Abstract While quiescent galaxies have comparable amounts of cool gas in their outer circumgalactic medium (CGM) compared to star-forming galaxies, they have significantly less interstellar gas. However, open questions remain on the processes causing galaxies to stop forming stars and stay quiescent. Theories suggest dynamical interactions with the hot corona prevent cool gas from reaching the galaxy, therefore predicting the inner regions of quiescent galaxy CGMs are devoid of cool gas. However, there is a lack of understanding of the inner regions of CGMs due to the lack of spatial information in quasar-sightline methods. We present integral-field spectroscopy probing 10–20 kpc (2.4–4.8 Re) around a massive quiescent galaxy using a gravitationally lensed star-forming galaxy. We detect absorption from Magnesium (MgII) implying large amounts of cool atomic gas (108.4–109.3 M⊙ with T~104 Kelvin), in comparable amounts to star-forming galaxies. Lens modeling of Hubble imaging also reveals a diffuse asymmetric component of significant mass consistent with the spatial extent of the MgII absorption, and offset from the galaxy light profile. This study demonstrates the power of galaxy-scale gravitational lenses to not only probe the gas around galaxies, but to also independently probe the mass of the CGM due to it’s gravitational effect.

Abstract Here we present a sample of 12 massive quiescent galaxy candidates at $$z\sim 3-4$$ z ∼ 3 - 4 observed with the James Webb Space Telescope (JWST) Near Infrared Spectrograph (NIRSpec). These galaxies were pre-selected from the Hubble Space Telescope imaging and 10 of our sources were unable to be spectroscopically confirmed by ground based spectroscopy. By combining spectroscopic data from NIRSpec with multi-wavelength imaging data from the JWST Near Infrared Camera (NIRCam), we analyse their stellar populations and their formation histories. We find that all of our galaxies classify as quiescent based on the reconstruction of their star formation histories but show a variety of quenching timescales and ages. All our galaxies are massive ( $$\sim 0.1-1.2\times 10^{11}$$ ∼ 0.1 - 1.2 × 10 11 M $$_\odot$$ ⊙ ), with masses comparable to massive galaxies in the local Universe. We find that the oldest galaxy in our sample formed $$\sim 1.0\times 10^{11}$$ ∼ 1.0 × 10 11 M $$_\odot$$ ⊙ of mass within the first few hundred million years of the Universe and has been quenched for more than a billion years by the time of observation at $$z\sim 3.2$$ z ∼ 3.2 ( $$\sim 2$$ ∼ 2 billion years after the Big Bang). Our results point to very early formation of massive galaxies requiring a high conversion rate of baryons to stars in the early Universe.

The baryon cycle is crucial for understanding galaxy formation, as gas inflows and outflows vary throughout a galaxy’s lifetime and affect its star formation rate. Despite the necessity of accretion for galaxy growth at high redshifts, direct observations of inflowing gas have proven elusive, especially at z ≳ 2. We present a spectroscopic analysis of a galaxy at redshift z = 2.45, which exhibits signs of inflow in several ultraviolet interstellar absorption lines, with no clear outflow signatures. The absorption lines are redshifted by ∼250 km s ^−1 with respect to the systemic redshift, and C iv shows a prominent inverse P-Cygni profile. Simple stellar population models suggest that this galaxy has a low metallicity (∼5% solar), with a very young starburst of age ∼4 Myr dominating the ultraviolet luminosity. The gas inflow velocity and nebular velocity dispersion suggest an approximate halo mass of order ∼10 ^11 M _⊙ , a regime in which simulations predict that bursty star formation is common at this redshift. We conclude that this system is likely in the beginning of a cycle of bursty star formation, where inflow and star formation rates are high, but where supernovae and other feedback processes have not yet launched strong outflows. In this scenario, we expect the inflow-dominated phase to be observable (e.g., with net redshifted interstellar medium absorption) for only a short timescale after a starburst onset. This result represents a promising avenue for probing the full baryon cycle, including inflows, during the formative phases of low-mass galaxies at high redshifts.

Background: The use of preoperative breast magnetic resonance imaging (MRI) after the diagnosis of breast cancer by mammography and/or ultrasound is inconsistent. Methods: After conducting a systematic review and meta-analysis comparing preoperative breast MRI versus no MRI, we reconvened to prepare a clinical practice guideline on this topic. Results: Based on the evidence that MRI improved recurrence, decreased the rates of reoperations (re-excisions or conversion mastectomy), and increased detection of synchronous contralateral breast cancer, we recommend that preoperative breast MRI should be considered on a case-by-case basis in patients diagnosed with breast cancer for whom additional information about disease extent could influence treatment. Based on stronger evidence, preoperative breast MRI is recommended in patients diagnosed with invasive lobular carcinoma for whom additional information about disease extent could influence treatment. For both recommendations, the decision to proceed with MRI would be conditional on shared decision-making between care providers and the patient, taking into account the benefits and risks of MRI as well as patient preferences. Based on the opinion of the Working Group, preoperative breast MRI is also recommended in the following more specific situations: (a) to aid in surgical planning of breast conserving surgery in patients with suspected or known multicentric or multifocal disease; (b) to identify additional lesions in patients with dense breasts; (c) to determine the presence of pectoralis major muscle/chest wall invasion in patients with posteriorly located tumours or when invasion of the pectoralis major muscle or chest wall is suspected; (d) to aid in surgical planning for skin/nipple-sparing mastectomies, autologous reconstruction, oncoplastic surgery, and breast conserving surgery with suspected nipple/areolar involvement; and (e) in patients with familial/hereditary breast cancer but who have not had recent breast MRI as part of screening or diagnosis.

