Southern Africa, and in particular its western part, Namibia, is considered a climate change hotspot by the IPCC and is at risk of severe temperature-related changes. As examples, models projections suggest by the end of the century: 1) Increase of surface temperature of +4 to +7°C; 2) destruction of the stratocumulus cloud deck along the western coast; and 3) decrease of the fog inland. The role of aerosols in this context is still not completely quantified, due to their high spatial and temporal variability, the many sources responsible for the complex mixture and the lack of continuous observations. Most of the past regional climate modelling (RCM) and intensive field campaigns focussed primarily on constraining the radiative effects of the seasonal biomass burning aerosols generally occurring in the end of the austral winter (August to October). However, the aerosol spectral optical properties, aerosol optical depth and the organic fractions are not studied enough to provide a firm understanding of regional aerosol load, interaction with radiation and interplays in relation to particle chemistry.In this work, we present the first analysis of new long-term ground-based aerosol observations conducted since april 2022 at Gobabeb Namib Research Institue (23°33’40‘’S, 15°02’24’’E) in Namibia. Gobabeb is located in the hyperarid Namib desert and it is under the influence of different air masses, transporting maritime and biomass burning aerosols, amongst others. The measurements of optical and physical properties of the aerosol mixtures are analysed to provide with the aerosol single scattering albedo and mass absorption, scattering and extinction efficiencies which are needed to evaluate the first Africa-based Earth system model by the Global Change Institute at the University of Witwatersrand in South Africa. The in situ surface data are complemented by the analysis of pre-existing observations such as those from the AERONET sunphotometers.

Plants have evolved unique morphological and developmental adaptations to cope with the abiotic stresses imposed by (hyper)arid environments. Such adaptations include the formation of rhizosheath–root system in which mutualistic plant–soil microbiome associations are established: the plant provides a nutrient-rich and shielded environment to microorganisms, which in return improve plant-fitness through plant growth promoting services. We hypothesized that the rhizosheath–root systems represent refuge niches and resource islands for the desert edaphic microbial communities. As a corollary, we posited that microorganisms compete intensively to colonize such “oasis” and only those beneficial microorganisms improving host fitness are preferentially selected by plant. Our results show that the belowground rhizosheath–root micro-environment is largely more hospitable than the surrounding gravel plain soil with higher nutrient and humidity contents, and cooler temperatures. By combining metabarcoding and shotgun metagenomics, we demonstrated that edaphic microbial biomass and community stability increased from the non-vegetated soils to the rhizosheath–root system. Concomitantly, non-vegetated soil communities favored autotrophy lifestyle while those associated with the plant niches were mainly heterotrophs and enriched in microbial plant growth promoting capacities. An intense inter-taxon microbial competition is involved in the colonization and homeostasis of the rhizosheath zone, as documented by significant enrichment of antibiotic resistance genes and CRISPR-Cas motifs. Altogether, our results demonstrate that rhizosheath–root systems are “edaphic mini-oases” and microbial diversity hotspots in hyperarid deserts. However, to colonize such refuge niches, the desert soil microorganisms compete intensively and are therefore prepared to outcompete potential rivals.

In a recent study, we found a distinct soil bacterial community associated with male and female plants of the desert gymnosperm Welwitschia mirabilis. In this subsequent study, we also found that the soil fungal community associated with Welwitschia differs between male and female plants, and between unvegetated areas and the soil under plants. Site location, pH, and soil moisture also had an important influence on the composition of the fungal community. A number of Ascomycota and Chytrid species were found to be distinct indicators of male and female plants, respectively, but there was no overall difference at the phylum level or in terms of diversity. The unvegetated areas between plants also differed in terms of several Ascomycota OTUs. Network connectivity of the fungal communities was found to be higher under both male and female Welwitschia plants than in unvegetated control areas. As with the bacterial community, it is unclear what processes produce the gender-distinct fungal community, and also the more general plant-associated community, and also what the effects on the biology of the plants are. One possibility behind the gender-related difference in fungal community is that there are differences in the production of pollen or nectar between the two plant genders, affecting the below-ground soil community.

