A great amount of knowledge on natural killer T cells (iNKTs) is now available, but a consensus about their activation mechanisms has not been reached. These cells recognize different glycolipid antigens through the CD1d molecule. Such antigens may be endogenous, derived from bacteria (foreign) and synthetic, the latter have been developed for clinical applications. There exists much interest in understanding how these different glycolipid compounds induce different types of polarization, but it has been difficult to reach a consensus due to the fact that responses depend on different factors such as: the nature of the molecule, the internalization process and the presentation of the glycolipids. Moreover, activation of iNKT cells is determined by the type and state of the antigen presenting cell, the co-stimulatory molecules, the transactivation mechanisms and the location of the glycolipid-CD1d complexes on the plasma membrane, such as the lipid rafts. This review explores the evidence about the factors that affect activation of iNKT cells in order to understand their immune-modulatory potential.

Introduction: Thyroid nodules are commonly found, and their incidence is increasing. The main objective during the evaluation of these nodules is to determine if they are benign or malignant. Materials and methods: We made a narrative review of the literature by searching Ovid, Medline and Lilacs from 1950 to 2010. Results: Most thyroid nodules are benign, but in 4% to 8% of cases they may be malignant, hence the importance of appropriate study and management. Nodules larger than 1 cm should undergo fine needle aspirate; however, the clinical features of the patient and the characteristics of the nodule on ultrasound assessment should always be taken into account. The following are risk factors associated with the presence of thyroid nodules: age over 45 years, female gender and smoking. The measurement of tumor markers in the initial study of patients with thyroid nodule is not recommended.Conclusion: Ultrasound and thyroid fine needle aspirate are the most important diagnostic methods in the evaluation of thyroid nodules, because they enable to make changes in therapeutic decisions thus decreasing the number of unnecessary thyroidectomies. Treatment and prognosis of thyroid nodules will depend on the patient´s risk factors and the cytological and pathological findings.

Se desarrolló un protocolo de propagación in vitro a partir de meristemos florales para Heliconia bihai cv. Lobster Salmón. Para el efecto se establecieron 250 yemas florales en diferentes tiempos utilizando NaClO al 1% durante 20 min. El medio de establecimiento Murashige y Skoog con mejores resultados fue BAP (2 mg/lt), AIA (1 mg/lt) y 0.1 g/lt de L-cisteína. La mayor produccion de brotes en multiplicación se obtuvo con 6 mg/lt de BAP y con un subcultivo posterior en medio con 2 mg/lt de BAP y 0.5 mg/lt de AIA, alcanzando una tasa de multiplicación de tres nuevas yemas. El enraizamiento de las plántulas in vitro se logró con 1.3 mg/lt de AIA. La aclimatación en vivero fue 92% de supervivencia. Se realizó un análisis de varianza (Anova) y un test de Tukey para las variables evaluadas las cuales mostraron diferencias estadísticas en cada fase. La propagación in vitro de heliconias a partir de yemas florales requiere mayor tiempo en la fase de establecimiento y multiplicación, no obstante permite mayores porcentajes de supervivencia y una menor contaminación, ofreciendo material de siembra de diferentes especies de esta planta de interés comercial.

10. Role of gene interactions in the pathophysiology of skeletal dysplasias: A case report in Colombia.

Genome association studies have shown that gene-gene interactions or epistasis play a crucial role in identifying the etiology, prognosis, and treatment response of many complex diseases beyond their main effects. Skeletal dysplasias are a heterogeneous group of congenital bone and cartilage disorders with a genetic and gen-gen interaction etiology. The current classification of skeletal dysplasias distinguishes 461 diseases in 42 groups, and the incidence of all skeletal dysplasias is more than 1 in every 5000 newborns. The objective is to present the case of a patient with four variants that generates gen-gen interactions in the skeletal dysplasia. A 1-year-old male patient was diagnosed with skeletal dysplasia based on prenatal ultrasound showing micromelia and pyelocalyceal dilation. Postnatal physical examination revealed body disproportion and involvement of other organs and systems. A sequencing study and deletions/duplications analysis were performed for 358 candidate genes associated with skeletal dysplasia. The GeneMANIA interface was used to evaluate the expression network of genes associated with each other for the gen-gen interaction. Four pathogenic variants were obtained two heterozygous variants with pathogenic significance in SLC26A , one heterozygous pathogenic variant in CLCN7 and another heterozygous pathogenic variant in CEP120. The GeneMANIA interface reveals 77.64% physical interactions, 8.01% co-expression, 5.37% prediction, 3.63% co-localization, 2.87% genetic interactions, 1.88% route of action, and 0.60% shared protein domains. These results suggest that the interaction between these genes affects the activity of the inorganic anion exchanger, leading to disorganization of collagen fibers, early mineralization, and decreased assembly of fibronectin in the bone extracellular matrix. Identifying gene-gene interactions is a fundamental step in understanding proper cell function and thus understanding the pathophysiology of many complex human diseases, improving diagnosis, and the possibilities of new personalized therapies. 12. The lignocellulolytic effect from newly wild white rot fungi isolated from Colombia