Inflammation serves as a pivotal defense mechanism orchestrated by the innate immune system to safeguard cellular health against adversities. Nonetheless, dysregulated inflammatory responses can precipitate chronic inflammatory ailments, notably autoimmune disorders. Central to this process are various pathways, with studies highlighting the pivotal role of transcription factors within the nuclear factor-kappa B (NF-κB) signaling pathway in disease onset and progression. This study concentrates on the p50 homodimer protein, a key transcription factor pivotal for the expression of proinflammatory cytokine genes. To explore potential inhibitors of p50, we conducted in silico procedures to investigate fifty-eight unexplored compounds, derived from plants indigenous to the Philippines. Initial screenings for compound feasibility, through drug-likeness analyses, yielded positive outcomes for 34 compounds. Subsequent docking analyses revealed six compounds exhibiting binding energies (ranging from −3.7 to −4.2 kcal/mol) akin to or lower than the positive control, dexamethasone (−3.7 kcal/mol). These compounds include eudesm-11-en-4α-O-β-D-3-tigoyloxy-6-deoxy-glucopyranoside, wadeiol, grandiflorolide, eudesm-11-en-4α-O-β-D-3-senecioyloxy-6-deoxyglucopyranoside, α-pinene-7β-O-β-D-2- acetylglucopyranoside, and (2aβ,3α,5aβ,6β,7α,8aα)-6-[2-(3-furanyl)ethyl]-2a,3,4,5,5a,6,7,8,8a,8b- decahydro-2a,3-dihydroxy-6,7,8b-trimethyl-2H-naphtho[1-8-bc]furan-2-one. Interaction analyses revealed a common engagement of amino acid residues within the p50 DNA binding pocket, notably Arg57, Tyr60, Glu63, Lys244, Ala245, Pro246, Lys275, Arg308, Gln309, and Phe310, through hydrogen bonding, van der Waals forces, alkyl, and pi–alkyl interactions. Pharmacophore analysis underscored aromatic rings, hydroxyl, methyl, and methylene groups as pivotal for non-covalent interactions with p50. Additionally, root mean square fluctuation (RMSF) analysis demonstrated minimal residue fluctuations in p50 upon ligand binding compared to the ligand-free protein structure. In conclusion, the six shortlisted compounds exhibiting comparable binding affinities with dexamethasone hold promise as potential anti-inflammatory agents targeting the NF-κB p50 homodimer.

We present the discovery of a unique Lyman-continuum (LyC) emitter at z = 3.088. The LyC emission was detected using the Hubble Space Telescope WFC3/UVIS F336W filter, covering a rest-frame wavelength range of 760–900 Å. The peak signal-to-noise ratio of LyC emission is 3.9 in an r = 0.″24 aperture and is spatially offset by 0.″29 ± 0.″04 (∼2.2 ± 0.3 kpc) from the peak of rest-UV emission (F606W). By combining imaging and spectroscopic data from the James Webb Space Telescope (JWST) JADES, FRESCO, and JEMS surveys, along with VLT/MUSE data from the MXDF survey, we estimate that the probability of random alignment with an interloper galaxy causing the LyC emission is less than 6 × 10 ^−5 . The interstellar medium (ISM) conditions in the galaxy are similar to those in other LyC emitters at high redshift ( $12+\mathrm{log}({\rm{O}}/{\rm{H}})={7.79}_{-0.05}^{+0.06}$ , $\mathrm{log}U=-{3.27}_{-0.12}^{+0.14}$ , O32 = 0.63 ± 0.03), although the single-peaked Ly α profile and lack of rest-UV emission lines suggest an optically thick ISM. Our observations indicate that LyC photons are leaking through a narrow cone of optically thin neutral ISM, most likely created by a past merger (as evidenced by medium-band F210M and F182M images). Using the constraints on escape fraction from individual leakers and a simple model, we estimate that the opening half-angle of ionization cones can be as small as 16° (2% ionized fraction) to reproduce some of the theoretical constraints on the average escape fraction for galaxies. The narrow opening angle required can explain the low number density of confirmed LyC leakers.

Over the past few years alone, the lensing community has discovered thousands of strong lens candidates, and spectroscopically confirmed hundreds of them. In this time of abundance, it becomes pragmatic to focus our time and resources on the few extraordinary systems, in order to most efficiently study the Universe. In this paper, we present such a system: DESI-090.9854-35.9683, a cluster-scale lens at z _l = 0.49, with seven observed lensed sources around the core, and additional lensed sources further out in the cluster. From the number and the textbook configuration of the lensed images, a tight constraint on the mass potential of the lens is possible. This would allow for detailed analysis on the dark and luminous matter content within galaxy clusters, as well as a probe into dark energy and high-redshift galaxies. We present our spatially resolved kinematic measurements of this system from the Very Large Telescope Multi Unit Spectroscopic Explorer, which confirm five of these source galaxies (in ascending order, at z _s = 0.962, 0.962, 1.166, 1.432, and 1.432). With previous Hubble Space Telescope imaging in the F140W and F200LP bands, we also present a simple flux-based lens model consisting of two power-law profiles that, for a cluster lens, well models the five lensed arc families with redshifts. We determine the mass to be M (< θ _E ) = 4.78 × 10 ^13 M _⊙ for the primary mass potential. From the model, we extrapolate the redshift of one of the two source galaxies not yet spectroscopically confirmed to be at ${z}_{{\rm{s}}}={4.52}_{-0.71}^{+1.03}$ .

Observed evolution of the total mass distribution with redshift is crucial to testing galaxy evolution theories. To measure the total mass distribution, strong gravitational lenses complement the resolved dynamical observations that are currently limited to z ≲ 0.5. Here we present the lens models for a pilot sample of seven galaxy-scale lenses from the ASTRO3D Galaxy Evolution with Lenses ( AGEL ) survey. The AGEL lenses, modeled using HST/WFC3-F140W images with Gravitational Lens Efficient Explorer ( GLEE ) software, have deflector redshifts in the range 0.3 < z _defl < 0.9. Assuming a power-law density profile with slope γ , we measure the total density profile for the deflector galaxies via lens modeling. We also measure the stellar velocity dispersions ( σ _obs ) for four lenses and obtain σ _obs from SDSS - BOSS for the remaining lenses to test our lens models by comparing observed and model-predicted velocity dispersions. For the seven AGEL lenses, we measure an average density profile slope of −1.95 ± 0.09 and a γ – z relation that does not evolve with redshift at z < 1. Although our result is consistent with some observations and simulations, it differs from other studies at z < 1 that suggest the γ – z relation evolves with redshift. The apparent conflicts among observations and simulations may be due to a combination of (1) systematics in the lensing and dynamical modeling; (2) challenges in comparing observations with simulations; and (3) assuming a simple power law for the total mass distribution. By providing more lenses at z _defl > 0.5, the AGEL survey will provide stronger constraints on whether the mass profiles evolve with redshift as predicted by current theoretical models.

