Active packaging that prolongs food shelf life, maintaining its quality and safety, is an increasing industrial demand, especially if integrated in a circular economy model. In this study, the fabrication and characterization of sustainable cellulose-based active packaging using food-industry waste and natural extracts as antioxidant agents was assessed. Grape marc, olive pomace and moringa leaf extracts obtained by supercritical fluid, antisolvent and maceration extraction in different solvents were compared for their antioxidant power and phenolic content. Grape and moringa macerates in acetone and methanol, as the most efficient and cost-effective extracts, were incorporated in the packaging as coatings or in-between layers. Both systems showed significant free-radical protection in vitro (antioxidant power 50%) and more than 50% prevention of ground beef lipid peroxidation over 16 days by indirect TBARS and direct in situ Raman microspectroscopy measurements. An innovative spectroscopic method that allows to chemically and structurally characterize viruses directly in suspension within few minutes was developed. A library of five different plant viruses was obtained combining dielectrophoresis (DEP), performed with a device specifically designed to capture and agglomerate virus particles, and Raman spectroscopy to provide a chemical fingerprint of virions. The tested viruses, purified from infected plants, were chosen for their economic impact on horticultural crops and for their different morphological and structural features. Using the Raman-DEP device, specific profiles for each virus were successfully obtained, relying on chemical differences occurring even with genetically similar viruses belonging to the same taxonomic species and morphologically indiscernible by transmission electron microscopy (TEM). Moreover, we investigated the potentiality of Raman-DEP to follow dynamic changes occurring upon heat treatment of tobacco mosaic virus (TMV) particles. Raman peak deviations linked to TMV coat protein conformation were observed upon treatment at temperatures equal or higher than 85°C, substantiating the rod-to-spherical shape transitions observed by TEM and the concomitant drastic loss of infectivity following plant inoculation. Grapevine is one of the most cultivated fruit plant among economically relevant species in the world. It is vegetatively propagated and can be attacked by more than 80 viruses with possible detrimental effects on crop yield and wine quality. Preventive measures relying on extensive and robust diagnosis are fundamental to guarantee the use of virus-free grapevine plants and to manage its diseases. New phenotyping techniques for non-invasive identification of biochemical changes occurring during virus infection can be used for rapid diagnostic purposes. Here, we have investigated the potential of Raman spectroscopy (RS) to identify the presence of two different viruses, grapevine fan leaf virus (GFLV) and grapevine rupestris stem pitting-associated virus (GRSPaV) in Vitis vinifera cv. Chardonnay. We showed that RS can discriminate healthy plants from those infected by each of the two viruses, even in the absence of visible symptoms, with accuracy up to 100% and 80% for GFLV and GRSPaV, respectively. Chemometric analyses of the Raman spectra followed by chemical measurements showed that RS could probe a decrease in the carotenoid content in infected leaves, more profoundly altered by GFLV infection. Transcriptional analysis of genes involved in the carotenoid pathway confirmed that this biosynthetic process is altered during infection. Antimicrobial peptides (AMPs) are widely studied as therapeutic agents due to their broad-spectrum efficacy against infections. However, their clinical use is hampered by the low in vivo bioavailability and systemic toxicity. Such limitations might be overcome by using appropriate drug delivery systems. Here, the preparation of a drug delivery system (DDS) by physical conjugation of an arginine-rich peptide and hydrothermal carbon nanoparticles (CNPs) has been explored, and its antimicrobial efficacy against Eschericia coli ( E. coli ) and Staphylococcus aureus investigated in comparison with the unloaded carrier and the free peptide. The mechanism of interaction between CNPs and the bacteria was investigated by scanning electron microscopy and a combined dielectrophoresis-Raman spectroscopy method for real-time analysis. In view of a possible systemic administration, the effect of proteins on the stability of the DDS was investigated by using albumin as a model protein. The peptide was bounded electrostatically to the CNPs surface, establishing an equilibrium modulated