1. Beam focusing and consequences for Doppler Backscattering measurements

Abstract

The phenomenon of beam focusing of microwaves in a plasma near a turning-point caustic is discussed in the context of the analytical solution to the Gaussian beam-tracing equations in the 2D linear-layer problem. The location of maximum beam focusing and the beam width at that location are studied in terms of the beam initial conditions. The analytic solution is used to study the effect of this focusing on Doppler backscattering (DBS). We find that the filter function that characterises the scattering intensity contributions along the beam path through the plasma is inversely proportional to the beam width, predicting enhanced scattering contributions from the beam focusing region. We show that the DBS signal enhancement for small incident angles between the beam path and the density gradient is due to beam focusing and not due to forward scattering. The analytic beam model is used to predict the measurement of the $k_y$ density-fluctuation wavenumber power spectrum via DBS, showing that the spectral exponent of the turbulent, intermediate-to-high $k_y$ density-fluctuation spectrum might be quantitatively measurable via DBS, but not the spectral peak corresponding to the driving scale of the turbulent cascade.

Abstract

Speech is a multiplexed signal displaying levels of complexity, organizational principles and perceptual units of analysis at distinct timescales. This critical acoustic signal for human communication is thus characterized at distinct representational and temporal scales, related to distinct linguistic features, from acoustic to supra-lexical. This chapter presents an overview of experimental work devoted to the characterization of the speech signal at different timescales, beyond its acoustic properties. The functional relevance of these different levels of analysis for speech processing is discussed. We advocate that studying speech perception through the prism of multi-time scale representations effectively integrates work from various research areas into a coherent picture and contributes significantly to increase our knowledge on the topic. Abstract

We report the first experimental detection of a zero-frequency fluctuation that is pumped by an Alfv\'en mode in a magnetically confined plasma. Core-localized bidirectional Alfv\'en modes of frequency inside the toroidicity-induced gap (and its harmonics) exhibit three-wave coupling interactions with a zero-frequency fluctuation. The observation of the zero-frequency fluctuation is consistent with theoretical and numerical predictions of zonal modes pumped by Alfv\'en modes, and is correlated with an increase in the deep core ion temperature, temperature gradient, and confinement factor $H_{89,P}$. Despite the energetic particle transport induced by the Alfv\'en eigenmodes, the generation of a zero-frequency fluctuation that can suppress the turbulence leads to an overall improvement of confinement.

Abstract

Gyrotrons are essential for electron cyclotron resonance heating (ECRH) in fusion reactors, making efficient operation crucial for advancing fusion energy. Past experiments revealed instability issues due to trapped electrons in the magnetron injection gun (MIG) region, causing undesired currents and operational failures. To address this, tight manufacturing tolerances are required for the MIG geometry [1]. We present initial findings of the TRapped Electrons eXperiment (T-REX) developed at the Swiss Plasma Center, designed to understand the physics of electron clouds in gyrotron MIGs. T-REX replicates MIG geometries, as well as their typical electric and magnetic fields, and it is supported by 2D Particle-in-Cell (PIC) simulations with the FENNECS code [2, 3]. The setup includes two coaxial electrodes in a vacuum chamber atop a superconducting magnet, with a central electrode biased to negative DC voltages and an outer one at ground, creating a radial electric field (1 to 2 MV/M) and an axial magnetic field (B < 0.4 T). This setup mimics Penning-Malmberg traps. We present the experimental device and first findings on current distribution and also qualitative comparison with FENNECS simulations [4]. Planned diagnostics include optical emission spectroscopy, phosphor screen imaging, Streak camera imaging, and potentially electric field distribution via the Stark effect. This research aims to enhance gyrotron performance and reliability in fusion energy systems.

Abstract

BlueMUSE is a blue-optimised, medium spectral resolution, panoramic integral field spectrograph under development for the Very Large Telescope (VLT). With an optimised transmission down to 350 nm, spectral resolution of R$\sim$3500 on average across the wavelength range, and a large FoV (1 arcmin$^2$), BlueMUSE will open up a new range of galactic and extragalactic science cases facilitated by its specific capabilities. The BlueMUSE consortium includes 9 institutes located in 7 countries and is led by the Centre de Recherche Astrophysique de Lyon (CRAL). The BlueMUSE project development is currently in Phase A, with an expected first light at the VLT in 2031. We introduce here the Top Level Requirements (TLRs) derived from the main science cases, and then present an overview of the BlueMUSE system and its subsystems fulfilling these TLRs. Abstract

Volume-phase holographic gratings (VPHGs) are widely used in astronomical spectrographs due to their adaptability and high diffraction efficiency. Most VPHGs in operation use dichromated gelatin as a recording material, whose performance is sensitive to the coating and development process, especially in the near-UV. In this letter, we present the characterization of two UV-blue VPHG prototypes for the BlueMUSE integral field spectrograph on the VLT, based on dichromated gelatin and the Bayfol$\circledR$HX photopolymer film as recording materials. Our measurements show that both prototypes meet the required diffraction efficiency and exhibit similar performance with a wavelength-average exceeding 70% in the 350-580 nm range. Deviations from theoretical models increase towards 350 nm, consistently with previous studies on similar gratings. We also report similar performances in terms spatial uniformity and grating-to-grating consistency. Likewise, no significant differences in wavefront error or scattered light are observed between the prototypes., Comment: 7 pages, 5 figures, 1 table. Abstract