We present an all-southern sky survey for Mg ii λλ 2796, 2803 doublet absorbers in 951 z < 4 active galactic nucleus/quasar spectra from the Australian Dark Energy Survey. The spectral resolution ranges from R = 1400 to 1700 over the wavelengths 3700–8800 Å. The survey has a 5 σ detection completeness of 50% and above for rest-frame equivalent widths W _r (2796) ≥ 0.3 Å. We studied 656 Mg ii absorption systems over the redshift range 0.33 ≤ z ≤ 2.19 with equivalent widths 0.3 ≤ W _r (2796) ≤ 3.45 Å. The equivalent width distribution is well fit by an exponential function with W _* = 0.76 ± 0.04 Å, and the redshift path density exhibits very little evolution. Overall, our findings are consistent with the large, predominantly northern sky, surveys of Mg ii absorbers. We developed and implemented a Monte Carlo model informed by a high-resolution Mg ii survey for determining the Mg ii mass density, Ω _Mg _II . We found Ω _Mg _II ∼ 5 × 10 ^−7 with no evidence of evolution over a ∼7 Gyr time span following Cosmic Noon. Incorporating measurements covering 2.0 ≤ z ≤ 6.4 from the literature, we extended our insights into Mg ii mass density evolution from the end of reionization well past the Cosmic Noon epoch. The presented Monte Carlo model has potential for advancing our knowledge of the evolution of mass densities of metal ions common to quasar absorption line studies, as it exploits the efficiency of large low-resolution surveys while requiring only small samples from expensive high-resolution surveys.

Bone autografts are frequently harvested for use in foot and ankle surgery. A commonly used harvest site is the iliac crest; however, because of known morbidity with this site, the tibia and calcaneus are attractive alternatives. There remains limited understanding regarding the osteogenic potential of autografts from each of these locations. In this review, we provided an update of the known data on bone autografts from the iliac crest, tibia, and calcaneus, focusing on the total cells harvested from each site as well as the presence of osteogenic osteoprogenitor cells. Work (labor), for the Visayan, is an essential expression of freedom which has a similar expression to JPII’s Philosophy of Labor.

Category: Midfoot/Forefoot Introduction/Purpose: Midfoot osteoarthritis affects up to 12% of the adult population greater than 50 years old. Arthrodesis is the traditional surgical procedure performed when nonoperative management fails but is a technically demanding procedure that requires prolonged postoperative immobilization and is not uncommonly complicated by nonunion or deep infection. The deep peroneal nerve (DPN) plays a significant role in afferent nociceptive midfoot joint pain perception, most consistently in the second and third tarsometatarsal joints and the naviculocuneiform joint. Early outcomes following DPN neurectomy have demonstrated promising results as a safe and effective treatment that allows early mobilization and weight bearing. The purpose of our study was to evaluate the mid-term patient satisfaction and pain relief after DPN neurectomy for treatment of chronic midfoot pain due to osteoarthritis. Methods: In this retrospective IRB-approved questionnaire-based study, we evaluated 49 out of 95 patients identified (56 feet) with an average follow-up of 34.6 (range, 9 to 51) months, who underwent DPN neurectomy (Figure 1) at our institution between September 2017 and February 2021. There were 39 women and 10 men, 42 unilateral (23 right, 19 left) and 7 bilateral procedures, with an average age of 67.9 (range, 35 to 88) years at the time of surgery. A questionnaire which included questions regarding post-surgical dorsal midfoot pain relief, surgical result satisfaction, and current functional limitations was administered via telephone. Demographic information, patient responses, and complications were recorded. Results: There was an 81.2% patient satisfaction with the result of the surgery in relieving their dorsal midfoot pain. Of the 49 patients, 84.9% would repeat the surgery under the same circumstances, 84.1% would recommend the surgery to a friend, 10.2% reported they wish they had undergone arthrodesis, 8.2% reported no pain relief in the first 6 months, and 55.8% reported current activity limitations because of dorsal midfoot pain. Six feet (10.7%) underwent a second procedure with an average postoperative time of 20.5 (range, 1 to 36) months. Complications include one patient with deep wound infection, one DPN neurectomy revision and burial into bone, one DPN neuroma excision, one midfoot exostosis excision, and two conversions to arthrodesis. Theoretic complications of neuroarthropathy, midfoot collapse, and development of lesser toe deformities were not observed. Conclusion: DPN neurectomy is a safe and effective surgical option for the management of chronic dorsal midfoot pain due to midfoot osteoarthritis after failed nonoperative management. At midterm follow-up, patients who underwent dorsal midfoot denervation reported acceptable satisfaction rates, similar to those reported for arthrodesis, with associated improved functional outcomes and few complications.

