3. Accuracy of Integrated Artificial Intelligence Grading Using Handheld Retinal Imaging in a Community Diabetic Eye Screening Program

Purpose: To evaluate mydriatic handheld retinal imaging performance assessed by point-of-care (POC) artificial intelligence (AI) as compared with retinal image graders at a centralized reading center (RC) in identifying diabetic retinopathy (DR) and diabetic macular edema (DME). Design: Prospective, comparative study. Subjects: Five thousand five hundred eighty-five eyes from 2793 adult patients with diabetes. Methods: Point-of-care AI assessment of disc and macular handheld retinal images was compared with RC evaluation of validated 5-field handheld retinal images (disc, macula, superior, inferior, and temporal) in identifying referable DR (refDR; defined as moderate nonproliferative DR [NPDR], or worse, or any level of DME) and vision-threatening DR (vtDR; defined as severe NPDR or worse, or any level of center-involving DME [ciDME]). Reading center evaluation of the 5-field images followed the international DR/DME classification. Sensitivity (SN) and specificity (SP) for ungradable images, refDR, and vtDR were calculated. Main Outcome Measures: Agreement for DR and DME; SN and SP for refDR, vtDR, and ungradable images. Results: Diabetic retinopathy severity by RC evaluation: no DR, 67.3%; mild NPDR, 9.7%; moderate NPDR, 8.6%; severe NPDR, 4.8%; proliferative DR, 3.8%; and ungradable, 5.8%. Diabetic macular edema severity by RC evaluation was as follows: no DME (80.4%), non–ciDME (7.7%), ciDME (4.4%), and ungradable (7.5%). Referable DR was present in 25.3% and vtDR was present in 17.5% of eyes. Images were ungradable for DR or DME in 7.5% by RC evaluation and 15.4% by AI. There was substantial agreement between AI and RC for refDR (κ = 0.66) and moderate agreement for vtDR (κ = 0.54). The SN/SP of AI grading compared with RC evaluation was 0.86/0.86 for refDR and 0.92/0.80 for vtDR. Conclusions: This study demonstrates that POC AI following a defined handheld retinal imaging protocol at the time of imaging has SN and SP for refDR that meets the current United States Food and Drug Administration thresholds of 85% and 82.5%, but not for vtDR. Integrating AI at the POC could substantially reduce centralized RC burden and speed information delivery to the patient, allowing more prompt eye care referral. 4. A Detailed Clinical Case of Localized Prostate Tumors Treated with Nanoparticle-Assisted Sub-Ablative Laser Ablation

AuroLase® Therapy—a nanoparticle-enabled focal therapy—has the potential to safely and effectively treat localized prostate cancer (PCa), preserving baseline functionality. This article presents a detailed case of localized PCa treated with AuroLase, providing insight on expectations from the diagnosis of PCa to one year post-treatment. AuroLase Therapy is a two-day treatment consisting of a systemic infusion of gold nanoshells (~150-nm hydrodynamic diameter) on Day 1, and sub-ablative laser treatment on Day 2. Multiparametric MRI (mpMRI) was used for tumor visualization, treatment planning, and therapy response assessment. The PCa was targeted with a MR/Ultrasound-fusion (MR/US) transperineal approach. Successful treatment was confirmed at 6 and 12 months post-treatment by the absence of disease in MR/US targeted biopsies. On the mpMRI, confined void space was evident, an indication of necrotic tissues encompassing the treated lesion, which was completely resolved at 12 months, forming a band-like scar with no evidence of recurrent tumor. The patient’s urinary and sexual functions were unchanged. During the one-year follow-up, changes on the DCE sequence and in the Ktrans and ADC values assist in qualitatively and quantitatively evaluating tissue changes. The results highlight the potential of gold-nanoparticle-enabled sub-ablative laser treatment to target and control localized PCa, maintain quality of life, and preserve baseline functionality.

Invasive alien species control is recognized worldwide as a priority action to preserve global biodiversity. However, a lack of general life history knowledge for threatened species can impede the effectiveness of conservation actions. Galápagos pink land iguanas (Conolophus marthae) are endemic to Wolf Volcano, Galápagos, Ecuador. These iguanas are threatened by invasive alien species, particularly feral cats, that may affect their small population size. To guarantee the long-term survival of C. marthae, the Galápagos National Park Directorate is considering, along with an ongoing campaign of feral cat control, the implementation of a head-start program. However, the success of this management strategy necessarily relies on the identification of pink iguana nesting grounds, which were still unknown at the onset of this study. We modeled the movement patterns of male and female iguanas during the reproductive season, using location data collected from custom-made remote tracking devices installed on adult pink iguanas in April 2021. We first calculated for each individual the vector of distances from its starting location, which was defined as net displacement. We then used net displacement as the response variable in a generalized additive mixed model with day of the year as the predictor. Based on the hypothesis that males and females may behaviorally differ after mating, we looked for female-specific migratory behavior suggesting females were moving toward nesting areas. The results obtained confirmed our hypothesis, as females exhibited a distinct migratory behavior, reaching a small plateau area inside of Wolf Volcano’s caldera and ca. 400 m below the volcano’s northern rim. Moreover, once inside the caldera, females displayed a more aggregated distribution pattern. The movement data obtained allowed Galápagos National Park rangers to locate individual pink iguana nests and subsequently to sight and collect the first observed hatchlings of the species. This work constitutes a necessary baseline to perform dedicated studies of pink iguana nests and emerging hatchling iguanas, which is an essential step toward the development of an effective head-start program.

In this review, we outline our current understanding of the mechanisms involved in the absorption, storage, and transport of dietary vitamin A to the eye, and the trafficking of rhodopsin protein to the photoreceptor outer segments, which encompasses the logistical backbone required for photoreceptor cell function. Two key mechanisms of this process are emphasized in this manuscript: ocular and systemic vitamin A membrane transporters, and rhodopsin transporters. Understanding the complementary mechanisms responsible for the generation and proper transport of the retinylidene protein to the photoreceptor outer segment will eventually shed light on the importance of genes encoded by these proteins, and their relationship on normal visual function and in the pathophysiology of retinal degenerative diseases.