Abstract The multivariate temporal response function (mTRF) is an effective tool for investigating the neural encoding of acoustic and complex linguistic features in natural continuous speech. In this study, we investigated how neural representations of speech features derived from natural stimuli are related to early signs of cognitive decline in older adults, taking into account the effects of hearing. Participants without ( $$n = 25$$ n = 25 ) and with ( $$n = 19$$ n = 19 ) early signs of cognitive decline listened to an audiobook while their electroencephalography responses were recorded. Using the mTRF framework, we modeled the relationship between speech input and neural response via different acoustic, segmented and linguistic encoding models and examined the response functions in terms of encoding accuracy, signal power, peak amplitudes and latencies. Our results showed no significant effect of cognitive decline or hearing ability on the neural encoding of acoustic and linguistic speech features. However, we found a significant interaction between hearing ability and the word-level segmentation model, suggesting that hearing impairment specifically affects encoding accuracy for this model, while other features were not affected by hearing ability. These results suggest that while speech processing markers remain unaffected by cognitive decline and hearing loss per se, neural encoding of word-level segmented speech features in older adults is affected by hearing loss but not by cognitive decline. This study emphasises the effectiveness of mTRF analysis in studying the neural encoding of speech and argues for an extension of research to investigate its clinical impact on hearing loss and cognition.

Abstract Root exudates contain specialised metabolites that shape the plant’s root microbiome. How host-specific microbes cope with these bioactive compounds, and how this ability affects root microbiomes, remains largely unknown. We investigated how maize root bacteria metabolise benzoxazinoids, the main specialised metabolites of maize. Diverse and abundant bacteria metabolised the major compound in the maize rhizosphere MBOA (6-methoxybenzoxazolin-2(3H)-one) and formed AMPO (2-amino-7-methoxy-phenoxazin-3-one). AMPO forming bacteria were enriched in the rhizosphere of benzoxazinoid-producing maize and could use MBOA as carbon source. We identified a gene cluster associated with AMPO formation in microbacteria. The first gene in this cluster, bxdA encodes a lactonase that converts MBOA to AMPO in vitro. A deletion mutant of the homologous bxdA genes in the genus Sphingobium, did not form AMPO nor was it able to use MBOA as a carbon source. BxdA was identified in different genera of maize root bacteria. Here we show that plant-specialised metabolites select for metabolisation-competent root bacteria. BxdA represents a benzoxazinoid metabolisation gene whose carriers successfully colonize the maize rhizosphere and thereby shape the plant’s chemical environmental footprint.

Abstract

High strength metastable β titanium alloys are widely employed in the aircraft industry due to their outstanding strength-to-weight ratio. While components can endure complex in-service mechanical loading, the anisotropy in tensile properties has been the subject of limited attention. In this study, its origin was investigated focusing on the role played by millimeter scale β grains as they were recently identified as a source of heterogeneous deformation. Tensile properties of Ti-10V-2Fe-3Al processed via different thermomechanical routes were assessed using multiple sampling directions. In particular, elongation values were observed to vary significantly depending on the testing direction. A combination of SEM, EBSD, µ-CT and in-situ DIC during tensile tests was employed to clarify the underlying causes of this behavior. Substantial differences in strain heterogeneity and localization were found related to features of β grains, including their crystallographic and morphologic orientations. Furthermore, multiple fracture mechanisms were observed to derive from the differences in deformation behavior, and eventually compete to trigger specimen failure. Elongation values are then determined by both the degree of strain heterogeneity and the operating fracture mechanisms. These findings provide a new understanding of the role of the microstructure in the tensile behavior of high strength metastable β titanium alloys.

Abstract

Abstract Chronic hepatitis B (CHB) is a major global health challenge. CHB can be controlled by antivirals but a therapeutic cure is lacking. CHB is characterized by limited HBV-specific T cell reactivity and functionality and expression of inhibitory receptors. The mechanisms driving these T cell phenotypes are only partially understood. Here, we created a single-cell RNA-sequencing dataset of HBV immune responses in patients to contribute to a better understanding of the dysregulated immunity. Blood samples of a well-defined cohort of 21 CHB and 10 healthy controls, including a subset of 5 matched liver biopsies, were collected. scRNA-seq data of total immune cells (55,825) plus sorted HBV-specific (1,963), non-naive (32,773) and PD1+ T cells (96,631) was generated using the 10X Genomics platform (186,123 cells) or the full-length Smart-seq2 protocol (1,069 cells). The shared transcript count matrices of single-cells serve as a valuable resource describing transcriptional changes underlying dysfunctional HBV-related T cell responses in blood and liver tissue and offers the opportunity to identify targets or biomarkers for HBV-related immune exhaustion.

Abstract

BackgroundMultiple Sclerosis (MS) is a common neurological disease among white populations of European origin. Frequencies among Latin Americans continue to be studied, however, epidemiologic, and clinical characterization studies lack from Central American and Caribbean countries. Ethnicity in these countries is uniformly similar with a prevalent Mestizo population.Methods and resultsData from January 2014 to December 2019 from Guatemala, El Salvador, Honduras, Nicaragua, Costa Rica, Panama, Dominican Republic, and Aruba on demographic, clinical, MRI and phenotypic traits were determined in coordinated studies: ENHANCE, a population-based, retrospective, observational study on incidence and clinical characteristics, and from the subgroup with MS national registries (Aruba, Dominican Republic, Honduras, and Panama), data on prevalence, phenotypes and demographics. Expanded Disability Status Scale (EDSS), and therapeutic schemes were included. ENHANCE data from 758 patients disclosed 79.8% of Mestizo ethnicity; 72.4% female; median age at onset 31.0 years and 33.2 at diagnosis. The highest incidence rate was from Aruba, 2.3–3.5 × 100,000 inhabitants, and the lowest, 0.07–0.15 × 100,000, from Honduras. Crude prevalence rates per 100,000 inhabitants fluctuated from 27.3 (Aruba) to 1.0 (Honduras). Relapsing MS accounted for 87.4% of cases; EDSS