The gold standard harvest site is the iliac crest; however there is known morbidity with this harvest site. Alternative harvest sites include the tibia and calcaneus which are easily accessible for lower extremity surgery. Controversy exists regarding the osteogenic potential of autografts from each location. We performed a systematic review of the known data on cellular autografts from the iliac crest, tibia, and calcaneus focusing on the total cells harvested from each site as well as the presence of the critically important osteogenic osteoprogenitor cells. We also reviewed autograft volume, autograft consistency, surgical techniques, and the morbidities and complications associated with each bone autograft harvest location. Methods: In accordance with the PRISMA 2020 guidelines, a literature search of the PubMed, Cochrane Library, Web of Science, and Google databases from inception through January 2022 was performed. The search terms included: 'autograft,' 'bone graft,' 'foot and ankle surgery,' 'iliac crest,' 'tibia,' 'calcaneus,' reamer irrigator aspirator,' 'RIA,' 'mesenchymal cells,' 'osteoprogenitor cells,' 'quantity,' 'number,' 'amount,' 'yield,' 'concentration,' 'surface antigens,' 'surface markers,' 'CD markers.' Studies were included if autologous bone graft was harvested from the iliac crest, femur, tibia, or calcaneus and the cells in the harvest underwent histologic evaluation, quantification, or CD surface antigen identification. We included studies with bone graft as well as bone marrow aspirates used as samples. The nucleated cell and osteoprogenitor cell yield was recorded from each location; and MSC CD markers present at each harvest site were extracted from the eligible studies. Results: 13 studies met our inclusion criteria. Eight studies performed cell quantification from the harvest sites. Notably these studies only provide counting of total nucleated cells without specific identification of the osteoprogenitors. Colony-forming units were also reported and their findings are reviewed. Total cellular material was on average greatest in the iliac crest, with intermediate counts in the proximal tibia, and lowest in the calcaneus autograft samples. Five studies identified mesenchymal stromal cell (MSC) cluster of differentiation (CD) antigens at the harvest sites of interest. CD markers confirm osteoprogenitor cells are present in each sample location. Total number of osteoprogenitor cells remains unknown, however authors have attempted to extrapolate this number based on number of total nucleated cells and other data. Conclusion: Osteoprogenitor cells can be harvested from the iliac crest, tibia, and calcaneus. Greater total nucleated cell yields were identified in harvests from the iliac crest with descending amounts moving location distally in the limb. Nucleated cells were counted to provident estimates of osteoprogenitor cells yields in most studies, with flow cytometry confirmation that osteoprogenitors are present. Strict quantification of osteoprogenitor cells remains unknown. Multiple studies compared cellular material from bone marrow aspirates versus bone graft harvests themselves. Studies with modern techniques are needed to identify the true number of osteoprogenitors in each location.

Category: Trauma; Ankle Introduction/Purpose: Supination-external rotation (SER) variant ankle fracture is common, and identifying the unstable SER variants requiring surgery has been controversial. Stress view radiographs are frequently obtained to assist in decision-making. Measurement of the medial clear space (MCS) is used to determine medial structure integrity. SER variant ankle injuries are characterized by talus external rotation, and posterolateral translation causing a posterior displaced distal fibula fracture, with possible medial and posterior injury at further subluxation events. We propose that the MCS measurement is flawed by measuring from the point of rotational pivoting. Measurement of lateral displacement of the lateral talus versus the tibial plafond will be a more sensitive measurement tool for an injury mechanism characterized by external rotation and posterolateral subluxation. Methods: IRB-approved, retrospective review of ankle injury radiographs for acute SER-variant isolated lateral malleolar fractures during a 3-year consecutive period. Patients were included if they had a gravity-stress view (GSV) evaluation. Patients were excluded for different fracture morphology, pediatric fractures, no GSV films, or prior history of ankle or foot arthritis or trauma. A priori power analysis was performed demonstrating 32 patients would be required to detect a change in 2mm. 102 patients underwent analysis. GSV were analyzed with measurements taken of the medial clear space (MCS), superior clear space (SCS), and we propose two novel measurements of lateral talus subluxation (LTS) by comparison to the anterior ankle syndesmosis marked by a vertical line at the tibial plafond. Standardized measurements were acquired by foot-and-ankle fellowship-trained orthopaedic surgeons and musculoskeletal radiologists. ICC data was obtained. Surgeons separately provided blinded decisions- for-surgery which patients appeared to have ankle instability requiring surgical stabilization. Results: Measurements were performed for 102 patients, there were no significant differences in baseline demographics. There was a statistically significant increase in mean MCS, SCS, and LTS within the operative group (MCS 7.3 vs 5, p=0.005, SCS 4.6 vs 3.9, p=0.02, LTS 6.1 vs 2.9, p =5mm had a sensitivity of 95% and specificity of 60% in our cohort, LTS >4mm demonstrating the same sensitivity (95%) with improved specificity to 75%. Receiver-operator-characteristic analysis for MCS vs LTS demonstrates an area under the curve (AUC) 0.786 for MCS >4 versus LTS (AUC) 0.918. Conclusion: The LTS measurement is superior to MCS measurement as a surrogate for medial ankle structure stability. We propose that ankle joint subluxation can be more accurately identified by measuring nearer to the point of maximal talar displacement in an external rotation and posterolaterally displacing injury pattern. The LTS measurement elevation >4mm was found to be superior to the MCS measurement >4mm in all categories, with high sensitivity and specificity, and superior performance in ROC analysis. We propose this will be a useful tool for clinicians when evaluating SER variant ankle fractures.

Virtual care in cancer care existed in a limited fashion globally before the COVID-19 pandemic, mostly driven by geographic constraints. The pandemic has required dramatic shifts in health care delivery, including cancer care. We conducted a systematic review of comparative studies evaluating virtual versus in-person care in patients with cancer. Embase, APA PsycInfo, Ovid MEDLINE, and the Cochrane Library were searched for literature from January 2015 to 6 August 2020. We adhered to PRISMA guidelines and used the modified GRADE approach to evaluate the data. We included 34 full-text publications of 10 randomized controlled trials, 13 non-randomized comparative studies, and 5 ongoing randomized controlled trials. Evidence was divided into studies that provide psychosocial or genetic counselling and those that provide or assess medical and supportive care. The limited data in this review support that in the general field of psychological counselling, virtual or remote counselling can be equivalent to in-person counselling. In the area of genetic counselling, telephone counselling was more convenient and noninferior to usual care for all outcomes (knowledge, decision conflict, cancer distress, perceived stress, genetic counseling satisfaction). There are few data for clinical outcomes and supportive care. Future research should assess the role of virtual care in these areas. Protocol registration: PROSPERO CRD42020202871.