by pH and albumin. The DDS exhibited antimicrobial activity toward the two bacterial strains, albeit lower as compared to the free peptide. The decrease in effectiveness toward E. coli was likely due to the rapid formation of a particle-induced extracellular matrix. In this work the quantification of antimicrobial properties of differently sized AgNPs immobilized on a surface was studied. Three different sizes of spheroidal AgNPs with a diameter of (6, 30 and 52) nm were synthetized and characterized with UV-vis, SEM, TEM and ICP-MS. The MIC (Minimal Inhibitory Concentration) and MBC (Minimal Bactericidal Concentration) against Escherichia coli were investigated. Then, the antibacterial efficacy (R) of amino-silanized glasses coated with different amounts of the three sizes of AgNPs were quantified by international standard ISO 22196 adapted protocol against E. coli, clarifying the relationship between size and antibacterial properties of immobilized AgNPs on a surface. The total amount of silver present on glasses with an R ∼ 6 for each AgNPs size was quantified with ICP-MS and this was considered the Surface MBC (SMBC), which were found to be (0.023, 0.026 and 0.034) μg/cm 2 for (6, 30 and 52) nm AgNPs, respectively. Thus, this study demonstrates that active surfaces with a bactericidal effect at least ≥ 99.9999 % could be obtained using an amount of silver almost 100 times lower than the MBC found for colloidal AgNPs. The immobilization reduces the aggregation phenomena normally occuring in liquid media, maximizing the exposed specific superficial area of the AgNPs and their direct contact with bacterial cells. The potential of surface-enhanced Raman scattering (SERS) has been investigated for the rapid analysis of two representative organotin compounds (OTCs): dibutyltin maleate (DTM) and tributyltin chloride (TBT), after migration tests from polyvinyl chloride (PVC), as a model food packaging material in aqueous food simulant (acetic acid 3% w/v). OTCs, often used as heat stabilizers additives for PVC, are classified as endocrine disrupting chemicals (EDCs) and their migration potential has to be controlled in compliance with the normative prescriptions for food contact materials. In this study, colloidal silver nanoparticles (AgNPs) were applied as liquid SERS substrate for direct-in-liquid analysis of food simulant after standardized migration tests of PVC samples spiked with OTCs. Promising results were obtained, reaching detection limits below the permitted limits for the considered OTCs (i.e. 0.15 mg/l): DTM and TBT were detected down to 0.01 mg/l and 0.08 mg/l, respectively. Calibration curves were calculated for standard solutions of DTM and TBT in the dynamic range between 0 and 1 mg/l (reduced χ 2  = 0.8), and 0.5-5 mg/l (reduced χ 2  = 0.2), respectively. Migrated TBT and DTM were detected in the food simulant, specifically identified and quantified by SERS, with a measurement uncertainty around 10% in all cases. In particular, it was found that TBT can migrate in higher amount compared to DTM when the PVC film is in contact with a slightly acidic matrix. These results were further confirmed by inductively coupled plasma-mass spectrometry and UV-Vis spectroscopy. A rapid Surface Enhanced Raman Spectroscopy (SERS) method to detect SO 2 in wine is presented, exploiting the preferential binding of silver nanoparticles (AgNPs) with sulfur-containing species. This interaction promotes the agglomeration of the AgNPs and inducing the formation of SERS "hot spots" responsible for SO 2 signals enhancement. For increasing SO 2 concentrations from 0 to100 mg/l in wine simulant, SERS intensity showed an increasing trend, following a Langmuir absorption function (R 2  = 0.94). Due to the wine matrix variability, a standard additions method was then employed for quantitative analysis in red and white wines. This method does not require the SO 2 separation but only a matrix pre-cleaning by solid phase extraction. The limit of detection (LOD) was defined for each wine tested, ranging from 0.6 mg/l to 9.6 mg/l. Seed-growth synthesis is a common strategy to prepare silver nanoplates, whose peculiar plasmonic features can be exploited for surface enhanced Raman scattering (SERS) applications. Here we describe the fabrication and characterization of SERS chips using a peculiar in situ seed growth method, yielding a dense layer of nano-objects directly on a glass slide. In this way, geometric features (i.e. shape and dimensions) of the nano-objects can be tuned by controlling the growth time, obtaining a high concentration of hot spots on the surface. In particular, the SERS response of four kinds of chips were investigated to