Abstract

Nonlinear multiscale gyrokinetic simulations of a Joint European Torus edge pedestal are used to show that electron-temperature-gradient (ETG) turbulence has a rich three-dimensional structure, varying strongly according to the local magnetic-field configuration. In the plane normal to the magnetic field, the steep pedestal electron temperature gradient gives rise to anisotropic turbulence with a radial (normal) wavelength much shorter than in the binormal direction. In the parallel direction, the location and parallel extent of the turbulence are determined by the variation in the magnetic drifts and finite-Larmor-radius (FLR) effects. The magnetic drift and FLR topographies have a perpendicular-wavelength dependence, which permits turbulence intensity maxima near the flux-surface top and bottom at longer binormal scales, but constrains turbulence to the outboard midplane at shorter electron-gyroradius binormal scales. Our simulations show that long-wavelength ETG turbulence does not transport heat efficiently, and significantly decreases overall ETG transport -- in our case by $\sim$40 \% -- through multiscale interactions., Comment: 17 pages, 14 figures

Abstract

Torpor or heterothermy is an energy‐saving mechanism used by endotherms to overcome harsh environmental conditions. During winter, the garden dormouse (Eliomys quercinus) hibernates with multiday torpor bouts and body temperatures of a few degrees Celsius, interrupted by brief euthermic phases. This study investigates gene expression within the hypothalamus, the key brain area controlling energy balance, adding information on differential gene expression potentially relevant to orchestrate torpor. A de novo assembled transcriptome of the hypothalamus was generated from garden dormice hibernating under constant darkness without food and water at 5 °C. Samples were collected during early torpor, late torpor, and interbout arousal. During early torpor, 765 genes were differentially expressed as compared with interbout arousal. Twenty‐seven pathways were over‐represented, including pathways related to hemostasis, extracellular matrix organization, and signaling of small molecules. Only 82 genes were found to be differentially expressed between early and late torpor, and no pathways were over‐represented. During late torpor, 924 genes were differentially expressed relative to interbout arousal. Despite the high number of differentially expressed genes, only 10 pathways were over‐represented. Of these, eight were also observed to be over‐represented when comparing early torpor and interbout arousal. Our results are largely consistent with previous findings in other heterotherms. The addition of a transcriptome of a novel species may help to identify species‐specific and overarching torpor mechanisms through future species comparisons.

Abstract The consensus molecular subtypes (CMS) of colorectal cancer (CRC) is the most widely-used gene expression-based classification and has contributed to a better understanding of disease heterogeneity and prognosis. Nevertheless, CMS intratumoral heterogeneity restricts its clinical application, stressing the necessity of further characterizing the composition and architecture of CRC. Here, we used Spatial Transcriptomics (ST) in combination with single-cell RNA sequencing (scRNA-seq) to decipher the spatially resolved cellular and molecular composition of CRC. In addition to mapping the intratumoral heterogeneity of CMS and their microenvironment, we identified cell communication events in the tumor-stroma interface of CMS2 carcinomas. This includes tumor growth-inhibiting as well as -activating signals, such as the potential regulation of the ETV4 transcriptional activity by DCN or the PLAU-PLAUR ligand-receptor interaction. Our study illustrates the potential of ST to resolve CRC molecular heterogeneity and thereby help advance personalized therapy.

Abstract

A linear response, local model for the DBS amplitude applied to gyrokinetic simulations shows that radial correlation Doppler reflectometry measurements (RCDR, Schirmer et al., Plasma Phys. Control. Fusion 49 1019 (2007)) are not sensitive to the average turbulence radial correlation length, but to a correlation length that depends on the binormal wavenumber $k_\perp$ selected by the Doppler backscattering (DBS) signal. Nonlinear gyrokinetic simulations show that the turbulence naturally exhibits a non-separable power law spectrum in wavenumber space, leading to a power law dependence of the radial correlation length with binormal wavenumber $l_r \sim C k_\perp^{-\alpha} (\alpha \approx 1)$ which agrees with the inverse proportionality relationship between the measured $l_r$ and $k_\perp $ in experiments (Fernandez-Marina et al., Nucl. Fusion 54 072001 (2014)). This offers the possibility of characterizing the eddy aspect ratio in the perpendicular plane to the magnetic field and motivates future use of a non-separable turbulent spectrum to quantitatively interpret RCDR and potentially other turbulence diagnostics. The radial correlation length is only measurable when the radial resolution at the cutoff location $W_n$ satisfies $W_n \ll l_r$, while the measurement becomes dominated by $W_n$ for $W_n \gg l_r$. This suggests that $l_r$ is likely inaccessible for electron-scale DBS measurements ($k_\perp\rho_s > 1$). 24. Improvements in Naturalistic Speech-in-Noise Comprehension in Middle-Aged and Older Adults after 3 Weeks of Computer-Based Speechreading Training

Abstract

Problems in understanding speech in noisy environments are characteristic for age-related hearing loss. Since hearing aids do not mitigate these communication problems in every case, potential alternatives in a clinical rehabilitation plan need to be explored. This study investigates whether a computer-based speechreading training improves audiovisual speech perception in noise in a sample of middle-aged and older adults (N = 62, 47-83 years) with 32 participants completing a speechreading training and 30 participants of an active control group completing a foreign language training. Before and after training participants performed a speech-in-noise task mimicking real-life communication settings with participants being required to answer a speaker's questions. Using generalized linear mixed-effects models we found a significant improvement in audiovisual speech perception in noise in the speechreading training group. This is of great relevance as these results highlight the potential of a low-cost and easy-to-implement intervention for a profound and widespread problem as speech-in-noise comprehension impairment.

