5. In Vitro and In Vivo Biocompatibility of Bacterial Cellulose.

Abstract

Bacterial cellulose is a unique biomaterial produced by various species of bacteria that offers a range of potential applications in the biomedical field. To provide a cost‐effective alternative to soft‐tissue implants used in cavity infills, remodeling, and subdermal wound healing, in vitro cytotoxicity and in vivo biocompatibility of native bacterial cellulose were investigated. Cytotoxicity was assessed using a metabolic assay on Swiss 3T3 fibroblasts and INS‐1832/13 rat insulinoma. Results showed no cytotoxicity, whether the cells were seeded over or under the bacterial cellulose scaffolds. Biocompatibility was performed on Sprague–Dawley rats (males and females, 8 weeks old) by implanting bacterial cellulose membranes subcutaneously for 1 or 12 weeks. The explanted scaffolds were then sliced and stained with hematoxylin and eosin for histological characterization. The first series of results revealed acute and chronic inflammation persisting over 12 weeks. Examination of the explants indicated a high number of granulocytes within the periphery of the bacterial cellulose, suggesting the presence of endotoxins within the membrane, confirmed by a Limulus amebocyte lysate test. This discovery motivated the development of non‐pyrogenic bacterial cellulose scaffolds. Following this, a second series of animal experiments was done, in which materials were implanted for 1 or 2 weeks. The results revealed mild inflammation 1 week after implantation, which then diminished to minimal inflammation after 2 weeks. Altogether, this study highlights that unmodified, purified native bacterial cellulose membranes may be used as a cost‐effective biomedical device provided that proper endotoxin clearance is achieved. Abstract

La cellulose bactérienne (CB) est un biomatériau unique produit par diverses espèces de bactéries qui s’adapte à une gamme d'applications potentielles dans le domaine biomédical. En particulier, cet ouvrage explore la cellulose bactérienne en tant qu’alternative économique aux implants de tissus mous utilisés pour combler les cavités, le remodelage et la guérison des plaies sous-cutanées. Pour se faire, la cytotoxicité in vitro et la biocompatibilité in vivo de la cellulose bactérienne native ont été étudiées. La cytotoxicité a été évaluée à l'aide d'un test métabolique sur des fibroblastes Swiss 3T3 et des insulinomes de rat INS-1 832/13. Les résultats ont montré l'absence de cytotoxicité, que les cellules aient été ensemencées sur ou sous les membranes de cellulose bactérienne. La biocompatibilité a été testée sur des rats Sprague-Dawley (mâles et femelles, âgés de 8 semaines) en implantant des membranes de cellulose bactérienne en sous-cutané pendant 1 ou 12 semaines. Les échafaudages explantés ont ensuite été coupés et colorés à l'H&E pour une caractérisation histologique. La première série de résultats a révélé une inflammation aiguë et chronique persistant sur 12 semaines. L'analyse des échantillons a indiqué un grand nombre de granulocytes dans la périphérie de la cellulose bactérienne, suggérant la présence d'endotoxines dans la membrane, confirmée par un test au lysat d'amébocytes de limule. Cette découverte a motivé le développement d'échafaudages de cellulose bactérienne non pyrogéniques. Suite à cela, une deuxième série d'expériences sur des animaux a été réalisée, au cours de laquelle les matériaux ont été implantés pendant 1 ou 2 semaines. Les résultats ont révélé une légère inflammation une semaine après l'implantation, qui a ensuite diminué pour devenir une inflammation minimale après deux semaines. Abstract

The origin of the diatoms still remains enigmatic. Their fossil record is scarce until the Late Cretaceous and great divergences exist between molecular data and the earliest fossil evidence. While molecular data indicate an origin during the Triassic or Early Jurassic, early fossil evidence is only from the Late Jurassic-Early Cretaceous. The discovery of diatoms in French mid-Cretaceous amber by the end of the 2000s already suggested a potential bias in the diatom fossil record as it made older many diatom lineages, the record of which hitherto began at the end of the Cretaceous. The Jurassic/Early Cretaceous fossil record of diatoms is extremely sparse and any new occurrence is important for retracing the evolutionary, palaeogeographical and palaeoenvironmental history of diatoms. Thai amber has yielded a new diatom specimen that has been attributed to the genus Hemiaulus. Fossil assemblages and sedimentological data indicate that Thai amber and its Hemiaulus specimen are Late Jurassic in age. This discovery represents the oldest hitherto known specimen of Hemiaulus and so extends the fossil record of the bipolar diatoms and of the genus Hemiaulus by several dozens of millions of years and brings closer the fossil evidence and molecular data (that estimated an origin of the bipolar diatoms about 150 Ma ago). It reinforces the hypothesis of a pre-Cretaceous fossil diatom records and also supports an origin of the diatoms in shallow coastal environments.