Salt pans or playas, which are saline-rich springs surrounded by halite evaporates in arid environments, have played an essential role in landscape erosion during the formation of the Namib Desert and are numerous in its central region. In this study, we used shotgun metagenomics to investigate the phylogenetic and functional capacities of the microbial communities from two salt pans (namely, Eisefeld and Hosabes) located in the Central Namib Desert, located in Southwest Africa. We studied the source and sink sediment mat communities of the saline streams, as well as those from two halites (crystallized structures on the stream margins). The microbial assemblages and potential functions were distinct in both niches. Independently from their localization (Eisfeld vs Hosabes and source vs sink), the sediment mat communities were dominated by members of theAlpha-andGamma-proteobacteriaclasses, while halites were Archaea-dominated and also contained high abundances of the extremely halophilic bacteriumSalinibactersp. (phylumBacteroidota). Photoheterotrophy and chemoheterotrophy were the principal lifestyles in both niches, with halite communities having a reduced diversity of metabolic pathways. Intense microbial-virus interactions in both niches were implied by the widespread detection of CRISPR-Cas defense systems. We identified a putatively novel clade of type II CRISPR-Cas systems, as well as novel candidate viral lineages of the class Caudoviricetes and of Halobacteriales-infecting haloviruses. Putative gene transfer agent-like sequences within theAlphaproteobacteriawere identified in the sediment mat communities. These horizontal gene transfer elements have the potential to drive genome plasticity and evolution of theAlphaproteobacteriain the Namib Desert salt pan microbiomes.ImportanceThe hyperarid Namib Desert is one of the oldest deserts on Earth. It contains multiple clusters of playas which are saline-rich springs surrounded by halite evaporites. Playas are of great ecological importance and their indigenous (poly)extremophilic microorganisms are potentially involved in the precipitation of minerals such as carbonates and sulfates and have been of great biotechnological importance. While there has been a considerable amount of microbial ecology research preformed on various Namib Desert edaphic microbiomes, little is known about the microbial communities inhabiting its multiple playas. In this work, we therefore provide a comprehensive taxonomic and functional potential characterization of the microbial, including viral, communities of sediment mats and halites from two distant Namib Desert, contributing towards a better understanding of the ecology of this biome.

The Namib Turbulence Experiment (NamTEX) was a multinational micrometeorological campaign conducted in the central Namib Desert to investigate three-dimensional surface layer turbulence and the spatiotemporal patterns of heat transfer between the subsurface, surface, and atmosphere. The Namib provides an ideal location for fundamental research that revisits some key assumptions in micrometeorology that are implicitly included in the parameterizations describing energy exchange in weather forecasting and climate models: homogenous flat surfaces, no vegetation, little moisture, and cloud-free skies create a strong and consistent diurnal forcing, resulting in a wide range of atmospheric stabilities. A novel combination of instruments was used to simultaneously measure variables and processes relevant to heat transfer: a 3-km fiber-optic distributed temperature sensor (DTS) was suspended in a pseudo-three-dimensional array within a 300 m × 300 m domain to provide vertical cross sections of air temperature fluctuations. Aerial and ground-based thermal imagers recorded high-resolution surface temperature fluctuations within the domain and revealed the spatial thermal imprint of atmospheric structures responsible for heat exchange. High-resolution soil temperature and moisture profiles together with heat flux plates provided information on near-surface soil dynamics. Turbulent heat exchange was measured with a vertical array of five eddy-covariance point measurements on a 21-m mast, as well as by collocated small- and large-aperture scintillometers. This contribution first details the scientific goals and experimental setup of the NamTEX campaign. Then, using a typical day, we demonstrate (i) the coupling of surface layer, surface, and soil temperatures using high-frequency temperature measurements, (ii) differences in spatial and temporal standard deviations of the horizontal temperature field using spatially distributed measurements, and (iii) horizontal anisotropy of the turbulent temperature field.

An intensive observation period was conducted in September 2017 in the central Namib, Namibia, as part of the project Namib Fog Life Cycle Analysis (NaFoLiCA). The purpose of the field campaign was to investigate the spatial and temporal patterns of the coastal fog that occurs regularly during nighttime and morning hours. The fog is often linked to advection of a marine stratus that intercepts with the terrain up to 100 km inland. Meteorological data, including cloud base height, fog deposition, liquid water path, and vertical profiles of wind speed/direction and temperature, were measured continuously during the campaign. Additionally, profiles of temperature and relative humidity were sampled during five selected nights with stratus/fog at both coastal and inland sites using tethered balloon soundings, drone profiling, and radiosondes. This paper presents an overview of the scientific goals of the field campaign; describes the experimental setup, the measurements carried out, and the meteorological conditions during the intensive observation period; and presents first results with a focus on a single fog event.

