Abstract

Twenty-two Assaf male lambs (29.2 ± 0.9 kg live weight and 89 ± 0.2 days of age), distributed in two experimental groups, were used to evaluate the use of either feed-grade conventional urea (Control diet; n = 11) or slow-release urea (SRU diet; n = 11) as sources of dietary nitrogen on animal performance, ruminal fermentation, blood acid-base status, plasmatic metabolic profile, and carcass and meat quality. Animals were housed individually and fed ad libitum. At the end of the fattening period (day 70), the animals were slaughtered to compare the fermentation patterns in ruminal digesta and to evaluate the carcass and meat characteristics. No statistically significant differences (p > 0.05) were observed between treatments in the dry matter intake, final live weight, average daily gain, and feed conversion rate. Regarding the ruminal fermentation parameters, the molar proportion of propionic acid was higher (p < 0.05) and that of butyric acid was lower (p < 0.05) with the SRU than with the Control diet. There were no significant differences (p > 0.05) between experimental treatments in the blood acid-base status and biochemical profile, except for the concentration of urea in plasma, which was significantly (p < 0. 05) greater in SRU than in Control lambs. No statistically significant differences were observed between treatments (p > 0.05) in the carcass and meat characteristics. In conclusion, the use of slow-release urea as a replacement for feed-grade conventional urea in the diet of Assaf fattening lambs, under the experimental conditions of this study, did not improve animal performance and increased the feeding costs.

Abstract

Specific IgA antibody has been shown to play an important role in resistance to gastrointestinal nematode (GIN) infections in sheep, particularly in Teladorsagia circumcincta parasitosis. In some breeds, negative associations have been shown between IgA levels and worm burden in experimentally infected sheep. In the present study, we have studied the relationship between IgA levels in naturally infected sheep (582 ewes in total; 193 younger than one year old and 389 older than one year old) and fecal egg count (FEC) in the Assaf, Castellana, and Churra breeds. ELISA assays were performed to measure IgA levels against the somatic antigen of T. circumcincta third larval stage (L3) and a 203-amino-acid fragment of the protein disulfide isomerase from the same GIN species. A multilevel random intercept model was developed to predict the infection risk according to age or breed. Spearman’s correlation rank was used for statistical analysis. The prediction model showed that breed was not an influential factor in this study, although the Assaf breed could be considered slightly more susceptible than the others. In addition, age affected the infection risk, with the young ewes more susceptible to infection than the adult groups, except for the Castellana breed, whose risk of infection was similar at all ages. The most significant positive association was found between FEC and IgA measured in the nasal secretions of young ewes using both antigens (Rho = 0.5; p = 0.00); the correlation of FEC with IgA in serum was moderately significant (Rho = 0.306; p = 0.00). Comparing both antigens, the protein disulfide isomerase antigen was less reactive than the somatic antigen from L3. In conclusion, under natural conditions, specific IgA against GIN was positively associated with FEC in sheep, with nasal secretions from young animals being the sample where this association is stronger, which, therefore, could be used as a marker of infection in further studies.

Abstract

The objective of the study was to evaluate the nutritional value of leaves from fve eucalyptus species (Eucalyptus astringens, E. leucoxylon, E. sideroxylon, E. lehmannii, E. camaldulensis), harvested in Tunisia (two locations) during the autumn, winter, and spring seasons. Chemical composition and digestibility (various in vitro procedures) and in vitro tannin biological activity were determined. The species E. lehmannii had the lowest neutral detergent fber (NDF) and lignin contents (252 and 92 g kg−1 dry matter (DM), respectively) and the highest nonfbrous carbohydrates content (553 g kg−1 DM), while the species E. leucoxylon and E. camaldulensis had the highest NDF concentrations (375 and 356 g kg−1 DM, respectively). Among the seasons, higher levels of NDF and lignin were observed in winter (349 and 129 g kg−1 DM, respectively). The species E. sideroxylon showed the highest gas production with polyethylene glycol addition and thus the highest biological tannin activity. The species E. lehmannii and E. camaldulensis had the highest metabolizable energy concentrations, with 6.20 and 6.07 MJ kg−1 DM, respectively. E. camaldulensis showed the highest (37.9%) and E. lehmannii the lowest (31.9%) organic matter digestibility. Foliage from eucalyptus trees are of limited energy and protein value as an alternative fodder for ruminants, with a substantial variability across species. No remarkable seasonal variations in chemical composition and digestibility of eucalyptus foliage were detected.