- Abstract
Lignocellulose is the most abundant natural polymer on earth. It is the main agro-industrial waste component, it has no added value, and in some cases is the main cause of environmental problems. The fungi can be used in the transformation of residues and different biotechnological processes. In the present research, seven strains of white rot fungi collected in Ibagué (Colombia) were isolated. The production of ligninolytic and cellulolytic enzymes as well as their capacity to transform lignocellulose from banana peel, rice husk and cocoa pod husk were evaluated. Strains of Pycnoporus sanguineus, Hexagonia glabraand Lentinus crinituswere selected from the enzymatic screening and showed high production of ligninolytic enzymes. The evaluation of agricultural waste degradation was carried out by Fourier-transform infrared spectroscopy and the results showed the ability of the tested fungi to, preferentially, degrade lignin or cellulose in each substrate. Based on the results, different biotechnological potentialities for each strain were proposed.

Tomato is considered a fruit with a high food demand across the world due to its high antioxidant capacity. Lycopene content in tomato has been widely studied, particularly in cherry-type tomatoes which are considered a high source of lycopene for humans. The aim of this study was to analyze the differences in terms of concentration and variation of diverse physicochemical parameters of quality (acidity, vitamin C, pH, °Brix, color coordinates L*, a* and b*) and, antioxidants like ß-carotene and lycopene in 11 accessions of cherry-type tomatoes, considering the advantages of antioxidants for human health. Multicriteria analysis and other tests showed substantial differences among the accessions. IAC401 was associated with the best physicochemical characteristics of a promissory cherry-type tomato for cultivation due to its association with functional food, followed by IAC426R and IAC1624. This study allowed an innovative association of physicochemical characteristics and antioxidant contents of cherry-type tomatoes through multicriteria analyses which enable the analysis, elimination, and proposal of promising accessions for their use in future crops which will promote the best intrinsic features of tomato to be used in the studied region. In this case accessions with the highest scores offer not only optimum amounts of antioxidants and highest levels of organic acids and sugars. [ABSTRACT FROM AUTHOR]