Functional structure and diversity of soil free-living nematodes in a desert environment depend on plant gender and sampling site. The objective of this study was to compare the composition, abundance and tropic group of soil free-living nematodes in the upper 0–10 cm soil layer under the male and female Acanthosicyos horridus Welw. ex Hook. f. plants and in the inter-shrub open areas (control) in the Namib Desert, Namibia in April 2015. Soil moisture, organic matter (OM) and pH was also analyzed. Free-living nematodes were extracted from 100 g soil using the Baermann funnel procedure, and total number of nematodes was counted under a microscope. Community composition and diversity of soil free-living nematodes were analyzed using 18S rDNA sequences. Results indicated that a total of 67 groups, including 64 species, 2 genera and 1 family were identified. Feeding behavior of 58 species were identified as follows: 15 bacteria-feeding species, 12 fungi-feeding species, 10 plant-parasite species, 5 omnivorous-predator species, 8 animal-parasite species, 5 invertebrate-parasite species and 3 non-free-living nematodes, known as marine species. Moreover, soil free-living nematodes were found to be affected by sampling locations and plant gender, and community composition and density of these nematodes were strongly influenced by soil OM content. Result confirmed that spatial location and plant cover were main factors influencing the diversity of soil free-living nematodes. Moreover, molecular tools were found to be very useful in defining the richness of soil non-free-living nematodes. In conclusion, the results elucidated the importance of biotic variables in determining the composition and abundance of soil free-living nematodes in the Namib Desert, Namibia.

Soil nematodes are fundamentally aquatic animals, requiring water to move, feed, and reproduce. Nonetheless, they are ubiquitous in desert soils because they can enter an anhydrobiotic state that allows them to persist when water is biologically unavailable. In the hyper-arid Namib Desert of Namibia, rain is rare, but fog routinely moves inland from the coast and supports plant and animal life. Very little is understood about how this fog may affect soil organisms. We investigated the role of fog moisture in the ecology of free-living, soil nematodes across an 87-km fog gradient in the gravel plains of the Namib Desert. We found that nematodes emerged from anhydrobiosis and became active during a fog event, suggesting that they can utilize fog moisture to survive. Nematode abundance did not differ significantly across the fog gradient and was similar under shrubs and in interplant spaces. Interplant soils harbor biological soil crusts that may sustain nematode communities. As fog declined along the gradient, nematode diversity increased in interplant soils. In areas where fog is rare, sporadic rainfall events can stimulate the germination and growth of desert ephemerals that may have a lasting effect on nematode diversity. In a 30-day incubation experiment, nematode abundance increased when soils were amended with water and organic matter. However, these responses were not evident in field samples, which show no correlations among nematode abundance, location in the fog gradient, and soil organic matter content. Soil nematodes are found throughout the Namib Desert gravel plains under a variety of conditions. Although shown to be moisture- and organic matter-limited and able to use moisture from the fog for activity, variation in fog frequency and soil organic matter across this unique ecosystem may be biologically irrelevant to soil nematodes in situ.

Rainfall is rare in hyperarid deserts but, when it occurs, it triggers large biological responses which are considered to be essential for the long-term maintenance of biodiversity. In such environments, microbial communities have major roles in nutrient cycling, but their functional responses to short-lived resource opportunities are poorly understood. We used whole community metatranscriptomic data to demonstrate structured and sequential functional responses in desiccated desert soils to a simulated rainfall event over a seven-day time frame. Rapid transcriptional activation of Actinobacteria, Alpha-proteobacteria and phage transcripts was followed by a marked increase in protist and myxobacterial activity, before returning to the original state. In functional terms, motility systems were activated in the early phases, whereas competition-toxicity systems increased in parallel to the activity from predators and the drying of soils. The dispersal-predation dynamic was identified as a central driver of microbial community responses to watering. Carbon fixation mechanisms that were active under dry condition were rapidly down-regulated in wetted soils, and only reactivated on a return to a near-dry state, suggesting a reciprocal balance between carbon fixation and fixed-carbon acquisition processes. Water addition induced a general reduction in the transcription of stress response genes, most prominently HSP20, indicating that this chaperone is particularly important for life in desiccated ecosystems. Based on these data, we propose a rainfall response model for desert soil microbiomes.