Abstract

Early feed restriction of lambs may program animals to achieve reduced feed efficiency traits as a consequence of permanent mitochondrial dysfunction. The hypothesis at the background of the present study is that dietary administration of L-Carnitine (a compound that promotes the activation and transportation of fatty acids into the mitochondria) during the fattening period of early feed restricted lambs can: (a) improve the biochemical profile of early feed restricted lambs, (b) improve feed efficiency, (c) modulate the ruminal and intestinal microbiota, and (d) induce changes in the gastrointestinal mucosa, including the immune status. Twenty-two newborn male Merino lambs were raised under natural conditions but separated from the dams for 9h daily to allow feed restriction during the suckling period. At weaning, lambs were assigned to a control group being fed ad libitum a complete pelleted diet during the fattening phase (CTRL, n=11), whereas the second group (CARN, n=11) received the same diet supplemented with 3g of L-Carnitine/kg diet. The results revealed that even though L-Carnitine was absorbed, feed efficiency was not modified by dietary L-Carnitine during the fattening period (residual feed intake, p > 0.05), whereas ruminal fermentation was improved [total short-chain fatty acids (SCFAs), 113 vs. 154mmol/l; p=0.036]. Moreover, a trend toward increased concentration of butyrate in the ileal content (0.568 vs. 1.194 mmol/100 ml SCFA; p = 0.074) was observed. Other effects, such as reduced heart weight, lower levels of markers related to muscle metabolism or damage, improved renal function, and increased ureagenesis, were detected in the CARN group. Limited changes in the microbiota were also detected. These findings suggest that L-Carnitine may improve ruminal fermentation parameters and maintain both the balance of gut microbiota and the health of the animals. Abstract

Important achievements have been made to improve feed efficiency of farm livestock, but molecular mechanisms determining feed utilization remain unclear. The objective of this work was to identify the reasons behind divergent feed efficiencies in fattening lambs. Twenty newborn male Assaf lambs were fed milk replacer (MR) ad libitum using an automatic feeder, and weaned progressively during the last 7 days of the artificial suckling period (50 days in total). Then, all the lambs were offered ad libitum a complete pelleted diet during the fattening phase. During the fattening period, lambs were weighed twice weekly and daily dry matter intake was measured. These data were used to estimate residual feed intake (RFI), residual weight gain (RWG), and residual intake and body weight gain (RIG). Animals were ranked according to RIG, and then assigned to two experimental groups (n = 10 per treatment): group EFFI including the most efficient lambs (with negative values for RIG) and group NEFFI with the least efficient lambs (positive values for RIG). The most relevant results revealed that divergent values for feed efficiency traits in these fattening lambs were not associated to differences in digestibility or ruminal fermentation parameters (P > 0.05). However, the NEFFI lambs presented lower concentration of glucose (95.4 vs. 88.2 mg/dL; P < 0.05) and cholesterol (62.8 vs. 55.1 mg/dL; P < 0.05) in the serum. These effects might be related to the lower gene expression of enzymes involved in key metabolic pathways in the liver (e.g., PDHA1OA, CYP3A4). Moreover, bone formation and growth in NEFFI lambs also might have been compromised by a lower expression of genes involved in protein synthesis, such as those encoding ribosomal proteins (e.g., RPL22L1), these results being aligned with the lower albumin concentration also observed in the serum of these animals (35.56 vs. 33.89 g/L; P < 0.05). Abstract

