This study aims to examine the functional morphology of the proventriculus of the broad breasted white turkey (BBWT) (Meleagris gallopavo, Linnaeus 1758) using gross anatomy, light microscopy, gross/histomorphometric analysis, and scanning and transmission electron microscopy. The proventriculus was characterized internally by many elevated papillae with a mound, leafy flower, and lotus flower-shapes. Each papilla was enclosed by concentrically organized mucosal folds with distinct or indistinct proventricular gland openings on its top. Longitudinal folds and grooves at the junction of the proventriculus with the esophagus exhibited various sized and shaped openings of esophageal glands with irregular outlines. Histologically, the surface epithelium of the proventriculus was covered by a thin layer of cuticle, particularly evident at its junction with the gizzard. The lamina epithelialis and propria, and secretory units of proventricular lobules were infiltrated by aggregations of lymphocytes and lymphoid follicles (nodules). Variably shaped glandular lobules of proventricular glands occupied the submucosa, surrounded by thin strands of smooth muscle fibers derived from muscularis mucosa. Triangular, cuboidal, or columnar-shaped secretory oxyntico-peptic cells lined the secretory units. Many telocytes (pyramidal or fusiform-shaped cell bodies with lengthy telopodes) were observed in interstitial tissue. Further, two types of argyrophilic endocrine cells were identified within the glandular epithelium. Emerging market differentiation for broiler meat from strains exhibiting a range of growth rates is necessitating comparative research on various physiological and production aspects of these strains. The objective of the present study was to compare select gastrointestinal, tibial, and plasma attributes in a sample of 48-day-old (50 male and 50 female) broilers obtained from fast-and slow-growing flocks maintained under similar feed and management regimens. Eight birds were randomly selected from a fast (B; representative of modern commercial strains) and each of the 4 slow-growing strains (SG; D, H, M, and E). The strains differed by estimated time to reach 2.2 kg bodyweight corresponding to 36, 50, 42, 44, and 50 D for B, D, H, M, and E, respectively. Blood samples were collected to determine plasma metabolites, and birds were subsequently euthanized, weighed, and necropsied for gizzard and small intestine weight, jejunal tissue for histomorphology, ceca digesta samples for concentration of short-chain fatty acids (SCFA) and left tibia for ash content. Gizzard was heavier (P < 0.01) for D, H, and M than that for B and E, whereas the small intestine was lighter (P < 0.01) for B, D, and H than for M and E. There were no (P > 0.05) strain differences on SCFA, jejunal villus height and crypt depth, plasma proteins, and electrolytes. Strains D, H, and M exhibited higher (P = 0.01) tibia ash concentration than B; E was intermediate and not different (P > 0.05) from any strain. Specifically, the tibia ash for B, D, H, SG 3, and E were 1.24, 1.44, 1.43, 1.49, and 1.39 g/kg BW, respectively. The B birds showed higher (P < 0.01) plasma concentrations of aspartate transaminase, creatine kinase, lactate dehydrogenase, and creatinine than SG strains. In conclusion, although B and some SG strains had lighter gastrointestinal tract indicative of energy efficiency, higher circulating plasma enzymes in B birds suggested impaired hepatic function. In this study, the effect of grinding or pelleting of rapeseed on the digestibility, apparent metabolisable energy (AME N ), growth performance and gizzard weight of broiler chickens was evaluated. In Exp. 1, four treatments were tested (240 broiler chickens, 20 replications, 3 birds per cage). Birds received either a basal diet or were fed with rapeseed of different sized particles (fine, medium and coarse ground, respectively). In Exp. 2, 10 treatments were assessed (360 chickens, 12 replications, 3 birds per cage), where five different diets (a basal diet and four diets with intact rapeseed, fine ground, medium ground or coarse ground rapeseed, respectively) were fed non-pelleted or pelleted. In Exp. 3, two treatments were compared (224 broiler chickens, 14 replications, 8 birds per floor pen): fine ground rapeseed (FG) and intact rapeseed (IS). Compared with medium and coarse ground rapeseed, fine grinding increased in Exp. 1 the AME N of rapeseed and the precaecal digestibility (PD) of dry matter and ether extract (EE) ( p < 0.05). In Exp. 2, the content of AME N and the PD of EE and crude protein (CP) increased as the degree of grinding increased when diets were not pelleted ( p < 0.01). In contrast, the significantly highest values for these parameters were measured after pelleting without effect of grinding, resulting in a significant interaction between pelleting and grinding ( p < 0.001). In Exp. 3, performance of chickens was not affected by dietary treatments, but gizzard weight and the PD of CP were higher when IS were included in the diet ( p < 0.05). The results suggested that fine grinding may be used to increase the feeding value of full-fat rapeseed for broiler chickens. Nevertheless, pelleting of diets with rapeseed seemed to be much more effective in the reduction of particle size than grinding.

Many studies have used grit (in the form of lead (Pb) pellets) presence in avian gizzards as an indicator of Pb shot exposure. However, due to nearly complete pellet absorption in gizzards or rapid passage of pellets, the absence of Pb shot presence in a gizzard does not confirm lack of Pb shot exposure. This study provides the basis for an additional technique to identify if elevated tissue Pb concentration is due to Pb shot exposure. Bioaccessibility of Pb and trace elements (Sb, As, and Sn) present in Pb shot were quantified to determine if any of these elements would be useful as a secondary marker of Pb shot exposure. An avian physiologically based extraction test (PBET) was used to determine pellet dissolution rate and bioaccessible concentrations of Pb, Sb, As, and Sn in a simulated gizzard environment. Of the three trace elements, only Sb concentrations (44-302 μg/mL) extracted into the gizzard solution were greater than environmental background levels (US soil average 0.48 μg/g); thus, no natural source likely provides this amount of Sb. Therefore, there is evidence that Sb can be extracted from Pb shot in bird gizzards at detectable concentrations (above natural background). While further studies are needed to delineate the mechanisms of absorption and distribution, this study lends credence to the hypothesis that Sb may be a useful marker of Pb shot exposure in biological tissues, particularly when Pb pellets are not present nor observed in avian tissues., Competing Interests: The authors have declared that no competing interests exist.

This study was performed to investigate the hypothesis that supplementation of processed lignocellulose (PL) in the diets of broilers has a positive effect on growing performance, pH value of gizzard, hepatic enzyme activity, immunologic indicators, histomorphological character of small intestine, and cecal microflora populations. A total of 720 one-day-old Ross 308 broiler chicks were allotted to 4 treatment groups and fed maize-soybean meal based diets. The basal diet was supplemented with PL with an amount of 0 kg (control), 0.5 kg, 1 kg, and 2 kg per ton feed. Growing performance parameters, were determined weekly until 35 D of age. Blood samples for enzyme activities and immunoglobulins, jejunum and cecum samples for histomorphological characters for villus growth, and microbial population were collected from 12 broilers from each group. At 35 D of age, body weight of broilers supplemented with 1 kg of PL was found to be the highest with a value of 2305.0 g, when compared to the broilers supplemented with control, 0,5 and 2 kg of PL groups (2154.0, 2201.0, and 2141.7 g, respectively, P = 0.001). An increased activity of aspartate amino transferase (AST) was observed in the control and 1 kg PL supplementation groups (633.6 and 597.4 IU/L, respectively), whereas alkaline phosphatase (ALP) activity was the highest in the control group (5404 IU/L, P < 0.05). Broilers in the control group had the lowest level of IgY and IgA (122.2 and 25.8 mg/dL, respectively, P < 0.05). Villus height increased by 22.0%, 40.7%, and 34.8% in 0.5, 1, and 2 kg PL supplementation groups, respectively, when compared to the control (P < 0.001). The processed lignocellulose supplemented as 1 kg of PL decreased the average count of Staphylococcaceae, E. coli, and Enterobacteriaceae, whereas it increased the population of Lactobacillus spp. in the cecum (P < 0.05). 1. A 2 × 2 factorial design was used to test the hypothesis that impaired intestinal starch digestibility is attributable to rapid passage of digesta from the gizzard to the intestine, and that, compared to steam pelleting, increasing the availability of starch through extrusion cooking may alleviate the potential negative effect of rapid digesta flow on starch utilisation. 2. Thus, 7-d-old-broiler chickens were distributed to 48 cages and given a wheat-based (WB) pelleted diet containing either coarse oat hulls (OH-Pel) or fine cellulose (Cel-Pel) until d 19 to stimulate divergent development of the gizzard. Thereafter, both groups were further subdivided and challenged with a WB diet containing cellulose in either pelleted (Cel-Pel) or extruded (Cel-Ext) form on d 20 and 22. Either excreta or intestinal contents were collected at time intervals after feeding and analysed for marker and starch. 3. OH-Pel increased gizzard size and holding capacity. No excessively high starch levels (maximum 25 g/kg) were detected in the excreta. However, 8 h feed-deprived birds given Cel-Pel and challenged with Cel-Pel exhibited higher starch excretion and showed large individual variation during the first 135 min of collection. 4. Contrary to the OH-Pel group, more digesta and starch passed to the jejunum at 1 and 2 h and ileum at 2 and 3 h after feeding for birds given Cel-Pel, resulting in lower jejunal and ileal starch digestibility. 5. Increased starch gelatinisation through extrusion processing significantly improved starch digestibility regardless of gizzard function. However, at 1, 2 and 3 h after feeding, more digesta was retained in the foregut of birds given Cel-Ext. 6. The current data showed that starch degradation rate is associated with the flow of digesta which is linked to gizzard development, and that enzymatic hydrolysis of intact starch granules may be limited with more rapid feed passage through the gut.