define the best SERS configuration in terms of size of the silver nano-objects, excitation wavelength and homogeneity of the SERS response. Silver nano-plates with a seeded growth time of 60 min demonstrated remarkable results both in terms of plasmonic enhancement, with an enhancement factor (EF) of 2 × 10 5 using a 532 nm laser excitation, and good homogeneity of the SERS response with intra- and inter-maps RSD of 10% and 5%, respectively. In order to demonstrate application of these chips for real sample analysis, an analytical procedure for the detection of a model pesticide, i.e. thiram fungicide, was developed and applied to its detection on green apples peels. Flexible and transparent substrates are emerging as low cost and easy-to-operate support for surface-enhanced Raman spectroscopy (SERS). In particular, in situ SERS detection approach for surface characterization in transmission modality can be efficiently employed for non-invasive analysis of non-planar surfaces. Here we propose a new methodology to fabricate a homogenous, transparent, and flexible SERS membrane by the assistance of a thin TiO 2 porous layer deposited on the PDMS surface, which supports the uniform loading of gold nanoparticles over large area. The substrate was first characterized for homogeneity, sensitivity and repeatability using a model molecule for SERS, i.e., 7-mercapto-4-methylcoumarin. Satisfactory intra-substrate uniformity and inter-substrates repeatability was achieved, showing an RSD of 10%, and an analytical sensitivity down to 10 nM was determined with an EF of 3.4 × 10 5 ± 0.4 × 10 5 . Furthermore, SERS detection of pyrimethanil (PMT), a commonly employed pesticide in crops for human consumption, was performed in situ, exploiting the optical transparency of the device, using both model surfaces and non-flat bio-samples. PMT contamination at the phytochemical concentration levels corresponding to commonly used infield doses was successfully detected on the surface of the yellow Ficus benjiamina leaves, supporting the use of this substrate for food safety in-field application.

The aim of this study was to evaluate the efficacy of a multi-analytical approach for origin authentication of cocoa bean shells (CBS). The overall chemical profiles of CBS from different origins were characterized using diffuse reflectance near-infrared spectroscopy (NIRS) and attenuated total reflectance mid-infrared spectroscopy (ATR-FT-IR) for molecular composition identification, as well as inductively coupled plasma-optical emission spectroscopy (ICP-OES) for elemental composition identification. Exploratory chemometric techniques based on Principal Component Analysis (PCA) were applied to each single technique for the identification of systematic patterns related to the geographical origin of samples. A combination of the three techniques proved to be the most promising approach to establish classification models. Partial Least Squares-Discriminant Analysis modelling of fused PCA scores of three independent models was used and compared with single technique models. Improved classification of CBS samples was obtained using the fused model. Global population forecasts dictate a rapid adoption of multifaceted approaches to fulfill increasing food requirements, ameliorate food dietary value and security using sustainable and economically feasible agricultural processes. Plant pathogens induce up to 25% losses in vegetable crops and their early detection would contribute to limit their spread and economic impact. As an alternative to time-consuming, destructive, and expensive diagnostic procedures, such as immunological assays and nucleic acid-based techniques, Raman spectroscopy (RS) is a nondestructive rapid technique that generates a chemical fingerprinting of a sample, at low operating costs. Here, we assessed the suitability of RS combined to chemometric analysis to monitor the infection of an important vegetable crop plant, tomato, by two dangerous and peculiarly different viral pathogens, Tomato yellow leaf curl Sardinia virus (TYLCSV) and Tomato spotted wilt virus (TSWV). Experimentally inoculated plants were monitored over 28 days for symptom occurrence and subjected to RS analysis, alongside with measuring the virus amount by quantitative real-time PCR. RS allowed to discriminate mock inoculated (healthy) from virus-infected specimens, reaching an accuracy of >70% after only 14 days after inoculation for TYLCSV and >85% only after 8 days for TSWV, demonstrating its suitability for early detection of virus infection. Importantly, RS also highlighted spectral differences induced by the two viruses, providing specific information on the infecting agent.

