Aims. We revised the treatment of interstellar dust in the KOSMA-t photo-dissociation region (PDR) model code to achieve a consistent description of the dust-related physics in the code. The detailed knowledge of the dust properties is then used to compute the dust continuum emission together with the line emission of chemical species. Methods. We coupled the KOSMA-τ PDR code with the multi component dust radiative transfer (MCDRT) code to to solve the frequency-dependent radiative transfer equations and the thermal balance equation in a dusty clump under the assumption of spherical symmetry, assuming thermal equilibrium in calculating the dust temperatures, neglecting non-equilibrium effects. We updated the calculation of the photoelectric heating and extended the parametrization range for the photoelectric heating toward high densities and UV fields. We revised the computation of the H2 formation on grain surfaces to include the Eley-Rideal effect, thus allowing for high-temperature H2 formation. Results. We demonstrate how the different optical properties, temperatures, and heating and cooling capabilities of the grains influence the physical and chemical structure of a model cloud. The most influential modification is the treatment of H2 formation on grain surfaces that allows for chemisorption. This increases the total H2 formation significantly and the connected H2 formation heating provides a profound heating contribution in the outer layers of the model clumps. The contribution of polycyclic aromatic hydrocarbons (PAH) surfaces to the photoelectric heating and H2 formation provides a boost to the temperature of outer cloud layers, which is clearly traced by high-J CO lines. Increasing the fraction of small grains in the dust size distribution results in hotter gas in the outer cloud layers caused by more efficient heating and cooler cloud centers, which is in turn caused by the more efficient FUV extinction. Based on [Ci] (1-0) observations and complementary Hi, 12/13CO (1-0), 12CO (2-1) and [Cii] data we estimated the column density and fraction of molecular hydrogen in GMCs along the major axis of the galaxy M33. We found that on average 75%±14% of the hydrogen is in molecular form. Roughly 40-45% of the H2 is both traced by [CII] and CO, while ~15% by the [Ci] lines. The found CO-dark H2 gas fractions are within the predictions by Wolfire et al. (2010) for a half solar metallicity, Z = 0.5Zʘ, as in M33. [ABSTRACT FROM AUTHOR]

Objectives: To investigate the burden of disease of myocarditis in Germany and identify similarities and differences in myocarditis with or without COVID-19., Methods: All patients hospitalized with myocarditis in Germany were included in this nationwide retrospective analysis. Data were retrieved from the Federal Statistical Office of Germany (DESTATIS) for the years from 2007 to 2022. The primary endpoint was hospital mortality., Results: A total of 88,159 patients hospitalized with myocarditis were analyzed. Annual cases increased from 5100 in 2007 to 6593 in 2022 (p < 0.001 for trend) with higher incidence during winter months. Incidence per 100,000 inhabitants was 6.2 in 2007 rising to 7.8 in 2022 (p < 0.001 for trend). Hospital mortality remained constant at an average of 2.44% (p = 0.164 for trend). From 2020 to 2022, 1547/16,229 (9.53%) patients were hospitalized with both, myocarditis and COVID-19 (incidence 0.62/100,000 inhabitants and 180/100,000 hospitalizations with COVID-19). These patients differed significantly in most patient characteristics and had a higher rate of hospital mortality compared to myocarditis without COVID-19 (12.54% vs. 2.26%, respectively, p < 0.001)., Conclusions: Myocarditis hospitalizations were slowly rising over the past 16 years with hospital mortality remaining unchanged. The pial vasculature is the sole source of blood supply to the neocortex. The brain is contained within the skull, a vascularized bone marrow with a unique anatomical connection to the brain meninges. Recent developments in tissue clearing have enabled detailed mapping of the entire pial and calvarial vasculature. However, what are the absolute flow rate values of those vascular networks? This information cannot accurately be retrieved with the commonly used bioimaging methods. Here, we introduce Pia-FLOW, a unique approach based on large-scale transcranial fluorescence localization microscopy, to attain hemodynamic imaging of the whole murine pial and calvarial vasculature at frame rates up to 1,000 Hz and spatial resolution reaching 5.4 µm. Using Pia-FLOW, we provide detailed maps of flow velocity, direction, and vascular diameters