7. APOE ε2-Carriers Are Associated with an Increased Risk of Primary Angle-Closure Glaucoma in Patients of Saudi Origin

This study investigated the association between apolipoprotein E (APOE) gene polymorphisms (rs429358 and rs7412) and primary angle-closure glaucoma (PACG) and pseudoexfoliation glaucoma (PXG) in a Saudi cohort. Genotyping of 437 DNA samples (251 controls, 92 PACG, 94 PXG) was conducted using PCR-based Sanger sequencing. The results showed no significant differences in the allele and genotype frequencies of rs429358 and rs7412 between the PACG/PXG cases and controls. Haplotype analysis revealed ε3 as predominant, followed by ε4 and ε2 alleles, with no significant variance in PACG/PXG. However, APOE genotype analysis indicated a significant association between ε2-carriers and PACG (odds ratio = 4.82, 95% CI 1.52–15.26, p = 0.007), whereas no notable association was observed with PXG. Logistic regression confirmed ε2-carriers as a significant predictor for PACG (p = 0.008), while age emerged as significant for PXG (p < 0.001). These findings suggest a potential role of ε2-carriers in PACG risk within the Saudi cohort. Further validation and larger-scale investigations are essential to elucidate the precise role of APOE in PACG pathogenesis and progression.

This retrospective study examines the clinical characteristics and underlying genetic variants that exist in a Leber congenital amaurosis (LCA) patient cohort evaluated at the inherited retinal disease (IRD) clinic at the University of Minnesota (UMN)/M Health System. Our LCA cohort consisted of 33 non-syndromic patients and one patient with Joubert syndrome. We report their relevant history, clinical findings, and genetic testing results. We monitored disease presentation utilizing ocular coherence tomography (OCT) and fundus autofluorescence (FAF). Electroretinogram testing (ERG) was performed in patients when clinically indicated. Next-generation sequencing (NGS) and genetic counseling was offered to all evaluated patients. Advanced photoreceptor loss was noted in 85.7% of the subjects. All patients who underwent FAF had findings of either a ring of macular hypo/hyper AF or peripheral hypo-AF. All patients had abnormal ERG findings. A diagnostic genetic test result was identified in 74.2% of the patients via NGS single-gene testing or panel testing. Two patients in our cohort qualified for Luxturna® and both received treatment at the time of this study. These data will help IRD specialists to understand the genetic variants and clinical presentations that characterize our patient population in the Midwest region of the United States.

11. Common Variants rs429358 and rs7412 in APOE Gene Are Not Associated with POAG in a Saudi Cohort

Adult-onset glaucoma, an age-related neurodegenerative disease, is very prevalent among the elderly Arabs of Saudi origin. This study investigated the association between apolipoprotein E (APOE) gene variants (rs429358 and rs7412) and primary open-angle glaucoma (POAG) in Arabs of Saudi origin. A case-control genetic association study involving 179 POAG patients and 251 controls utilized Sanger sequencing to genotype APOE gene variants. The allele frequencies and genotype distributions for rs429358 and rs7412 did not show significant associations with POAG. The haplotype analysis revealed apoε3 (87.6% and 87.4%) as the most prevalent, followed by ε4 (2.8% and 3.6%) and ε2 (9.6% and 8.9%) in the controls and POAG patients, respectively. Although the ε2/ε3 genotype and ε2-carriers displayed a more than two-fold increased risk, statistical significance was not reached. Notably, these polymorphisms did not affect clinical markers, such as intraocular pressure and cup/disc ratio. The logistic regression analysis demonstrated no significant influence of age, sex, rs429358, or rs7412 polymorphisms on POAG. In conclusion, within the Saudi cohort, APOE variants (rs429358 and rs7412) do not appear to be associated with POAG and are not substantial risk factors for its development. However, additional population-based studies are required to validate these findings.

12. Association between Polymorphism rs61876744 in PNPLA2 Gene and Keratoconus in a Saudi Cohort

The genetic etiology of Keratoconus (KC) in Middle Eastern Arabs of Saudi origin is still unclear. A recent genome-wide study identified two significant loci in the region of PNPLA2 (rs61876744) and CSNK1E (rs138380) for KC that may be associated with KC in the Saudi population. In addition, polymorphisms in the apolipoprotein E (APOE) gene, namely, rs429358 and rs7412, responsible for APOE allelic variants ε2, ε3, and ε4, may influence KC via oxidative stress mechanism(s). Thus, we investigated the possible association of polymorphisms rs61876744, rs138380, rs429358, rs7412, and APOE genotypes in KC patients of the Saudi population. This study included 98 KC cases and 167 controls. Polymorphisms rs6187644 and rs138380 were genotyped using TaqMan assays, and rs429358 and rs7412 were genotyped via Sanger sequencing. Although the allele frequency of rs61876744(T) in PNPLA2 was a protective effect against KC (odds ratio (OR) = 0.64, 95% confidence interval (CI) = 0.44–0.93), the p-value (p = 0.020) was not significant for multiple testing correction (p = 0.05/4 = 0.015). However, rs6187644 genotype showed a modestly significant protective effect in the dominant model (OR = 0.53, 95% CI = 0.32–0.88, p = 0.013). Polymorphisms rs138380, rs429358, and rs7412 showed no significant allelic or genotype association with KC. However, the ε2-carriers (ε2/ε2 and ε2/ε3 genotypes) exhibited a greater than 5-fold increased risk of KC, albeit non-significantly (p = 0.055). Regression analysis showed no significant effect of age, gender, and the four polymorphisms on KC. Our results suggest that polymorphism rs6187644 in PNPLA2 might be associated with KC in the Middle Eastern Arabs of Saudi origin but warrant a large-scale association analysis at this locus.