Migratory birds undergo impressive body remodelling over the course of an annual cycle. Prior to long-distance flights, red knots ( Calidris canutus islandica) reduce gizzard mass while increasing body mass and pectoral muscle mass. Although body mass and pectoral muscle mass are functionally linked via their joint effects on flight performance, gizzard and pectoral muscle mass are thought to be independently regulated. Current hypotheses for observed negative within-individual covariation between gizzard and pectoral muscle mass in free-living knots are based on a common factor (e.g. migration) simultaneously affecting both traits, and/or protein limitation forcing allocation decisions. We used diet manipulations to generate within-individual variation in gizzard mass and test for independence between gizzard and pectoral muscle mass within individuals outside the period of migration and under conditions of high protein availability. Contrary to our prediction, we observed a negative within-individual covariation between gizzard and pectoral muscle mass. We discuss this result as a potential outcome of an evolved mechanism underlying body remodelling associated with migration. Although our proposed mechanism requires empirical testing, this study echoes earlier calls for greater integration of studies of function and mechanism, and in particular, the need for more explicit consideration of the evolution of mechanisms underlying phenotypic design.

A 21-day experiment was conducted to investigate the effects of wheat particle size, insoluble fibre source and whole wheat (WW) inclusion on the gizzard musculature, intestinal architecture and nutrient utilisation of broiler chickens. The following seven treatments, all in mash form, were tested: control diet using finely ground wheat (Fine); control diet using medium-ground wheat (Medium); control diet using coarsely ground wheat (Coarse); fine diet diluted with 10 g/kg lignocellulose (LC); fine diet diluted with 30 g/kg oat hulls (OH); fine diet diluted with 30 g/kg wood shavings (WS); and ground wheat in the fine diet replaced with 50 and 100 g/kg WW during the second (day 8-14) and third (day 15-21) weeks of the study respectively. Except for the LC diet, all treatments increased (p < 0.05) the thickness of the caudodorsal thick muscle of gizzard compared to the fine diet. The highest (p < 0.05) caudoventral thin and cranioventral thick muscle diameters in the gizzard walls were achieved with the OH and WS diets. Feeding OH, WS and WW diets improved (p < 0.05) starch digestibility compared with the fine diet, while medium, coarse and LC diets had no effect (p > 0.05). All diets containing fibre sources (LC, OH and WS) reduced (p < 0.05) the energy digestibility compared to the fine diet. Birds fed OH and WS diets gained the lowest (p < 0.05) weights. Feeding LC, OH, WS and WW diets increased (p < 0.05) the feed per gain compared with the fine diet. The current results suggest that there is a potential for coarse particles, insoluble fibre sources and WW feeding to improve the gizzard musculature and digestibility of some nutrients, with the effect being more pronounced with the inclusion of structural fibre sources., (© 2018 Blackwell Verlag GmbH.)

1. The current study was conducted to evaluate the influence of high phytase doses and xylanase, individually and in combination, on performance, blood inositol and real-time gastric pH in broilers fed wheat-based diets. 2. In a 42-d experiment, a total of 576 male Ross 308 broiler chicks were allocated to 4 dietary treatments. Treatments consisted of a 2 × 2 factorial arrangement, with 500 or 2500 FTU/kg phytase and 0 or 16 000 BXU/kg xylanase, fed in two phases (starter 0-21; grower 21-42 d). Heidelberg pH capsules were administered to 8 birds from each treatment group, pre- and post-diet phase change, with readings captured over a 5.5-h period. 3. At 21 and 42 d, birds fed 500 FTU/kg phytase without xylanase had on average 127 and 223 g lower weight gain than all other treatments, respectively (P < 0.05). At 21 d, feed conversion ratio (FCR) was reduced (P < 0.01) by 2500 FTU/kg phytase or xylanase; however, 42-d FCR was unaffected by enzyme treatment. Inositol content of plasma was twice that of the erythrocyte (P < 0.001), with 2500 FTU/kg phytase tending to increase (P = 0.07) inositol content in both blood fractions. 4. Across all treatments, capsule readings ranged from pH 0.54 to 4.84 in the gizzard of broilers. Addition of 2500 FTU/kg phytase to the grower diet reduced (P < 0.05) average gizzard pH from 2.89 to 1.69, whilst feeding xylanase increased (P < 0.001) gizzard pH from 2.04 to 2.40. In contrast, digital probe measurements showed no effect of xylanase on gizzard pH, while addition of 2500 FTU/kg phytase increased (P = 0.05) pH compared to 500 FTU/kg phytase with or without xylanase. 5. These findings suggested that xylanase and high phytase doses have opposite effects on real-time gastric pH, while similarly improving performance of broilers.

The digestive system of a chicken is simple, short, and extremely efficient. In the upper gastro-intestinal tract (GIT) or foregut, in particular when feed is not continuously available, moistening of feed occurs in the crop followed by chemical and physical digestion in the proventriculus and gizzard, respectively, as a preparation for nutrient absorption which occurs in the distal portions of the GIT-duodenum, jejunum, ileum, and to a lower extent in the ceca and colon. Broiler husbandry practices in the past have focused on achieving high intakes of readily digestible nutrients and largely failed to consider the ancestral purpose of the foregut. Besides holding important digestive functions, the rich bacterial communities in both the crop and the gizzard transform these organs in barriers preventing the invasion of the GIT by pathogens. However, the amount of time feed spends in the foregut-and therefore the environment in this gut segment-is extremely variable and can be manipulated by different feeding practices, such as feed structure and composition, frequency of feeding, and/or light regimes.With further progress in terms of performance and health of broiler chickens being sought, and following decades of research focusing on distal GIT segments, it is about time to explore how the foregut can contribute to achieve these goals. This review revisits morphological, physiological, and microbiological characteristics of the foregut; explores the importance of this GIT portion as feed intake regulator; and discusses how husbandry and feeding practices such as lighting regimes and feed structure may be adapted to activate the crop and the gizzard, which results in performance and health improvements. Finally, interactions of these feeding practices with exogenous enzymes are discussed and suggestions for further research are made.

We analyzed the gizzards, and grits retained in the gizzards of 41 cranes that migrated to the Izumi Plain during the winter of 2015/2016 and died there, either due to accident or disease. These included 31 Hooded Cranes (Grus monacha) and 10 White-naped Cranes (G. vipio). We determined body weight, gizzard weight, total grit weight and number per gizzard, and size, shape, and surface roundness of the grits. Average gizzard weights were 92.4 g for Hooded Cranes and 97.1 g for White-naped Cranes, and gizzard weight positively correlated with body weight in both species. Average total grit weights per gizzard were 19.7 g in Hooded Cranes and 25.7 g in White-naped Cranes, and were significantly higher in the latter. Average percentages of body weight to grit weight were 0.8% in Hooded Cranes and 0.5% in White-naped Cranes. Average grit number per gizzard was 693.5 in Hooded Cranes and 924.2 in White-naped Cranes, and were significantly higher in the latter. The average grit size was 2.8 mm in both species. No differences were found in the shape and surface roundness of grits between the two species. To the best of our knowledge, this is the first study on the grits retained in the gizzards of Hooded and White-naped Cranes.

The purpose of this study was to ascertain the composition and origin of certain gastroliths or gizzard stones from broiler breeders (Gallus gallus domesticus). Fourier transform infrared spectroscopy (FTIR) and X-ray powder diffraction techniques were used as analysis tools. The main components resulted to be organic fibre and minerals referred to a pegmatite rock (quartz, feldspar albite and haematite) used as grit. Although the generally accepted purpose of gastroliths in chicken is the crushing and grinding of foodstuff in the ventriculus, the importance of mineral uptake from grit of iron and other essential elements such as Al, Ca, K, Cl, P, Ti, Mn, Mg, Cu, Zn and Zr (in ppm) as a secondary effect of the abrasion and dissolution of gastroliths should not be disregarded., (© 2017 Blackwell Verlag GmbH.)

