14. Microsporidian heavy infection in a batch of salted and dried cod.

Abstract

The aim of this work is the description and characterization of a severe microsporidian infection in a batch of salted and dried cod. Particularly, the case involves a batch of approximately 800 kg obtained from Gadus macrocephalus (Food and Agriculture Organization Zone 61 - Northwest Pacific Ocean), which, after rehydration and sectioning operations, underwent routine company checks before packaging. In about 20% of the samples, the presence of whitish nodules with a diameter ranging from 1 to 2 mm was observed on the surface of the fillets and in cross-section. The lesions ranged from a few units to 10 per cm2. Some samples were subjected to fresh microscopic observation with the stereomicroscope, confirming the nodular nature of the lesions, which were often confluent, alternating with empty spaces, giving the tissue a honeycombing aspect. The histological examination at low magnification allowed us to observe the heavy vacuolization of nodular lesions irregularly surrounded by a spongy-like wall. The observation at higher magnification of other sections allowed us to identify intra-myofibrillar cists containing presumptive microsporidian elements. The tissue damage derived from the technological processes and gravity of lesions did not allow a morphological characterization of presumptive protozoans. The molecular examination of the nodular lesions and the analysis of the sequence of an 897 bp fragment of the small subunit 16S rRNA revealed 100% identity with Microsporidium theragrae (GenBank accession number MT928885-89) first isolated from the skeletal muscles of Gadus chalcogrammus specimens from the Sea of Okhotsk. Abstract

Chemical composition and in vitro antimicrobial activity of parsley [Petroselinum crispum (Mill.) Fuss] and ginger (Zingiber officinale Roscoe) essential oils from store-bought parsley plants and ginger rhizomes were evaluated. A sensory analysis was performed on aged cheese slices treated with a mix of each essential oil and seed oil. Parsley essential oil was selected as the best compromise between sensory influence and antimicrobial activity and tested to evaluate its antifungal effects on cheese slices experimentally contaminated with Aspergillus flavus. Monoterpenes and α-zingiberene were the most abundant compounds in parsley and ginger essential oil, respectively. Both essential oils showed in vitro antimicrobial activity against different fungal species while only a bacterial strain (Enterococcus faecalis) was sensible to ginger essential oil. Only the cheese slices treated with parsley essential oil were appreciated by the panellists and its use has been effective in preventing the A. flavus growth in cheese. [ABSTRACT FROM AUTHOR]

Abstract

This study aimed at identifying species of staphylococci isolated from the oral cavity of dogs and cats, in Algeria, and to determine their antibiotic susceptibility.Oral swabs were collected from 70 healthy animals (35dogs and 35cats) and were grown in Mannitol Salt Agar medium.Isolates were identified using API staph commercial kits and then confirmed with MALDI-TOF MS associated with SARAMIS software.Coagulase-positive staphylococci (CoPS) were tested for antibiotic susceptibility by disk diffusion method.On the 70 sampled animals, 59 were carriers of staphylococci in the oral cavity and more than one species was detected in 11 of them. Seventy (70) staphyloccoccal isolates were obtained belonging to 10 defferent species. Coagulase-negative staphylococci (CoNS) were most prevalent (81.42% ),of which the dominant species was S. xylosus (40%).Other coagulase-negative species,such as S, simulans (14.28%), S ,sciuri (11.42%) , S. saprophyticus (10%),S. capitis (2.85%),S, cohnii subsp cohnii (1.42%), and S, epidermidis (1.42%) were also isolated.The remaining n.13 CoPS included : S. intermedius (2.85%),S .pseudintermedius (8.57%) and S. intermedius group (SIG) (4.28%). No strain of S. aureus was found.Results of antimicrobial resistance showed that 61.53% of CoPS isolates were resistant to at least two drugs.The highest rate of resistance was observed against penicillin,ampicillin and tetracycline (53.84% for each drug),while amoxicillin-clavulanate was active on most isolates.In Algeria,Stray dogs and cats are carriers of many staphylococci species in the oral cavity, including multidrug resistant CoPS, which could be transmitted to humans through bites.

Abstract

Pesticide usage is becoming increasingly necessary to escalate agricultural productivity and meet food production needs. However, it harms in different degrees all living organisms, plants, and animals, whether terrestrial or aquatic. Soil microorganisms, are microbes belonging to microorganisms, are the first to be specifically affected by pesticides. This study aims to evaluate the impact of two herbicides, paraquat and glyphosate, on symbiotic nitrogen-fixing bacteria. Our study was carried out in the greenhouse. Bituminaria bituminosa plants were inoculated with four different nitrogen-fixing bacteria, Pantoea agglomerans, Rhizobium nepotum, Rhizobium radiobacter, and Rhizobium tibeticum, and then treated with varying herbicide concentrations were selected according to the doses recommended by the National Office of Food Safety (ONSSA) and according to a survey conducted among farmers in the Meknes region-Morocco, (0.05, 0.1, 5.4, 10.8 g/L glyphosate and 0.05, 0.1, 2, 4 g/L paraquat). After 6 months after sowing, the following parameters were evaluated: nodule number, nodule mass, nodule weight, nodule dry, and fresh weight, nitrogen content, and symbiotic efficiency. At higher doses (5.4, 10.8 g/L for glyphosate and 2, 4 g/L for paraquat), both herbicides decreased the number and the size of nodules, the weight of nodules, nitrogen content of Bituminaria bituminosa and symbiotic efficiency of the four different nitrogen-fixing bacteria studied. The effect of herbicides increased as the used concentration increased. The current research demonstrates that the decreased growth of herbicide-treated plants was caused by herbicides' direct effects on rhizobia rather than herbicides' indirect effects on Bituminaria bituminosa.