The distribution of dietary vitamin A/all-trans retinol/ROL throughout the body is critical for maintaining retinoid function in peripheral tissues and for retinoid delivery to the eye in the support of visual function. In the circulation, all-trans-retinol bound to the RBP4 protein is transported and sequestered into target tissues for long-term storage. Two membrane receptors that facilitate all-trans retinol uptake from RBP4 have been proposed. While it is well established that the membrane receptor, STRA6, binds to circulatory RBP4 for ROL transport into the eye, the second vitamin A receptor, RBPR2, which is expressed in non-ocular tissues, is less characterized. Based on the structural homology between these two RBP4 receptors, published literature, and from our recent work in Rbpr2−/− deficient mice, we hypothesized that RBPR2 might also have high-binding affinity for RBP4 and this mechanism facilitates ROL transport. Herein, we aimed to elucidate the membrane topology and putative RBP4 binding residues on RBPR2 to understand its physiological function for retinoid homeostasis. Using in silico analysis and site-directed mutagenesis, we identified a potential RBP4 binding domain on RBPR2. We employed an in vitro cell-based system and confirmed that mutations of these residues on RBPR2 affected its binding to exogenous RBP4 and subsequently vitamin A uptake. Using Surface Plasmon Resonance assays, we analyzed both the binding affinities and kinetic parameters of wild-type RBPR2 and individual mutants affecting the RBPR2-RBP4 binding domain with its physiological ligand RBP4. These studies not only revealed a putative RBP4 binding domain on RBPR2 but also provided new structural, biochemical, and critical information on its proposed role in RBP4 binding for ROL transport and retinoid homeostasis.

Aim In a retrospective and exploratory case-control study, we examined the genetic association of two common polymorphisms in the 3’ untranslated region (UTR) of DICER1 (rs3742330) and DROSHA (rs10719) genes in primary open-angle glaucoma (POAG) and primary angle-closure glaucoma (PACG), and its related clinical phenotypes in a Saudi cohort. Methods DNA genotyping was performed using TaqMan real-time PCR assays in 500 participants, including 152 POAG, 102 PACG, and 246 non-glaucomatous controls. Statistical analyses were performed to examine the association(s). Results Allele and genotype frequency of rs3742330 and rs10719 did not vary significantly in POAG and PACG compared to controls. No significant deviation was observed from Hardy-Weinberg Equilibrium (p > 0.05). Gender stratification revealed no significant allelic/genotype association with glaucoma types. Also, these polymorphisms showed no significant genotype effect on clinical markers such as intraocular pressure, cup/disc ratio, and the number of antiglaucoma medications. Logistic regression showed no effect of age, sex, rs3742330, and rs10719 genotypes on the risk of disease outcome. We also examined a combined allelic effect of rs3742330 (A>G) and rs10719 (A>G). However, none of the allelic combinations significantly affected POAG and PACG. Conclusions The 3’ UTR polymorphisms rs3742330 and rs10719 of DICER1 and DROSHA genes are not associated with POAG and PACG or its related glaucoma indices in this Middle-Eastern cohort of Saudi Arab ethnicity. However, there is a need to validate the results on a broader population and other ethnicities.

15. The G Protein-Coupled Receptor, VPAC1, Mediates Vasoactive Intestinal Peptide-Dependent Functional Homeostasis of the Gut Microbiota

Background and Aims: Vasoactive intestinal peptide (VIP) is a neuropeptide involved in the regulation of feeding behavior and circadian rhythms, metabolism, and immunity. Previous studies revealed the homeostatic effects of VIP signaling on the gut microbiota. VIP-deficient mice demonstrate a gut microbiota dysbiosis characterized by reduced α-diversity and decreased relative abundance (RA) of Gram-positive Firmicutes. However, the mechanism by which VIP signaling affects changes in the microbiota is unknown. Methods: To investigate the role of the 2 cognate G protein-coupled receptors for VIP (VPAC1 and VPAC2) in VIP-mediated homeostasis of the microbiota, fecal samples from VPAC1- and VPAC2-deficient, heterozygous, and wild-type littermate mice were assessed via targeted amplicon sequencing. Their microbiota profiles were additionally compared with microbiota from VIP-deficient, heterozygous, and wild-type littermates, where genotype-dependent changes in the composition and predicted function of each cohort were compared. Results: While wild-type mice in each line differed in α-diversity and β-diversity, consistent changes in both metrics were observed in VIP-deficient and VPAC1-deficient mice. This includes a dramatic reduction in α-diversity, increased RA of Proteobacteria and Bacteroidetes, and decreased RA of Lachnospiraceae, Ruminococcaceae, Muribaculaceae, and Rikenellaceae. Specific amplicon sequence variants and predicted functions found to differ significantly based on VIP or VPAC1 genotype were concordant in their directions of change. Multiplatform predicted functional profiling suggested a defective VIP-VPAC1 axis was associated with reduced amino acid degradation along with reduced quinol and quinone biosynthesis. Furthermore, alterations in predicted functions include increased sugar degradation, nitrate reduction, and fatty acid biosynthetic pathways, among other changes. Conclusion: We conclude that VIP signaling through VPAC1 is critical for the maintenance of normal function of the gut microbiota.

Abstract Background Hyperimmune plasma raised against β‐1→6‐poly‐N‐acetyl glucosamine (PNAG HIP) mediates more opsonophagocytic killing of Rhodococcus equi (R equi) than does R equi hyperimmune plasma (RE HIP) in vitro. The relative efficacy of PNAG HIP and RE HIP to protect foals against R equi pneumonia, however, has not been evaluated. Hypothesis Transfusion with PNAG HIP will be superior to RE HIP in foals for protection against R equi pneumonia in a randomized, controlled, blinded clinical trial. Animals Four hundred sixty Quarter Horse and Thoroughbred foals at 5 large breeding farms in the United States. Methods A randomized, controlled, blinded clinical trial was conducted in which foals were transfused within 24 hours after birth with 2 L of either RE HIP or PNAG HIP. Study foals were monitored through weaning for clinical signs of pneumonia by farm veterinarians. The primary outcome was the proportion of foals that developed pneumonia after receiving each type of plasma. Results The proportion of foals that developed pneumonia was the same between foals transfused with RE HIP (14%; 32/228) and PNAG HIP (14%; 30/215). Conclusions and Clinical Importance Results indicate that PNAG HIP was not superior to a commercially available, United States Department of Agriculture‐licensed RE HIP product for protecting foals against R equi pneumonia under field conditions.