In our previous work \cite{PTA2} we calculated the overlap reduction function for the tensor polarization without employing the short wavelength approximation, this was done by obtaining a power series of nested sums which is valid for all gravitational wave frequencies and pulsar distances. In this work we generalize the power-series expansion method to vector and scalar polarizations. We have compared our expression for the breathing and vector modes with previous literature. We present for the first time analytic expressions for the overlap reduction function of the longitudinal mode for all angles between the pulsar pairs.

Abstract

Hibernation is a period of metabolic suppression utilized by many small and large mammal species to survive during winter periods. As the underlying cellular and molecular mechanisms remain incompletely understood, our study aimed to determine whether skeletal muscle myosin and its metabolic efficiency undergo alterations during hibernation to optimize energy utilization. We isolated muscle fibers from small hibernators, Ictidomys tridecemlineatus and Eliomys quercinus and larger hibernators, Ursus arctos and Ursus americanus. We then conducted loaded Mant-ATP chase experiments alongside X-ray diffraction to measure resting myosin dynamics and its ATP demand. In parallel, we performed multiple proteomics analyses. Our results showed a preservation of myosin structure in U. arctos and U. americanus during hibernation, whilst in I. tridecemlineatus and E. quercinus, changes in myosin metabolic states during torpor unexpectedly led to higher levels in energy expenditure of type II, fast-twitch muscle fibers at ambient lab temperatures (20 °C). Upon repeating loaded Mant-ATP chase experiments at 8 °C (near the body temperature of torpid animals), we found that myosin ATP consumption in type II muscle fibers was reduced by 77–107% during torpor compared to active periods. Additionally, we observed Myh2 hyper-phosphorylation during torpor in I. tridecemilineatus, which was predicted to stabilize the myosin molecule. This may act as a potential molecular mechanism mitigating myosin-associated increases in skeletal muscle energy expenditure during periods of torpor in response to cold exposure. Altogether, we demonstrate that resting myosin is altered in hibernating mammals, contributing to significant changes to the ATP consumption of skeletal muscle. Additionally, we observe that it is further altered in response to cold exposure and highlight myosin as a potentially contributor to skeletal muscle non-shivering thermogenesis.

Abstract

Seasonal animal dormancy is widely interpreted as a physiological response for surviving energetic challenges during the harshest times of the year (the physiological constraint hypothesis). However, there are other mutually non-exclusive hypotheses to explain the timing of animal dormancy, that is, entry into and emergence from hibernation (i.e. dormancy phenology). Survival advantages of dormancy that have been proposed are reduced risks of predation and competition (the ‘life-history’ hypothesis), but comparative tests across animal species are few. Using the phylogenetic comparative method applied to more than 20 hibernating mammalian species, we found support for both hypotheses as explanations for the phenology of dormancy. In accordance with the life-history hypotheses, sex differences in hibernation emergence and immergence were favored by the sex difference in reproductive effort. In addition, physiological constraint may influence the trade-off between survival and reproduction such that low temperatures and precipitation, as well as smaller body mass, influence sex differences in phenology. We also compiled initial evidence that ectotherm dormancy may be (1) less temperature dependent than previously thought and (2) associated with trade-offs consistent with the life-history hypothesis. Thus, dormancy during non-life-threatening periods that are unfavorable for reproduction may be more widespread than previously thought.

Abstract

The increasing use of chemicals requires a better understanding of their presence and dynamics in the environment, as well as their impact on ecosystems. The aim of this study was to validate the first steps of an innovative multi-omics approach based on metabolomics and 16S metabarcoding data for analyses of the fate and impact of contaminants in Mediterranean lagoons. Semi-targeted analytical procedures for water and sediment matrices were implemented to assess chemical contamination of the lagoon: forty-six compounds were detected, 28 of which could be quantified in water (between 0.09 and 47.4 ng/L) and sediment (between 0.008 and 26.3 ng/g) samples using the UHPLC-MS/MS instrument. In addition, a non-targeted approach (UHPLC-HRMS) using four different sample preparation protocols based on solid/liquid extractions or an automated pressurized fluid extraction system (EDGE®) was carried out to determine the protocol with the best metabolome coverage, efficiency and reproducibility. Solid/liquid extraction using the solvent mixture acetonitrile/methanol (50/50) was evaluated as the best protocol. Microbial diversity in lagoon sediment was also measured after DNA extraction using five commercial extraction kits. Our study showed that the DNeasy PowerSoil Pro Qiagen kit (Promega, USA) was the most suitable for assessing microbial diversity in fresh sediment.