which can serve as ground-truth data for further studies, advancing our understanding of brain fluid dynamics. Genetically-encoded dopamine (DA) sensors enable high-resolution imaging of DA release, but their ability to detect a wide range of extracellular DA levels, especially tonic versus phasic DA release, is limited by their intrinsic affinity. Here we show that a human-selective dopamine receptor positive allosteric modulator (PAM) can be used to boost sensor affinity on-demand. The PAM enhances DA detection sensitivity across experimental preparations (in vitro, ex vivo and in vivo) via one-photon or two-photon imaging. In vivo photometry-based detection of optogenetically-evoked DA release revealed that DETQ administration produces a stable 31 minutes window of potentiation without effects on animal behavior. The use of the PAM revealed region-specific and metabolic state-dependent differences in tonic DA levels and enhanced single-trial detection of behavior-evoked phasic DA release in cortex and striatum. Our chemogenetic strategy can potently and flexibly tune DA imaging sensitivity and reveal multi-modal (tonic/phasic) DA signaling across preparations and imaging approaches., (© 2024. Recanalization is the mainstay of ischemic stroke treatment. However, even with timely clot removal, many stroke patients recover poorly. Leptomeningeal collaterals (LMCs) are pial anastomotic vessels with yet-unknown functions. We applied laser speckle imaging, ultrafast ultrasound, and two-photon microscopy in a thrombin-based mouse model of stroke and fibrinolytic treatment to show that LMCs maintain cerebral autoregulation and allow for gradual reperfusion, resulting in small infarcts. In mice with poor LMCs, distal arterial segments collapse, and deleterious hyperemia causes hemorrhage and mortality after recanalization. In silico analyses confirm the relevance of LMCs for preserving perfusion in the ischemic region. Accordingly, in stroke patients with poor collaterals undergoing thrombectomy, rapid reperfusion resulted in hemorrhagic transformation and unfavorable recovery. Thus, we identify LMCs as key components regulating reperfusion and preventing futile recanalization after stroke. Large-scale imaging of brain activity with high spatio-temporal resolution is crucial for advancing our understanding of brain function. The existing neuroimaging techniques are largely limited by restricted field of view, slow imaging speed, or otherwise do not have the adequate spatial resolution to capture brain activities on a capillary and cellular level. To address these limitations, we introduce fluorescence localization microscopy aided with sparsely-labeled red blood cells for cortex-wide morphological and functional cerebral angiography with 4.9 µm spatial resolution and 1 s temporal resolution. When combined with fluorescence calcium imaging, the proposed method enables extended recordings of stimulus-evoked neuro-vascular changes in the murine brain while providing simultaneous multiparametric readings of intracellular neuronal activity, blood flow velocity/direction/volume, and vessel diameter. Owing to its simplicity and versatility, the proposed approach will become an invaluable tool for deciphering the regulation of cortical microcirculation and neurovascular coupling in health and disease., (© 2024. Leptomeningeal collaterals (LMCs) connect the main cerebral arteries and provide alternative pathways for blood flow during ischaemic stroke. This is beneficial for reducing infarct size and reperfusion success after treatment. However, a better understanding of how LMCs affect blood flow distribution is indispensable to improve therapeutic strategies. Here, we present a novel in silico approach that incorporates case-specific in vivo data into a computational model to simulate blood flow in large semi-realistic microvascular networks from two different mouse strains, characterised by having many and almost no LMCs between middle and anterior cerebral artery (MCA, ACA) territories. This framework is unique because our simulations are directly aligned with in vivo data. Moreover, it allows us to analyse perfusion characteristics quantitatively across all vessel types and for networks with no, few and many LMCs. We show that the occlusion of the MCA directly caused a redistribution of blood that was characterised by increased flow in LMCs. Interestingly, the improved perfusion of MCA-sided microvessels after dilating LMCs came at the cost of a reduced blood supply in other brain areas. This effect was enhanced in regions close to the watershed line and when the number of LMCs was increased.