Sepsis, a complex clinical syndrome resulting from a serious infection, is a major healthcare problem associated with high mortality. Sex-related differences in the immune response to sepsis have been proposed but the mechanism is still unknown. Purinergic signaling is a sex-specific regulatory mechanism in immune cell physiology. Our studies have shown that blocking the ADP-receptor P2Y12 but not P2Y1 receptor was protective in male mice during sepsis, but not female. We now hypothesize that there are sex-related differences in modulating P2Y12 or P2Y1 signaling pathways during sepsis. Male and female wild-type (WT), P2Y12 knock-out (KO), and P2Y1 KO mice underwent sham surgery or cecal ligation and puncture (CLP) to induce sepsis. The P2Y12 antagonist ticagrelor or the P2Y1 antagonist MRS2279 were administered intra-peritoneally after surgery to septic male and female mice. Blood, lungs and kidneys were collected 24 hours post-surgery. Sepsis-induced changes in platelet activation, secretion and platelet interaction with immune cells were measured by flow cytometry. Neutrophil infiltration in the lung and kidney was determined by a myeloperoxidase (MPO) colorimetric assay kit. Sepsis-induced platelet activation, secretion and aggregate formation were reduced in male CLP P2Y12 KO and in female CLP P2Y1 KO mice compared with their CLP WT counterpart. Sepsis-induced MPO activity was reduced in male CLP P2Y12 KO and CLP P2Y1 KO female mice. CLP males treated with ticagrelor or MRS2279 showed a decrease in sepsis-induced MPO levels in lung and kidneys, aggregate formation, and platelet activation as compared to untreated male CLP mice. There were no differences in platelet activation, aggregate formation, and neutrophil infiltration in lung and kidney between female CLP mice and female CLP mice treated with ticagrelor or MRS2279. In human T lymphocytes, blocking P2Y1 or P2Y12 alters cell growth and secretion in vitro in a sex-dependent manner, supporting the data obtained in mice. In conclusion, targeting purinergic signaling represents a promising therapy for sepsis but drug targeting purinergic signaling is sex-specific and needs to be investigated to determine sex-related targeted therapies in sepsis.

Due to the presence of flat Chern bands, moir\'e transition metal dichalcogenide (TMD) bilayers are a platform to realize strongly correlated topological phases of fermions such as fractional Chern insulators. TMDs are also known to host long-lived excitons, which inherit the topology of the underlying Chern bands. For the particular example of MoTe$_2$/WSe$_2$ heterobilayers we perform a time-dependent Hartree-Fock calculation to identify a regime in the phase diagram where the excitons themselves form a topological flat band. Much of modern cosmology relies on the Cosmological Principle, the assumption that the Universe is isotropic and homogeneous on sufficiently large scales, but it remains worthwhile to examine cosmological models that violate this principle slightly. We examine a class of such spacetimes that maintain homogeneity but break isotropy through their underlying local spatial geometries. These spacetimes are endowed with one of five anisotropic model geometries of Thurston's geometrization theorem, and their evolution is sourced with perfect fluid dust and cosmological constant. We show that the background evolution of these spacetimes induces fluctuations in the observed cosmic microwave background (CMB) temperature with amplitudes coupled to the curvature parameter $\Omega_K$. In order for these fluctuations to be compatible with the observed CMB angular power spectrum, we find $|\Omega_K| \lesssim 10^{-5}$ is required in all five geometries, with two geometries requiring $|\Omega_K| \lesssim 10^{-10}$. The EXperiment for Cryogenic Large-Aperture Intensity Mapping (EXCLAIM) is a balloon-borne telescope designed to survey star formation over cosmological time scales using intensity mapping in the 420 - 540 GHz frequency range. EXCLAIM uses a fully cryogenic telescope coupled to six on-chip spectrometers featuring kinetic inductance detectors (KIDs) to achieve high sensitivity, allowing for fast integration in dark atmospheric windows. The telescope receiver is cooled to $\approx$ 1.7 K by immersion in a superfluid helium bath and enclosed in a superfluid-tight shell with a meta-material anti-reflection coated silicon window. In addition to the optics and the spectrometer package, the receiver contains the magnetic shielding, the cryogenic segment of the spectrometer readout, and the sub-Kelvin cooling system. A three-stage continuous adiabatic demagnetization refrigerator (CADR) keeps the detectors at 100 mK while a $^4$He sorption cooler provides a 900 mK thermal intercept for mechanical suspensions and coaxial cables. The baryon cycle is crucial for understanding galaxy formation, as gas inflows and outflows vary throughout a galaxy's lifetime and affect its star formation rate. Despite the necessity of accretion for galaxy growth at high redshifts, direct observations of inflowing gas have proven elusive especially at $z\gtrsim2$. We present spectroscopic analysis of a galaxy at redshift $z=2.45$ which exhibits signs of inflow in several ultraviolet interstellar absorption lines, with no clear outflow signatures. The absorption lines are redshifted by $\sim$250 km sec$^{-1}$ with respect to the systemic redshift, and C IV shows a prominent inverse P-Cygni profile. Simple stellar population models suggest that this galaxy has a low metallicity ($\sim$5\% solar), with a very young starburst of age $\sim$4 Myr dominating the ultraviolet luminosity. The gas inflow velocity and nebular velocity dispersion suggest an approximate halo mass of order $\sim 10^{11}M_{\odot}$, a regime in which simulations predict that bursty star formation is common at this redshift. We conclude that this system is likely in the beginning of a cycle of bursty star formation, where inflow and star formation rates are high, but where supernovae and other feedback processes have not yet launched strong outflows. In this scenario, we expect the inflow-dominated phase to be observable (e.g., with net redshifted ISM absorption) for only a short timescale after a starburst onset. This result represents a promising avenue for probing the full baryon cycle, including inflows, during the formative phases of low-mass galaxies at high redshifts., Comment: 12 pages, 4 figures

We solve the optimal extraction problem for e-graphs by first showing a connection between e-graphs and cyclic monotone Boolean circuits, then solving the weighted satisfiability problem for such circuits. The solution is a parameterized algorithm based on treewidth. Additionally, we show how the circuit view of e-graphs allows us to apply simplification techniques that are not possible when operating directly on e-graphs. While the core parameterized algorithm may be adapted to work directly on e-graphs, the simplification results show why the circuit view is helpful.