Abstract Background Despite centuries of research, debate remains on the scaling of metabolic rate to mass especially for intraspecific cases. The high variation of body mass within brown bears presents a unique opportunity to study the intraspecific effects of body mass on physiological variables. The amplitude of metabolic rate reduction in hibernators is dependent on body mass of the species. Small hibernators have high metabolic rates when euthermic but experience a drastic decrease in body temperature during torpor, which is necessary to reach a very low metabolic rate. Conversely, large hibernators, such as the brown bear (Ursus arctos), show a moderate decrease in temperature during hibernation, thought to be related to the bear’s large size. We studied body mass, abdominal body temperature, heart rate, and accelerometer-derived activity from 63 free-ranging brown bears (1–15 years old, 15–233 kg). We tested for relationships between body mass and body temperature, heart rate, and hibernation duration. Results The smallest individuals maintained lower body temperatures during hibernation, hibernated longer, and ended hibernation later than large bears. Unlike body temperature, winter heart rates were not associated with body mass. In summer, the opposite pattern was found, with smaller individuals having higher body temperature and daytime heart rates. Body mass was associated with body temperature in the winter hypometabolic state, even in a large hibernating mammal. Smaller bears, which are known to have higher thermal conductance, reached lower body temperatures during hibernation. During summer, smaller bears had higher body temperatures and daytime heart rates, a phenomenon not previously documented within a single mammalian species. Conclusion We conclude that the smallest bears hibernated more deeply and longer than large bears, likely from a combined effect of basic thermodynamics, the higher need for energy savings, and a lower cost of warming up a smaller body.

We report on a detailed study of magnetic fluctuations in the JET pedestal, employing basic theoretical considerations, gyrokinetic simulations, and experimental fluctuation data, to establish the physical basis for their origin, role, and distinctive characteristics. We demonstrate quantitative agreement between gyrokinetic simulations of microtearing modes (MTMs) and two magnetic frequency bands with corresponding toroidal mode numbers n=4 and 8. Such disparate fluctuation scales, with substantial gaps between toroidal mode numbers, are commonly observed in pedestal fluctuations. Here we provide a clear explanation, namely the alignment of the relevant rational surfaces (and not others) with the peak in the omega star profile, which is localized in the steep gradient region of the pedestal. We demonstrate that a global treatment is required to capture this effect. Nonlinear simulations suggest that the MTM fluctuations produce experimentally-relevant transport levels and saturate by relaxing the background electron temperature gradient, slightly downshifting the fluctuation frequencies from the linear predictions. Scans in collisionality are compared with simple MTM dispersion relations. At the experimental points considered, MTM growth rates can either increase or decrease with collision frequency depending on the parameters thus defying any simple characterization of collisionality dependence.

37. Toroidal and slab ETG instability dominance in the linear spectrum of JET-ILW pedestals

Local linear gyrokinetic simulations show that electron temperature gradient (ETG) instabilities are the fastest growing modes for $k_y \rho_i \gtrsim 0.1$ in the steep gradient region for a JET pedestal discharge (92174) where the electron temperature gradient is steeper than the ion temperature gradient. Here, $k_y$ is the wavenumber in the direction perpendicular to both the magnetic field and the radial direction, and $\rho_i$ is the ion gyroradius. At $k_y \rho_i \gtrsim 1$, the fastest growing mode is often a novel type of toroidal ETG instability. This toroidal ETG mode is driven at scales as large as $k_y \rho_i \sim (\rho_i/\rho_e) L_{Te} / R_0 \sim 1$ and at a sufficiently large radial wavenumber that electron finite Larmor radius effects become important; that is, $K_x \rho_e \sim 1$, where $K_x$ is the effective radial wavenumber. Here, $\rho_e$ is the electron gyroradius, $R_0$ is the major radius of the last closed flux surface, and $1/L_{Te}$ is an inverse length proportional to the logarithmic gradient of the equilibrium electron temperature. The fastest growing toroidal ETG modes are often driven far away from the outboard midplane. In this equilibrium, ion temperature gradient instability is subdominant at all scales and kinetic ballooning modes are shown to be suppressed by $\mathbf{ E} \times \mathbf{ B} $ shear. ETG modes are very resilient to $\mathbf{ E} \times \mathbf{ B}$ shear. Abstract

Abstract Background Torpor is an energy saving strategy achieved by substantial reductions of metabolic rate and body temperature that enables animals to survive periods of low resource availability. During hibernation (multiday torpor), the frequency of periodic rewarming—characterised by high levels of oxidative stress—is associated with shortening of telomeres, a marker of somatic maintenance. Objectives In this study, we determined the impact of ambient temperature on feeding behaviour and telomere dynamics in hibernating garden dormice (Eliomys quercinus) over winter. This obligate hibernator prepares for hibernation by accumulating fat stores but can also feed during hibernation. Methodology Food intake, torpor pattern, changes in telomere length, and body mass change were assessed in animals housed at experimentally controlled temperatures of either 14 °C (i.e., a mild winter) or 3 °C (i.e., a cold winter) over 6 months. Results When hibernating at 14 °C, dormice experienced 1.7-fold more frequent and 2.4-fold longer inter-bout euthermia, and spent significantly less time torpid, compared to animals hibernating at 3 °C. Higher food intake enabled individuals to compensate for increased energetic costs when hibernating at milder temperatures (14 °C vs. 3 °C), to buffer body mass loss and thus increase winter survival. Interestingly, we observed a significant increase of telomere length over the entire hibernation period, irrespective of temperature treatment. Conclusion We conclude that higher temperatures during winter, if associated with sufficient food availability, can have a positive effect on the individual’s energy balance and somatic maintenance. These results suggest that winter food availability might be a crucial determinant for the survival of the garden dormouse in the context of ever-increasing environmental temperatures.