13 páginas, 8 figuras, The ventral surfaces of translucent rocks from hot desert pavements often harbor hypolithic microbial communities, which are mostly dominated by cyanobacteria. The Namib Desert fog belt supports extensive hypolithic colonization of quartz rocks, which are also colonized by lichens on their dorsal surfaces. Here, we aim to evaluate whether lichens colonize the ventral surface of the rocks (i.e., show hypolithic lifestyle) and compare the bacterial composition of these coastal hypolithic communities with those found inland. Fungal DNA barcoding and fungal and bacterial Illumina metabarcoding were combined with electron microscopy to characterize the composition and spatial structure of hypolithic communities from two (coastal and inland) areas in the Namib Desert. We report, for the first time, the structure and composition of lichen-dominated hypolithic communities found in the coastal zone of the Namib Desert with extensive epilithic lichen cover. Lichen modified areoles with inverted morphology of the genus Stellarangia (three lineages) and Buellia (two lineages) were the main components of these hypolithic communities. Some of these lineages were also found in epilithic habitats. These lichen-dominated hypolithic communities differed in structural organization and bacterial community composition from those found in inland areas. The hypolithic lichen colonization characterized here seems not to be an extension of epilithic or biological soil crust lichen growths but the result of specific sublithic microenvironmental conditions. In a xeric environment, plant ecophysiological adaptation helps determine soil microbial-community structure beneath plant canopies. We compared the plant-associated microbiota of the dioecious perennial plant Acanthosicyos horridus (Nara) in the Namib Desert in relation to soil parameters, climate, and plant gender. Abiotic soil attributes (soil moisture, total organic matter, salinity) and plant gender were among the main factors correlating with soil microbial-community biomass, basal respiration, and substrate-induced respiration rates. These varied between an extremely arid Far East (FE) site and a less arid site in the sandy, saline delta ecosystem. A. horridus gender was correlated with soil microbiome composition and activity, and varied between the relatively humid and more xeric environments. This study highlighted the apparent role of plant dimorphism in determining soil biotic composition and diversity in a desert ecosystem; however, the cause of the relationship between plant gender and microbial community remains uncertain. Possible explanations include gender-related variation in the plant itself, a link to certain abiotic soil conditions that incidentally influence plant gender, or a combination of both. This is the first example of gender-related differences in microbiota reported in a plant from an arid environment, and only the second example from the plant world in general.

Plant clusters govern soil microbiology in desert systems. Female Acanthosicyos horridus plants in the Namib Desert are preferentially accessed by grazers, especially during fruit availability. We hypothesized that this differential grazing affects the taxonomic diversity of soil bacterial populations. Sampling was carried out at three locations on a northwest-to-southeast transect starting at the Kuiseb Delta, near the shores of the Atlantic in the western Namib Desert, and extending inland over a distance of 140 km. Analyses of the soil samples showed that proximity to the sea had a strong impact on soil salinity. Soil pH and organic matter content were generally not significantly correlated with the presence or absence of plants and varied little and non-uniformly along the transect. A. horridus presence led to distinct under-canopy bacterial-community diversity in contrast with the non-vegetated spaces between shrubs. However, plant gender has only a marginal, statistically insignificant impact on the bacterial-diversity properties, thus not supporting our hypothesis. Therefore, the taxonomic diversity of the bacterial community in Namib Desert soils vegetated with A. horridus is primarily governed by the presence or absence of plants and by proximity to the ocean.

In hyper-arid soil environments, photosynthetic microorganisms are largely restricted to hypolithic (sub-lithic) habitats: i.e., on the ventral surfaces of translucent pebbles in desert pavements. Here, we combined fluorometric, spectroscopic, biochemical and metagenomic approaches to investigate in situ the light transmission properties of quartz stones in the Namib Desert, and assess the photosynthetic activity of the underlying hypolithic cyanobacterial biofilms. Quartz pebbles greatly reduced the total photon flux to the ventral surface biofilms and filtered out primarily the short wavelength portion of the solar spectrum. Chlorophylls d and f were not detected in biofilm pigment extracts; however, hypolithic cyanobacterial communities showed some evidence of adaptation to sub-lithic conditions, including the prevalence of genes encoding Helical Carotenoid Proteins, which are associated with desiccation stress. Under water-saturated conditions, hypolithic communities showed no evidence of light stress, even when the quartz stones were exposed to full midday sunlight. This initial study creates a foundation for future in-situ and laboratory exploration of various adaptation mechanisms employed by photosynthetic organisms forming hypolithic microbial communities.