Introduction Total knee arthroplasty is a common operation performed to relieve pain and restore functional activity. While overall widely successful, a subset of patients has continued pain postoperatively with no identifiable cause. Neuroma formation has been identified as a possible contributor to this unexplained pain, often necessitating an additional procedure for neuroma removal. The purpose of our study was to evaluate if prophylactic neurectomy could reduce the occurrence of postoperative pain. Methods A total of 112 patients were compared, 44 control patients and 68 neurectomy patients. Demographic information, Numerical rating pain scale (NRS) and Knee Society Scores (KSS) were collected pre- and post-operatively. Patients were additional asked if they were overall satisfied with the operation. Results There were no differences between groups with respect to age (Median: 71 vs 69 years, p = 0.28), male sex (41% vs 44%, p = 0.85), or body mass index (Median: 32.2 vs 31.3, p = 0.80). When comparing the degree of change following surgery there were no statistically significant differences observed in NRS pain scores (Median change: −7 vs −6, p = 0.89) or KSS scores (Median change: +44 vs +40, p = 0.14). Similarly, there was no statistically significant difference in overall patient-reported satisfaction with the knee replacement (82.5% vs 86.6%, p = 0.59). Conclusion We did not find a statistically significant difference in NRS, KSS, or overall patient satisfaction between the prophylactic neurectomy and control patient groups. Larger studies with evaluation of the nerve diameter will be needed to determine which patients are at risk for symptomatic neuroma development following total knee arthroplasty.

Objective: To evaluate antioxidant, cytotoxic, and anti-venom capacity of crude bark extracts of Alstonia parvifolia Merr. Methods: Gas chromatography-mass spectrometry (GC-MS) and energy dispersive X-ray analyses were accomplished to characterize the chemical constituents of Alstonia parvifolia. Biochemical characterization was evaluated using an inhibitory phospholipase A2 (PLA2) assay, DPPH, and cytotoxicity assays. Using the constituents listed in the GC-MS analyses, molecular docking was conducted to inspect the binding energies between the chosen compounds and selected PLA2 isoforms. Results: GC-MS analyses showed that the Alstonia parvifolia crude extract consisted predominantly of acetylmarinobufogenin (14.89%), γ-sitosterol (10.44%), 3-O-methyl-D-glucose (5.88%), 3,5-dimethoxy-4-hydroxyphenylacetic acid (5.30%), (2α,5α)-17-methoxyaspidofractinin-3-one (AFM) (4.08%), and 2,3,5,6,7,8,9-heptahydro-1-phenyl-5-(p-chlorophenylimino)-1H-benzo[e][1],[4]thiazepine (HPT) (1.37%). The principal elemental components of Alstonia parvifolia were Ca (4.012%) and K (1.496%), as exhibited by energy dispersive X-ray examination. Alstonia parvifolia showed significant free radical scavenging ability (IC50: 0.287 mg/mL) and was non-cytotoxic to normal HDFn cells (IC50 >100 μg/mL). Moreover, Alstonia parvifolia was favorably cytotoxic to MCF-7 (IC50: 4.42 μg/mL), followed by H69PR, HT-29, and THP-1, with IC50 values of 4.94, 5.07, and 6.27 μg/mL, respectively. Alstonia parvifolia also displayed notable inhibition against PLA2 activity of Naja philippinensis Taylor venom with IC50 of (15.2 ± 1.8) μg/mL. Docking and cluster analyses projected negative binding energies from AFM (-6.36 to -9.68 kcal/mol), HPT (-7.38 to -9.77 kcal/ mol), and acetylmarinobufogenin (-7.22 to -9.59 kcal/mol). These calculations were for the particular interactions of Alstonia parvifolia constituents to PLA2 homologues where the utmost affinity was detected in HPT owing to the dipole interactions with amino acid residues. Conclusions: The bark extract of Alstonia parvifolia shows great potential as an anti-venom agent due to its low cytotoxic profile, remarkable PLA2 inhibition, and docking binding energies between its bioactive constituents and PLA2 homologues.

We study the kinematics of the interstellar medium (ISM) viewed “down the barrel” in 20 gravitationally lensed galaxies during cosmic noon ( z = 1.5–3.5). We use moderate-resolution spectra ( R ∼ 4000) from Keck’s Echellette Spectrograph and Imager and Magellan/MagE to spectrally resolve the ISM absorption in these galaxies into ∼10 independent elements and use double Gaussian fits to quantify the velocity structure of the gas. We find that the bulk motion of gas in this galaxy sample is outflowing, with average velocity centroid $\left\langle {v}_{\mathrm{cent}}\right\rangle =-141$ km s ^−1 (±111 km s ^−1 scatter) measured with respect to the systemic redshift. A total of 16 out of the 20 galaxies exhibit a clear positive skewness, with a blueshifted tail extending to ∼ −500 km s ^−1 . We examine scaling relations in outflow velocities with galaxy stellar mass and star formation rate, finding correlations consistent with a momentum-driven wind scenario. Our measured outflow velocities are also comparable to those reported for FIRE-2 and TNG50 cosmological simulations at similar redshift and galaxy properties. We also consider implications for interpreting results from lower-resolution spectra. We demonstrate that while velocity centroids are accurately recovered, the skewness, velocity width, and probes of high-velocity gas (e.g., v _95 ) are subject to large scatter and biases at lower resolution. We find that R ≳ 1700 is required for accurate results for the gas kinematics of our sample. This work represents the largest available sample of well-resolved outflow velocity structure at z > 2 and highlights the need for good spectral resolution to recover accurate properties.

Gravitational lenses can magnify distant galaxies, allowing us to discover and characterize the stellar populations of intrinsically faint, quiescent galaxies that are otherwise extremely difficult to directly observe at high redshift from ground-based telescopes. Here, we present the spectral analysis of two lensed, quiescent galaxies at z ≳ 1 discovered by the ASTRO 3D Galaxy Evolution with Lenses survey: AGEL 1323 ( M _* ∼ 10 ^11.1 M _⊙ , z = 1.016, μ ∼ 14.6) and AGEL 0014 ( M _* ∼ 10 ^11.5 M _⊙ , z = 1.374, μ ∼ 4.3). We measured the age, [Fe/H], and [Mg/Fe] of the two lensed galaxies using deep, rest-frame-optical spectra (S/N ≳40 Å ^−1 ) obtained on the Keck I telescope. The ages of AGEL 1323 and AGEL 0014 are ${5.6}_{-0.8}^{+0.8}$ Gyr and ${3.1}_{-0.3}^{+0.8}$ Gyr, respectively, indicating that most of the stars in the galaxies were formed less than 2 Gyr after the Big Bang. Compared to nearby quiescent galaxies of similar masses, the lensed galaxies have lower [Fe/H] and [Mg/H]. Surprisingly, the two galaxies have comparable [Mg/Fe] to similar-mass galaxies at lower redshifts, despite their old ages. Using a simple analytic chemical evolution model connecting the instantaneously recycled element Mg with the mass-loading factors of outflows averaged over the entire star formation history, we found that the lensed galaxies may have experienced enhanced outflows during their star formation compared to lower-redshift galaxies, which may explain why they quenched early.

