The papers in this special issue, Geographies of Religion and Spirituality: Pilgrimage beyond the ‘Officially Sacred,’ are placed in the context of a comprehensive theoretical overview of the role that the sacred plays in shaping, conducting, controlling, and contesting pilgrimage. As scholarship examining the lived experiences of travelers has demonstrated, pilgrimages need not necessarily be religious in nature, nor be officially sanctioned. Rather, if pilgrimages are perceived as ‘hyper-meaningful’ by the practitioner, the authors in this special issue argue that a common denominator of all of these journeys is the perception of sacredness—a quality that is opposed to profane, everyday life. Separating the social category of ‘religion’ from the ‘sacred,’ these articles employ an interdisciplinary approach to theorize sacredness, its variability, and the ways in which it is officially recognized or condemned. Thus, the authors pay particular attention to the authorizing processes that religious and temporal power centers employ to either promote, co-opt, or stave off, such popular manifestations of devotion, focusing on three ways: through tradition, text or institutionalized norms. Referencing examples from across the globe, and linking them to the varied contributions in this special issue, this introduction complexifies the ways in which pilgrims, central authorities, locals and other stakeholders on the ground appropriate, negotiate, shape, contest, or circumvent the powerful forces of the sacred. Delving ‘beyond the officially sacred,’ this collective examination of pilgrimages, both well-established and new; religious and secular; authorized and not; the contributions to this special issue, as well as this Introduction, examines the interplay of a transcendent sacred for pilgrims and tourists so as to provide a blueprint for how work in the geography of religion and the fields of pilgrimage and religious tourism may move forward.

In Demospongiae (phylum Porifera) the formation of the siticeous skeleton, composed of spicules, is an energetically expensive reaction. The present study demonstrates that primmorphs from the demosponge Suberites domuncula express the gene for arginine kinase after exposure to exogenous silicic acid. The deduced sponge arginine kinase sequence displays the two characteristic domains of the ATP:guanido phosphotransferases ; it can be grouped to the 'usual' mono-domain 40kDa guanidino kinases (arginine kinases). Phylogenetic studies indicate that the metazoan guanidino kinases evolved from this ancestral sponge enzyme ; among them are also the 'unusual' two-domain 80 kDa guanidino kinases. The high expression level of the arginine kinase gene was already measurable 1 day after Summary addition of silicic acid by northern blot, as well as by in situ hybridization analysis. Parallel determinations of enzyme activity confirmed that high levels of arginine kinase are present in primmorphs that had been exposed for 1-5 days to silicic acid. Finally, transmission electron-microscopical studies showed that primmorphs containing high levels of arginine kinase also produce siliceous spicules. These data highlight that silicic acid is an inorganic morphogenetic factor that induces the expression of the arginine kinase, which in turn probably catalyzes the reversible transfer of high-energy phosphoryl groups.

In some sponges peculiar proteins called silicateins catalyze silica polymerization in ordered structures, and their study is of high interest for possible biotechnological applications in the nanostructure industry. In this work we describe the isolation and the molecular characterization of silicatein from spicules of Petrosia ficiformis, a common Mediterranean sponge, and the development of a cellular model (primmorphs) suitable for in vitro studies of silicatein gene regulation. The spicule of P. ficiformis contains an axial filament composed of 2 insoluble proteins, of 30 and 23 kDa. The 23-kDa protein was characterized, and the full-length cDNA was cloned. The putative amino acid sequence has high homology with previously described silicateins from other sponge species and also is very similar to cathepsins, a cystein protease family. Finally, P. ficiformis primmorphs express the silicatein gene, suggesting that they should be a good model for biosilicification studies.

Recently, the thermosensing pathway in sponges (Porifera) was elucidated. The thermosensor triggering this cascade is a heat-activated cation channel, with the phytohormone abscisic acid (ABA), cyclic ADP-ribose (cADPR) and calcium acting as intracellular messengers, similarly to the drought-stress signaling cascade in higher plants. Here, we investigated the functional effects downstream of the temperature-signaling pathway in Axinella polypoides (Porifera, Demonspongiae). Short-term stimulation followed by long-term depression of amino acid incorporation, oxygen consumption and water filtration were observed after exposure of the sponge to a brief heat stress or to micromolar ABA. These effects could be prevented by the targeted interruption of the signaling pathway either at the level of the cation channel thermosensor or at the level of the cADPR-induced intracellular calcium increase. Moreover, release of cyclase activity into the sea water and generation of extracellular cADPR were observed following brief heat stress. Intact sponge cells were sensitive to extracellular cADPR and addition of purified cyclase increased sponge respiration similarly to heat stress. This is the first observation of functional effects exerted on Metazoa by the phytohormone ABA: conservation of the ABA/cADPR stress-signaling cascade points to its early evolution in a common precursor of modern Metazoa and Metaphyta. The functional effects induced by extracellular cyclase/cADPR suggest an evolutionary origin of cADPR as an ancient stress hormone in Porifera.

Epoxy-ZnO composites are prepared via UV-induced cationic polymerization. Complete unsoluble materials are achieved after 10min of irradiation. Thermal analyses shows an increase of T-g in the presence of the ZnO attributable to a strong hindrance effect of the polymer chain mobility. Electrical characterization of the cured materials shows the decrease of the real part of permittivity with frequency. Both permittivity and AC conductivity values increase with nanofiller content, especially for the highest contents. This is described as typical behavior of nanocomposites made from polymers and fillers with higher permittivity and conductivity values, in accordance with previous data reported in literature for similar composites.