Deficient management of replacement animals in the farm during early developmental windows may promote adverse programming effects on reproductive traits and subsequent transmission to the next generation. In this sense, DNA methylation profiles allow researchers to decode epigenetic regulation mechanisms in mammals and identify novel candidate genes correlated with phenotype differences in both dams and offspring. Therefore, improving knowledge in the field of epigenetics and intergenerational effects caused by prenatal and postnatal early nutritional events (e.g., feed restriction) is crucial for refining strategies dedicated to animal breeding. In this study, we determined differences in the global blood methylation patterns, biochemical profile, and metabolome of ewe lambs (F1) born from either early feed restricted dams (F0-RES) or fed ad libitum (F0-ADL). Our data show that functional categories such as those related to cellular processes, phosphorylation, nervous system, immunity response, or reproductive function were enriched significantly in the F1-RES lambs due to differences in the methylation of genes in these categories. These F1-RES lambs did not show differences in feed efficiency during the replacement period but presented higher levels of insulin and triglycerides and reduced concentration of progesterone, whereas the metabolome profile demonstrated variations in the bile acid composition when compared with the F1-ADL lambs. Taken together, all these results suggest that intergenerational effects caused by early feed restriction of dams (F0) may persist in the F1 female lambs with negative consequences on genes involved in cellular processes and reproductive traits.

Abstract

Banana pseudo-stem (BPS), which is rich in fibre and polyphenols, is a potential functional ingredient for the food industry. In this study, BPS was added at concentrations of 1.5, 3.0, and 4.5 g/kg to a minced pork batter to evaluate its performance as a filler and to pork burger patties to evaluate its performance as a natural antioxidant. The effects of BPS were compared with those of carrageenan and ascorbate, which are a conventional binder and antioxidant, respectively. The performance of BPS was similar to that of carrageenan in terms of the cooking yield and texture of the cooked batter. BPS reduced the brightness of fresh patties and appeared to reduce oxidative discolouration during the frozen storage of raw patties. Moreover, BPS reduced the levels of thiobarbituric acid reactive substances (TBARS) during the refrigerated and frozen storage of cooked patties. A greater decrease in TBARS formation was observed with 4.5 g BPS/kg compared with 0.5 g sodium ascorbate/kg during refrigerated storage. In contrast to ascorbate, BPS promoted the presence of lipid-derived volatile compounds induced by thermal breakdown in the headspace of cooked patties. Nonetheless, this effect was reduced as the amount of BPS in the patties increased. In cooked minced meat products, BPS could increase cooking yields and lipid oxidative stability during storage and might result in a more intense flavour.

Abstract

Changes in the levels of pH, lactic acid, acetic acid, headspace volatile compounds, and spoilage odour intensity during a 35 day refrigerated storage period in two sets of a Balkan-style fresh sausage, a control sausage (CON) and a sausage containing an aqueous hop extract (HOP), packaged under a 20% CO2 and 80% N2 atmosphere were evaluated. Storage resulted in progressive sausage acidification and increased the levels of acetic acid, 1-methylbutanol, ethyl acetate, ethyl hexanoate, and 2-ethylhexanol; all of which are associated with anaerobic microbial metabolism under sugar restricted conditions. Storage decreased the levels of hexanal, heptanal, pentanol, and garlic-derived organosulfur compounds. Hop extract showed oxygen scavenging abilities, and decreased the levels of the volatile compounds derived from lipid auto-oxidation while contributing to the presence of specific terpene compounds. The use of hop extract did not improve the shelf life of sausages packed under anaerobic atmosphere. The spoilage odour appeared in both types of sausages on the 14th day, and it was considered strong from day 21 onwards.