Desert and semi-desert plants are often associated with a distinct soil biota under the plants and close to the root system. We aimed to understand if similar effects could be found in the taxonomically isolated desert gymnosperm Welwitschia mirabilis in the Namib Desert, and whether this island effect varied with climate and with gender of plants. We took soil cores adjacent to the plants in environments ranging from extreme desert to arid shrubland, and in nearby control sites between the plants. Soil chemistry was analysed, and deoxyribonucleic acid was extracted and sequenced for the bacterial 16s region. Soil under the plants was richer in organic C, N and moisture. Despite the range of climates, the soil around Welwitschia plants was consistently associated with a particular bacterial community composition that was distinct from samples further away. Compared to unvegetated control patches, bacterial diversity close to the plants was reduced. In the plant-associated soil community, there was a clear gender effect across all sites with a distinct community composition and greater diversity under male plants. It is unclear what differences in the soil environment might be producing these gender-associated differences, which provide an additional dimension to the fertile island effect.

Studies of microbial biogeography are often convoluted by extremely high diversity and differences in microenvironmental factors such as pH and nutrient availability. Desert endolithic (inside rock) communities are exceptionally simple ecosystems that can serve as a tractable model for investigating long-range biogeographic effects on microbial communities. We conducted a comprehensive survey of endolithic sandstones using high-throughput marker gene sequencing to characterize global patterns of diversity in endolithic microbial communities. We also tested a range of abiotic variables in order to investigate the factors that drive community assembly at various trophic levels. Macroclimate was found to be the primary driver of endolithic community composition, with the most striking difference witnessed between hot and polar deserts. This difference was largely attributable to the specialization of prokaryotic and eukaryotic primary producers to different climate conditions. On a regional scale, microclimate and properties of the rock substrate were found to influence community assembly, although to a lesser degree than global hot versus polar conditions. We found new evidence that the factors driving endolithic community assembly differ between trophic levels. While phototrophic taxa were rigorously selected for among different sites, heterotrophic taxa were more cosmopolitan, suggesting that stochasticity plays a larger role in heterotroph assembly. This study is the first to uncover the global drivers of desert endolithic diversity using high-throughput sequencing. We demonstrate that phototrophs and heterotrophs in the endolithic community assemble under different stochastic and deterministic influences, emphasizing the need for studies of microorganisms in context of their functional niche in the community.

The Namib Desert is one of the world's only truly coastal desert ecosystem. Until the end of the 1st decade of the twenty-first century, very little was known of the microbiology of this southwestern African desert, with the few reported studies being based solely on culture-dependent approaches. However, from 2010, an intense research program was undertaken by researchers from the University of the Western Cape Institute for Microbial Biotechnology and Metagenomics, and subsequently the University of Pretoria Centre for Microbial Ecology and Genomics, and their collaborators, led to a more detailed understanding of the ecology of the indigenous microbial communities in many Namib Desert biotopes. Namib Desert soils and the associated specialized niche communities are inhabited by a wide array of prokaryotic, lower eukaryotic and virus/phage taxa. These communities are highly heterogeneous on both small and large spatial scales, with community composition impacted by a range of macro- and micro-environmental factors, from water regime to soil particle size. Community functionality is also surprisingly non-homogeneous, with some taxa retaining functionality even under hyper-arid soil conditions, and with subtle changes in gene expression and phylotype abundances even on diel timescales. Despite the growing understanding of the structure and function of Namib Desert microbiomes, there remain enormous gaps in our knowledge. We have yet to quantify many of the processes in these soil communities, from regional nutrient cycling to community growth rates. Despite the progress that has been made, we still have little knowledge of either the role of phages in microbial community dynamics or inter-species interactions. Furthermore, the intense research efforts of the past decade have highlighted the immense scope for future microbiological research in this dynamic, enigmatic and charismatic region of Africa.

