Blockchain technology offers unique features, such as transparency, the immutability of data, and the capacity to establish trust without a central authority. Such characteristics can be leveraged to support the collaboration among several different software systems operating within the healthcare ecosystem, while ensuring data integrity and make electronic health records (EHRs) more easily accessible. To provide a solution based on blockchain technology, this paper has evaluated the main issues that arise when large amounts of data are expected, i.e., mainly cost and performance. A balanced approach that maximizes the benefits and mitigates the constraints of the blockchain has been designed. The proposed decentralized application (dApp) architecture employs a hybrid storage strategy that involves storing medical records locally, on users’ devices, while utilizing blockchain to manage an index of these data. The dApp clients facilitate interactions among participants, leveraging a smart contract to enable patients to set authorization policies, thereby ensuring that only designated healthcare providers and authorized entities have access to specific medical records. The blockchain data-immutability property is used to validate data stored externally. This solution significantly reduces the costs related to the utilization of the blockchain, while retaining its advantages, and improves performance, since the majority of data are available off-chain.

Abstract In this paper we analyse the effects of information flows in cryptocurrency markets. We first define a cryptocurrency trading network, i.e. the network made using cryptocurrencies as nodes and the Granger causality among their weekly log returns as links, later we analyse its evolution over time. In particular, with reference to years 2020 and 2021, we study the logarithmic US dollar price returns of the cryptocurrency trading network using both pairwise and high-order statistical dependencies, quantified by Granger causality and O-information, respectively. With reference to the former, we find that it shows peaks in correspondence of important events, like e.g., Covid-19 pandemic turbulence or occasional sudden prices rise. The corresponding network structure is rather stable, across weekly time windows in the period considered and the coins are the most influential nodes in the network. In the pairwise description of the network, stable coins seem to play a marginal role whereas, turning high-order dependencies, they appear in the highest number of synergistic information circuits, thus proving that they play a major role for high order effects. With reference to redundancy and synergy with the time evolution of the total transactions in US dollars, we find that their large volume in the first semester of 2021 seems to have triggered a transition in the cryptocurrency network toward a more complex dynamical landscape. Our results show that pairwise and high-order descriptions of complex financial systems provide complementary information for cryptocurrency analysis.

We face a decentralized renewable energy production scenario, where a large number of small energy producers, i.e., prosumers, contribute to a common distributor entity, who resells energy directly to end-users. A major challenge for the distributor is to ensure power stability, constantly balancing produced vs consumed energy flows. In this context, being able to provide quick restore actions in response to unpredictable unbalancing events is a must, as fluctuations are the norm for renewable energy sources. To this aim, the high scalability and diversity of sources are crucial requirements for the said balancing to be actually manageable. In this study, we explored the challenges and benefits of adopting a blockchain-based software architecture as a scalable, trustless interaction platform between prosumers’ smart energy meters and the distributor. Our developed prototype accomplishes the energy load balancing service via smart contracts deployed in a real blockchain network with an increasing number of simulated prosumers. We show that the blockchain-based application managed to react in a timely manner to energy unbalances for up to a few hundred prosumers.

Abstract Online platforms implement digital reputation systems in order to steer individual user behaviour towards outcomes that are deemed desirable on a collective level. At the same time, most online platforms are highly decentralised environments, leaving their users plenty of room to pursue different strategies and diversify behaviour. We provide a statistical characterisation of the user behaviour emerging from the interplay of such competing forces in Stack Overflow, a long-standing knowledge sharing platform. Over the 11 years covered by our analysis, we represent the interactions between users and topics as bipartite networks. We find such networks to display nested structures akin to those observed in ecological systems, demonstrating that the platform’s user base consistently self-organises into specialists and generalists, i.e., users who focus on narrow and broad sets of topics, respectively. We relate the emergence of these behaviours to the platform’s reputation system with a series of data-driven models, and find specialisation to be statistically associated with a higher ability to post the best answers to a question. We contrast our findings with observations made in top-down environments—such as firms and corporations—where generalist skills are consistently found to be more successful.

The increased penetration of Distributed Energy Resources (DERs) in electricity markets has given rise to a new category of energy players, called Aggregators, whose role is to ensure fair remuneration for energy supplied by DERs, and support the smooth feeding of the intermittent energy produced into the distribution network. This paper presents a software solution, described as a design pattern, that governs the interaction between an Aggregator and DERs, leveraging blockchain technology to achieve a higher degree of decentralization, data integrity and security, through a properly designed, blockchain-based, smart contract. Thus, the proposed solution reduces the reliance on intermediaries acting as authorities, while affording transparency, efficiency and trust to the energy exchange process. Thanks to the underlying blockchain properties, generated events are easily observable and cannot be forged or altered. However, blockchain technology has inherent drawbacks, i.e., mainly the cost of storage and execution, hence our solution provides additional strategies for limiting blockchain usage, without undermining its strengths. Moreover, the design of our smart contract takes care of orchestrating the players, and copes with their potential mutual disagreements, which could arise from different measures of energy, providing an automatic decision process to resolve such disputes. The overall approach results in lower fees for running smart contacts supporting energy players and in a greater degree of fairness assurance.