Telomere dynamics in hibernating species are known to reflect seasonal changes in somatic maintenance. Throughout hibernation, the periodic states of rewarming, known as inter-bout euthermia or arousals, are associated with high metabolic costs including shortening of telomeres. In the active season, if high energetic resources are available, telomere length can be restored in preparation for the upcoming winter. The mechanism for telomere elongation has not been clearly demonstrated, although the action of the ribonucleoprotein complex, telomerase, has been implicated in many species. Here we tested for levels of telomerase activity in the garden dormouse (Eliomys quercinus) at different seasonal time points throughout the year and across ages from liver tissues of male juveniles to adults. We found that telomerase is active at high levels across seasons (during torpor and inter-bout euthermia, plus in the active season) but that there was a substantial decrease in activity in the month prior to hibernation. Telomerase levels were consistent across age groups and were independent of feeding regime and time of birth (early or late born). The changes in activity levels that we detected were broadly associated with changes in telomere lengths measured in the same tissues. We hypothesise that i) telomerase is the mechanism used by garden dormice for maintenance of telomeres and that ii) activity is kept at high levels throughout the year until pre-hibernation when resources are diverted to increasing fat reserves for overwintering. We found no evidence for a decrease in telomerase activity with age or a final increase in telomere length which has been detected in other hibernating rodents.

Abstract

Healthy aging is often associated with speech comprehension difficulties in everyday life situations despite a pure-tone hearing threshold in the normative range. Drawing on this background, we used a multidimensional approach to assess the functional and structural neural correlates underlying age-related temporal speech processing while controlling for pure-tone hearing acuity. Accordingly, we combined structural magnetic resonance imaging and electroencephalography, and collected behavioral data while younger and older adults completed a phonetic categorization and discrimination task with consonant-vowel syllables varying along a voice-onset time continuum. The behavioral results confirmed age-related temporal speech processing singularities which were reflected in a shift of the boundary of the psychometric categorization function, with older adults perceiving more syllable characterized by a short voice-onset time as /ta/ compared to younger adults. Furthermore, despite the absence of any between-group differences in phonetic discrimination abilities, older adults demonstrated longer N100/P200 latencies as well as increased P200 amplitudes while processing the consonant-vowel syllables varying in voice-onset time. Finally, older adults also exhibited a divergent anatomical gray matter infrastructure in bilateral auditory-related and frontal brain regions, as manifested in reduced cortical thickness and surface area. Notably, in the younger adults but not in the older adult cohort, cortical surface area in these two gross anatomical clusters correlated with the categorization of consonant-vowel syllables characterized by a short voice-onset time, suggesting the existence of a critical gray matter threshold that is crucial for consistent mapping of phonetic categories varying along the temporal dimension. Taken together, our results highlight the multifaceted dimensions of age-related temporal speech processing characteristics, and pave the way toward a better understanding of the relationships between hearing, speech and the brain in older age.

Abstract

Fusion performance in tokamaks hinges critically on the efficacy of the Edge Transport Barrier (ETB) at suppressing energy losses. The new concept of fingerprints is introduced to identify the instabilities that cause the transport losses in the ETB of many of today's experiments, from widely posited candidates. Analysis of the Gyrokinetic-Maxwell equations, and gyrokinetic simulations of experiments, find that each mode type produces characteristic ratios of transport in the various channels: density, heat and impurities. This, together with experimental observations of transport in some channel, or, of the relative size of the driving sources of channels, can identify or determine the dominant modes causing energy transport. In multiple ELMy H-mode cases that are examined, these fingerprints indicate that MHD-like modes are apparently not the dominant agent of energy transport; rather, this role is played by Micro-Tearing Modes (MTM) and Electron Temperature Gradient (ETG) modes, and in addition, possibly Ion Temperature Gradient (ITG)/Trapped Electron Modes (ITG/TEM) on JET. MHD-like modes may dominate the electron particle losses. Fluctuation frequency can also be an important means of identification, and is often closely related to the transport fingerprint. The analytical arguments unify and explain previously disparate experimental observations on multiple devices, including DIII-D, JET and ASDEX-U, and detailed simulations of two DIII-D ETBs also demonstrate and corroborate this.

This paper compares the gyrokinetic instabilities and transport in two representative JET pedestals, one (pulse 78697) from the JET configuration with a carbon wall (C) and another (pulse 92432) from after the installation of JET's ITER-like Wall (ILW). The discharges were selected for a comparison of JET-ILW and JET-C discharges with good confinement at high current (3 MA, corresponding also to low $\rho_*$) and retain the distinguishing features of JET-C and JET-ILW, notably, decreased pedestal top temperature for JET-ILW. A comparison of the profiles and heating power reveals a stark qualitative difference between the discharges: the JET-ILW pulse (92432) requires twice the heating power, at a gas rate of $1.9 \times 10^{22}e/s$, to sustain roughly half the temperature gradient of the JET-C pulse (78697), operated at zero gas rate. This points to heat transport as a central component of the dynamics limiting the JET-ILW pedestal and reinforces the following emerging JET-ILW pedestal transport paradigm, which is proposed for further examination by both theory and experiment. ILW conditions modify the density pedestal in ways that decrease the normalized pedestal density gradient $a/L_n$, often via an outward shift of the density pedestal. This is attributable to some combination of direct metal wall effects and the need for increased fueling to mitigate tungsten contamination. The modification to the density profile increases $\eta = L_n/L_T$ , thereby producing more robust ion temperature gradient (ITG) and electron temperature gradient driven instability. The decreased pedestal gradients for JET-ILW (92432) also result in a strongly reduced $E \times B$ shear rate, further enhancing the ion scale turbulence. Collectively, these effects limit the pedestal temperature and demand more heating power to achieve good pedestal performance.