Abstract

Bovine respiratory disease (BRD) is still a serious concern in feedlots, where it exerts a negative effect on farm productivity. There is a shortage of studies focused on the evaluation of BRD-associated lesions at the slaughterhouse in clinically healthy animals. The objective of this work was to investigate the prevalence and type of subclinical pneumonic lesions in slaughtered beef cattle, according to the age range and management system, and its impact on carcass weight. A total of 1101 beef cattle intended for human consumption were examined at slaughter. Information on age, sex, management system and carcass weight was recorded. The presence and type of pneumonia were evaluated according to gross and microscopic findings and etiological agents by PCR. Lung pneumonic lesions appeared in 17.9% of animals and were predominant among veal calves. According to the type, chronic catarrhal pneumonia prevailed in the majority of animals, and mixed and extensively reared cattle were more likely to suffer acute fibrinous pneumonia. The presence of pneumonic lesions was associated with a significant decrease in carcass weight that had more of an impact in veal male calves coming from intensive systems. Bacterial infections were the predominant infectious agent and the only cause of acute fibrinous pneumonia, while viruses were infrequent and only found in lesions with chronic catarrhal pneumonia. This study shows the importance of BRD in beef feedlots upon production values and points out the feasibility of slaughterhouse assessment of pneumonia as a method for the evaluation of BRD significance.

Abstract

Intrauterine growth restriction (IUGR) is a key developmental programming factor which might impair both the feed efficiency of lambs and meat quality, since it deeply impacts skeletal muscle and adipose tissue development. To determine the effect of birth weight on the growth performance, ruminal parameters, digestibility, non-carcass components, carcass traits, physicochemical characteristics and fatty acid profile of meat, two experimental groups (six animals in each group) of male Merino lambs with different body weights (BW) at birth (low BW (LW; 3.88 ± 0.281 kg) and high BW (HW; 5.80 ± 0.647 kg)) were used. The lambs were penned with their corresponding ewe during the natural suckling period, being weaned at 15 kg. Then, the lambs were penned individually and offered a complete pelleted diet during the fattening period. All the animals were slaughtered when they reached 27 kg of BW. After weaning, both daily dry matter intake (578 vs. 615 g/day; p = 0.021) and average daily gain (141 vs. 190 g/day; p = 0.004) were significantly lower in LW lambs, and a higher feed:gain ratio was recorded for this group (3.98 vs. 3.45; p = 0.008). Carcass traits did not show differences (p > 0.05) between both groups of lambs, except for higher chilling losses for the LW group (3.29 vs. 2.69%; p = 0.012). Additionally, higher amounts of kidney knob and channel fat were observed for LW lambs (85.4 vs. 152 g; p = 0.028). Apart from a higher hardness of meat in LW lambs (152 vs. 189 Newtons, p = 0.040), no other differences (p > 0.05) were observed in the physicochemical traits of this product; however, the meat of LW lambs tended (p = 0.057) to contain more total fatty acid content with a higher (p = 0.041) proportion of saturated fatty acids than the meat of HW lambs. Abstract

Despite the importance of atmospheric methane as a potent greenhouse gas and the significant contribution from ruminant enteric fermentation on methane emissions at a global scale, little effort has been made to consider the influence that different plant-based natural diets have on methane emissions in grazing systems. Heathland is an ericaceous dwarf-shrub-dominated habitat widespread across the northern hemisphere, in Europe, provides valuable ecosystem services in areas with poor soils, such as water flow regulation, land-based carbon skin, energy reservoir and habitat of key game species. We (i) measured methane emissions from red deer (Cervus elaphus) and sheep (Ovis aries) fed mixed diets of natural grass plus ericaceous species (either Calluna vulgaris or Vaccinium myrtillus) using open-circuit respiration chambers; and (ii) modelled the results to estimate methane emissions from red deer and sheep populations inhabiting heathland habitats across Europe under different scenarios of grass-based mixed diets with varying proportions of ericaceous species. Our results indicated that methane emissions per unit of digestible organic matter intake decreased as the proportion of ericaceous species in diet increased, but this relationship was complex because of the significant interaction between the proportion of ericaceous species in the diet and digestible organic matter intake. According to our estimates red deer and sheep populations across European heathlands produce 129.7 kt·y methane (se = 1.79) based on a hypothetical grass-ericaceous species mixed diet containing 30% of ericaceous species; this is 0.5% of total methane emissions from human activity across Europe (24,755 kt·y), and a reduction in methane emissions of 63.8 kt·y against the same deer and sheep populations, if assumed to consume a grass-only diet. Abstract