The aim of this study was to evaluate cytotoxicity (WST-1 assay), LDH release (LDH assay) and genotoxicity (Comet assay) of three engineered TiO 2 -NPs with different shapes (bipyramids, rods, platelets) in comparison with two commercial TiO 2 -NPs (P25, food grade). After NPs characterization (SEM/T-SEM and DLS), biological effects of NPs were assessed on BEAS-2B cells in presence/absence of light. The cellular uptake of NPs was analyzed using Raman spectroscopy. The cytotoxic effects were mostly slight. After light exposure, the largest cytotoxicity (WST-1 assay) was observed for rods; P25, bipyramids and platelets showed a similar effect; no effect was induced by food grade. No LDH release was detected, confirming the low effect on plasma membrane. Food grade and platelets induced direct genotoxicity while P25, food grade and platelets caused oxidative DNA damage. No genotoxic or oxidative damage was induced by bipyramids and rods. Biological effects were overall lower in darkness than after light exposure. Considering that only food grade, P25 and platelets (more agglomerated) were internalized by cells, the uptake resulted correlated with genotoxicity. In conclusion, cytotoxicity of NPs was low and affected by shape and light exposure, while genotoxicity was influenced by cellular-uptake and aggregation tendency., (Copyright © 2019 Elsevier Ltd. Elemental carbon nanomaterials (ECNMs) are redox active agents that can be exploited to purposely modify the redox balance of cells. Both pro- or antioxidant properties have been reported. However, to the best of our knowledge, there are not comprehensive studies exploring both properties on the same material in view of a potential application in medicine. At the same time, the effect of the bulk structure on the pro/antioxidant properties is poorly known. Here, carbon nanoparticles (CNPs) derived by glucose with definite size and shape have been prepared, and their redox properties evaluated in cell free systems in the dark or following activation with a Near Infrared (NIR) laser beam (945 nm, 1.3 W/cm 2 ). We found that, when irradiated with NIR, CNPs efficiently generate heat and singlet oxygen ( 1 O 2 ), a property that can be exploited for dual photo-thermal (PT)/photodynamic (PD) therapy in cancer. On the other hand, in the absence of photo-activation, CNPs react with both oxidant (hydroxyl radicals) and antioxidant (glutathione) species. When tested on a murine macrophages cell line (RAW 264.7) CNPs were clearly antioxidant. Furthermore, albeit efficiently internalized, CNPs do not exert cytotoxic effect up to 80 µg/ml and do not exacerbate TNF-α-mediated inflammation. Biomedical devices are complex products requiring numerous assembly steps along the industrial process chain, which can carry the potential of surface contamination. Cleanliness has to be analytically assessed with respect to ensuring safety and efficacy. Although several analytical techniques are routinely employed for such evaluation, a reliable analysis chain that guarantees metrological traceability and quantification capability is desirable. This calls for analytical tools that are cascaded in a sensible way to immediately identify and localize possible contamination, both qualitatively and quantitatively. In this systematic inter-comparative approach, we produced and characterized sodium dodecyl sulfate (SDS) films mimicking contamination on inorganic and organic substrates, with potential use as reference materials for ambient techniques, i.e., ambient mass spectrometry (AMS), infrared and Raman spectroscopy, to reliably determine amounts of contamination. Non-invasive and complementary vibrational spectroscopy techniques offer a priori chemical identification with integrated chemical imaging tools to follow the contaminant distribution, even on devices with complex geometry. AMS also provides fingerprint outputs for a fast qualitative identification of surface contaminations to be used at the end of the traceability chain due to its ablative effect on the sample. To absolutely determine the mass of SDS, the vacuum-based reference-free technique X-ray fluorescence was employed for calibration. Convex hip liners were deliberately contaminated with SDS to emulate real biomedical devices with an industrially relevant substance. Implementation of the aforementioned analytical techniques is discussed with respect to combining multimodal technical setups to decrease uncertainties that may arise if a single technique approach is adopted. Tip-Enhanced Raman Spectroscopy (TERS) is a topographic and chemical analysis technique with nanoscale resolution, consisting of the combination of Scanning Probe Microscopy (SPM) and Localized Surface Plasmon Resonance (LSPR) for the enhancement of Raman scattering in the vicinity of the probe. The quantification of spatial resolution represents an important issue, and, as of now, standards for calibration are not available. In the present work a candidate reference sample for TERS measurements was fabricated. It consists of a flat, conductive gold surface with a nanometric grating of a self-assembled monolayer of Raman-active organic molecules fabricated by an optimized Electron Beam Lithography (EBL) method to replicate established SPM calibration standards. Controlling the location and the distribution of hot spots is a crucial aspect in the fabrication of surface-enhanced Raman spectroscopy (SERS) substrates for bio-analytical applications. The choice of a suitable method to tailor the dimensions and the position of plasmonic nanostructures becomes fundamental to provide SERS substrates with significant signal enhancement, homogeneity and reproducibility. In the present work, we studied the influence of the long-range ordering of different flexible gold-coated Si nanowires arrays on the SERS activity. The substrates are made by nanosphere lithography and metal-assisted chemical etching. The degree of order is quantitatively evaluated through the correlation length (ξ) as a function of the nanosphere spin-coating speed. Our findings showed a linear increase of the SERS signal for increasing values of ξ, coherently with a more ordered and dense distribution of hot spots on the surface. The substrate with the largest ξ of 1100 nm showed an enhancement factor of 2.6 · 10 3 and remarkable homogeneity over square-millimetres area. The variability of the signal across the substrate was also investigated by means of a 2D chemical imaging approach and a standard methodology for its practical calculation is proposed for a coherent comparison among the data reported in literature.