Abstract

Strained cyclic organic molecules, such as arynes, cyclic alkynes and cyclic allenes, have intrigued chemists for more than a century with their unusual structures and high chemical reactivity1. The considerable ring strain (30-50 kilocalories per mole)2,3 that characterizes these transient intermediates imparts high reactivity in many reactions, including cycloadditions and nucleophilic trappings, often generating structurally complex products4. Although strategies to control absolute stereochemistry in these reactions have been reported using stoichiometric chiral reagents5,6, catalytic asymmetric variants to generate enantioenriched products have remained difficult to achieve. Here we report the interception of racemic cyclic allene intermediates in a catalytic asymmetric reaction and provide evidence for two distinct mechanisms that control absolute stereochemistry in such transformations: kinetic differentiation of allene enantiomers and desymmetrization of intermediate π-allylnickel complexes. Computational studies implicate a catalytic mechanism involving initial kinetic differentiation of the cyclic allene enantiomers through stereoselective olefin insertion, loss of the resultant stereochemical information, and subsequent introduction of absolute stereochemistry through desymmetrization of an intermediate π-allylnickel complex. These results reveal reactivity that is available to cyclic allenes beyond the traditional cycloadditions and nucleophilic trappings previously reported, thus expanding the types of product accessible from this class of intermediates. Additionally, our computational studies suggest two potential strategies for stereocontrol in reactions of cyclic allenes. Combined, these results lay the foundation for the development of catalytic asymmetric reactions involving these classically avoided strained intermediates.

Abstract

Tokamak à configuration variable (TCV), recently celebrating 30 years of near-continual operation, continues in its missions to advance outstanding key physics and operational scenario issues for ITER and the design of future power plants such as DEMO. The main machine heating systems and operational changes are first described. Then follow five sections: plasma scenarios. ITER Base-Line (IBL) discharges, triangularity studies together with X3 heating and N2 seeding. Edge localised mode suppression, with a high radiation region near the X-point is reported with N _2 injection with and without divertor baffles in a snowflake configuration. Negative triangularity (NT) discharges attained record, albeit transient, β _N ∼ 3 with lower turbulence, higher low-Z impurity transport, vertical stability and density limits and core transport better than the IBL. Positive triangularity L-Mode linear and saturated ohmic confinement confinement saturation, often-correlated with intrinsic toroidal rotation reversals, was probed for D, H and He working gases. H-mode confinement and pedestal studies were extended to low collisionality with electron cyclotron heating obtaining steady state electron iternal transport barrier with neutral beam heating (NBH), and NBH driven H-mode configurations with off-axis co-electron cyclotron current drive. Fast particle physics. The physics of disruptions, runaway electrons and fast ions (FIs) was developed using near-full current conversion at disruption with recombination thresholds characterised for impurity species (Ne, Ar, Kr). Different flushing gases (D2, H2) and pathways to trigger a benign disruption were explored. The 55 kV NBH II generated a rich Alfvénic spectrum modulating the FI fas ion loss detector signal. NT configurations showed less toroidal Alfvén excitation activity preferentially affecting higher FI pitch angles. Scrape-off layer and edge physics. gas puff imaging systems characterised turbulent plasma ejection for several advanced divertor configurations, including NT. Combined diagnostic array divertor state analysis in detachment conditions was compared to modelling revealing an importance for molecular processes. Divertor physics. Internal gas baffles diversified to include shorter/longer structures on the high and/or low field side to probe compressive efficiency. Divertor studies concentrated upon mitigating target power, facilitating detachment and increasing the radiated power fraction employing alternative divertor geometries, optimised X-point radiator regimes and long-legged configurations. Smaller-than-expected improvements with total flux expansion were better modelled when including parallel flows. Peak outer target heat flux reduction was achieved (>50%) for high flux-expansion geometries, maintaining core performance ( H _98 > 1). A reduction in target heat loads and facilitated detachment access at lower core densities is reported. Real-time control. TCV’s real-time control upgrades employed MIMO gas injector control of stable, robust, partial detachment and plasma β feedback control avoiding neoclassical tearing modes with plasma confinement changes. Machine-learning enhancements include trajectory tracking disruption proximity and avoidance as well as a first-of-its-kind reinforcement learning-based controller for the plasma equilibrium trained entirely on a free-boundary simulator. Finally, a short description of TCV’s immediate future plans will be given.

Abstract

Within the 9th European Framework programme, since 2021 EUROfusion is operating five tokamaks under the auspices of a single Task Force called ‘Tokamak Exploitation’. The goal is to benefit from the complementary capabilities of each machine in a coordinated way and help in developing a scientific output scalable to future largre machines. The programme of this Task Force ensures that ASDEX Upgrade, MAST-U, TCV, WEST and JET (since 2022) work together to achieve the objectives of Missions 1 and 2 of the EUROfusion Roadmap: i) demonstrate plasma scenarios that increase the success margin of ITER and satisfy the requirements of DEMO and, ii) demonstrate an integrated approach that can handle the large power leaving ITER and DEMO plasmas. The Tokamak Exploitation task force has therefore organized experiments on these two missions with the goal to strengthen the physics and operational basis for the ITER baseline scenario and for exploiting the recent plasma exhaust enhancements in all four devices (PEX: Plasma EXhaust) for exploring the solution for handling heat and particle exhaust in ITER and develop the conceptual solutions for DEMO. The ITER Baseline scenario has been developed in a similar way in ASDEX Upgrade, TCV and JET. Key risks for ITER such as disruptions and run-aways have been also investigated in TCV, ASDEX Upgrade and JET. Experiments have explored successfully different divertor configurations (standard, super-X, snowflakes) in MAST-U and TCV and studied tungsten melting in WEST and ASDEX Upgrade. The input from the smaller devices to JET has also been proven successful to set-up novel control schemes on disruption avoidance and detachment.