Motivated by the rich properties and various applications of recurrence relations, we consider the extension of traditional recurrence relations to matrices, where we use matrix multiplication and the Kronecker product to construct matrix sequences. We provide a sharp condition, which when satisfied, guarantees that any fixed-depth matrix recurrence relation defined over a product (with respect to matrix multiplication) will converge to the zero matrix. We also show that the same statement applies to matrix recurrence relations defined over a Kronecker product. Lastly, we show that the dual of this condition, which remains sharp, guarantees the divergence of matrix recurrence relations defined over a consecutive Kronecker product. These results completely determine the stability of nontrivial fixed-depth complex-valued recurrence relations defined over a consecutive product.

In this paper, we present such a system: DESI-090.9854-35.9683, a cluster-scale lens at $z_{\rm l} = 0.49$, with seven observed lensed sources around the core, and additional lensed sources further out in the cluster. From the number and the textbook configuration of the lensed images, a tight constraint on the mass potential of the lens is possible. This would allow for detailed analysis on the dark and luminous matter content within galaxy clusters, as well as a probe into dark energy and high-redshift galaxies. We present our spatially resolved kinematic measurements of this system from the Very Large Telescope Multi Unit Spectroscopic Explorer, which confirm five of these source galaxies (in ascending order, at $z_{\rm s} = 0.962, 0.962, 1.166, 1.432,$ and $1.432$). With previous Hubble Space Telescope imaging in the F140W and F200LP bands, we also present a simple two power-law profile flux-based lens model that, for a cluster lens, well models the five lensed arc families with redshifts. We determine the mass to be $M(< \theta_{\rm E}) = 4.78\times10^{13} M_{\odot}$ for the primary mass potential. From the model, we extrapolate the redshift of one of the two source galaxies not yet spectroscopically confirmed to be at $z_{\rm s}=4.52^{+1.03}_{-0.71}$., Comment: 14 pages, 6 figures

Conway Checkers is a game played with a checker placed in each square of the lower half of an infinite checkerboard. Pieces move by jumping over an adjacent checker, removing the checker jumped over. Conway showed that it is not possible to reach row 5 in finitely many moves by weighting each cell in the board by powers of the golden ratio such that no move increases the total weight. Other authors have considered the game played on many different boards, including generalising the standard game to higher dimensions. We work on a board of arbitrary dimension, where we allow a cell to hold multiple checkers and begin with m checkers on each cell. We derive an upper bound and a constructive lower bound on the height that can be reached, such that the upper bound almost never fails to be equal to the lower bound. We also consider the more general case where instead of jumping over 1 checker, each checker moves by jumping over k checkers, and again show the maximum height reachable lies within bounds that are almost always equal., Comment: 17 pages, 7 figures

Language Model (LM) agents for cybersecurity that are capable of autonomously identifying vulnerabilities and executing exploits have the potential to cause real-world impact. Policymakers, model providers, and other researchers in the AI and cybersecurity communities are interested in quantifying the capabilities of such agents to help mitigate cyberrisk and investigate opportunities for penetration testing. Toward that end, we introduce Cybench, a framework for specifying cybersecurity tasks and evaluating agents on those tasks. We include 40 professional-level Capture the Flag (CTF) tasks from 4 distinct CTF competitions, chosen to be recent, meaningful, and spanning a wide range of difficulties. Each task includes its own description, starter files, and is initialized in an environment where an agent can execute bash commands and observe outputs. Since many tasks are beyond the capabilities of existing LM agents, we introduce subtasks, which break down a task into intermediary steps for more gradated evaluation; we add subtasks for 17 of the 40 tasks. To evaluate agent capabilities, we construct a cybersecurity agent and evaluate 7 models: GPT-4o, Claude 3 Opus, Claude 3.5 Sonnet, Mixtral 8x22b Instruct, Gemini 1.5 Pro, Llama 3 70B Chat, and Llama 3.1 405B Instruct. Without guidance, we find that agents are able to solve only the easiest complete tasks that took human teams up to 11 minutes to solve, with Claude 3.5 Sonnet and GPT-4o having the highest success rates. Finally, subtasks provide more signal for measuring performance compared to unguided runs, with models achieving a 3.2\% higher success rate on complete tasks with subtask-guidance than without subtask-guidance. While quiescent galaxies have comparable amounts of cool gas in their outer circumgalactic medium (CGM) compared to star-forming galaxies, they have significantly less interstellar gas. However, open questions remain on the processes causing galaxies to stop forming stars and stay quiescent . Theories suggest dynamical interactions with the hot corona prevent cool gas from reaching the galaxy, therefore predicting the inner regions of quiescent galaxy CGMs are devoid of cool gas. However, there is a lack of understanding of the inner regions of CGMs due to the lack of spatial information in quasar-sightline methods. We present integral-field spectroscopy probing 10--20~kpc (2.4--4.8 R\textsubscript{e}) around a massive quiescent galaxy using a gravitationally lensed star-forming galaxy. We detect absorption from Magnesium (MgII) implying large amounts of cool atomic gas (10\textsuperscript{8.4} -- 10\textsuperscript{9.3} M\textsubscript{$\odot$} with T$\sim$10\textsuperscript{4} Kelvin), in comparable amounts to star-forming galaxies. Lens modeling of Hubble imaging also reveals a diffuse asymmetric component of significant mass consistent with the spatial extent of the MgII absorption, and offset from the galaxy light profile. This study demonstrates the power of galaxy-scale gravitational lenses to not only probe the gas around galaxies, but to also independently probe the mass of the CGM due to it's gravitational effect., Comment: accepted nature communications physics

31. Real-world validation of safe reinforcement learning, model predictive control and decision tree-based home energy management systems