This study investigates the developmental morphology changes on the proventriculus and gizzard of African ostrich chicks using gross anatomy, and light and transmission electronic microscopy. The body weight; the weight of the proventriculus and gizzard; the thickness of simple tubular glands, muscularis mucosae, compound tubular glands, and muscular layer of the proventriculus; and the thickness of the gizzard glands and muscularis mucosae were measured on postnatal d one, 45, 90, and 334. Under transmission electronic microscopy, the oxynticopeptic cells, mucous cells, and endocrine cells were observed in the proventriculus on postnatal d one, 45, and 90. Our results revealed that the weights of the proventriculus and gizzard (relative to the body weight) both peaked on d 90, respectively, and declined thereafter. The thicknesses of the simple tubular glands, muscularis mucosae, compound tubular glands, muscular layer of the proventriculus, and gizzard glands increased with increasing bird age. However, the thickness of the muscularis mucosae in the gizzard peaked on d 90 and gradually decreased thereafter. The result of transmission electron microscopy revealed that the glands in the submucosa layer of the proventriculus did not differ from other cell types, aside from an increase in cell diameter and an increase in cytoplasmic content. A significant increase in the number of mitochondria and smooth endoplasmic reticulum was noted, as well as an in granules secreted by endocrine cells. Housing and feeding practices of wild birds for conservation management of biodiversity or restocking play a crucial role in determining the survival rates of animals when released into nature. Failure in coping with the environment might be one of the main flaws captive animals can experience when put into natural habitat. The present investigation aimed at exploring feeding habits and related morphometric traits of gizzard with respective content from wild partridges in comparison with captive ones. A total of 52 hunted wild Sardinian adult partridges (Alectoris barbara barbara Bonnaterre, 1790) were used. By comparison, 42 captive adult partridges reared in cages were enrolled. From each animal, the morphology of gizzard was investigated and respective content analysed for gross composition and taxonomical determination of fractions. Wet sieving analysis of each gizzard content was carried out (four-sieve towers with different mesh sizes: 1 mm, 500 μm, 250 μm and 125 μm), and relative and absolute weight of fresh filled and empty gizzards were recorded. Thickness of muscular layer of gizzard wall was measured by stereomicroscope. Carcass weight significantly (p < 0.05) differed between captive vs. wild partridges (478 ± 21 and 305 ± 35 g respectively). Post-mortem inspection highlighted gross morphological differences of gizzards between the two groups. Fresh weight of empty gizzards was 6.37 ± 0.80 vs. 11.25 ± 1.82 g, with average pH values of digesta 4.97 ± 0.11 vs. 4.38 ± 0.28 in captive vs. wild partridges respectively. Gizzard content from wild partridges accounted a 61.7% vs. 38.3% of biological vs. non-biological material proportions (DM basis). The non-biological material was mostly represented by lithic fragments and minerals (quartz, feldspar, calcite and mica) with specific peculiarities in terms of granulometry and morphometry. Digestive capacity often limits food intake rate in animals. Many species can flexibly adjust digestive organ mass, enabling them to increase intake rate in times of increased energy requirement and/or scarcity of high-quality prey. However, some prey species are defended by secondary compounds, thereby forcing a toxin limitation on the forager's intake rate, a constraint that potentially cannot be alleviated by enlarging digestive capacity. Hence, physiological flexibility may have a differential effect on intake of different prey types, and consequently on dietary preferences. We tested this effect in red knots (Calidris canutus canutus), medium-sized migratory shorebirds that feed on hard-shelled, usually mollusc, prey. Because they ingest their prey whole and crush the shell in their gizzard, the intake rate of red knots is generally constrained by digestive capacity. However, one of their main prey, the bivalve Loripes lucinalis, imposes a toxin constraint due to its symbiosis with sulphide-oxidizing bacteria. We manipulated gizzard sizes of red knots through prolonged exposure to hard-shelled or soft foods. We then measured maximum intake rates of toxic Loripes versus a non-toxic bivalve, Dosinia isocardia. We found that intake of Dosinia exponentially increased with gizzard mass, confirming earlier results with non-toxic prey, whereas intake of Loripes was independent of gizzard mass. Using linear programming, we show that this leads to markedly different expected diet preferences in red knots that try to maximize energy intake rate with a small versus a large gizzard. Intra- and inter-individual variation in digestive capacity is found in many animal species. Hence, the here proposed functional link with individual differences in foraging decisions may be general. We emphasize the potential relevance of individual variation in physiology when studying trophic interactions.

Two 49 d floor pen studies were conducted to evaluate the effects of litter type and dietary coarse ground corn (CC) inclusion on broiler live performance, gastrointestinal tract (GIT) development, and litter characteristics. Experiment 1 was a 2×2 factorial arrangement of 2 genders (male or female) and 2 CC levels (0 or 50%). From 15 to 35 d, the addition of CC decreased feed intake (P<0.01) and BW gain (P<0.05) of males but not females. The inclusion of CC decreased feed intake (P<0.01) and BW gain (P<0.01) from 0 to 49 d but improved adjusted feed conversion ratio (AdjFCR) from 35 to 49 d (P<0.05). Male broilers exhibited better live performance than females during the study as evidenced by greater feed intake (P<0.01) and BW gain (P<0.01), and improved FCR (P<0.01), but with increased mortality (P<0.05). The inclusion of CC increased relative gizzard weight (P<0.01) and decreased relative proventriculus weight (P<0.01) at 49 d. Experiment 2 was a 2×2 factorial arrangement of 2 CC levels (0 or 50%) and 2 litter types (ground old litter or new wood shavings litter). The inclusion of CC decreased feed intake throughout the experiment without affecting final BW when only males were used and improved FCR after 25 d (P<0.01). New litter improved FCR from 1 to 14 d (P<0.01). At 49 d, the birds fed the CC diet had reduced excreta nitrogen (P<0.05) and litter moisture (P<0.05). In conclusion, 50% CC inclusion initially produced negative effects on live performance that became positive as BW increased. The effects of CC became evident at an earlier age for males. 1. A total of 2200 White Leghorn layers were used to study the effect of feeding coarse maize on productive performance, gizzard weight, apparent metabolisable energy (AME) and egg quality parameters. 2. The experiment was a completely randomised design with 5 treatments, each being replicated 5 times (88 birds per replicate). Dietary treatments included a control diet with 600 g/kg of ground maize (6 mm sieve) and experimental diets with 150, 300, 450 or 600 g/kg coarse maize (10 mm sieve) replacing (w/w) ground maize. Diets, in mash form, were offered from 39 to 62 weeks of age. 3. Over the entire experimental period, dietary treatments had no effect on any of the production parameters, except on feed intake. A quadratic effect was observed for feed intake, where intake increased at 150 g/kg coarse maize inclusion. At higher inclusion levels of coarse maize, feed intake was similar to that of the control diet. But the differences in daily intake between dietary treatments were only around 1 g/bird and may not be of any biological significance. 4. Dietary treatments had no effect on gizzard weight, AME or egg quality. 5. The results indicate that coarse maize could completely replace maize in layer diets with no adverse effect on egg production, egg quality and dietary energy utilisation. In the current work, although the proportion of coarse particles (over 1 mm) increased with increasing inclusion levels of coarse maize, the differences in the percentage of particles above 1 mm between the fine control (0 g/kg coarse maize) and coarsest (600 g/kg coarse maize) diets was only 15%, and this may limit the value of the findings reported herein.

We carried out an observational and experimental study to decipher how resource characteristics, in interaction with the predator's phenotype, constrain a fitness-determining performance measure, i.e. refuelling in a migrant bird. Two subspecies of red knot (Calidris canutus rogersi and C. c. piersmai) use northern Bohai Bay, Yellow Sea, China, for the final prebreeding stopover, during their 10,000-15,000 km long migrations between wintering and breeding areas. Here, they feed on small bivalves, especially 2-7 mm long Potamocorbula laevis. With an average stay of 29 days, and the need to store 80 g of fat for the onward flights to high-Arctic breeding grounds, red knots need to refuel fast. Using existing knowledge, we expected them to achieve this on the basis of (1) prey with high flesh to shell mass ratios, (2) large gizzards to crush the ingested molluscs, or (3) a combination of the two. Rejecting all three predictions, we found that red knots staging in Bohai Bay had the smallest gizzards on record (4.9 ± 0.8 g, mean ± s.e.m., N = 27), and also found that prey quality of P. laevis is much lower than predicted for the measured gizzard size (i.e. 1.3 rather than the predicted 4.5 kJ g(-1) dry shell mass, DM(shell)). The estimated handling time of P. laevis (0.2 s) is much shorter than the observed time between two prey ingestions (0.7 s), indicating that prey handling time is no constraint. Based on field observations of dropping rates and on indoor digestion trails, the shell processing rate was estimated at 3.9 mg DM(shell) s(-1), i.e. three times higher the rate previously predicted for red knots eating as fast as they can with the measured gizzard size. This is explained by the small and easily crushed P. laevis enabling high processing rates. As P. laevis also occurred in high densities, the metabolizable energy intake rate of red knots with small gizzards at 5 J s(-1) was as high as at northward staging sites elsewhere in the world. Currently, therefore, food characteristics in Bohai Bay are such that red knots can refuel fast whilst economizing on the size of their gizzard. These time-stressed migrants thus provide an elegant example of symmorphosis.