Background: The Internet is often the first resource used by applicants to evaluate fellowship programs. However, information on these websites can be often incomplete, inaccessible, and/or inaccurate. The primary objective of this study was to examine key factors that orthopedic foot and ankle fellowship applicants use to rank programs. The secondary objective was to assess both the accessibility and availability of the information on orthopedic foot and ankle fellowship program websites. Methods: A Qualtrics survey was distributed via e-mail to those who matched into an orthopedic foot and ankle fellowship position from years 2008-2020. A comprehensive list of orthopedic foot and ankle fellowship programs was created. Program websites were evaluated for accessibility as well as the quality of recruitment and educational content. Results: There were a total of 114 survey responses out of 644 invites (17.7%). The most important factors for establishing a rank list were operative experience, current faculty, and program reputation. Eighty-five percent (41/48) of orthopedic foot and ankle fellowship websites were directly accessible using Google. On average, accessible orthopedic foot and ankle fellowship websites contained only 57% (11.5/20) of the content deemed desirable. Conclusion: Orthopedic foot and ankle websites are widely accessible and have higher recruitment and educational quality content scores compared with previously published data. The most important factors for establishing a rank list are consistent with previous literature. Those who ranked operative experience as one of the most important factors when establishing a rank list did not complete more operative cases than those who did not. Level of Evidence: Level IV.

Over the past few years alone, the lensing community has discovered thousands of strong lens candidates, and spectroscopically confirmed hundreds of them. In this time of abundance, it becomes pragmatic to focus our time and resources on the few extraordinary systems, in order to most efficiently study the Universe. In this paper, we present such a system: DESI-090.9854-35.9683, a cluster-scale lens at z l = 0.49, with seven observed lensed sources around the core, and additional lensed sources further out in the cluster. From the number and the textbook configuration of the lensed images, a tight constraint on the mass potential of the lens is possible. This would allow for detailed analysis on the dark and luminous matter content within galaxy clusters, as well as a probe into dark energy and high-redshift galaxies. We present our spatially resolved kinematic measurements of this system from the Very Large Telescope Multi Unit Spectroscopic Explorer, which confirm five of these source galaxies (in ascending order, at z s = 0.962, 0.962, 1.166, 1.432, and 1.432). With previous Hubble Space Telescope imaging in the F140W and F200LP bands, we also present a simple flux-based lens model consisting of two power-law profiles that, for a cluster lens, well models the five lensed arc families with redshifts. We determine the mass to be M(< θ E) = 4.78 × 1013 M ⊙ for the primary mass potential. From the model, we extrapolate the redshift of one of the two source galaxies not yet spectroscopically confirmed to be at z s = 4.52 − 0.71 + 1.03 .

Andrographis paniculata (Burm f.) Nees is a tropical plant native to Southeast Asia that has been used as an effective remedy for a wide variety of illnesses in traditional Chinese and Ayurvedic medicine. The antimicrobial activity of its crude extract had been shown to be due to its quorum quenching activity. The study determined the effect of purified extracted compounds from the leaf of A. paniculata, namely: andrographolide, 14-deoxyandrographolide, 14-deoxy-12-hydroxyandrographolide and neoandrographolide on quorum sensing-mediated virulence mechanisms in clinical isolates of metallo-β-lactamase (MβL)-producing Pseudomonas aeruginosa. Their effect on the expression of the lasR gene, which codes for LasR, a transcription activator protein of the quorum sensing system in P. aeruginosa was also determined using RT-qPCR. All the pure compounds significantly decreased the biofilm formation, protease production and swarming motility of the P. aeruginosa isolates compared to the untreated controls (p < 0.05). Results of the RT-qPCR assay showed that all compounds significantly downregulated the expression of lasR compared to the untreated control (p < 0.05), supporting the position that the lower virulence activities of the treated group were due to quorum quenching activity of the pure compounds. Multiple comparisons using Tukey's HSD analysis revealed that the means of the relative expression of lasR of the isolates treated with the different compounds were not significantly different from each other (p > 0.05), suggesting equal potencies. Results show the potential of the isolated pure compounds from A. paniculata for use as antimicrobial agents as a result of their quorum quenching activities.

The data included in this article are related to research paper entitled “Efficient fortification of folic acid in rice through ultrasonic treatment and absorption”. These datasets compile the folic acid uptake expressed in concentration and the effects of folic acid fortification on the physical properties of brown and milled rice. We reported the folic acid uptake of rice in increasing fortificant concentration through soaking, one-step, and stepwise fortification protocols. In addition, the data on the effects of fortification on the color, pasting, and textural properties of brown and milled rice were also presented.

