Lysosomal storage diseases are inherited and acquired disorders characterized by dysfunctional lysosomes. Intracytoplasmic accumulation of undegraded substrates leads to impaired cellular function and death. Several plant species are toxic to livestock because of the presence of indolizidine alkaloids, including swainsonine, which cause a storage disease. Swainsonine-induced nervous disease (i.e., locoism) of sheep and cattle is well recognized in several parts of the world, particularly in the western United States and in parts of Australia. Spontaneous intoxication by Astragalus garbancillo var. garbancillo was suspected in a group of 70 llamas (Lama glama) in Jujuy Province, northwestern Argentina. The animals grazed an area dominated by stands of A. garbancillo var. garbancillo. Clinical signs were staggering, ataxia, hypermetria, and progressive weight loss. The clinical course in individual animals was ~50 d. The main microscopic changes were Purkinje cell degeneration, necrosis, and loss, associated with intracytoplasmic vacuolation, meganeurite formation, and Wallerian degeneration. Specific positive labeling for ubiquitin was observed in axonal spheroids. Composite leaf and stem samples of A. garbancillo var. garbancillo analyzed by high-performance liquid chromatography contained 0.03% swainsonine. Based on the microscopic lesions, clinical history, and plant analysis, a diagnosis was made of storage disease caused by consumption of swainsonine-containing A. garbancillo var. garbancillo.

Este trabalho inclui os estudos clínicos e patológicos da doença de armazenamento lisossomal induzida pelo consumo espontâneo de Sida carpinifolia. A enfermidade foi observada em três rebanhos, que juntos eram compostos por 51 caprinos, dos quais, 25 foram afetados e 11 necropsiados. Nos três surtos, S. carpinifolia era a vegetação predominante nos piquetes ocupados pelos animais. Clinicamente, a doença caracterizou-se por distúrbios neurológicos que consistiam de ataxia, hipermetria, posturas anormais, tremores musculares afetando principalmente as regiões da cabeça e pescoço, dificuldade para ingestão de alimentos e quedas freqüentes. Estes sinais clínicos eram exacerbados pela movimentação. Em alguns animais, embora com um quadro clínico estabilizado, as alterações neurológicas persistiram durante 24 meses após sua retirada dos piquetes infestados por S. carpinifolia. A doença foi reproduzida administrando-se S. carpinifolia, in natura ou seca à sombra, para 3 caprinos. Um caprino recebeu Sida rhombifolia, ad libidum, por 40 dias e não desenvolveu alterações clínicas ou patológicas. Na necropsia não havia alterações. Microscopicamente, as principais alterações foram distensão e vacuolização citoplasmáticas em neurônios e, em menor intensidade, em células da glia do sistema nervoso central. Alterações similares foram observadas em células acinares pancreáticas, hepatócitos, células tubulares renais, células foliculares epiteliais da tireóide e macrófagos de órgãos linfóides. Nos animais que não mais ingeriam S. carpinifolia por períodos de um mês ou mais, observou-se uma diminuição da vacuolização citoplasmática de neurônios, que apresentavam citoplasma eosinofílico e aspecto enrugado. Nestes casos, notou-se também desaparecimento neuronal especialmente em células de Purkinje e gliose local. Em cortes cerebelares, esta doença de armazenamento foi caracterizada como ?-manosidose pelo estudo histoquímico por lectinas. Os vacúolos nas células de Purkinje reagiram fortemente com as lectinas Concanavalia ensiformis, Triticum vulgaris e Triticum vulgaris succinilado. O padrão obtido neste estudo é similar ao encontrado em intoxicação por plantas que apresentam swainsoniana como princípio tóxico.

Abstract Background Cystic ovarian disease is an important cause of infertility that affects bovine, ovine, caprine and porcine species and even human beings. Alterations in the ovarian micro-environment of females with follicular cysts could alter the normal processes of proliferation and programmed cell death in ovarian cells. Thus, our objective was to evaluate apoptosis and proliferation in ovarian cystic follicles in rats in order to investigate the cause of cystic follicle formation and persistence. Methods We compared the number of in situ apoptotic cells by TUNEL assay, expression of active caspase-3 and members of Bcl-2 family by immunohistochemistry; and cell proliferation by the expression of the proliferation markers: PCNA and Ki-67. Results The proliferation index was low in granulosa of tertiary and cystic follicles of light exposed rats when compared with tertiary follicles of control animals, while in theca interna only cystic follicles presented low proliferation index when compared with tertiary follicles (p < 0.05). The granulosa of cysts exhibited a similar cell DNA fragmentation to early atretic follicles. In the granulosa and theca interna, active caspase-3 shown similar immunostaining levels in tertiary and cystic follicles (p < 0.05). The granulosa cells presented high expression of Bcl-2, Bcl-xL and Bcl-w in the tertiary and cystic follicles with diminishing intensity in the atretic follicles, except with Bcl-w where the intensity was maintained in the atretic follicles (p < 0.05). The expression of Bax was weak in the healthy and cystic follicles. In the theca interna, Bcl-2 expression was the same as the pattern found in the granulosa; no differences were found between tertiary and cystic follicles from both groups for Bcl-xL and Bcl-w. The expression of Bax in this layer was higher in the tertiary follicles of the treated animals (p < 0.05) while the values for cystic follicles were similar to those in the tertiary follicles of controls. The theca externa showed low expression of the pro and anti-apoptotic proteins. Conclusion These results show that the combination of weak proliferation indices and low apoptosis observed in follicular cysts, could explain the cause of the slow growth of cystic follicles and the maintenance of a static condition without degeneration, which leads to their persistence. These alterations may be due to structural and functional modifications that take place in these cells and could be related to hormonal changes in animals with this condition.