Thirty-two lambs were finished on a total mixed ration (TMR) pelleted alone (00SS) or including sunflower soap stock (SS): 15SS (15 g SS per kg TMR), 30SS (30 g SS per kg TMR) and 60SS (60 g SS per kg TMR). Lambs (8 per group) were slaughtered at 27 kg live weight. Colour evolution, lipid oxidation, microbial growth and detection of diarrhoeagenic Escherichia coli were studied in meat samples. SS in the diet reduced meat lipid oxidation (P < 0.05), but had no significant effect (P > 0.05) on the cholesterol oxidation products. Meat from 60SS and control animals (00SS) revealed greater discoloration (P < 0.05) than 15SS. SS supplementation did not affect the microbiological populations, whereas high doses of SS seemed to increase the proliferation of diarrhoeagenic E. coli on day 14 (P < 0.10). These results suggest that the rate of SS inclusion in the diet of fattening lambs should not be above 15 g SS per kg TMR.

Abstract

Fresh sausages are highly perishable, and the preservatives allowed in these types of meat preparations are limited. Balkan-style fresh sausages were prepared in triplicate without antimicrobials (Control), with an aqueous hops extract (30 mL/kg), with Zataria multiflora Boiss essential oil (1 mL/kg), or a combination of both (15 and 0.5 mL/kg, respectively), and refrigerator-stored under a 20% CO2 and 80% N2 atmosphere. The spoilage microbial growth, i.e., lactic acid bacteria (LAB), Brochothrix thermosphacta, Enterobacteriaceae, Micrococcaceae, molds and yeasts, the pH value, and the production of biogenic amines in the sausages were monitored weekly and compared with a control sausage during a 35-day storage period. Furthermore, 349 colonies of presumptive LAB (isolated from the De Mann, Rogose-Sharpe agar plates) were identified using a MALDI-TOF-based method. Growth levels to ≈ 9 Log colony forming units (CFU) per g were reached by LAB, with a predominance of Lactobacillus sakei. Enterobacteriaceae and B. thermosphacta also showed significant growth (up to 6 Log CFU/g). Biogenic amine levels increased, and tyramine values overcame 250 mg/kg. The study could not demonstrate a significant effect of antimicrobial source treatments in any of the characteristics studied, and thus, the shelf-life of sausages.

Abstract

Thirty-six Assaf male lambs (29.4 ± 3.10 kg body weight (BW)) were used to study the feasibility of including urea (at 0, 0.6 or 0.95% of dry matter for Control, Urea1, and Urea2 diets, respectively) in substitution of soybean meal in fattening diets. Animals were individually penned and feed intake was recorded daily. Blood samples were taken at days 35 and 63 of the experimental period to determine the acid-base status and the biochemical profile. At the end of the experiment (nine weeks), lambs were slaughtered, ruminal contents were collected and carcass and meat quality were evaluated. There were not differences (p > 0.05) among treatments in dry matter intake, animal performance, ruminal fermentation pattern, and carcass and meat parameters. Serum albumin concentration was higher and concentration of HCO3 and total CO2 in blood were lower in Urea2 compared to Urea1 and Control lambs. These results, together with the tendency to lower (p = 0.065) blood pH in this group might suggest a moderate metabolic acidosis. Partial replacement of soybean meal with urea did not impair growth rate in heavy fattening Assaf lambs (from 29 to 50 kg body weight), reduced feeding costs and had no adverse effects on feed efficiency, rumen fermentation and carcass and meat quality.