Objetivo. Determinar la estructura trófica del ensamblaje de insectos acuáticos asociados a ocho quebradas de la ecorregión cafetera colombiana. Materiales y métodos. Se colectaron insectos acuáticos en ocho quebradas protegidas por corredores de bosque en la cuenca del río La Vieja. Los taxa encontrados fueron asignados a grupos dietarios con base en una clasificación regional, generada a partir de la revisión del contenido estomacal de los insectos, en quebradas boscosas de la cuenca del río Otún. Resultados. En total se colectaron 2019 individuos distribuidos en 73 grupos taxonómicos, de los cuales 60 fueron clasificados en grupos dietarios. Los colectores fueron el grupo más abundante (55%), seguido por los fragmentadores (31%) y los depredadores (10%). Los raspadores sólo representaron el 0,05% de la muestra y el 3,95% restante no pudo ser clasificado por falta de información. Conclusiones. La dominancia de colectores y fragmentadores refleja la importancia de la materia orgánica particulada gruesa (hojarasca), como recurso alimentario para la entomofauna. La similitud de la estructura trófica entre esta comunidad y otras estudiadas en quebradas similares, sugiere la posibilidad de un patrón común para las quebradas andinas. Se evidenció la falta de información sobre ecología trófica de insectos acuáticos tropicales; 50% de los taxa colectados no presentaron reportes de este tipo en el trópico y para el 20% no existe información en el trópico ni en la zona templada. Objective. To determine the trophic structure of the aquatic insect assembly associated to eight streams in the Colombian coffee-growing ecoregion. Materials and methods. Aquatic insects were collected in eight forested streams located in La Vieja river basin. The taxa collected were assigned to dietary groups according to a regional classification based on the gut content analysis of aquatic insects associated to forested streams of the Otún river basin. Results. 2019 individuals belonging to 73 taxa were collected and 60 were classified into dietary groups. The most abundant group was collectors (55%), followed by shredders (31%) and predators (10%). Scrapers represented only 0.05% of the sample and the remaining 3,95% could not be classified due to lack of information. Conclusions. The dominance of collectors and shredders reveals the importance of coarse particulate organic matter (leaf litter) as a food resource for the insect fauna. Similarities between the trophic structure of this community and other communities studied in similar streams, suggest the possibility of a common pattern for Andean streams. This study evidenced the lack of knowledge on trophic ecology of tropical aquatic insects; 50% of the taxa collected did not have this kind of information for the tropics and 20% had no information neither for the tropics nor temperate zones. Objetivo. Determinar a estrutura trófica da assembléia de insetos aquáticos associados a oito córregos da ecorregião cafetera colombiana. Materiais e métodos. Foram coletados insetos aquáticos em oito córregos protegidos por corredores florestais na bacia do rio La Vieja. Os taxa encontrados foram designados a grupos de dieta baseados numa classificação regional, gerada a partir da análise do conteúdo estomacal dos insetos, em córregos da bacia do rio Otún. Resultados. Foram coletados 2.019 indivíduos pertencentes a 73 grupos taxonômicos, dos quais 60 foram classificados em grupos alimentares. Os coletores foram o grupo mais abundante (55%), seguido por fragmentadores (31%) e predadores (10%). Os raspadores representaram apenas 0,05% da amostra e os restantes 3,95% não puderam ser classificados devido à falta de informação. Conclusões. O predomínio de coletores e fragmentadores reflete a importância da matéria orgânica particulada grossa (serrapilheira), como recurso alimentar para a fauna de insetos. A semelhança entre a estrutura trófica desta comunidade e outras estudadas em córregos similares, sugerem a possibilidade de um padrão comum para córregos andinos. Este estudo revelou a falta de informação sobre a ecologia alimentar de insetos aquáticos tropicais, 50% dos taxa coletados não apresentaram nenhum registro deste tipo no trópico, e para o 20% não há informação nem nos trópicos nem na zona temperada.

19. Effect of restoration of riparian corridors on the biotic and abiotic characteristics of streams in cattle ranching areas of La Vieja river catchment in Colombia

Con el fin de determinar el efecto de actividades de restauración sobre las características de quebradas en la cuenca del río La Vieja, Colombia, se evaluó la calidad del agua, las características del hábitat y el estado de los macroinvertebrados acuáticos en cuatro de estos ambientes. En cada quebrada se seleccionó un tramo de 100 m que se evaluó en el momento de hacer la intervención y en los dos años siguientes. Se notó una recuperación inicial de la vegetación nativa en el área ribereña y se disminuyó la turbiedad y la demanda bioquímica de oxígeno en el agua. Se incrementaron características deseables en las quebradas como los sustratos gruesos y las piscinas. En cuanto a los macroinvertebrados, los taxa dominantes fueron Mollusca (Basommatophora, Sphaeriidae) y Diptera. Hubo una recuperación en el porcentaje de Ephemeroptera, Plecoptera y Trichoptera, grupos considerados sensibles a la perturbación del hábitat. Los resultados obtenidos demuestran que el aislamiento de las quebradas permite el inicio de la recuperación de las características del hábitat y los macroinvertebrados, pero para detectar cambios notables es necesario hacer evaluaciones a largo plazo y estimular el crecimiento de árboles y arbustos que compitan con las gramíneas exóticas presentes en la zona. Four streams of La Vieja river catchment, Colombia, were monitored in order to evaluate the effect of restoration activities on the habitat characteristics, water quality, and macroinvertebrates. A 100 m reach in each stream was selected and monitored previous to the intervention and for the next two years. An initial recovery of the native vegetation in the riparian area was noticed. Turbidity and biochemical oxygen demand were reduced and desirable habitat characteristics, such as coarse substrates and pools were increased. Predominant macroinvertebrate taxa were Mollusca (Basommatophora, Sphaeriidae) and Diptera, but Ephemeroptera, Plecoptera, and Trichoptera, groups considered sensible to perturbation, increased slightly through the years. The results demonstrate that the isolation of streams stimulates the restoration of the aquatic habitat and macroinvertebrates, but a longer period of evaluation is needed to detect more remarkable changes. Additional actions are needed to allow the growth of trees that compete with the present exotic pasture species.