Although with a long track record of success, limited work has been dedicated to this procedures effect on the first tarsometatarsal (TMT-1) joint. The purpose of this study is to evaluate the changes in TMT-1 joint contact stress in the diseased and corrected state following an opening wedge osteotomy of the proximal metatarsal. In addition, the effect of a distal soft tissue release (DSTR) was evaluated as it related to both radiographic correction and TMT-1 joint contact stress. Methods: Seven fresh-frozen cadaveric below knee specimens (mean age: 69yrs) with hallux valgus deformities (mean HVA: 31.7+-12.0degs) were obtained for the study. The specimen was loaded up to 400N on an MTS servo hydraulic load frame with the tibia at 90-degree to the neutrally position foot. Joint contact characteristics at TMT-1 joint were measured with a Tekscan pressure sensor (Model6900, 1100psi). A standard proximal metatarsal osteotomy was performed. Various sized metal wedges (3, 5, 7 mm) with locking plates and screws were inserted in the osteotomy for correction. Following initial tests, a complete distal soft tissue release (DSTR) was performed and the specimens were retested. Additionally, dorsoplantar weight bearing (400N) radiograph was obtained for each condition to measure intermetatarsal (IMA) and hallux valgus (HVA) angles. Category: Bunion; Midfoot/Forefoot; Other Introduction/Purpose: Previous studies have demonstrated success in using autogenous bone graft for arthrodesis in patients with previously failed surgeries of the hallux. These patients have several causes for pain and dysfunction preoperatively, including shortened first ray, nonunions, and poor hallux alignment. In this study, a consecutive series of 36 patients (38 procedures) were treated with a patellar wedge interposition structural allograft to salvage bone loss from great toe arthrodesis malunion, painful joint replacement, failed osteotomy, or infection of the great toe metatarsophalangeal (MP) joint with shortening of the first ray. The goals of the surgery were to restore length to the first ray and provide a stable MP joint fusion to relieve pain. Methods: The 38 treated toes were followed for at least one year and were evaluated for preoperative and postoperative American Orthopaedic Foot & Ankle Society (AOFAS) MP scores, subjective patient outcome scores, and clinically successful fusion of the hallux. Results: At a minimum one-year follow-up, all but two feet healed with a solid fusion, and all patients reported good or excellent outcomes. AOFAS MP scores averaged 43.5 preoperatively and 77.2 postoperatively. Three patients with infection as cause for nonunion of the initial procedure were treated with staged procedures, including the use of a temporary antibiotic spacer and mini external fixator; all three healed without recurrent infection. One patient had a fracture of her allograft following her interposition arthrodesis, but fused successfully after a second interposition arthrodesis surgery. Two patients developed a nonunion of the revision arthrodesis. Conclusion: In conclusion, the use of an interposition patellar wedge allograft can restore length to the first ray and provide successful salvage of arthrodesis nonunions and bone loss from failed hemi and total joint implants of the great toe MP joint.

Small extracellular vesicles (sEV) are nano-sized (40-150 nm), membrane-encapsulated vesicles that are released by essentially all cells into the extracellular space and function as intercellular signaling vectors through the horizontal transfer of biologic molecules, including microRNA (miRNA) and other small non-coding RNA (ncRNA), that can alter the phenotype of recipient cells. sEV are present in essentially all extracellular biofluids, including serum, urine and saliva, and offer a new avenue for discovery and development of novel biomarkers of various disease states and exposures. The objective of this study was to systematically interrogate similarities and differences between sEV ncRNA derived from saliva, serum and urine, as well as cell-free small ncRNA (cf-ncRNA) from serum. Saliva, urine and serum were concomitantly collected from 4 healthy donors to mitigate potential bias that can stem from interpersonal and temporal variability. sEV were isolated from each respective biofluid, along with cf-RNA from serum. sEV were isolated from the respective biofluids via differential ultracentrifugation with a 30% sucrose cushion to minimize protein contamination. Small RNA-sequencing was performed on each sample, and cluster analysis was performed based on ncRNA profiles. While some similarities existed in terms of sEV ncRNA cargo across biofluids, there are also notable differences in ncRNA class and ncRNA secretion, with sEV in each biofluid bearing a unique ncRNA profile, including major differences in composition by ncRNA class. We conclude that sEV ncRNA cargo varies according to biofluid, so thus should be carefully selected and interpreted when designing or contrasting translational or epidemiological studies.

We present spectroscopic confirmation of candidate strong gravitational lenses using the Keck Observatory and Very Large Telescope as part of our ASTRO 3D Galaxy Evolution with Lenses ( AGEL ) survey. We confirm that (1) search methods using convolutional neural networks (CNNs) with visual inspection successfully identify strong gravitational lenses and (2) the lenses are at higher redshifts relative to existing surveys due to the combination of deeper and higher-resolution imaging from DECam and spectroscopy spanning optical to near-infrared wavelengths. We measure 104 redshifts in 77 systems selected from a catalog in the DES and DECaLS imaging fields ( r ≤ 22 mag). Combining our results with published redshifts, we present redshifts for 68 lenses and establish that CNN-based searches are highly effective for use in future imaging surveys with a success rate of at least 88% (defined as 68/77). We report 53 strong lenses with spectroscopic redshifts for both the deflector and source ( z _src > z _defl ), and 15 lenses with a spectroscopic redshift for either the deflector ( z _defl > 0.21) or source ( z _src ≥ 1.34). For the 68 lenses, the deflectors and sources have average redshifts and standard deviations of 0.58 ± 0.14 and 1.92 ± 0.59 respectively, and corresponding redshift ranges of z _defl = 0.21–0.89 and z _src = 0.88–3.55. The AGEL systems include 41 deflectors at z _defl ≥ 0.5 that are ideal for follow-up studies to track how mass density profiles evolve with redshift. Our goal with AGEL is to spectroscopically confirm ∼100 strong gravitational lenses that can be observed from both hemispheres throughout the year. The AGEL survey is a resource for refining automated all-sky searches and addressing a range of questions in astrophysics and cosmology.