Two experiments were conducted from 0 to 21 d of age and evaluated diets containing combinations of fine or coarse ground corn (557 or 1,387 μm, respectively), whole sorghum, 15% corn distillers dried grains with solubles (DDGS), or a prebiotic-type product containing yeast cell wall, lactose, citric acid, and other fermentable carbohydrates. In experiment 1, feed efficiency was decreased (P < 0.001) after the first week of age for broilers fed diets containing whole sorghum, whereas broilers receiving diets with 15% DDGS had increased feed efficiency (P < 0.03) compared with those receiving no DDGS. In the second experiment, BW gain was increased (P < 0.03) after the first week of age for broilers fed diets containing the prebiotic and DDGS compared with their respective controls. In experiment 1, the diet containing sorghum yielded the highest AMEn value (P < 0.03). In experiment 2, diets containing the combination of the prebiotic + DDGS yielded higher AMEn values (P < 0.004) at 7 and 21 d compared with diets containing no combination. The effects of diet on amino acid digestibility were generally small and inconsistent in both experiments. In experiment 1, broilers fed the coarse corn or whole sorghum diets had increased (P < 0.0001) relative gizzard weights compared with broilers fed the fine corn diet. Also, diets containing DDGS yielded increased relative gizzard weights (P < 0.05) compared with diets containing no DDGS. In experiment 2, there was a decrease (P < 0.03) in cecal Escherichia coli when the combination of the coarse ground corn, prebiotic, and DDGS was fed in comparison with broilers receiving no prebiotic or DDGS. These results indicate that diets containing coarsely ground corn or whole sorghum in combination with DDGS can be fed to broilers with no long-term adverse effects on growth performance and nutrient digestibility and that these ingredients can have beneficial effects on AMEn, gizzard size, and cecal microflora in some instances.

We report the results of spatiotemporal mapping of the spontaneous actions of component muscles of the gizzard and associated structures in ex vivo preparations with combined superfusion and vascular perfusion. Ongoing spontaneous contraction of cranial and caudal thin muscles occurred at a frequency of 2.2 ± 0.1 cycles per minute. Contractions of M. tenuis craniodorsalis with mean duration of 2.8 ± 0.2 s commenced ventrally adjacent to the distal limit of the proventriculus and progressed dorsally at 2.02 ± 0.03 mm•s(-1) in a concerted front. Near simultaneous contraction of M. tenuis caudoventralis of mean duration of contraction of 4.7 ± 0.7 s commenced dorsally and progressed ventrally at a similar rate (2.1 ± 0.1 mm•s(-1)) and in a similar manner. Contraction of the caudoventralis preceded that of craniodorsalis (mean 1.1 ± 0.15 s). Contraction of the 2 tenuis muscles was synchronous with the first component peak of the cyclic increase in lumen pressure and with distension of the crassus musculature. Contraction of the M. crassus caudodorsalis muscle coincided with the second component peak and was followed by distension of the tenuis musculature. The latter commenced before the relaxation of the tenuis muscles. Contractions of the crassus muscle propagated rapidly at right angles to the orientation of the muscle fibers at a faster velocity than that of the tenuis musculature. The durations of the component peaks in lumen pressure indicated that the duration of crassus contraction was similar to that of the tenuis musculature.

Aim: We have tested the effect of feed structure and feeding regime to prevent the spread of the zoonotic pathogen Campylobacter jejuni in broiler chicken flocks., Methods and Results: Birds were offered two types of feed, control diet and a diet supplemented with 15% oat/barley hulls for structure. In addition, the birds were either fed ad libitum or intermittent. One bird in each treatment group was infected with a three-strain-mix of Camp. jejuni, and the spread of Camp. jejuni within the group was investigated. Feed structure increased the gizzard weight, delayed the spread of Camp. jejuni within the group and reduced the relative amount of Camp. jejuni in the caecum compared with the control diet., Conclusions: Our results show that stimulating the bird's natural barriers is a novel and promising intervention strategy to reduce the spread of Camp. jejuni in chicken flocks., Significance and Impact of the Study: Preventing Camp. jejuni in broiler chicken flocks is essential to ensure food safety because this bacterium is transferred to chicken carcasses during the slaughter process and readily survive in unprocessed poultry products. Two experiments were conducted to test the following hypothesis: exposing broiler chickens to coarse insoluble fiber in the diet will result in enhanced gizzard function and performance, improved adaptability to an intermittent feeding program, and an increase in the occurrence of reverse peristalsis. In experiment 1, 102 Ross 308 broiler chickens were either intermittently or ad libitum fed a basal diet, the basal diet diluted with 15% coarse hulls (consisting of equal weights of hulls from oats and barley), or the basal diet diluted with 15% of the same hulls finely ground in a 2 × 3 factorial arrangement with 17 individually caged birds per treatment. Birds fed ad libitum had access to feed continuously for 18 h/d, whereas those on intermittent feeding had restricted access to feed from 7 d of age. From 18 d of age, the restrictive-feeding program consisted of four 1-h meals and one 2-h meal per day. In experiment 2, 156 broiler chickens in 12 pen cages with wood shaving-lined floors were exposed to 1 of 4 treatment groups with 3 pens/treatment: intermittent or ad libitum feeding of a basal diet and intermittent or ad libitum feeding of a coarse hull diet, as described above. At 31 and 32 d of age, birds in experiment 1 were inoculated with chromium EDTA via the cloaca. There was no interaction between diet and feeding regimen. The addition of hulls increased gizzard weight and content and lowered (P < 0.001) gizzard pH, but it had no effect on the ability of the birds to handle intermittent feeding. Despite the dilution with coarse hulls, weight gain and the gain:feed ratio were not affected, which could partly be explained by an increased (P < 0.001) starch digestibility. Dietary reflux was confirmed by the presence of chromium in all intestinal tract sections. Broilers exhibited reverse peristaltic contractions of sufficient magnitude to propel the marker from the cloaca to the gizzard.

Gizzard motility was compared between chickens from D(+) (high digestion efficiency) and D(-) (low digestion efficiency) genetic lines selected for divergent digestion efficiency, using strain gauge transducers. Motility was recorded continuously during 24 h in 6 birds per line. Two stimuli, a meal distribution after a feed-deprivation period and lighting after a dark period, were tested during the recording period. A functional test with intravenous injection of serotonin performed at the end of the recording day often resulted in a sharp reduction in gizzard motility, without a significant difference between lines. Compared with D(+) birds, gizzard activity in D(-) birds remained high during fasting or dark periods (P < 0.0006), which reduced the effects of stimuli in D(-) birds. So, coordination between bird activity and gizzard motility tended to be reduced in D(-) compared with D(+) birds (P = 0.0018). This coordination was observed to be positively correlated (P = 0.011) with the relative weight (g/kg of BW) of the stomach (gizzard + proventriculus). This experiment pointed out differences in gizzard motility between D(+) and D(-) chicken lines in terms of response to environmental stimuli, characterized by a failure in the gizzard relaxation process in D(-) birds during rest periods.