Surface Enhanced Raman Scattering (SERS) supported by gold nanoparticles (AuNPs) was applied to detect and quantify residues of pyrimethanil on pome fruits, a widely used fungicide in horticultural species. Spheroidal AuNPs with different size were fabricated and compared in this study. The analytical procedure was set up on a silicon dioxide flat substrate to standardize SERS methodology. A Raman mapping strategy was exploited to increase signal reproducibility and to minimize bias due to different local surface morphologies. Univariate and multivariate regressions were compared for calibration. Multivariate PLS approach demonstrated acceptable repeatability and method stability (RMSECV = 4.79 ppm; RMSEP = 4.31 ppm) in the range 0-40 mg kg -1 , providing higher accuracy and intra-day repeatability with a mean percentage error of 18.7% and 32.8% for PLS and univariate calibration, respectively. There is a strong need in the medical device industry to decrease failure rates of biomedical devices by reducing the incidence of defect structures and contaminants during the production process. The detection and identification of defect structures and contaminants is crucial for many industrial applications. The present study exploits reference-free X-ray fluorescence (XRF) analysis as an analytical tool for the traceable characterization of surface contaminants of medical devices, in particular N,N'-ethylene-bis (stearamide), an ubiquitous compound used in many industrial applications as a release agent or friction reduction additive. Reference-free XRF analysis as primary method has been proven to be capable of underpinning all other applied methods since it yields the absolute mass deposition of the selected N,N'-ethylene-bis (stearamide) contaminant whilst X-ray absorption fine structure analysis determines the chemical species. Ambient vibrational spectroscopy and mass spectroscopy methodologies such as Fourier transform infrared, Raman, and secondary ion mass spectroscopy have been used in this systematic procedure providing an extensive range of complementary analyses. The calibration procedure described in this paper was developed using specially designed and fabricated model systems varying in thickness and substrate material. Furthermore, typical real medical devices such as both a polyethylene hip liner and a silver-coated wound dressing have been contaminated and investigated by these diverse methods, enabling testing of this developed procedure. These well-characterized samples may be used as calibration standards for bench top instrumentation from the perspective of providing traceable analysis of biomaterials and surface treatments. In this paper, a Fourier Transform Raman spectroscopy method, to authenticate the provenience of wine, for food traceability applications was developed. In particular, due to the specific chemical fingerprint of the Raman spectrum, it was possible to discriminate different wines produced in the Piedmont area (North West Italy) in accordance with i) grape varieties, ii) production area and iii) ageing time. In order to create a consistent training set, more than 300 samples from tens of different producers were analyzed, and a chemometric treatment of raw spectra was applied. A discriminant analysis method was employed in the classification procedures, providing a classification capability (percentage of correct answers) of 90% for validation of grape analysis and geographical area provenance, and a classification capability of 84% for ageing time classification. Ammonia in gastric juice is considered a potential biomarker for Helicobacter pylori infection and as a factor contributing to gastric mucosal injury. High ammonia concentrations are also found in patients with chronic renal failure, peptic ulcer disease, and chronic gastritis. Rapid and specific methods for ammonia detection are urgently required by the medical community. Here we present a method to detect ammonia directly in gastric juice based on Fourier transform infrared spectroscopy. The ammonia dissolved in biological liquid samples as ammonium ion was released in air as a gas by the shifting of the pH equilibrium of the ammonium/ammonia reaction and was detected in line by a Fourier transform infrared spectroscopy system equipped with a gas cell for the quantification. The method developed provided high sensitivity and selectivity in ammonia detection both in pure standard solutions and in a simulated gastric juice matrix over the range of diagnostic concentrations tested. Preliminary analyses were also performed on real gastric juice samples from patients with gastric mucosal injury and with symptoms of H. pylori infection, and the results were in agreement with the clinicopathology information. The whole analysis, performed in less than 10 min, can be directly applied on the sample without extraction procedures and it ensures high specificity of detection because of the ammonia fingerprint absorption bands in the infrared spectrum. This method could be easily used with endoscopy instrumentation to provide information in real time and would enable the endoscopist to improve and integrate gastroscopic examinations.