Recent advancements in machine learning based energy management approaches, specifically reinforcement learning with a safety layer (OptLayerPolicy) and a metaheuristic algorithm generating a decision tree control policy (TreeC), have shown promise. However, their effectiveness has only been demonstrated in computer simulations. This paper presents the real-world validation of these methods, comparing against model predictive control and simple rule-based control benchmark. The experiments were conducted on the electrical installation of 4 reproductions of residential houses, which all have their own battery, photovoltaic and dynamic load system emulating a non-controllable electrical load and a controllable electric vehicle charger. The results show that the simple rules, TreeC, and model predictive control-based methods achieved similar costs, with a difference of only 0.6%. The reinforcement learning based method, still in its training phase, obtained a cost 25.5\% higher to the other methods. Additional simulations show that the costs can be further reduced by using a more representative training dataset for TreeC and addressing errors in the model predictive control implementation caused by its reliance on accurate data from various sources. The OptLayerPolicy safety layer allows safe online training of a reinforcement learning agent in the real-world, given an accurate constraint function formulation. The proposed safety layer method remains error-prone, nonetheless, it is found beneficial for all investigated methods. The TreeC method, which does require building a realistic simulation for training, exhibits the safest operational performance, exceeding the grid limit by only 27.1 Wh compared to 593.9 Wh for reinforcement learning.

The Jovian atmosphere contains a wide diversity of vortices, which have a large range of sizes, colors and forms in different dynamical regimes. The formation processes for these vortices is poorly understood, and aside from a few known, long-lived ovals, such as the Great Red Spot, and Oval BA, vortex stability and their temporal evolution are currently largely unknown. In this study, we use JunoCam data and a citizen-science project on Zooniverse to derive a catalog of vortices, some with repeated observations, through May 2018 to Sep 2021, and analyze their associated properties, such as size, location and color. We find that different colored vortices (binned as white, red, brown and dark), follow vastly different distributions in terms of their sizes and where they are found on the planet. We employ a simplified stability criterion using these vortices as a proxy, to derive a minimum Rossby deformation length for the planet of $\sim1800$ km. If launched, STARI will be the first mission to control a 3-D CubeSat formation to the few mm-level, reflect starlight over 10s to 100s of meters from one spacecraft to another, control tip-tilt with sub-arcsecond stability, and validate end-to-end performance by injecting light into a single-mode fiber. The Probe far-Infrared Mission for Astrophysics (PRIMA) is a proposed space observatory which will use arrays of thousands of kinetic inductance detectors (KIDs) to perform low- and moderate-resolution spectroscopy throughout the far-infrared. The detectors must have noise equivalent powers (NEPs) at or below 0.1 aW/sqrt(Hz) to be subdominant to noise from sky backgrounds and thermal noise from PRIMA's cryogenically cooled primary mirror. Using a Radio Frequency System on a Chip for multitone readout, we measure the NEPs of detectors on a flight-like array designed to observe at a wavelength of 210 microns. We find that 92% of the KIDs measured have an NEP below 0.1 aW/sqrt(Hz) at a noise frequency of 10 Hz., Comment: 7 pages, 7 figures

Wire Arc Additive Manufacturing (WAAM) is a metal 3D printing technology that deposits molten metal wire on a substrate to form desired geometries. Articulated robot arms are commonly used in WAAM to produce complex geometric shapes. However, they mostly rely on proprietary robot and weld control software that limits process tuning and customization, incorporation of third-party sensors, implementation on robots and weld controllers from multiple vendors, and customizable user programming. This paper presents a general open-source software architecture for WAAM that addresses these limitations. The foundation of this architecture is Robot Raconteur, an open-source control and communication framework that serves as the middleware for integrating robots and sensors from different vendors. Based on this architecture, we developed an end-to-end robotic WAAM implementation that takes a CAD file to a printed WAAM part and evaluates the accuracy of the result. The major components in the architecture include part slicing, robot motion planning, part metrology, in-process sensing, and process tuning. The current implementation is based on Motoman robots and Fronius weld controller, but the approach is applicable to other industrial robots and weld controllers. The capability of the WAAM tested is demonstrated through the printing of parts of various geometries and acquisition of in-process sensor data for motion adjustment.

36. Measuring the Impact of New Risk Factors Within Survival Models

Survival is poor for patients with metastatic cancer, and it is vital to examine new biomarkers that can improve patient prognostication and identify those who would benefit from more aggressive therapy. In metastatic prostate cancer, two new assays have become available: one that quantifies the number of cancer cells circulating in the peripheral blood, and the other a marker of the aggressiveness of the disease. It is critical to determine the magnitude of the effect of these biomarkers on the discrimination of a model-based risk score. To do so, most analysts frequently consider the discrimination of two separate survival models: one that includes both the new and standard factors and a second that includes the standard factors alone. However, this analysis is ultimately incorrect for many of the scale-transformation models ubiquitous in survival, as the reduced model is misspecified if the full model is specified correctly. To circumvent this issue, we developed a projection-based approach to estimate the impact of the two prostate cancer biomarkers. The results indicate that the new biomarkers can influence model discrimination and justify their inclusion in the risk model; however, the hunt remains for an applicable model to risk-stratify patients with metastatic prostate cancer.

The Sun drives a supersonic wind which inflates a giant plasma bubble in our very local interstellar neighborhood, the heliosphere. It is bathed in an extremely variable background of energetic ions and electrons which originate from a number of sources. Solar energetic particles (SEPs) are accelerated in the vicinity of the Sun, whereas shocks driven by solar disturbances are observed to accelerate energetic storm particles (ESPs). Moreover, a dilute population with a distinct composition forms the anomalous cosmic rays (ACRs) which are of a mixed interstellar-heliospheric origin. Particles are also accelerated at planetary bow shocks. Medical imaging segmentation is a highly active area of research, with deep learning-based methods achieving state-of-the-art results in several benchmarks. However, the lack of standardized tools for training, testing, and evaluating new methods makes the comparison of methods difficult. To address this, we introduce the Medical Imaging Segmentation Toolkit (MIST), a simple, modular, and end-to-end medical imaging segmentation framework designed to facilitate consistent training, testing, and evaluation of deep learning-based medical imaging segmentation methods. MIST standardizes data analysis, preprocessing, and evaluation pipelines, accommodating multiple architectures and loss functions. This standardization ensures reproducible and fair comparisons across different methods. We detail MIST's data format requirements, pipelines, and auxiliary features and demonstrate its efficacy using the BraTS Adult Glioma Post-Treatment Challenge dataset. In this work, we introduce Gemma 2, a new addition to the Gemma family of lightweight, state-of-the-art open models, ranging in scale from 2 billion to 27 billion parameters. In this new version, we apply several known technical modifications to the Transformer architecture, such as interleaving local-global attentions (Beltagy et al., 2020a) and group-query attention (Ainslie et al., 2023). We also train the 2B and 9B models with knowledge distillation (Hinton et al., 2015) instead of next token prediction. The resulting models deliver the best performance for their size, and even offer competitive alternatives to models that are 2-3 times bigger. We release all our models to the community.