All living birds are toothless, constituting by far the most diverse toothless vertebrate clade, and are striking examples of evolutionary success following tooth loss. In recent years, an unprecedented number of Mesozoic birds have been described, illustrating the evolution of dentition reductions. Simultaneously, major advances in experimental embryology have yielded new results concerning avian edentulism. Reviewing these lines of evidence, we propose hypotheses for its causes, with a prominent role for the horny beak during development. A horny beak and a muscular gizzard functionally 'replaced' dentition for food acquisition and processing, respectively. Together with edentulism itself, these features and others contributed to the later success of birds, as a result of their high performance or additional functionality working in concert in these complex organisms., (Copyright © 2011 Elsevier Ltd. Two experiments were carried out to study the effect of intermittent feeding, diet structure, and their interaction on the performance, feeding pattern, and digestive adaptation of broiler chickens. In experiment 1, 40 Cobb broiler chickens were fed, either ad libitum or intermittently, a commercial starter diet diluted with 150 g/kg of ground or whole wheat in a 2 × 2 factorial arrangement. Ad libitum feeding consisted of continuous access to feed in a room with 18 h of light and 6 h of complete darkness, whereas birds on intermittent feeding had restricted access to feed from 7 d of age, with 4 one-hour feeding bouts/d and one 2-h feeding bout/d from d 14. No interaction was found between feed structure and feeding regimen for any of the measurements. Although BW gain was not significantly different among any of the treatments, birds given whole wheat or fed intermittently had significantly better feed efficiency than those given ground wheat and fed ad libitum. Inclusion of whole wheat instead of ground wheat also markedly increased (P < 0.001) the AME(n) value and fecal starch digestibility. In experiment 2, 60 Ross broiler chickens were trained to meal feeding on a wheat-based diet containing a commercial phytase, and 5 birds were killed every 15 min after having access to feed, to collect crop contents. Phytate was gradually degraded in the crop, with a 50% reduction in inositol 6-phosphate after a 100-min retention time. In conclusion, these results indicate that broiler chickens have a remarkable ability to maintain growth rate during intermittent feeding because the crop is used as an intermediate storage organ, and that improvements in feed efficiency may occur, among others through increased efficacy of exogenous enzymes. Even though stimulation of the gizzard through a coarse feed structure improves feed efficiency, it does not appear to affect the ability of the bird to handle intermittent feeding.

Mucins are high molecular weight glycoproteins which constitute the major component of the mucus layer and are produce by many epithelial tissues in vertebrates. Osteopontin (OPN) is an adhesive phosphorylated glycoprotein that is expressed by a broad range of tissues and cells. Although gastric mucins MUC1, MUC5AC, MUC6 and OPN have been widely used in histological studies and in diagnostic pathology in order to diagnose gastric carcinomas, their localizations in the stomach of quail have not yet been studied. In this study, the localizations of MUC1, MUC5AC, MUC6 and OPN in the proventriculus and gizzard of Japanese quail during the post-hatching period were compared at light microscope levels by applying immunohistochemical methods. In all ages studied, the immunoreactivity of MUC5AC was present in the lining epithelium of both folds and superficial proventricular glands in the proventriculus, whereas MUC1, MUC6 and OPN reactivity was found in the oxynticopeptic cells of profound proventricular glands. In addition, some cells in the fold epithelium of the proventriculus showed a positive reaction to OPN. The immunoreactivity of MUC1 in gizzard was different from that of MUC5AC. Although MUC5AC was expressed in the cells of both the surface epithelium and profound glands of the gizzard, MUC1 was only localized in the profound glands of the gizzard. However, MUC6 and OPN immunoreactivity was absent in the gizzard. The results indicated that the differences between the localizations of MUC1, MUC5AC, MUC6 and OPN in quail proventriculus and gizzard may be a reflection of functional differences of stomach parts. Although the biological significances of the expressions of MUC1, MUC5AC, MUC6 and OPN in the quail stomach remains unknown, these notable glycoproteins may be associated with barrier function, host defence, and/or secretion.

The water-holding capacity (WHC), and toughness (shear force) of chicken gizzard were evaluated during postmortem storage for 4.5, 7, 12, 24, 48, 72 and 96 h at 4 degrees C. Degradation of the cytoskeletal proteins desmin, talin and vinculin were monitored by sodium dodecyl sulfate--polyacrylamide gel electrophoresis and Western blotting during the same designated storage period. The WHC of the gizzards decreased significantly from 12 h to 72 h of storage, but by 96 h the WHC was restored to the level measured after storage for 12 h. The shear force value of the gizzards increased rapidly until 12 h and then decreased until 24 h, with a further slight decrease by 48 h. Degradation products of desmin, talin and vinculin appeared at 96 h, 12 h and 48 h postmortem, respectively. The intensity of immunolabeling for desmin, talin and vinculin after storage for 96 h decreased to 51%, 25% and 52% of the initial value. The appearance of desmin degradation products was accompanied by an increase in WHC. This suggests that the postmortem degradation of desmin is involved in the increase of WHC in chicken gizzard during storage at 4 degrees C, and talin and vinculin may be involved.

An experiment was conducted to study the effects of type of cereal (60% corn or 60% rice), heat processing of the cereal (raw or steam-cooked), and fiber source (control without hulls, 3% oat hulls, and 3% soy hulls) on gizzard pH and total tract apparent retention (TTAR) of nutrients in broilers at different ages. There were 12 treatments arranged factorially (2x2x3) and 6 replicates (8 chicks) per treatment. The 2 basal diets were based on soy protein concentrate and fish meal and had a neutral detergent fiber content of 3.5% for the rice and 7.3% for the corn diet. To address some concerns about the expansion of genetically engineered pharmaceutical and industrial crops to outdoor plantings and potential impacts on the human food supply, we determined whether commercial agriculture seeds of maize or corn Zea mays L., barley Hordeum vulgare L., safflower Carthamus tinctorius L. and rice Oryza sativa L. are digested or pass viably through the digestive tract, or are transported externally, by captive mallard ducks Anas platyrhynchos L., ring-necked pheasants Phasianus colchicus L., red-winged blackbirds Agelaius phoeniceus (L.) and rock pigeons Columba livia Gmelin (with the exception of whole maize seeds which were too large to feed to the blackbirds). These crop seeds, whether free-fed or force-fed, did not pass through the digestive tract of these bird species. The birds nonetheless did retain viable seeds in the esophagus/crop and gizzard for several hours. For example, after foraging for 6 h, mallards had retained an average of 228 +/- 112 barley seeds and pheasants 192 +/- 78 in the esophagus/crop, and their germination rates were 93 and 50%, respectively. Birds externally transported seeds away from the feeding location, but in only four instances were seeds found attached to their muddy feet or legs and in no case to feathers. Risk of such crop seeds germinating, establishing and reproducing off site after transport by a bird (externally or internally) or movement of a carcass by a predator, will depend greatly on the crop and bird species, location, environmental conditions (including soil characteristics), timing, and seed condition.

Polished pebbles occasionally found within skeletons of giant herbivorous sauropod dinosaurs are very likely to be gastroliths (stomach stones). Here, we show that based on feeding experiments with ostriches and comparative data for relative gastrolith mass in birds, sauropod gastroliths do not represent the remains of an avian-style gastric mill. Feeding experiments with farm ostriches showed that bird gastroliths experience fast abrasion in the gizzard and do not develop a polish. Relative gastrolith mass in sauropods (gastrolith mass much less than 0.1% of body mass) is at least an order of magnitude less than that in ostriches and other herbivorous birds (gastrolith mass approximates 1% of body mass), also arguing against the presence of a gastric mill in sauropods. Sauropod dinosaurs possibly compensated for their limited oral processing and gastric trituration capabilities by greatly increasing food retention time in the digestive system. Gastrolith clusters of some derived theropod dinosaurs (oviraptorosaurs and ornithomimosaurs) compare well with those of birds, suggesting that the gastric mill evolved in the avian stem lineage.

Yanornis martini is an Early Cretaceous basal ornithurine bird. Its fish-eating diet was previously recognized from a discrete mass of disarticulated fish remains discovered in its abdominal region. A new complete and articulated specimen of Yanornis martini preserves abundant in-situ gastroliths such as have been associated with a herbivorous diet. We suggest that the occurrence of gastroliths in this specimen, fish remains in a second, and the lack of gastroliths in three others, is consistent with diet-switching in Yanornis martini. Incompatibility of the preserved data with explanations of the grit as an artifact of preservation or result of accidental ingestion is discussed. This discovery indicates the earliest presence of intermittent diet change (and associated gizzard plasticity) observed in extant birds seasonally and in response to changes in available food sources.