48. Changes in volatile compounds in a fresh lamb sausage refrigerated stored under anaerobic modified atmosphere

Abstract

Fresh sausages are commonly packaged in anaerobic modified atmospheres and stored under refrigeration in order to achieve a long shelf life, i.e. few weeks. During storage and due to the internal conditions of fresh sausages (pH, aw, nutrient availability) there is a progressive development of facultative anaerobic bacteria, i.e. lactic acid bacteria or enterobacteria, which in the course of time results in product spoilage. The main reason for this is the production volatile compounds from microbial metabolism which can lead on the generation of undesirable flavours. In this study, the volatile compounds in a fresh Bosnian-style lamb sausage during refrigerated storage under 20% CO2 and 80% N2 atmosphere were weekly monitored for 35 days. Sausages were prepared in triplicate and volatiles were determined by head space solid-phase microextraction (SPME) coupled with gas chromatography-mass spectrometry. The microbial origin volatiles detected at day 0 (stuffing into casings) were diacetyl, hexanol and ethylhexanol. At day 7 of storage appeared 3-methyl-butanal, 3-methyl-butanol and acetoin, at day 14 ethyl acetate, ethylhexanoate and benzene acetaldehyde, and at day 28 butanediol. Most of them tended to increase during storage until a point where their concentrations stabilise. Relevant increases were observed for diacetyl from day 0 to 7, 1-hexanol from day 14 to 21 (then tended to decrease) and acetoin from day 21 to 28. Moreover, ethyl acetate, acetoin, 3-methyl-butanol, ethylhexanoate and ethylhexanol increased from the day where they were detected to day 28. Meanwhile, butanediol and benzene acetaldehyde increased until the end of storage. Volatiles presumably originated from lipid oxidation, such as hexanal, 1-octen-3-ol or heptanal, were also detected and their concentration tended to be constant or decrease with time. Se compararon las características de la canal y de la carne de corderos pesados de raza assaf con diferente eficiencia alimentaria, parámetro relacionado con la cantidad de alimento que consume cada cordero para alcanzar una determinada ganancia de peso. Los resultados obtenidos sugieren que, en las condiciones del presente estudio, la eficiencia con que los animales utilizan el mismo alimento no guarda relación ni con la composición de la canal ni de la carne, pero sí influye negativamente en el rendimiento comercial.

Abstract

Thirty Assaf lambs, divided into three groups, equilibrated for live body weight (29.8 ± 1.88 kg), were used to study the effect of different dietary levels of protein on feed intake, animal performance and carcass characteristics. Three total mixed rations including barley straw, barley, corn, soybean meal, urea, molasses, soy oil, bicarbonate, vitamin-mineral premix, with a 15:85 forage:concentrate ratio, were formulated in order to supply 135 (LP group), 155 (MP group) and 175 (HP group) g of crude protein/kg dry matter. Animals were housed individually and had free access to fresh water. Feed was offered ad libitum and daily intake was recorded. Lambs were weighed once a week and they were slaughtered at the end of the experimental period (63 days). Carcasses were weighed before and after chilling for 24 hours at 4ºC and the following parameters were recorded or calculated: hot and cold carcass weight (HCW, CCW), chilling losses, carcass commercial yield and proportions of carcass joints. Data were subjected to analysis of variance, considering the protein content in the diet as fixed effect. Linear and quadratic trends were evaluated using orthogonal polynomial contrasts. Dry mater intake (DMI) was not affected (P>0.05) by dietary protein level, although DMI was numerically greater in HP than in LP group (1256, 1401 and 1464 g DM/day for LP, MP and HP groups, respectively). Average daily gain (221, 268 and 297 g/day for LP, MP and HP, respectively) and feed to gain conversion rate (5.82, 5.32 and 4.97 g DMI/g ADG for LP, MP and HP, respectively) increased (P<0.01) and decreased (P<0.01), respectively, with increasing level of dietary protein. There was a linear effect (P<0.05) of dietary protein level on CCW, with the highest (26.1 kg) and lowest (22.8 kg) values for HP and LP groups, respectively.
