Drypetes klainei Pierre ex Pax is used in Cameroon by Baka people in the wound healing process and for the treatment of burns. In a previous paper we demonstrated the ability of both water (WE) and defatted methanol (DME) extracts to accelerate scratch wound closure in fibroblast cultures, thus validating the traditional use of D. klainey stem bark in the treatment of skin lesions. In this work we carried out a bioassay-guided fractionation of the most active DME, which exhibited in vitro efficacy in accelerating wound healing process, in order to isolate and identify the compound/s responsible for the assessed biological activity. HPLC was used for the metabolite profiling of DME and fractions (analytical) and for the isolation of the bioactive compound (semi-preparative). MS analyses and NMR spectroscopy were used for identifying the isolated compound. The abilities of treatments in accelerating wound healing were studied on murine fibroblasts in terms of cell viability and cell migration (scratch wound-healing assay). The results obtained allowed to unambiguously identify the isolated bioactive compound as nigracin, a known phenolic glycoside firstly isolated and characterized from bark and leaves of Populus nigra in 1967. However, this is the first time that nigracin is identified in the Drypetes genus and that a wound healing activity is demonstrated for this molecule. Specifically, we demonstrated that nigracin significantly stimulates fibroblast growth and improves cell motility and wound closure of fibroblast monolayer in a dose-dependent manner, without any toxicity at the concentrations tested, and is still active at very low doses. This makes the molecule particularly attractive as a possible candidate for developing new therapeutic options for wound care.

Nel pesco l’epoca di maturazione è un target importante per l’innovazione varietale. È un carattere quantitativo, cioè determinato dalla somma dell’azione di più geni. Le regioni di DNA associate ad un particolare carattere quantitativo vengono definite QTL (Quantitative Trait Loci). Finora sono stati individuati alcuni QTL in diversi cromosomi (2, 6 e soprattutto 4). Studi recenti hanno individuato proprio all’interno del cromosoma 4 un gene fortemente associato all’epoca di maturazione negli individui di due progenie ottenute dall’incrocio di varietà assai distanti fra loro. Grazie a tale associazione è stato trovato un marcatore molecolare (“marker”) in grado di discriminare, con buona approssimazione, gli individui a maturazione precoce da quelli a maturazione tardiva. La disponibilità di questo marcatore consentirebbe ai breeder di sviluppare nuove cultivar che maturano nel periodo desiderato e sarebbe quindi potenzialmente in grado di servire per una selezione mirata all’epoca di maturazione. Per valutare il marcatore è stata fatta una verifica su un gruppo di 140 varietà di pesche e nettarine della collezione di pesco del Crea di Forlì, comprendenti varietà tradizionali e moderne a maturazione molto differenziata, nell’arco di tre mesi.

Peach (Prunus persica L.) fruits contain several health-promoting phytochemicals. Among these, carotenoids, in addition to being involved in determining flesh color, play a relevant role in cell protection against oxidative stress. Nevertheless, antioxidant activity (AA) of peach carotenoids so far has not been investigated in as much detail as phenols. In the present study, for the first time, AA of peach carotenoid extracts was evaluated using the innovative lipoxygenase/4-nitroso-N,N-dimethylaniline (LOX/RNO) method, able to simultaneously detect different antioxidant mechanisms and synergistic antioxidant interactions, as well as using the well-known ORAC and TEAC assays. In particular, extracts were obtained from fruits collected in S4 development stage from two yellow-fleshed (Armking and Redhaven) and three white-fleshed (Silverking, Caldesi 2000, IFF331) varieties. The LOX/RNO method gave high AA values (10-150 μmol eq. Trolox/g f.w.), about 85-1900-fold higher than ORAC and TEAC methods. Moreover, the ratio between AA values, measured by the LOX/RNO method, of yellow- and white-fleshed peaches resulted equal to 14, but only 2.6 and 3.6 for ORAC and TEAC, respectively. Results of this study indicate that the LOX/RNO method, measuring high AA values and easily discriminating among samples, is an advisable tool to assess the AA of the carotenoid component in peach., Advances in Horticultural Science, Vol 29 No 2-3 (2015)

In pesco, il colore della polpa è un carattere monogenico, con il fenotipo bianco dominante su quello giallo; si sa inoltre che la sua espressione dipende da un enzima che degrada i carolenoidi della polpa. In questo lavoro, il gene che codifica per una carotenoide diossigenasi (ccd4) è stato studiato per verificarne l'influenza nella determinazione del colore della polpa. Le analisi sono state condotte su mutanti chimerici con settori del mesocarpo bianchi o gialli; sono poi state estese a un pool di cultivar e a una popolazione F1 segregante. La presenza di almeno un allele di ccd4 funzionale è coerentemente associata al colore ancestrale della polpa; al contrario, il fenotipo giallo ha avuto origine dall'insorgenza di almeno tre indipendenti mutazioni che inattivano la funzionalità di questa diossigenasi, impedendo quindi la degradazione dei carotenoidi. Sono state inoltre caratterizzate alcune retro-mutazioni che riattivano le funzioni di ccd4 e ripristinano il colore bianco ancestrale. I risultati mostrano che ccd4 è il gene che controlla il colore della polpa in pesco; la sua espressione causa la degradazione dei carotenoidi nei genotipi a polpa bianca, mentre i genotipi a polpa gialla appaiono come conseguenza della sua inattivazione.

Italy is one of the main European sweet cherry producers and it has a rich germplasm, including many minor local varieties that have not been well studied or used in breeding programmes. Sweet cherry is self-incompatible, with few exceptions; its incompatibility is controlled by a multi-allelic S locus, which is gametophytically expressed. The knowledge of S-alleles and cross-incompatibility groups of sweet cherry genotypes and cultivars is important for growers and breeders. In this work we analysed 48 traditional sweet cherry varieties and cultivars, mainly from Calabria and Emilia Romagna regions, together with eight Sicilian cultivars and the standard set of reference genotypes proposed by the European Collaborative Programme for Genetic Resources (ECPGR) Prunus group (Clarke and Tobutt, 2009), using molecular markers. Two consensus primer pairs for the incompatibility (S) locus, and eight microsatellite (SSR) primers, recommended by the ECPGR, were used in multiplexed reactions to characterise the accessions. Twelve different S-alleles were detected. S3, S6 and S13 alleles were the most common. Accessions were assigned to incompatibility groups. The chosen SSRs were able to discriminate most of the accessions and to discover duplicates and synonyms. Our results are useful for efficiently design orchards and planning crosses for future breeding programmes. The usefulness of some accessions for breeding is discussed.