Abstract

La découverte de trois dépôts en 2011 dans la plaine alluviale de l’Oise à Ribécourt-Dreslincourt (Oise), lors d’un diagnostic suivi d’une fouille, a permis d’apporter un nouvel éclairage sur la pratique des dépôts non funéraires. Enfouis à 5 m environ les uns des autres, l’un (F29) est constitué d’un objet unique, à savoir une hache enfouie verticalement, tandis que les deux autres (F28 et F36) ont livré à la fois des parures, des outils et des armes. Dans deux des dépôts (F28 et F29), tous les objets sont en alliage cuivreux, mais le troisième a aussi livré de l’ambre, de l’or et du verre. Tous trois sont attribués au Bronze moyen atlantique 2 d’après la typologie des objets et une datation radiocarbone.L’état d’enfouissement des objets est variable : tous sont complets, mais cinq ont été intentionnellement manipulés avant d’être déposés. Certains semblent avoir peu ou pas servi, tandis que d’autres ont clairement été portés ou employés. On notera aussi la présence étonnante de ratés de fonte. Ces dépôts matérialisent la circulation d’individus, d’objets et d’idées entre le sud de l’Angleterre et le nord de la France, mais aussi avec d’autres régions plus éloignées (Allemagne, Suisse, Baltique, Proche-Orient).Fait particulièrement original et rarement observé jusqu’à présent : un poteau était associé à chacun des dépôts. Érigés comme des marqueurs spatiaux, ils indiquent que ces dépôts n’ont pas nécessairement tous été enterrés pour être cachés et qu’il a pu exister une volonté de les mettre en évidence.Dans deux des trois dépôts (F28 et F36) se distinguent deux lots d’objets séparés par une couche de sédiments de plusieurs centimètres vide de mobilier. Des matériaux organiques pouvaient s’y trouver initialement (textiles, fourrures…). Néanmoins, combiné à la présence des poteaux, ce niveau stérile soulève la question de réouvertures des dépôts pour les compléter, y prélever des objets ou intervenir sur ces derniers. On parlerait alors de dépôts polyphasés, qui pourraient dès leur origine être prévus pour être rouverts. Ces observations viennent alimenter les discussions autour de cette pratique des dépôts non funéraires et participent à en renouveler la lecture.

Abstract

International audience; Cretaceous ambers have been discovered in France since the beginning of the 18th century. The best known are those from south-western France which are Late Albian-Early Cenomanian in age, but there are other important amber deposits in other regions. Here, we summarise the data on one of these other Cretaceous amber regions, the Sarthe Department. These deposits have been mentioned in the literature since the end of the 18th century, but they have remained relatively unknown. The material, that has been studied during the 1970's and 1980's, yielded a well-diversified arthropod fauna (72 arthropod specimens, including arachnids, cockroaches, bugs, beetles, flies, wasps...) dated to late Early-Middle Cenomanian. In the last decade, 4 types of bacteria, a possible testate amoeba and fungal remains were also found. A re-examination of the historical collections of the Sarthe amber, housed in the "Musée Vert" (Le Mans, France), allows to estimate the geographical extent of the amber deposits in the Sarthe Department. The study of the microfossils of these samples provides new data on their palaeoenvironment.

Abstract

During pathogenesis, the ascomycete Botrytis cinerea secretes a range of cell-wall-degrading enzymes such as polygalacturonases, glucanases and proteases. We report the identification of a new member of the G1 family of proteases, BcACP1, which is secreted by B. cinerea during infection. The production of BcACP1 correlates with the acidification of the plant tissue, and transcriptional analysis of the Bcacp1 gene showed that it is only expressed under acidic growth conditions. Using a transcriptional reporter system, we showed that pH regulation of Bcacp1 is not mediated by the canonical PacC transcription factor binding site. Like other G1 proteases, BcACP1 is produced as a pro-enzyme. Trapping of the zymogen form allowed investigation of its maturation process. Evidence is presented for an autocatalytic proteolysis of the enzyme that is triggered by acidic pH. Environmental pH therefore controls Bcacp1 production at both the transcriptional and post-translational level.

46. Exceptional preservation of marine diatoms in upper Albian amber

Abstract

Late Albian amber from Charente-Maritime (southwestern France) contains the first known marine diatoms preserved in a fossil resin. Approximately 70 inclusions were assignable to the genera Basilicostephanus, Coscinodiscus, Hemiaulus, Melosira, Paralia, Skeletonema, Stephanopyxis, Trochosira, ?Aulacoseira, and to the order Rhizosoleniales. Some of them are represented by several species. This diatom assemblage is mainly composed of colonial planktonic genera, which are typical for coastal shallow waters. The newly found amber inclusions extend the fossil record of four genera and one order from the Late Cretaceous and support certain molecular phylogenetic assumptions regarding the diversification of marine diatoms in the Early Cretaceous. The unusual introduction of diatom shells from the beach or sea by wind, spray, or high tide onto the resin flows was possible because the amber forest grew close to the seashore.
