Background. Efficient analysis of large image data produced in greenhouse phenotyping experiments is often challenged by a large variability of optical plant and background appearance which requires advanced classification model methods and reliable ground truth data for their training. In the absence of appropriate computational tools, generation of ground truth data has to be performed manually, which represents a time-consuming task. Methods. Here, we present a efficient GUI-based software solution which reduces the task of plant image segmentation to manual annotation of a small number of image regions automatically pre-segmented using k-means clustering of Eigen-colors (kmSeg). Results. Our experimental results show that in contrast to other supervised clustering techniques k-means enables a computationally efficient pre-segmentation of large plant images in their original resolution. Thereby, the binary segmentation of plant images in fore- and background regions is performed within a few minutes with the average accuracy of 96–99% validated by a direct comparison with ground truth data. Conclusions. Primarily developed for efficient ground truth segmentation and phenotyping of greenhouse-grown plants, the kmSeg tool can be applied for efficient labeling and quantitative analysis of arbitrary images exhibiting distinctive differences between colors of fore- and background structures.

The present thesis deals with the numerical modeling of facial tissue biomechanics for the prediction of the patient's postoperative appearance in cranio-, dento-maxillofacial surgery planning. Typically, craniofacial surgery interventions consist in the rearrangement of facial bones, which induces the deformation of surrounding soft tissue. The postoperative patient's appearance has to be predicted on the basis of 3D geometrical models derived from tomographic data and the prescribed displacements of relocated bone structures. The linear elastic approach known from the previous works and widely used in soft tissue modeling is generally limited by the assumption of small deformations and implies a substantial error by ad hoc calculations of large deformations. The investigations carried out in this work lead to the development of a more accurate and flexible non- linear elastic approach. Different tissue types are modeled as a homogeneous, isotropic, quasi-incompressible St. Venant-Kirchhoff material characterized by two elastic constants, the Young modulus describing the material stiffness and the Poisson ratio describing the material compressibility. The numerical solution of the associated discrete boundary value problem on tetrahedral grids is obtained via the finite element method (FEM). Since the non-linear elastic FEM is in general very expensive, an adaptive numerical scheme for the efficient and robust computation of both small and large deformations with user-defined precision is developed. The modelling approach has been validated by the direct comparison between the simulation outcome and the patient's postoperative facial outline in a number of clinical studies. In addition to the static soft tissue prediction, we present a new approach for the estimation of individual facial emotion expressions. We assume natural physiological mechanism of facial expressions that is the impact of contracting muscles on remaining facial tissue. Using this technique, single muscle actions as well as complex individual facial emotion expressions can be estimated., In der vorliegenden Arbeit wurden theoretische und experimentelle Untersuchungen zur Entwicklung eines numerischen Modells des Gesichtsweichgewebes durchgeführt, mit dem die Vorhersage des postoperativen Erscheinungsbildes des Patienten bei der Planung chirurgischer Eingriffe in der Mund-, Kiefer-, Gesichtschirurgie (MKG) möglich ist. Typischerweise bestehen die MKG-Eingriffe in einer korrigierenden Umstellung der Gesichtsknochen, die zu einer Verformung des umliegenden Weichgewebes führt. Die primäre Aufgabe ist es, die Deformation des Gesichtsweichgewebes anhand eines dreidimensionalen geometrischen Modells der Patientenanatomie und der Randbedingungen, die ausschließlich in Form der Randverschiebungen der umpositionierten Knochenteile gegeben sind, zu bestimmen. Die bisherigen Ansätze zur vereinfachten Modellierung der Weichgewebebiomechanik basieren auf der linear elastischen Approximation, die grundsätzlich auf den Bereich kleiner Deformationen beschränkt ist und bei der Berechnung großer Deformationen einen erheblichen Fehler impliziert. Die in dieser Arbeit durchgeführten Untersuchungen haben zur Entwicklung eines nichtlinear elastischen Modells geführt, bei dem verschiedene Gewebetypen als ein stückweise homogenes, isotropes, quasi-inkompressibles St. Venant-Kirchhoff Material mit zwei Elastizitätskonstanten, dem Elastizitätsmodul und der Querkontraktionszahl beschrieben werden. Zur numerischen Lösung der zugehörigen diskreten Randwertprobleme auf dem Tetrahedergitter wurde die Finite Elemente Methode (FEM) angewendet. Da die nichtlinear elastische FEM im allgemeinen aufwendig ist, wurde ein adaptives numerisches Schema entwickelt, um eine effiziente und robuste Performanz bei der Berechnung sowohl kleiner als auch großer Deformationen mit vorgegebener Genauigkeit zu erzielen. Der gewählte Modellierungsansatz wurde in einer Reihe von klinisch relevanten Studien durch den direkten Vergleich zwischen Simulationsergebnissen und postoperativen Daten validiert. Neben der statischen Weichgewebevorhersage präsentieren wir ein neues Verfahren zur Abschätzung individueller Gesichtsmimik. Dabei gehen wir von einem natürlichen physiologischen Mechanismus der Gesichtsmimik aus, d.h. die Verformung des Gesichtsweichgewebes unter der Einwirkung der kontrahierenden mimischen Muskulatur. Mit Hilfe des entwickelten numerischen Modells können sowohl Einzelmuskelaktionen als auch komplexe Gesichtsausdrücke einer Person abgeschätzt werden.