An experiment was carried out to study the effect of different forms of wheat (airtight silo stored whole wheat, conventionally stored whole wheat, and ground wheat included in pellets) and dietary xylanase addition on production results and gastrointestinal characteristics of broiler chickens. Ileal viscosity, pancreatic digestive enzyme activities, and the composition and activity of the intestinal microflora were considered as response parameters. Differences between the 2 types of whole wheat with respect to the various measured parameters were marginal, whereas distinct differences were found between pellet-fed birds and birds receiving whole wheat. Whole wheat feeding improved feed conversion ratio and reduced water consumption (P < 0.001). Compared with pellets, whole wheat increased the relative weight of pancreas and gizzard and the dry matter concentration of gizzard content (P < 0.001). Whole wheat feeding reduced the pH in the gizzard contents (P < 0.01) and increased ileal viscosity. The addition of xylanase reduced ileal viscosity in birds receiving whole wheat to the same level as in pellet-fed birds. Whole wheat feeding resulted in lower activities of amylase in pancreatic tissue (P = 0.054), whereas xylanase addition increased chymotrypsin (P = 0.030) and lipase activities (P = 0.052). Whole wheat feeding resulted in lower intestinal numbers of lactose-negative enterobacteria (P < 0.05) and tended to reduce the ileal and cecal numbers of Clostridium perfringens (P < or = 0.08). It is concluded that whole wheat feeding stimulates gizzard function, which in turn prevents potentially pathogenic bacteria from entering the intestinal tract.

We are using the tissue-specific splicing of myosin phosphatase targeting subunit (MYPT1) as a model to investigate smooth muscle phenotypic diversity. We previously identified a U-rich intronic enhancer flanking the 5' splice site (IE1), and a bipartite exonic enhancer/suppressor, that regulate splicing of the MYPT1 central alternative exon. Here we show that T-cell inhibitor of apoptosis (TIA-1) and T-cell inhibitor of apoptosis-related (TIAR) proteins bind to the IE1. Co-transfection of TIA expression vectors with a MYPT1 mini-gene construct increase splicing of the central alternative exon. TIA proteins do not enhance splicing when the palindromic exonic splicing enhancer (ESE) is mutated, indicating that TIAs are necessary but not sufficient for splicing. The ESE specifically binds SRp55 and SRp20 proteins, supporting a model in which both SR and TIA proteins binding to their cis-elements are required for the recruitment of the splicing complex to a weak 5' splice site. Inactivation of TIA proteins in the DT40 cell line (TIA-1(-/-)TIAR(+/-)) reduced the splicing of the central alternative exon of the endogenous MYPT1 as well as stably transfected MYPT1 minigene constructs. Splicing of the MYPT1 3' alternative exon and the MLC(17) alternative exon were unaffected, suggesting that TIA proteins regulate a subset of smooth muscle/nonmuscle alternative splicing reactions. Finally, reduced RNA binding and reduced expression of the TIA and SR proteins in phasic (gizzard) smooth muscle around hatching coincided with the switch from exon inclusion to exon skipping, suggesting that loss of TIA and SR enhancer activity may play a role in the developmental switch in MYPT1 splicing.

Recently, it has been hypothesized that myosin light chain (MLC) phosphatase is activated by cGMP-dependent protein kinase (PKG) via a leucine zipper-leucine zipper (LZ-LZ) interaction through the C-terminal LZ in the myosin-binding subunit (MBS) of MLC phosphatase and the N-terminal LZ of PKG (Surks, H. K., Mochizuki, N., Kasai, Y., Georgescu, S. P., Tang, K. M., Ito, M., Lincoln, T. M., and Mendelsohn, M. E. (1999) Science 286, 1583-1587). Alternative splicing of a 3'-exon produces a LZ+ or LZ- MBS, and the sensitivity to cGMP-mediated smooth muscle relaxation correlates with the relative expression of LZ+/LZ- MBS isoforms (Khatri, J. J., Joyce, K. M., Brozovich, F. V., and Fisher, S. A. (2001) J. Biol. Chem. 276, 37250 -37257). In the present study, we determined the effect of LZ+/LZ- MBS isoforms on cGMP-induced MLC20 dephosphorylation. Four avian smooth muscle MBS-recombinant adenoviruses were prepared and transfected into cultured embryonic chicken gizzard smooth muscle cells. The expressed exogenous MBS isoforms were shown to replace the endogenous isoform in the MLC phosphatase holoenzyme. The interaction of type I PKG (PKGI) with the MBS did not depend on the presence of cGMP or the MBS LZ. However, direct activation of PKGI by 8-bromo-cGMP produced a dose-dependent decrease in MLC20 phosphorylation (p<0.05) only in smooth muscle cells expressing a LZ+ MBS. These results suggest that the activation of MLC phosphatase by PKGI requires a LZ+ MBS, but the binding of PKGI to the MBS is not mediated by a LZ-LZ interaction. Thus, the relative expression of LZ+/LZ- MBS isoforms could explain differences in tissue sensitivity to NO-mediated vasodilatation.

Aiming to interpret functionally the large variation in gizzard masses of red knots Calidris canutus, we experimentally studied how the digestive processing rate is influenced by the size of the gizzard. During their non-breeding season, red knots feed on hard-shelled molluscs, which they ingest whole and crush in their gizzard. In three experiments with captive birds we tested predictions of the hypothesis that gizzard size, via the rate of shell crushing and processing, constrains intake rate in red knots (against the alternative idea that external handling times constrain intake rate). Gizzard size within individual birds was manipulated by varying the hardness of the diet on offer, and was confirmed by ultrasonography. The results upheld the "shell-crushing hypothesis" and rejected the "handling time hypothesis". Intake rates on with-shell prey increased with gizzard size, and decreased with shell mass per prey. Intake rates on soft (without shell) prey were higher than on with-shell prey and were unaffected by gizzard size. Offering prey that were heavily shelled relative to their flesh mass led to energy intake rates that were marginally sufficient to balance the daily energy budget within the time that is naturally available in a tidal system. We predicted the optimal gizzard sizes that are required to either (1) balance energy income with energy expenditure, or (2) to maximise net daily energy intake. The gizzard mass of free-living red knots in the Wadden Sea is such that it maximises daily net energy intake in spring when fuelling for migration, while it balances energy budget throughout the remainder of the year.

1. An experiment was conducted to study the effects of inclusion of oat hulls in diets based on whole or ground wheat for broilers. Effects of wood shavings on layers were investigated in a further experiment. 2. Inclusion of oat hulls in wheat-based broiler diets did not affect weight gain. Feed conversion efficiency (FCE), corrected for insoluble fibre contents, was improved by oat hull inclusion. 3. Gizzard size increased with inclusion of oat hulls, whole wheat, wood shavings and grit. 4. Starch digestibility was significantly increased by inclusion of oat hulls for broilers, and by wood shavings for layers. 5. Wood shavings and whole wheat did not affect bile acid concentration of gizzard contents. However, the total amount of bile acids in gizzard increased with access to wood shavings due to an increase in the weight of gizzard contents, indicating an increased gastroduodenal reflux. 6. Fibre concentration was considerably higher in the gizzard contents than in the feed. 7. Duodenal particle size decreased with access to grit. No effect of hulls or whole wheat inclusion was found, indicating that all particles are ground to a certain critical size before leaving the gizzard.

1. The effects of whole grains of wheat on the digestive tract of broiler chickens was studied. A complete pelleted feed was compared with free choice feeding of whole wheat and a pelleted protein concentrate, given from 7 to 29 d of age. 2. Pepsin activity in proventriculus tissue was lower in whole wheat-fed birds than in complete diet-fed birds. The weight (g/kg body weight) of the gizzard was higher in whole wheat-fed birds and its contents had a lower pH. 3. In the intestine, there were no differences in deoxyribonucleic acid (DNA) concentration, protein/DNA, ribonucleic acid (RNA)/DNA, RNA/protein ratios or alkaline phosphatase activity expressed per tissue weight. The weight (g/kg body weight) of the duodenum was lower in whole wheat-fed birds and its contents had a higher pH. Also the activities of alkaline phosphatase and leucine aminopeptidase in the duodenum, and maltase in the ileum, expressed per unit of bird weight, were lower in whole wheat-fed birds. 4. These results suggest that whole grain feeding increases the chemical (pepsin in proventriculus) and physical (gizzard muscle) functionality of the upper part of the digestive tract but decreases the digestive capacity of the intestine. Higher gizzard functionality may play a positive role in the control of bacterial populations. The lower digestive enzyme activities in the intestine may be detrimental in situations of mucosal deterioration caused by intestinal disease.

The magnitude of agonist-induced Ca(2+) sensitization of force is tissue-dependent, but an explanation for this diversity is unknown. Ca(2+) sensitization is thought to involve a G-protein-mediated inhibition of myosin light chain phosphatase activity by phosphorylation of the myosin-targeting subunit (MYPT). The MYPT has two isoforms that differ by a central insert, which lies near this phosphorylation site. Expression of MYPT isoforms is both developmentally regulated and tissue-specific. We hypothesized that the presence or absence of the central insert determines the magnitude of agonist-induced Ca(2+) sensitization. Throughout development, the chicken aorta exclusively expresses the splice-in MYPT isoform, and guanosine 5'-O-(thiotriphosphate) (GTPgammaS) produces a significant force enhancement. Early during development, the chicken gizzard expresses the splice-in MYPT isoform, and GTPgammaS produced a Ca(2+) sensitization. In the gizzard coincident with the shift in expression from the splice-in to splice-out MYPT isoform, GTPgammaS no longer produced force enhancement. In addition, adenosine 5'-O-(thiotriphosphate) (ATPgammaS) phosphorylated only adult gizzard tissue, the only tissue that did not demonstrate a Ca(2+) sensitization. These results suggest that the relative expression of splice-in/splice-out MYPT isoforms determines the magnitude of agonist-induced force enhancement and that MYPT phosphorylation is not required for Ca(2+) sensitization.