International audience; Atmospheric aerosols contribute significantly to the uncertainty in radiative forcing effects that influence the climate as well as posing a significant health risk to humans. The climatic implications of aerosols are dependent on many variables, including aerosol size, shape, chemical composition, and position in the atmospheric column. The radiative impact of aerosols transported over the west coast of southern Africa has been found, in particular, to be complicated by the aforementioned aerosol properties. This study investigated the columnar optical properties of aerosols over Gobabeb, Namibia (23.5621° S, 15.0409° E, 405 m asl) using sunphotometer data between December 2014 and November 2015. Aerosol mean optical depth AOD500 had its maximum and minimum values in 2015 August (0.37±0.30) and June (0.06±0.02), respectively. The Angström parameter was mostly above unity during the study period and indicated the prevalence of fine particles for the most part of the year with maximum and minimum values observed in August 2015 (1.44±0.19) and December 2014 (0.57±0.19), respectively. The columnar water vapor was highest in January (2.62±0.79) and lowest in June (0.76±0.27). The volume size distribution showed the fine particles having a mean radius of about 0.16 μm and the coarse mode had variation in sizes with a radius ranging between 3 μm and 7 μm. The single scattering albedo at visible wavelengths ranged between 0.83 and 0.91. The phase function was high at small angles but minimum at about 140° in all seasons. The radiative forcing showed a heating effect in all seasons with maximum and minimum in winter (9.41 Wm-2) and autumn (3.64 Wm-2), respectively. Intercomparison of the sunphotometer data with the Moderate Resolution Imaging Spectroradiometer (MODIS) showed that the satellite sensor overestimates the aerosol loading compared to the ground-based sunphotometer measurements. Both sets of observations were better correlated during the spring and winter seasons than for the summer and autumn.

In this book chapter, we focus on the research performed to date on the microbiology and microbial ecology of the Namib Desert, one of the oldest hyperarid deserts on Earth. Microbial communities consist of complex assemblages of interacting microorganisms, ranging from viruses to small eukaryotes (such as nematodes and springtails) and these microbial community members have all been studied, to a certain extent, in the Namib Desert. It is worth noting that most of the microbial ecology research performed in the Namib Desert has taken place in the central Namib Desert, in the vicinity of the Gobabeb Research and Training Center ( http://www.gobabebtrc.org/ ). Nevertheless, in this chapter, we have attempted to review all the scientific literature available on the microbiology and microbial ecology of the Namib Desert.

Hot desert surface soils are characterized by extremely low water activities for large parts of any annual cycle. It is widely assumed that microbial processes in such soils are very limited. Here we present the first metatranscriptomic survey of microbial community function in a low water activity hyperarid desert soil. Sequencing of total mRNA revealed a diverse and active community, dominated by Actinobacteria. Metatranscriptomic analysis of samples taken at different times over 3 days indicated that functional diel variations were limited at the whole community level, and mostly affected the eukaryotic subpopulation which was induced during the cooler night hours. High levels of transcription of chemoautotrophic carbon fixation genes contrasted with limited expression of photosynthetic genes, indicating that chemoautotrophy is an important alternative to photosynthesis for carbon cycling in desiccated desert soils. Analysis of the transcriptional levels of key N-cycling genes provided strong evidence that soil nitrate was the dominant nitrogen input source. Transcriptional network analyses and taxon-resolved functional profiling suggested that nutrient acquisition processes, and not diurnal environmental variation, were the main drivers of community activity in hyperarid Namib Desert soil. While we also observed significant levels of expression of common stress response genes, these genes were not dominant hubs in the co-occurrence network.

Here we present the first metatranscriptomic survey of microbial community function in a low water activity hyperarid desert soil. Sequencing of total mRNA revealed a diverse and active community, dominated byActinobacteria. Metatranscriptomic analysis of samples taken at different times over three days indicated that most functions did not fluctuate on a diel basis, except for a eukaryotic subpopulation which was induced during the cooler night hours. High levels of transcription of chemoautotrophic carbon fixation genes contrasted with limited expression of photosynthetic genes, indicating that chemoautotrophy is an important alternative to photosynthesis for carbon cycling in desiccated desert soils. Analysis of the transcriptional levels of key N-cycling genes provided strong evidence that soil nitrate was the dominant nitrogen input source. Transcriptional network analyses and taxon-resolved functional profiling suggested that nutrient acquisition processes, and not diurnal environmental variation, were the main drivers of community activity in hyperarid Namib Desert soil. While we also observed significant levels of expression of common stress response genes, these genes were not dominant hubs in the co-occurrence network.