Solanum glaucophyllum, a toxic plant known for its calcinogenic effects, causes enzootic calcinosis in ruminant and monogastric animals. We describe an outbreak of enzootic calcinosis that occurred in a herd of 110 horses grazing pastureland heavily contaminated with S. glaucophyllum in Buenos Aires province, Argentina. Ten horses developed clinical signs, and 6 horses died. Clinical signs included abnormal gait (stiff-legged action, short strides), stiffness, thoracolumbar kyphosis, reluctance to move, wide stance, chronic weight loss, weakness, recumbency, and difficulty standing. Autopsy of 2 horses revealed severe mineralization of the aorta, pulmonary arteries, heart, and lungs, consistent with enzootic calcinosis. Although horses usually have very selective grazing behavior, under food restriction conditions, they can ingest the toxic plants and can develop the disease. Enzootic calcinosis should be considered as a differential diagnosis in horses grazing S. glaucophyllum–invaded pasturelands with compatible clinical signs and lesions. [ABSTRACT FROM AUTHOR]

The article discusses a study analyzing the presence of cytoskeletal proteins in the follicular wall in the normal and cystic ovaries of the sows and proves that the development of ovarian cysts in sows is related to the changes in the cytoskeletal proteins CK and vimentin.

Background: The spread of the invasive snail Pomacea canaliculata is expanding the rat lungworm disease beyond its native range. Their toxic eggs have virtually no predators and unusual defenses including a neurotoxic lectin and a proteinase inhibitor, presumably advertised by a warning coloration. We explored the effect of egg perivitellin fluid (PVF) ingestion on the rat small intestine morphology and physiology. Methodology/Principal Findings: Through a combination of biochemical, histochemical, histopathological, scanning electron microscopy, cell culture and feeding experiments, we analyzed intestinal morphology, growth rate, hemaglutinating activity, cytotoxicity and cell proliferation after oral administration of PVF to rats. PVF adversely affects small intestine metabolism and morphology and consequently the standard growth rate, presumably by lectin-like proteins, as suggested by PVF hemaglutinating activity and its cytotoxic effect on Caco-2 cell culture. Short-term effects of ingested PVF were studied in growing rats. PVF-supplemented diet induced the appearance of shorter and wider villi as well as fused villi. This was associated with changes in glycoconjugate expression, increased cell proliferation at crypt base, and hypertrophic mucosal growth. This resulted in a decreased absorptive surface after 3 days of treatment and a diminished rat growth rate that reverted to normal after the fourth day of treatment. Longer exposure to PVF induced a time-dependent lengthening of the small intestine while switching to a control diet restored intestine length and morphology after 4 days. Conclusions/Significance: Ingestion of PVF rapidly limits the ability of potential predators to absorb nutrients by inducing large, reversible changes in intestinal morphology and growth rate. The occurrence of toxins that affect intestinal morphology and absorption is a strategy against predation not recognized among animals before. Remarkably, this defense is rather similar to the toxic effect of plant antipredator strategies. This defense mechanism may explain the near absence of predators of apple snail eggs. A hunted free-ranging female red deer (Cervus elaphus) from a region near the Nahuel Huapi National Park, Northern Patagonia, Argentina, had a focally extensive peribronchial lymphoid proliferative lesion in the lung characterized by formation of multiple follicles, with prominent germinal centers lacking mantle zone cells and antigen-related polarity. On examination of immunohistochemically stained tissues, a predominance of B cells (cluster of differentiation [CD]20 positive) with only a few scattered T cells (CD3 positive) were present. The histologic and immunohistochemical characteristics are consistent with follicular lymphoma, which is frequently seen in human beings and less frequently in domestic animals. Commerson's dolphins Cephalorhynchus c. commersonii are the small cetacean most frequently found stranded along the coast of Tierra del Fuego, Argentina, but little is known about their pathologies. We examined the postcranial axial skeleton of 425 Commerson's dolphins collected in the period 1974-2011 for the presence of pathologies. Miscellaneous lesions were detected in 107 (25.2%) of the specimens. Among them, idiopathic hyperostosis was the most frequent pathology (73.8%), followed by spondyloarthropathy (40.2%). Traumata (25.2%), spondylitis (10.3%) and osteoarthritis (11.2%) occurred less frequently. The type of lesion was statistically associated with the region of the vertebral column. Idiopathic hyperostosis occurred significantly more frequently in the lumbar vertebrae than in other parts of the vertebral column, while spondyloarthropathy was more frequent in the thoracic and caudal vertebrae. Both conditions were predominantly seen in mature dolphins. Osteoarthritis affected the transverse processes of the thoracic vertebrae of 6 males, 3 females and 3 individuals of undetermined sex, all mature. Healed and unhealed fractures were observed in the ribs of 27 specimens and in the transverse processes of 3 others. Spondylitis of various degrees of severity affected the cervical, thoracic, lumbar and caudal regions of 11 individuals. Together these data indicate that bone lesions and traumata commonly occur in Commerson's dolphins, likely causing pain and morbidity in severe cases.