Objective: To explore cytotoxicity of Synsepalum dulcificum (S. dulcificum) Daniell (Sapotaceae) on human colon cancer (HCT-116 and HT-29), human monocytic leukemia (THP-1) and normal (HDFn) cell lines, and its effect on the expression of early apoptotic genes, c-fos and c-jun. Methods: Leaf, stem and berry of S. dulcificum were separately extracted by using 2 solvents, 10% ethanol (EtOH) and 80% methanol (MeOH). PrestoBlue® cell viability assay and qRT-PCR assay were conducted to examine the above objectives respectively. Results: Stem MeOH, stem EtOH, and berry EtOH extracts of S. dulcificum were cytotoxic to HCT-116 and HT-29 human colon cancer cells. For HCT-116, IC50 values of these 3 extracts were not significantly different (P>0.05) from that of the positive control bleomycin (IC50 of 33.57 μg/mL), while for HT-29, IC50 values of these 3 extracts were significantly lower (P

Abstract Electron-donors can impart charge to the surface of transition metal dichalcogenide (TMD) films while interacting with the film via a weak physisorption bond, making them ideal for vapor and gas sensors. We expose monolayer MoS2 and MoSe2 films to strong electron-donor chemical vapor analytes. After analyzing the resultant behavior and taking into consideration doping effects, we conclude that exposure to strong electron-donors could be a method of inducing the semiconductor-metal 2H-1T TMD phase transition. We find that the conductance response to strong electron donors in both monolayer MoS2 and MoSe2 FET devices ceases after moderate exposure, with final value of the conductance being on order of that expected for the 1T phase. Full device relaxation back to a semiconducting state is accomplished by annealing in vacuum at 400 °C. We also examine chemically exposed TMD films intermittently interrogated with Raman and photoluminescence spectroscopy. We observe the appearance of weak characteristic 1T phase Raman features for MoS2 and we observed a quenching of the photoluminescence of both TMD films that is recoverable with annealing. Considering all of our data together, the effects cannot be described by doping alone. Additionally, our results suggest a mechanism for a new type of passive chemical vapor sensor.

We consider, for a positive integer $k$, induced subgraphs in which each component has order at most $k$. Such a subgraph is said to be $k$-divided. We show that finding large induced subgraphs with this property is NP-complete. We also consider a related graph-coloring problem: how many colors are required in a vertex coloring in which each color class induces a $k$-divided subgraph. We show that the problem of determining whether some given number of colors suffice is NP-complete, even for $2$-coloring a planar triangle-free graph. Lastly, we consider Ramsey-type problems where graphs or their complements with large enough order must contain a large $k$-divided subgraph. We study the asymptotic behavior of "$k$-divided Ramsey numbers". We conclude by mentioning a number of open problems.

Objective: To investigate if the protective effect of α-tocopherol against the impact of ethanol on brain morphogenesis involved the activity of the selenoproteins phospholipid hydroperoxide glutathione peroxidase (PHGPx; GPx4) and selenoprotein P (SelPP) that have roles against oxidative stress.Methods: Forty female mice were randomly assigned into natural control (CON), positive control (ETOH), low-, medium-, and high-α-tocopherol-supplemented-ethanol groups (LTOC, MTOC, HTOC, respectively). CON received drinking water without ethanol while ETOH, LTOC, MTOC and HTOC groups received 20% ethanol in drinking water. The supplemented groups were given respective dosages of α-tocopherol, 0.410, 0.819, and 1.640 mg/g body weight, at day 14 before mating onwards to the day 9 of gestation. At 10.5 ED of gestation (1 100 h), the pregnant females were sacrificed by cervical dislocation and the embryos were harvested. Total RNA were extracted, cDNA synthesis and qRT-PCR analyses were carried out.Results: The level of expression of PHGPx in the positive control was significantly lower than that of the natural control. Among the three α-tocopherol-supplemented groups, only the medium dose-group was significantly higher than the positive control. The level of expression of SelPP in the positive control was significantly lower than those of the natural control, the low- and medium-dose-tocopherol supplemented groups. In the high dose-α-tocopherol supplemented group, the level of expression was not significantly different from the positive control but significantly lower than the natural control.Conclusions: The activity of the selenoproteins PHGPx and SelPP are involved in the internetwork of antioxidative enzymes with vitamin E when given up to a medium dose only and is one of the possible pathways of shielding embryonic development against the impact of ethanol on brain morphogenesis. This study strengthens the impact of dietary α-tocopherol and Selenium supplement during the critical period of pregnancy.

After failure of nonoperative management, midfoot arthrodesis has been favored as the primary operative treatment producing acceptable outcomes with associated potentially long postoperative immobilization and 17% major complications rate. The aim of this study was to evaluate the pain and functional outcomes of dorsal midfoot surgical denervation by deep peroneal neurectomy as a safe alternative for management of dorsal midfoot pain. Methods: In this retrospective study, 18 patients (21 feet) who underwent dorsal denervation of the midfoot by deep peroneal neurectomy by a single surgeon were evaluated. There were 13 women and 5 men with mean age 70.4 (range, 47 to 88) at the time of surgery. Indications for the procedure include painful midfoot arthritis, lack of radiographic collapse, intact plantar protective sensation, failed nonoperative management for 6 months, and greater than 75% transient pain relief from preoperative diagnostic block of deep peroneal nerve. Preoperative and postoperative (3 months, 6 months) Visual Analog Scale (VAS), Short- Form 36 (SF-36), and Foot and Ankle Outcome Score (FAOS) were reviewed. Complications were recorded. Category: Professional, Patient Care Introduction/Purpose: Differences exist in the training backgrounds of medical providers who treat foot and ankle disorders. Considerable overlap and similarities also exist between podiatric and orthopaedic surgeons, though patients may be unaware of the differences. It is not known to what extent professional training influences how patients seek care. The purpose of this study is to understand patients’ knowledge of the differences in professional training background between podiatry and orthopedic surgery and to determine which factors are important to patients when selecting a provider. Methods: Patient survey data was gathered from Froedtert Memorial Lutheran Hospital and the Mayo Clinic. A 27-question survey was administered to new patients who were referred to the foot and ankle service in an orthopedic department at both institutions. Survey questions included data on patient demographics, patient opinion, and knowledge of differences between podiatry, orthopedics, and other foot and ankle providers. Patients were grouped by provider preference. Univariate and multivariate regressions were used to characterize the study population and determine provider preference. Significance was determined through t-tests, Fisher’s Exact test, and chi-square tests. Results: Of the 169 patients who completed the entire survey, 99 chose “orthopedic surgeon” as their provider of preference for any foot or ankle injury. Between the groups, there was no significant difference in age, healthcare affiliation, previous podiatric visits, level of education, and perceived knowledge about the differences between the two specialties (Table 1). For patients who listed podiatry as their preference, they were less likely to expect their doctor to have completed residency (76.2% vs. 90.7-94.9%, p=0.03).