Aim To assess spatial patterns of genetic and species-level diversity for Namib Desert Collembola using mitochondrial DNA cytochrome c oxidase subunit I (COI) gene sequences. Location Namib Desert gravel plains. Taxon Collembola (springtails). Methods A total of 77 soil samples were collected along NE-SW (60 km) and E-W (160 km) transects from within a 4,000 km2 area of the Namib Desert gravel plains. We extracted 434 springtails from the 37 samples which contained Collembola and sequenced them at the COI gene locus. In the absence of specific taxonomic keys and previous genetic data for these taxa, we used Generalized Mixed Yule Coalescent (GMYC) analyses to provide putative species-level designations. Results We obtained 341 successful COI sequences, 175 of which were unique haplotypes. GMYC analyses identified 30 putative species, with up to 28% sequence divergence (uncorrected p-distance). The distribution of genetic variants was disjunct, with 97% of haplotypes and 70% of "GMYC species" found only at single sites. Main conclusions Dispersal events, although rare, may be facilitated by environmental events such as prevailing onshore winds or occasional flow of rainwater to the coast. We conclude that the high genetic diversity we observed is the result of ancient springtail lineages, patchy distribution of suitable habitats, and limited dispersal (gene flow) among habitable locations.

There is increasing interest in using &ldquo, real-world pedagogy&rdquo, to train students in ways that make them better able to contribute toward a more sustainable society. While there is a robust body of literature on the competencies that students need as sustainability professionals, there is a lack of specific guidance in the literature on how to teach for competency development or on how to structure a program or course to support competency development. Our research addresses this gap in the literature through a description and autoethnographic reflection on the design and early implementation of a &ldquo, real-world&rdquo, course. The course is from the Environmental Studies Program at Dartmouth College (Hanover, NH, USA), but it takes place in the environs of the Gobabeb Research and Training Centre in the Namib Desert of Namibia and in nearby Topnaar settlements. Our research objective was to articulate strategies to address the primary pedagogical challenges that we faced during the design and first five iterations of the course. These include: How do we frame this course and communicate it to students in a way that is understandable and works within the particular context and constraints of the course? Can we provide students with a coherent framework that helps them to understand the approach and also provides a platform for thoughtful consideration, acquisition, and retention of appropriate competencies? How do we develop collaborations with our community partners that are ethical and effective? How do we frame these real-world experiences in a way that allows for students to integrate their experience with the theory and broader empiricism they learn on campus? To address these pedagogic challenges, we framed the course as a research-based course, more specifically community-based research (CBR), conducted in a social-ecological system (SES). We developed lower-level strategies for implementing this framing, including preparing students for collaborative research, encouraging student ownership of their learning, linking theory to research, and thoughtfully navigating time constraints. Furthermore, program-level and student-level engagement with community have been critical for avoiding becoming &ldquo, helicopter researchers.&rdquo, Drawing on our personal reflections and those of our community partners, we conclude with a discussion of emergent outcomes and the next steps for continual improvement and adaptation.

The hyper-arid central region of the Namib Desert is characterized by quartz desert pavement terrain that is devoid of vascular plant covers. In this extreme habitat the only discernible surface covers are epilithic lichens that colonize exposed surfaces of quartz rocks. These lichens are highly susceptible to disturbance and so field surveys have been limited due to concerns about disturbing this unusual desert feature. Here we present findings that illustrate how non-destructive surveys based upon advanced photogrammetry techniques can yield meaningful and novel scientific data on these lichens. We combined 'structure from motion analysis,' computer vision and GIS to create 3-dimensional point clouds from two-dimensional imagery. The data were robust in its application to estimating absolute lichen cover. An orange Stellarangia spp. assemblage had coverage of 22.8% of available substrate, whilst for a black Xanthoparmelia spp. assemblage coverage was markedly lower at 0.6% of available substrate. Hyperspectral signatures for both lichens were distinct in the near-infra red range indicating that Xanthoparmelia spp. was likely under relatively more moisture stress than Stellarangia spp. at the time of sampling, and we postulate that albedo effects may have contributed to this in the black lichen. Further transformation of the data revealed a colonization preference for west-facing quartz surfaces and this coincides with prevailing winds for marine fog that is the major source of moisture in this system. Furthermore, a three-dimensional 'fly through' of the lichen habitat was created to illustrate how the application of computer vision in microbiology has further potential as a research and education tool.