1. Pelleted diets, incorporating whole or ground triticale or wheat in the pellets, were fed to broiler chickens and the performance, gastro-intestinal development and health of those chickens recorded. 2. The use of whole triticale in the pelleted food produced similar bodyweight responses to when ground triticale was incorporated in the food. Food conversion efficiency was enhanced when whole triticale was used compared to ground triticale and was similar to when an exogenous enzyme was added to the ground triticale diet. The incorporation of whole wheat into the pelleted food produced similar production responses to the use of ground wheat. 3. The gastro-intestinal development of the birds was greater when they were fed the whole grain diet as gizzard weights were increased by the incorporation of whole grain into the diet. Gastro-intestinal development was thought to be related to the presence of large fibre particles in the food. 4. Proventricular dilatation and mortality due to ascites were reduced by feeding pelleted diets containing whole grain, which was possibly related to the early development of the gastro-intestinal tract relative to total body development. 5. It is suggested that the current use of exogenous enzyme additions to broiler diets may be reduced by incorporating whole grain into pelleted diets for broiler chickens.

The ability to change organ size reversibly can be advantageous to birds that perform long migrations. During winter, red knots (Calidris canutus) feed on shellfish and carry a muscular gizzard that weighs 10% of their body mass. Gizzard size decreases when these birds eat soft foods, e.g. while breeding in the tundra. We studied the reversibility and time course of such changes using ultrasonography. Two groups of shellfish-adapted knots (N=9 and N=10) were fed alternately a hard and a soft food type. Diet switches elicited rapid reversible changes. Switches from hard to soft food induced decreases to 60% of initial gizzard mass within 8.5 days, while switches to hard food induced increases in gizzard mass to 147% within 6.2 days. A third group of knots (N=11), adapted to soft food for more than 1 year, initially had very small gizzards (25% of the mass of shellfish-adapted gizzards), but showed a similar capacity to increase gizzard size when fed shellfish. This is the first non-invasive study showing rapid digestive organ adjustments in non-domesticated birds.

1. A study was conducted to evaluate the individual and combined effects of aflatoxin B1 (AF), ochratoxin A (OA) and T-2 toxin (T-2) on performance, organ morphology serum biochemistry and haematology of broiler chickens and the efficacy of esterified-glucomannan (E-GM), a cell wall derivative of Saccharomyces cerevisiae1026 in their counteraction. 2. Two dietary inclusion rates of AF (0 and 0.3 mg/kg), OA (0 and 2 mg/kg), T-2 (0 and 3 mg/kg) and E-GM (0 and 1 g/kg) were tested in a 2 x 2 x 2 x 2 factorial manner on a total of 960 broiler chickens from 1 to 35 d of age in an open sided deep litter pen house. 3. Body weight and food intake were depressed by all the mycotoxins, OA being the most toxic during early life. 4. Weights of kidney and adrenals were increased by AF and OA. Liver weight was increased by AF (17.8%), while OA increased gizzard weight (14.6%) and reduced bone ash content (8.1%). T-2 toxin showed no effect on these variables. 5. Serum cholesterol content was decreased and activity of serum gamma glutamyl transferase (GGT) was increased by AF and OA while serum protein content was decreased by AF. These effects were more pronounced at 21 d than at 35 d of age. Inconsistent responses were seen in the other variables: blood urea nitrogen (BUN) content, activities of serum alanine amino transferase and aspertate amino transferase. Blood haemoglobin content was depressed by AF and T-2, whereas blood coagulation time was prolonged by OA. 6. Significant interactions were observed between any 2 toxins for their additive effects on body weight, food intake, bone ash content and serum GGT activity at 21 d. Conversely, antagonistic interactions were observed among any 2 of the toxins for their effects on variables such as serum protein and serum cholesterol content. Simultaneous feeding of all 3 mycotoxins did not show increased toxicity above that seen with any 2. 7. Esterified-glucomannan increased body weight (2.26%) and food intake (1.6%), decreased weights of liver (32.5%) and adrenals (18.9%) and activity of serum GGT (8.7%), and increased serum protein (14.7%), cholesterol (21.9%), BUN (20.8%) and blood haemoglobin (3.1%) content, indicating its possible beneficial effect on mycotoxicosis in broiler chickens.

We have attempted to determine whether muscarinic stimulation induces RhoA/ROCK-mediated Ca2+ sensitization of contractions in chicken gizzard smooth muscles. rhoA is a small GTP-binding protein, and ROCK is a rhoA-associated coiled coil-forming serine/threonine kinase. The relationship between the cytosolic Ca2+ level ([Ca2+]i) and muscle force in the presence of a high K+ concentration was not different from that in the presence of carbachol. Verapamil inhibited muscle force in proportion to the decrease in [Ca2+]i in both the muscle stimulated with high K+ and that stimulated with carbachol. In addition, Y-27632 (10 microM), a ROCKs inhibitor, had no effect on the contractions. In the alpha-toxin-permeabilized muscles, Ca2+ induced a greater contraction in the presence of guanosine 5'-O-(3-thiotriphosphapte) (GTP[gamma-S]), whereas carbachol with GTP was not effective. The GTP[gamma-S]-induced Ca2+ sensitization was completely inhibited by Clostridium botulinum exoenzyme C3. Western blot analysis revealed both rhoA and ROCKII in the muscle extract. In addition, reverse transcriptase polymerase chain reaction (RT-PCR) analysis revealed the expression of both ROCKI and ROCKII mRNAs. These results suggest that Ca2+ sensitization in the chicken gizzard is elicited via a rhoA/ROCKs pathway, and that this pathway may be responsible for the augmentation of contraction by GTP[gamma-S] in the permeabilized muscles. If such a pathway does exist, however, carbachol-induced contraction may not be coupled to it, which explains the absence of Ca2+ sensitization in the intact chicken gizzard stimulated by carbachol.

Smooth muscle is generally grouped into two classes of differing contractile properties. Tonic smooth muscles show slow rates of force activation and relaxation and slow speeds of shortening (V(max)) but force maintenance, whereas phasic smooth muscles show poor force maintenance but have fast V(max) and rapid rates of force activation and relaxation. We characterized the development of gizzard and aortic smooth muscle in embryonic chicks to identify the cellular determinants that define phasic (gizzard) and tonic (aortic) contractile properties. Early during development, tonic contractile properties are the default for both tissues. The gizzard develops phasic contractile properties between embryonic days (ED) 12 and 20, characterized primarily by rapid rates of force activation and relaxation compared with the aorta. The rapid rate of force activation correlates with expression of the acidic isoform of the 17-kDa essential myosin light chain (MLC(17a)). Previous data from in vitro motility assays (Rover AS, Frezon Y, and Trybus KM. J Muscle Res Cell Motil 18: 103-110, 1997) have postulated that myosin heavy chain (MHC) isoform expression is a determinant for V(max) in intact tissues. In the current study, differences in V(max) did not correlate with previously published differences in MHC or MLC(17a) isoforms. Rather, V(max) was increased with thiophosphorylation of the 20-kDa regulatory myosin light chain (MLC(20)) in the gizzard, suggesting that a significant internal load exists. Furthermore, V(max) in the gizzard increased during postnatal development without changes in MHC or MLC(17) isoforms. Although the rate of MLC(20) phosphorylation was similar at ED 20, the rate of MLC(20) dephosphorylation was significantly higher in the gizzard versus the aorta, correlating with expression of the M130 isoform of the myosin binding subunit in the myosin light chain phosphatase (MLCP) holoenzyme. These results indicate that unique MLCP and MLC(17) isoform expression marks the phasic contractile phenotype.