A rapid and sensitive method to detect melamine in liquid milk based on Surface Enhanced Raman Scattering (SERS) spectroscopy is presented, exploiting the selective binding of gold nanoparticles (AuNPs) with this analyte. This interaction promotes the aggregation of the AuNPs inducing a huge enhancement of the melamine signals in the Raman spectrum due to the formation of SERS "hot spots". An external standard calibration method was employed for quantitative analysis and the method was validated for linearity, sensitivity, repeatability and recovery. A good linearity (R(2)=0.99) was found in the concentration range of 0.31-5.0 mg l(-1) in milk with a limit of detection of 0.17 mg l(-1). This method does not require a long extraction procedure (total analysis time can be lower than 30 min) and can be reliably used for melamine detection in milk matrix in accordance with the European law limits., (Copyright © 2014 Elsevier Ltd. Surface-relief diffraction gratings and planar diffraction gratings directly written on nanoporous silicon layers using 514 nm continuous-wave lasers at very low power (less than 20 mW) were demonstrated. Diffraction-based biosensing application to detect arachidonic acid was experimentally demonstrated at incident light wavelength of 632.8 nm. A comparison in sensing applications was made between the two types of gratings to show the distinct advantage of the planar grating with selective functionalization. Laser-written planar gratings enable directly immobilizing biomolecules in the laser oxidized area of nanoporous silicon, resulting in a new patterned functionalization technique for biosensing applications. The functionalization technique can not only simplify the functionalization procedure in biosensing but also it has potential to increase the sensitivity of sensors by accurately defining grating patterns using the laser direct writing technique.

Three-dimensional submicrometric structures and biomolecular patterns have been fabricated on a porous silicon film by an electron beam-based functionalization method. The immobilized proteins act as a passivation layer against material corrosion in aqueous solutions. The effects' dependence on the main parameters of the process (i.e., the electron beam dose, the biomolecule concentration, and the incubation time) has been demonstrated.

A porous silicon biosensor based on P450 enzyme for arachidonic acid detection was developed. A new transduction method is presented with a simultaneous measurement of refractive index and fluorescence intensity changes when the analyte is binding to an enzyme on the porous silicon surface. A fluorophore bound to a cysteine residue in an allosteric position of the haem domain (BMP) of cytochrome P450 BM3 enhances its fluorescence intensity upon interaction with its substrate arachidonic acid, involved in diseases such as Alzheimer's, liver cancer and cellular inflammation processes. BMP has been anchored on porous silicon surface and the new transduction method has been successfully exploited to develop a biosensor for arachidonic acid, reaching a detection limit of 10 μM arachidonic acid in a dynamic range of 10-200 μM. Moreover, the change of the refractive index has been also monitored at the same time, displaying a higher detection limit of 30 μM. Preliminary test were also conducted in plasma proving the high specificity and selectivity of the sensor even in presence of interferents in the range of 50-100 μM. Here we suggest these two detection systems could be used simultaneously to increase the accuracy and the dynamic range of the sensor avoiding a false positive response., (Copyright © 2011 Elsevier B.V.
