomia is less frequent among diabetic mothers with phosphoglucomutase (PGM1) genotype PGM1*1/*2 than among mothers with other PGM1 genotypes. In gestational diabetes the association is more evident among younger women than among older women. Diabetic women with the PGM1*1/*2 genotype show a reduced proportion of heterozygous PGM1*1/*2 offspring. The phenomenon is much more evident among women under 28 years of age and does not depend on the quality of metabolic control. The data suggest that when both mother and fetus share the PGM1*1/*2 genotype, the deleterious effects of a diabetic environment on fetal development are more severe, leading to an early loss of zygotes. Abstract

Objective: In the present paper we have searched for a possible cooperative effect of p53 codon 72 and ACP1 genetic polymorphisms on cancer grade. Methods: 69 women with endometrium cancer and 696 adults without cancer have been studied. P53 codon 72 and ACP1 genotypes were determined by DNA analysis. Results: The analysis has shown a cooperative interaction between ACP1 and p53 codon 72 concerning the effect on cancer grade: the proportion of the joint genotype *Arg/*Arg carrying the low activity *A/*A and *A/*B ACP1 genotypes is lower in high grade than in low grade. The proportion of gametic type *A/*Arg is significantly lower in high than in low grade cancer. Conclusion: The combination of the effect of low activity ACP1 that may favor cellular growth with the increase of apoptosis due to *Arg allele could act against the progression towards forms with immature and undifferentiated cells protecting from severe manifestation of cancer.

Abstract

Background: The role of adenosine as cardio protective factor is well established. Adenosine deaminase (ADA) contributes to the control of adenosine concentration in body fluids and, as ecto-enzyme, to the regulation of adenosine receptors activity. ADA and adenosine receptors expressions have been found down regulated in heart failure. We have studied the relationship between genetic variability within the ADA gene and left ventricular ejection fraction (LVEF). Methods: The genotypes of three polymorphic sites (SNPs) within the ADA gene have been determined in 346 patients admitted to the hospital for cardiovascular diseases. Informed consent was obtained by the patients to participate to the study that was approved by the Council of Department of Biomedicine and Prevention. The three polymorphic sites in the ADA gene are called ADA1, ADA2 and ADA6. Each locus shows two alleles called ADA1*1 and ADA1*2, ADA2*1 and ADA2*2, ADA6*1 and ADA6*2 respectively. Results: The joint “ADA2*2 carrier/ADA6*1/*1 genotype” shows a statistically significant lower value of LVEF as compared to other joint genotypes (p=0.004). Such association is statistically significant in subjects with coronary artery disease only. Conclusions: The study of polymorphic sites of ADA gene could allow to detect subjects with higher risk of cardiac failure following infarction.