20. Cytotoxic activity of ethanolic extracts of a selection of macromycetes

AbstractMacromycetes are an important factory of bioactive compounds, which have shown to have therapeutic use given their specific cytotoxicity, turning them into new and natural source of pharmaceuticals for treatments against several diseases. The purpose of this paper was to collect new data on the chemical and biological knowledge of the mycobiota that grows wild in the city of Ibagué (Colombia), in order to try to understand the cytotoxicity of these organisms. Based on abundance and chemotaxonomic criteria and in order to evaluate their biological activity, basidiomes of the following species were collected: Pycnoporus sanguineus, Phellinus gilvus, Pleurotus tubarius, Lenzites elegansand Ganoderma australe. An ethanolic maceration of basidiomes was used as base to perform cytogenotoxicity tests on Allium cepa. The extracts of P. sanguineusand G. australeinhibited A. cepamitosis in 58.92 % at 8000 mg l–1and 53.81% at 2000 mg l–1, respectively. The extracts of the studied species have shown capacity to cause some genetic changes associated to the concentrations evaluated. This study shows that there is an important potential in wild macromycete biodiversity in the region of Tolima (Colombia), making them potential candidates for future research on clinical essays for the pharmaceutical industry.

- Abstract
AbstractDownregulation of the expression of enzymes involved in lignin biosynthesis in alfalfa (Medicago sativa L.) has been shown to result in reduced cell wall recalcitrance and increased saccharification. Here we describe changes in the extractability of hemicellulosic and pectic polysaccharides in cell walls of alfalfa downregulated in the expression of hydroxycinnamoyl CoA:shikimate hydroxycinnamoyl transferase (HCT). Xylan and pectic polysaccharides are more readily solubilized from the cell walls of HCT downregulated transgenic lines than from empty vector control plants, suggesting alterations in lignin-polysaccharide interactions in the transgenic plants. Our studies show that reduced recalcitrance of HCT downregulated lines is also correlated with increased xylan extractability.

- Abstract
Objetivo. Determinar la estructura trófica del ensamblaje de insectos acuáticos asociados a ocho quebradas de la ecorregión cafetera colombiana. Materiales y métodos. Se colectaron insectos acuáticos en ocho quebradas protegidas por corredores de bosque en la cuenca del río La Vieja. Los taxa encontrados fueron asignados a grupos dietarios con base en una clasificación regional, generada a partir de la revisión del contenido estomacal de los insectos, en quebradas boscosas de la cuenca del río Otún. Resultados. En total se colectaron 2019 individuos distribuidos en 73 grupos taxonómicos, de los cuales 60 fueron clasificados en grupos dietarios. Los colectores fueron el grupo más abundante (55%), seguido por los fragmentadores (31%) y los depredadores (10%). Los raspadores sólo representaron el 0,05% de la muestra y el 3,95% restante no pudo ser clasificado por falta de información. Conclusiones. La dominancia de colectores y fragmentadores refleja la importancia de la materia orgánica particulada gruesa (hojarasca), como recurso alimentario para la entomofauna. La similitud de la estructura trófica entre esta comunidad y otras estudiadas en quebradas similares, sugiere la posibilidad de un patrón común para las quebradas andinas. Se evidenció la falta de información sobre ecología trófica de insectos acuáticos tropicales; 50% de los taxa colectados no presentaron reportes de este tipo en el trópico y para el 20% no existe información en el trópico ni en la zona templada.