Equipped with a camera, Bluetooth radio, inertial measurement unit (IMU), and an onboard battery, IRIS meets the small size, weight, and power (SWaP) requirements for ring devices. IRIS is context-aware, adapting its gesture set to the detected device, and can last for 16-24 hours on a single charge. IRIS leverages the scene semantics to achieve instance-level device recognition. In a study involving 23 participants, IRIS consistently outpaced voice commands, with a higher proportion of participants expressing a preference for IRIS over voice commands regarding toggling a device's state, granular control, and social acceptability. Traditional cosmic ray filtering algorithms used in X-ray imaging detectors aboard space telescopes perform event reconstruction based on the properties of activated pixels above a certain energy threshold, within 3x3 or 5x5 pixel sliding windows. This approach can reject up to 98% of the cosmic ray background. However, the remaining unrejected background constitutes a significant impediment to studies of low surface brightness objects, which are especially prevalent in the high-redshift universe. The main limitation of the traditional filtering algorithms is their ignorance of the long-range contextual information present in image frames. This becomes particularly problematic when analyzing signals created by secondary particles produced during interactions of cosmic rays with body of the detector. Such signals may look identical to the energy deposition left by X-ray photons, when one considers only the properties within the small sliding window. Additional information is present, however, in the spatial and energy correlations between signals in different parts of the frame, which can be accessed by modern machine learning (ML) techniques. In this work, we continue the development of an ML-based pipeline for cosmic ray background mitigation. Our latest method consist of two stages: first, a frame classification neural network is used to create class activation maps (CAM), localizing all events within the frame; second, after event reconstruction, a random forest classifier, using features obtained from CAMs, is used to separate X-ray and cosmic ray features. The method delivers >40% relative improvement over traditional filtering in background rejection in standard 0.3-10keV energy range, at the expense of only a small (<2%) level of lost X-ray signal. Future strategic X-ray satellite telescopes, such as the probe-class Advanced X-ray Imaging Satellite (AXIS), will require excellent soft energy response in their imaging detectors to enable maximum discovery potential. In order to characterize Charge-Coupled Device (CCD) and Single Electron Sensitive Read Output (SiSeRO) detectors in the soft X-ray region, the X-ray Astronomy and Observational Cosmology (XOC) group at Stanford has developed, assembled, and commissioned a 2.5-meter-long X-ray beamline test system. The beamline is designed to efficiently produce monoenergetic X-ray fluorescence lines in the 0.3-10 keV energy range and achieve detector temperatures as low as 173 K. We present design and simulation details of the beamline, and discuss the vacuum, cooling, and X-ray fluorescence performance achieved. The Advanced X-ray Imaging Satellite (AXIS) is a NASA probe class mission concept designed to deliver arcsecond resolution with an effective area ten times that of Chandra (at launch). The AXIS focal plane features an MIT Lincoln Laboratory (MIT-LL) X-ray charge-coupled device (CCD) detector working in conjunction with an application specific integrated circuit (ASIC), denoted the Multi-Channel Readout Chip (MCRC). While this readout ASIC targets the AXIS mission, it is applicable to a range of potential X-ray missions with comparable readout requirements. Designed by the X-ray astronomy and Observational Cosmology (XOC) group at Stanford University, the MCRC ASIC prototype (MCRC-V1.0) uses a 350 nm technology node and provides 8 channels of high speed, low noise, low power consumption readout electronics. Each channel implements a current source to bias the detector output driver, a preamplifier to provide gain, and an output buffer to interface directly to an analog-to-digital (ADC) converter. The MCRC-V1 ASIC exhibits comparable performance to our best discrete electronics implementations, but with ten times less power consumption and a fraction of the footprint area. In a total ionizing dose (TID) test, the chip demonstrated a radiation hardness equal or greater to 25 krad, confirming the suitability of the process technology and layout techniques used in its design. The next iteration of the ASIC (MCRC-V2) will expand the channel count and extend the interfaces to external circuits, advancing its readiness as a readout-on-a-chip solution for next generation X-ray CCD-like detectors. Future strategic X-ray astronomy missions will require unprecedentedly sensitive wide-field imagers providing high frame rates, low readout noise and excellent soft energy response. To meet these needs, our team is employing a multi-pronged approach to advance several key areas of technology. Our first focus is on advanced readout electronics, specifically integrated electronics, where we are collaborating on the VERITAS readout chip for the Athena Wide Field Imager, and have developed the Multi-Channel Readout Chip (MCRC), which enables fast readout and high frame rates for MIT-LL JFET (junction field effect transistor) CCDs. Second, we are contributing to novel detector development, specifically the SiSeRO (Single electron Sensitive Read Out) devices fabricated at MIT Lincoln Laboratory, and their advanced readout, to achieve sub-electron noise performance. Hardware components set the stage for performance, but their efficient utilization relies on software and algorithms for signal and event processing. Our group is developing digital waveform filtering and AI methods to augment detector performance, including enhanced particle background screening and improved event characterization. Single electron Sensitive Read Out (SiSeRO) is a novel on-chip charge detection technology that can, in principle, provide significantly greater responsivity and improved noise performance than traditional charge coupled device (CCD) readout circuitry. The SiSeRO, developed by MIT Lincoln Laboratory, uses a p-MOSFET transistor with a depleted back-gate region under the transistor channel; as charge is transferred into the back gate region, the transistor current is modulated. With our first generation SiSeRO devices, we previously achieved a responsivity of around 800 pA per electron, an equivalent noise charge (ENC) of 4.5 electrons root mean square (RMS), and a full width at half maximum (FWHM) spectral resolution of 130 eV at 5.9 keV, at a readout speed of 625 Kpixel/s and for a detector temperature of 250 K. Importantly, since the charge signal remains unaffected by the SiSeRO readout process, we have also been able to implement Repetitive Non-Destructive Readout (RNDR), achieving an improved ENC performance. In this paper, we demonstrate sub-electron noise sensitivity with these devices, utilizing an enhanced test setup optimized for RNDR measurements, with excellent temperature control, improved readout circuitry, and advanced digital filtering techniques. We are currently fabricating new SiSeRO detectors with more sensitive and RNDR-optimized amplifier designs, which will help mature the SiSeRO technology in the future and eventually lead to the pathway to develop active pixel sensor (APS) arrays using sensitive SiSeRO amplifiers on each pixel. Digital image manipulation has become increasingly accessible and realistic with the advent of generative AI technologies. Recent developments allow for text-guided inpainting, making sophisticated image edits possible with minimal effort. This poses new challenges for digital media forensics. For example, diffusion model-based approaches could either splice the inpainted region into the original image, or regenerate the entire image. In the latter case, traditional image forgery localization (IFL) methods typically fail. This paper introduces the Text-Guided Inpainting Forgery (TGIF) dataset, a comprehensive collection of images designed to support the training and evaluation of image forgery localization and synthetic image detection (SID) methods. The TGIF dataset includes approximately 80k forged images, originating from popular open-source and commercial methods; SD2, SDXL, and Adobe Firefly. Using this data, we benchmark several state-of-the-art IFL and SID methods. Whereas traditional IFL methods can detect spliced images, they fail to detect regenerated inpainted images. Moreover, traditional SID may detect the regenerated inpainted images to be fake, but cannot localize the inpainted area. Finally, both types of methods fail when exposed to stronger compression, while they are less robust to modern compression algorithms, such as WEBP. As such, this work demonstrates the inefficiency of state-of-the-art detectors on local manipulations performed by modern generative approaches, and aspires to help with the development of more capable IFL and SID methods. The standard cosmological model, which assumes statistical isotropy and parity invariance, predicts the absence of correlations between even-parity and odd-parity observables of the cosmic microwave background (CMB). Contrary to these predictions, large-angle CMB temperature anomalies generically involve correlations between even-$\ell$ and odd-$\ell$ angular power spectrum $C_\ell$, while recent analyses of CMB polarization have revealed non-zero equal-$\ell$ $EB$ correlations. These findings challenge the conventional understanding, suggesting deviations from statistical isotropy, violations of parity, or both. Cosmic topology, which involves changing only the boundary conditions of space relative to standard cosmology, offers a compelling framework to potentially account for such parity-violating observations. Topology inherently breaks statistical isotropy, and can also break homogeneity and parity, providing a natural paradigm for explaining observations of parity-breaking observables without the need to add parity violation to the underlying microphysics. Our investigation delves into the harmonic space implications of topology for CMB correlations, using as an illustrative example $EB$ correlations generated by tensor perturbations under both parity-preserving and parity-violating scenarios. Consequently, these findings not only challenge the foundational assumptions of the standard cosmological model but also open new avenues for exploring the topological structure of the Universe through CMB observations., Comment: 20 pages, 4 figures