Patterning of the gut into morphologically distinct regions results from the appropriate factors being expressed in strict spatial and temporal patterns to assign cells their fates in development. Often, the boundaries of gene expression early in development correspond to delineations between different regions of the adult gut. For example, Bmp4 is expressed throughout the hindgut and midgut, but is not expressed in the early gizzard. Ectopic BMP4 in the gizzard caused a thinning of the muscularis. To understand this phenotype we examined the expression of the receptors transducing BMP signaling during gut development. We find that the BMP receptors are differentially expressed in distinct regions of the chicken embryonic gut. By using constitutively activated versions of the BMP type I receptors, we find that the BMP receptors act similarly to BMP4 in the gizzard when ectopically expressed. We show that the mesodermal thinning seen upon ectopic BMP signaling is due to an increase in apoptosis and a decrease in proliferation within the gizzard mesoderm. The mesodermal thinning is characterized by a disorganization and lack of differentiation of smooth muscle in the gizzard mesoderm. Further, ectopic BMP receptors cause an upregulation of Nkx2.5, the pyloric sphincter marker, similar to that seen with ectopic BMP4. This upregulation of Nkx2.5 is a cell-autonomous event within the mesoderm of the gizzard. We also find that Nkx2.5 is necessary and sufficient for establishing aspects of pyloric sphincter differentiation.

1. The effect of diet form (mash, cold-pelleted, steam-conditioned/pelleted, wet mash, whole wheat with balancer pellet, restricted pellet) and enzyme inclusion (Avizyme 1300, absent, present) was studied in 2 trials using individually caged, male broilers from 14 to 42 d. Bird performance, viscosity of ileal contents and diet metabolisability (AME) were measured. 2. The performance of mash-fed birds was significantly poorer than for the other treatments in relation to dry matter intake, liveweight gain and gain:food. This was not due to reduced diet AME content. 3. There was no significant effect of heat treatment on any of the variables measured, although viscosity of ileal contents was increased by 30% as compared to the cold-pelleted diet. 4. Gain:food was improved with wet-mash feeding in comparison to the dry mash treatment but it was concluded that this was not due to any intrinsic improvement in diet quality, but rather to voluntary food restriction on introduction of the wet food. 5. Whole wheat feeding improved gain:food and diet AME content by 3% as compared to the complete diets and caused approximately a 50% increase in gizzard weight as compared with the pelleted diets. 6. Food enzyme inclusion did not improve performance although a significant improvement in diet AME content was observed with enzyme inclusion in trial 1.

Lead poisoning by shot-pellet ingestion is an important cause of death in waterfowl from wetlands that have high densities of lead-shot pellets and low availabilities of grit in their sediments. To evaluate grit supplementation when mixed with baited grain as a method of reducing shot ingestion in waterfowl, 24 mallards (Anas platyrhynchos) were kept in captivity to study the relationship between steel-shot, grit and grain ingestion in four experiments. Steel-shot ingestion was more related to seeking grit than to feeding on grain, and it did not differ between two types of grain offered (rice or sorghum). Grit ingestion increased with grit supplementation and shot ingestion was only slightly reduced. Calcareous grit used in the experiments had a shorter half-life (1.4 days) in the gizzard than values reported for siliceous grit (3.1 days).

1. Two experiments were carried out to investigate the addition of 1 3 kg water per kg air-dry mash diets containing high proportions (600 to 700 g/kg) of ground cereal grains (wheat, barley or oats) on broiler performance and the structure and function of the gastro-intestinal tract. 2. Chicks at the age of 7 d were fed on the wheat-, barley- or oats-based diets in the dry or wet forms for 35 d. Food and water intakes were recorded daily while body weight was measured weekly. Two birds from each treatment were killed each week to measure gut size and the viscosity of gut contents. Tissue samples from various digestive segments were histo-morphologically examined to determine the thickness of tissue layers, size of tissue glands, villa heights, crypt depths and thickness of tunica muscularis. Crypt cell proliferation rate (CCPR) for each segment was also determined using a metaphase arrest technique. 3. The results from both experiments showed that wetting food significantly (P<0.05) increased food intake, total water intake and body weight gain of broiler chickens. The body weight gains of birds were proportional to their food intakes so that the efficiency of food utilisation was similar for all treatments. Dry matter retention of food tended to increase in birds given wet food from 7 to 21 d but not thereafter, compared to the dry-fed birds. Although water intake from the water bottle was significantly (P<0.05) reduced in birds given wet food, total water intakes from the water bottle plus that from food were significantly (P<0.05) higher in the wet-fed birds than in the dry-fed birds. The ratio of total water to dry food intake was, however, similar in both feeding regimens. 4. The fresh empty weight of the gut was increased by wet-feeding while its relative weight to body weight and the length of gut was not affected by dietary treatments. Significantly greater development of the tissue glands in the proventriculus and gizzard was observed in the birds given wet food; this was associated with the reduced thickness of the muscular layer of these segments. An increase in villus height was also observed in duodenum, small intestine, caeca and colon of birds given wet food, compared to those given dry food. CCPR was significantly (P<0.05) reduced by wet-feeding throughout the digestive tract. This was associated with a significant decrease in the mean viscosity of the gut contents and the concentration of volatile fatty acids (VFAs) in the caeca. 5. Wetting diets based on cereal grains caused a significant improvement in the performance of broiler chickens. The mechanism of the beneficial effects of wet feeding could be attributed to the decreased viscosity of gut contents; the greater development of the layer of villi in the digestive segments and the reduced CCPR in the crypts of the epithelium.

Small intestinal refluxes (SIRs) were monitored in the gastrointestinal tract of Nicholas turkeys via implanted strain gage transducers and radiographic observations. It was determined that reflux of ingesta from the small intestine into the gizzard was a result of a single antiperistaltic contraction. Radiographic observation indicated that the antiperistaltic contraction originated in the ileum 88% of the time and the duodenum 12% of the time. Each antiperistaltic contraction was preceded by a single peristaltic contraction. No gastric contractions occurred during SIRs. Fasting for 12 to 24 hr significantly increased the frequency of occurrence of SIRs in the Nicholas turkey (P < 0.0001) but had no significant effect on the velocity of the peristaltic and antiperistaltic contractions or on recovery time to pre-SIR gastric contractile frequency. The pooled mean velocity of peristaltic contractions was determined to be 10.55 +/- 3.68 cm/sec, and that of antiperistaltic contractions was determined to be 12.12 +/- 3.69 cm/sec. The pooled mean recovery time to pre-SIR gastric contraction frequency was 28.31 +/- 8.25 sec. It has been suggested that the SIR may be a mechanism of maximizing nutrient utilization by the turkey.

1. Routes of yolk utilisation and some aspects of intestinal digestion and absorption were determined in the hatching poult and compared to the chick. 2. Transfer from yolk to blood was observed pre-hatch and up to 72 h post-hatch. From hatch the transport pattern was similar to the chick. 3. Transport from the yolk sac into the intestine via the yolk stalk was observed up to 120 h after hatch. Secretion was initially to the proximal ileum and reflux occurred, transporting contents to the proximal small intestine and gizzard. Antiperistaltic movements increased after hatch and secretion continued for longer post-hatch than in the chick. 4. In situ uptake of glucose per unit of duodenum did not change with age, whereas uptake of methionine and oleic acid decreased with age. In contrast, in the chick glucose and methionine uptake capacity increased slightly between hatch and 7 d. 5. The lipid class distribution of intestinal contents resembled that of yolk close to hatch, however, some lipolysis was observed in the duodenum. With age the proportion of free fatty acids increased rapidly, first in the duodenum then in the ileum and finally in the caecum. Yolk in the distal small intestine close to hatch did not appear to be utilised.

Several studies were conducted to determine whether suppression of in vivo limestone solubilization was partially responsible for the reduction of shell quality under conditions of high environmental temperatures. In Experiment 1, excreta from hyperthermic and thermoneutral hens fed three levels of Ca (3.5, 4.3, and 5.2%) at two cycling environmental temperatures (averaging 28.3 and 26.1 C) were analyzed for percentage in vivo limestone solubilization. Hens in Experiment 2 received diets containing 3.9% Ca at 32.8 and 18.3 C. Rate of feed passage and gastrointestinal pH were also measured in Experiment 2. Experiment 3 evaluated the influence of temperature (22.2 and 30.0 C) and feed consumption on percentage limestone solubilization when Ca intake was held constant. In vivo limestone solubilization was influenced by Ca level in Experiment 1, but not by temperature. Hyperthermic hens solubilized a higher percentage of limestone than thermoneutral hens in Experiment 2, but it was concluded that this was due to a difference in Ca consumption and not due to temperature directly. When Ca intake was held constant in Experiment 3, there was no difference in limestone solubilization at the temperatures tested. Rate of feed passage was 16.6% slower in the hyperthermic hens. Crop, proventriculus, and upper small intestine pH were similar at each temperature, but gizzard pH was significantly lower in the hyperthermic hens. It was concluded that the high environmental temperatures used in the present studies did not suppress in vivo limestone solubilization.
