Cancer is a major cause of mortality in humans; often, rather than the primary tumor, it is the presence of metastases that are the cause of death. Extracellular vesicles (EVs) are small structures released by both normal and cancer cells; regarding the latter, they have been demonstrated to modulate almost all cancer-related processes, such as invasion, angiogenesis induction, drug resistance, and immune evasion. In the last years, it has become clear how EVs are widely involved in metastatic dissemination as well as in pre-metastatic niche (PMN) formation. Indeed, in order to achieve a successful metastatic process, i.e., penetration by cancer cells into distant tissues, the shaping of a favorable environment into those distant tissue, i.e., PMN formation, is mandatory. This process consists of an alteration that takes place in a distant organ and paves the way for the engraftment and growth of circulating tumor cells derived from the tumor primary site. This review focuses on the role of EVs in pre-metastatic niche formation and metastatic dissemination, also reporting the last studies suggesting the EVs role as biomarkers of metastatic diseases, possibly in a liquid biopsy approach. Multiple myeloma (MM) is an incurable hematologic neoplasm, whose poor prognosis is deeply affected by the propensity of tumor cells to localize in the bone marrow (BM) and induce the protumorigenic activity of normal BM cells, leading to events associated with tumor progression, including tumor angiogenesis, osteoclastogenesis, and the spread of osteolytic bone lesions. The interplay between MM cells and the BM niche does not only rely on direct cell-cell interaction, but a crucial role is also played by MM-derived extracellular vesicles (MM-EV). Here, we demonstrated that the oncogenic NOTCH receptors are part of MM-EV cargo and play a key role in EV protumorigenic ability. We used in vitro and in vivo models to investigate the role of EV-derived NOTCH2 in stimulating the protumorigenic behavior of endothelial cells and osteoclast progenitors. Importantly, MM-EV can transfer NOTCH2 between distant cells and increase NOTCH signaling in target cells. MM-EV stimulation increases endothelial cell angiogenic ability and osteoclast differentiation in a NOTCH2-dependent way. Indeed, interfering with NOTCH2 expression in MM cells may decrease the amount of NOTCH2 also in MM-EV and affect their angiogenic and osteoclastogenic potential. Finally, we demonstrated that the pharmacologic blockade of NOTCH activation by γ-secretase inhibitors may hamper the biological effect of EV derived by MM cell lines and by the BM of MM patients. These results provide the first evidence that targeting the NOTCH pathway may be a valid therapeutic strategy to hamper the protumorigenic role of EV in MM as well as other tumors.

The role of platelets in haemostasis has long been known, but understanding of these cells’ involvement in wound healing/tissue repair is more recent and has given rise to a multitude of translational studies. Tissue repair processes consist of complex, regulated interactions between cells modulated by biologically active molecules, most of which are growth factors released by activated platelets: this aspect represents the rationale on which the use of platelet derivatives for clinical purposes is based. In the last years, many in vitro studies have focused on the mechanisms of action by which these growth factors affect the biological activities of cells, thus supporting tissue healing. Although limited by some drawbacks (two-dimensional in vitro monocultures cannot replicate the tissue architecture and organisation of organs or the continuous interplay between different cell types), in vitro studies do have the advantages of giving rapid results and allowing precise control of platelet concentrations and other parameters. This review offers an updated overview of the data obtained from the most recent bench-top studies focused on the effects of platelet derivatives on a wide variety of human cells, highlighting their possible impact for in vivo applications. The heterogeneity of the data obtained so far is very evident. This can be explained by the different experimental settings used in each study, which may be the cause of the variability in clinical outcomes. In fact, in vitro studies suggest that the composition of platelet derivatives and the method used for their production and activation (or not) and the platelet concentration used can have profound effects on the final results.