Nuclear Experiment - Abstract
The PHENIX experiment measured the centrality dependence of two-pion Bose-Einstein correlation functions in $\sqrt{s_{_{NN}}}=200$~GeV Au$+$Au collisions at the Relativistic Heavy Ion Collider at Brookhaven National Laboratory. The data are well represented by L\'evy-stable source distributions. The extracted source parameters are the correlation-strength parameter $\lambda$, the L\'evy index of stability $\alpha$, and the L\'evy-scale parameter $R$ as a function of transverse mass $m_T$ and centrality. The $\lambda(m_T)$ parameter is constant at larger values of $m_T$, but decreases as $m_T$ decreases. The L\'evy scale parameter $R(m_T)$ decreases with $m_T$ and exhibits proportionality to the length scale of the nuclear overlap region. The L\'evy exponent $\alpha(m_T)$ is independent of $m_T$ within uncertainties in each investigated centrality bin, but shows a clear centrality dependence. At all centralities, the L\'evy exponent $\alpha$ is significantly different from that of Gaussian ($\alpha=2$) or Cauchy ($\alpha=1$) source distributions. Comparisons to the predictions of Monte-Carlo simulations of resonance-decay chains show that in all but the most peripheral centrality class (50%-60%), the obtained results are inconsistent with the measurements, unless a significant reduction of the in-medium mass of the $\eta'$ meson is included. In each centrality class, the best value of the in-medium $\eta'$ mass is compared to the mass of the $\eta$ meson, as well as to several theoretical predictions that consider restoration of $U_A(1)$ symmetry in hot hadronic matter., Comment: 401 authors from 75 institutions, 20 pages, 15 figures, 2 tables. v1 is version submitted to Physical Review C. Electrons in a quantum Hall fluid can bind with an integer number of vortices to form composite fermions and composite bosons. We show that the quantum Hall bilayer at filling $\nu=\frac{1}{4}+\frac{3}{4}$ with interlayer separation $d$ can be well-described in terms of these composite particles. At small $d$ the system can be understood as interlayer paired electrons and holes, whereas at large $d$ the system is best understood in terms of composite fermions with four vortices attached to each electron. By computing the overlaps of trial wavefunctions with the ground state from exact diagonalization, we find that as $d$ increases, the number of vortices that attach to each electron increases. We also construct trial states for two types of excitation, the Goldstone mode and a meron excitation. These two trial states have good overlaps with the lowest excited states in the exact diagonalization spectrum for small and intermediate $d$ respectively., Comment: 6 pages, 2 figures (3 pages, 4 figures in supplement)