In the last two decades, the amyloid hypothesis, i.e., the abnormal accumulation of toxic Aβ assemblies in the brain, has been considered the mainstream concept sustaining research in Alzheimer’s Disease (AD). However, the course of cognitive decline and AD development better correlates with tau accumulation rather than amyloid peptide deposition. Moreover, all clinical trials of amyloid-targeting drug candidates have been unsuccessful, implicitly suggesting that the amyloid hypothesis needs significant amendments. Accumulating evidence supports the existence of a series of potentially dangerous relationships between Aβ oligomeric species and tau protein in AD. However, the molecular determinants underlying pathogenic Aβ/tau cross interactions are not fully understood. Here, we discuss the common features of Aβ and tau molecules, with special emphasis on: (i) the critical role played by metal dyshomeostasis in promoting both Aβ and tau aggregation and oxidative stress, in AD; (ii) the effects of lipid membranes on Aβ and tau (co)-aggregation at the membrane interface; (iii) the potential of small peptide-based inhibitors of Aβ and tau misfolding as therapeutic tools in AD. Although the molecular mechanism underlying the direct Aβ/tau interaction remains largely unknown, the arguments discussed in this review may help reinforcing the current view of a synergistic Aβ/tau molecular crosstalk in AD and stimulate further research to mechanism elucidation and next-generation AD therapeutics.

Pike (1945) classified the world languages into two types of rhythmic/prosodic patterns: stress-timed and syllable-timed. According to this classification, stress-timed languages, like English and German, tend to have isochronous interstress intervals, while syllable-timed languages, like Italian and Spanish, tend to have equal syllable duration. Ladefoged (1975) added the mora-timed type, in which isochrony is maintained at the level of the mora, a sub-syllabic constituent that includes either onset and nucleus, or a coda. Japanese is often referred to as a mora-timed language (Otake 2015): the mora is the psychological prosodic unit in spoken language, and the metric unit of traditional poetry (Bloch 1950). The syllabaries, in which each grapheme corresponds to a mora, make this prosodic segmentation clear. However, previous experimental studies have claimed that the mora is not a perfect isochronous unit (Warner and Arai 2001). The aim of this paper is to present the rhythm-prosodic system of the Japanese language giving a precise description of its prosodic units -- the mora and the syllable--, and to provide empirical quantitative data on the duration of mora in spontaneous Japanese. The dataset used in the present study is a portion of the Corpus of Spontaneous Japanese called Core, consisting of about 45 hours of extensively annotated speech. The variation of the average duration of the mora has been analysed on the basis of linguistic parameters, such as the typology of mora and the phonotactic structure of the word in which it is included, and of extra-linguistic parameters, such as the typology of speech.

Abstract We investigate Bitcoin network observing transactions broadcasted into the network during a week from 04/05/2016 and then monitoring their inclusion into the blockchain during the following seven months.We unveil that 42% of the transactions are still not included in the Blockchain after 1 h from their appearance and 20% of the transactions are still not included in the Blockchain after 30 days, therefore revealing a great inefficiency in the Bitcoin system. However, we observe that most of these “forgotten” transactions have low values and in terms of transferred value the system is less inefficient with 93% of the transactions value being included into the Blockchain within 3 h and 98.8% within a day. The fact that a sizeable fraction of transactions is not processed timely casts serious doubts on the usability of the Bitcoin Blockchain for reliable time-stamping purposes. It also calls for a debate about the right systems of incentives which a peer-to-peer unintermediated system should introduce to promote efficient transaction recording

In the few last years, nanosystems have emerged as a potential therapeutic approach to improve the efficacy and selectivity of many drugs. Cyclodextrins (CyDs) and their nanoparticles have been widely investigated as drug delivery systems. The covalent functionalization of CyD polymer nanoparticles with targeting molecules can improve the therapeutic potential of this family of nanosystems. In this study, we investigated cross-linked γ- and β-cyclodextrin polymers as carriers for doxorubicin (ox) and oxaliplatin (Oxa). We also functionalized γ-CyD polymer bearing COOH functionalities with arginine-glycine-aspartic or arginine moieties for targeting the integrin receptors of cancer cells. We tested the Dox and Oxa anti-proliferative activity in the presence of the precursor polymer with COOH functionalities and its derivatives in A549 (lung, carcinoma) and HepG2 (liver, carcinoma) cell lines. We found that CyD polymers can significantly improve the antiproliferative activity of Dox in HepG2 cell lines only, whereas the cytotoxic activity of Oxa resulted as enhanced in both cell lines. The peptide or amino acid functionalized CyD polymers, loaded with Dox, did not show any additional effect compared to the precursor polymer. Finally, studies of Dox uptake showed that the higher antiproliferative activity of complexes correlates with the higher accumulation of Dox inside the cells. The results show that CyD polymers could be used as carriers for repositioning classical anticancer drugs such as Dox or Oxa to increase their antitumor activity.