Abstract

In 2021 JET exploited its unique capabilities to operate with T and D–T fuel with an ITER-like Be/W wall (JET-ILW). This second major JET D–T campaign (DTE2), after DTE1 in 1997, represented the culmination of a series of JET enhancements—new fusion diagnostics, new T injection capabilities, refurbishment of the T plant, increased auxiliary heating, in-vessel calibration of 14 MeV neutron yield monitors—as well as significant advances in plasma theory and modelling in the fusion community. DTE2 was complemented by a sequence of isotope physics campaigns encompassing operation in pure tritium at high T-NBI power. Carefully conducted for safe operation with tritium, the new T and D–T experiments used 1 kg of T (vs 100 g in DTE1), yielding the most fusion reactor relevant D–T plasmas to date and expanding our understanding of isotopes and D–T mixture physics. Furthermore, since the JET T and DTE2 campaigns occurred almost 25 years after the last major D–T tokamak experiment, it was also a strategic goal of the European fusion programme to refresh operational experience of a nuclear tokamak to prepare staff for ITER operation. The key physics results of the JET T and DTE2 experiments, carried out within the EUROfusion JET1 work package, are reported in this paper. Progress in the technological exploitation of JET D–T operations, development and validation of nuclear codes, neutronic tools and techniques for ITER operations carried out by EUROfusion (started within the Horizon 2020 Framework Programme and continuing under the Horizon Europe FP) are reported in (Litaudon et al Nucl. Fusion accepted), while JET experience on T and D–T operations is presented in (King et al Nucl. Fusion submitted).

Abstract

Recent results from MAST Upgrade are presented, emphasising understanding the capabilities of this new device and deepening understanding of key physics issues for the operation of ITER and the design of future fusion power plants. The impact of MHD instabilities on fast ion confinement have been studied, including the first observation of fast ion losses correlated with Compressional and Global Alfvén Eigenmodes. High-performance plasma scenarios have been developed by tailoring the early plasma current ramp phase to avoid internal reconnection events, resulting in a more monotonic q profile with low central shear. The impact of m / n = 3/2, 2/1 and 1/1 modes on thermal plasma confinement and rotation profiles has been quantified, and scenarios optimised to avoid them have transiently reached values of normalised beta approaching 4.2. In pedestal and ELM physics, a maximum pedestal top temperature of ∼350 eV has been achieved, exceeding the value achieved on MAST at similar heating power. Mitigation of type-I ELMs with n = 1 RMPs has been observed. Studies of plasma exhaust have concentrated on comparing conventional and Super-X divertor configurations, while X-point target, X-divertor and snowflake configurations have been developed and studied in parallel. In L-mode discharges, the separatrix density required to detach the outer divertors is approximately a factor 2 lower in the Super-X than the conventional configuration, in agreement with simulations. Detailed analysis of spectroscopy data from studies of the Super-X configuration reveal the importance of including plasma-molecule interactions and D _2 Fulcher band emission to properly quantify the rates of ionisation, plasma-molecule interactions and volumetric recombination processes governing divertor detachment. In H-mode with conventional and Super-X configurations, the outer divertors are attached in the former and detached in the latter with no impact on core or pedestal confinement.

Abstract

JET, the world’s largest operating tokamak with unique Be/W wall and tritium handling capability, completed a Deuterium-Tritium (D-T) campaign in 2021 (Maggi et al 29th Fusion Energy Conf. ) following a decade of preparatory experiments, dedicated enhancements, technical rehearsals and training (Horton et al 2016 Fusion Eng. Des. 109–111 925). Operation with tritium raises significant technical, safety and scientific challenges not encountered in standard protium or deuterium operation. This contribution describes the tritium operational requirements, pulses and technical preparations, new operating procedures, lessons learned and details on the achieved operational availability and performance. The preparation and execution of the recent JET tritium experiments benefitted from the previous experience in 1991 (Preliminary Tritium Experiment), 1997 (DTE1 campaign) and 2003 (Trace Tritium Campaigns) and consisted of the following five phases: technical rehearsals and scenario preparation, tritium commissioning, 100% tritium campaign, D-T campaign (DTE2), tritium clean-up. Following the clean-up JET resumed normal operation and is currently undertaking a further D-T campaign (DTE3).

Abstract

This paper reports the first experiment carried out in deuterium–tritium addressing the integration of a radiative divertor for heat-load control with good confinement. Neon seeding was carried out for the first time in a D–T plasma as part of the second D–T campaign of JET with its Be/W wall environment. The technical difficulties linked to the re-ionisation heat load are reported in T and D–T. This paper compares the impact of neon seeding on D–T plasmas and their D counterpart on the divertor detachment, localisation of the radiation, scrape-off profiles, pedestal structure, edge localised modes and global confinement.