The present study shows the results of experimental analyses of field cone penetration tests as well as calibration chamber mini-piezocone tests on soils of intermediate permeability (silts, clayey and sandy silts). The penetration rate varied across over three orders of magnitude to provide information on partially drained and undrained tip resistance, excess pore water pressure and friction sleeve. Whilst previous experimental researches essentially focused on tip resistance and pore water pressure measurements, it is worthwhile underlying that the present study is one of the first experimental studies that explored the effect of penetration rate on sleeve friction measurements. As the penetration rate is reduced, moving from the undrained conditions to the fully drained conditions, friction sleeve systematically decreases, together with the expected results in terms of increasing tip resistance and decreasing excess porewater pressure. The obtained experimental database of penetration measurements on intermediate soils can be added to the previous worldwide collected data to develop a new general interpretation procedure for cone tests in transitional soils. Besides, numerical analyses have been carried out by using the Finite Element Method. The Updated Lagrangian technique has been adopted to simulate the large strain penetration process. Both the Modified Cam Clay constitutive model and the Mohr Coulomb model have been used to compare numerical simulation results with, respectively, the experimental results on kaolin clay (Randolph and Hope, 2004; Schneider et al., 2007) and those obtained with the present study. The problem of piezocone miss-interpretation in case of transitional soils, such as loose silt mixture has been dealt with an empirical methodology, based on the calibration of the Soil Behaviour Type index using soil characteristics inferred from reference boreholes. Moreover, a new approach has been proposed to overcome miss-interpretation of piezocone test results for soil layers belonging to vadose zones in which the effective stress state is controlled by suction. In addition to that, the applied method has suggested a procedure, based on piezocone measurements, to estimate the effective stress state in the case of a homogeneous soil layer in which a vadose zone above the water table is present., Die vorliegende Studie zeigt die Ergebnisse experimenteller Analysen von Feldkegel-Penetrationstests sowie von Kalibrierkammer Miniatur-Piezocone Tests an Böden mittlerer Permeabilität (schluffige, lehmige und sandige Schluffsteine). Die Durchdringungsrate variierte über drei Größenordnungen, um Informationen über teilweise abgelassenen und undrainierten Spitzenwiderstand, überhöhten Porenwasserdruck und Reibungshülsen zu erhalten. Während sich bisherige experimentelle Forschungen im Wesentlichen auf den Spitzenwiderstand und Porenwasserdruckmessungen konzentrierten, ist die vorliegende Studie eine der ersten experimentellen Studien, die den Einfluss der Penetrationsrate auf die Messung von Reibungshülsen untersucht. Wenn die Durchdringungsrate verringert wird, von der undrainierten Bedingungen zu den vollständig abgelassenen Bedingungen gehend, nimmt die Reibungshülse systematisch ab, zusammen mit den erwarteten Ergebnissen im Hinblick auf einen erhöhten Spitzenwiderstand und einen abnehmenden Porenwasserdruck. Die entstandene experimentelle Datenbank für Penetrationsmessungen in Zwischenböden kann zu den zuvor weltweit gesammelten Daten hinzugefügt werden, um ein neues, allgemeines Interpretationsverfahren für Kegeltests in Übergangsböden zu entwickeln. Darüber hinaus wurden numerische Analysen mit der Finiten-Elementen-Methode durchgeführt. Die aktualisierte Lagrange-Methode wurde angewandt, um den Durchdringungsprozess großer Dehnungen zu simulieren. Sowohl das (modified) Cam Clay-Modell als auch das Mohr-Coulomb-Modell wurden verwendet, um numerische Simulationsergebnisse mit den experimentellen Ergebnissen an Kaolin (Randolph und Hope, 2004; Schneider et al., 2007) bzw. denen der Gegenwart zu vergleichen Studie. Das Problem der Piezocone Fehlinterpretation bei Übergangsböden wie losem Schlickgemisch wurde durch eine empirische Methode behandelt, basierend auf der Kalibrierung des Bodentyps unter Verwendung von aus Referenzbohrungen abgeleiteten Bodeneigenschaften. Außerdem wurde ein neuer Ansatz angeregt, um die Fehlinterpretation der Piezocone-Testergebnisse für Bodenschichten, die zur ungesättigten Zone gehören, in denen der effektive Spannungszustand durch Saugspannung gesteuert wird, zu beheben. Darüber hinaus hat das angewandte Verfahren eine Methode vorgestellt, das auf Piezocone-Messungen beruht, um den effektiven Spannungszustand bei einer homogenen Bodenschicht zu einzuschätzen, in der sich eine ungesättigte Zone oberhalb des Wasserspiegels befindet.

Com este trabalho pretendo analisar, através de uma revisão da literatura, os fatores que influenciam a escolha das várias metodologias de restauração adesiva, direta e indireta, em compósito nos dentes definitivos posteriores tratados endodonticamente. Uma vez submetido ao tratamento endodôntico, o dente sofre mudanças irreversíveis do ponto de vista anatómico, bioquímico e biomecânico, tornando-o mais suscetível à fratura e condicionando as futuras escolhas protéticas. As resinas compostas, usadas com técnicas adesivas, são os materiais de escolha para este tipo de restauração. Vão oferecer preparos conservadores, exercer ação de reforço em tecidos dentários residuais, reparáveis e podem dar excelente estética; em virtude dum módulo de elasticidade semelhante à dentina, garantem excelente desempenho biomecânico e permitem que o clínico possa realizar mais tarde uma restauração protética. Fundamental para um bom resultado final é necessário que o clínico realize um exame clínico meticuloso, para poder escolher as técnicas mais apropriadas. De acordo com os princípios da bio economia, a inserção de espigão endocanalar é limitada aos casos de perda coronária grave, (quando é necessário ancorar a restauração na raiz). Objetivo Avaliar o sucesso a longo prazo do tratamento com técnica direta (conservadora), ou técnica indireta (inlay, onlay ou overlay) em dentes tratados endodonticamente.

- Abstract
The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. We now update these “MISEV2014” guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles ("MISEV") guidelines for the field in 2014. We now update these "MISEV2014" guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Finally, a checklist is provided with summaries of key points.

- Abstract
The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles ("MISEV") guidelines for the field in 2014. We now update these "MISEV2014" guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Finally, a checklist is provided with summaries of key points.

Although the incidence of ovarian cancer is low (i.e., less than 5% in European countries), it is the most lethal gynecologic malignancy and typically has a poor prognosis. To ensure optimal survival, it is important to diagnose this condition when the pathology is confined to the ovary. However, this is difficult to achieve because the first specific symptoms appear only during advanced disease stages. To date, the biomarker mainly used for the diagnosis and prognosis of ovarian cancer is CA125; however, this marker has a low sensitivity and specificity and is associated with several other physiological and pathological conditions. No other serum ovarian cancer markers appear to be able to replace or complement CA125, and the current challenge is therefore to identify novel markers for the early diagnosis of this disease. For this purpose, studies have focused on the microvesicles (MVs) released from tumor cells. MVs may represent an ideal biomarker because they can be easily isolated from blood, and they have particular features (mainly regarding microRNA profiles) that strongly correlate with ovarian cancer stage and may be effective for early diagnosis.
