Long-term observations of deep convective cloud (DCC) vertical velocity and mass flux were collected during the Observations and Modelling of the Green Ocean Amazon (GoAmazon2014/5) experiment. Precipitation echoes from a surveillance weather radar near Manaus, Brazil, are tracked to identify and evaluate the isolated DCC lifecycle evolution during the dry and wet seasons. A radar wind profiler (RWP) provides precipitation and air motion profiles to estimate the vertical velocity, mass flux, and mass transport rates within overpassing DCC cores as a function of the tracked cell lifecycle stage. The average radar reflectivity factor (Z), DCC area (A), and surface rainfall rate (R) increased with DCC lifetime as convective cells were developing, reached a peak as the cells matured, and decreased thereafter as cells dissipated. As the convective cells mature, cumulative DCC properties exhibit stronger updraft behaviors with higher upward mass flux and transport rates above the melting layer (compared with initial and later lifecycle stages). In comparison, developing DCCs have the lowest Z associated with weak updrafts, as well as negative mass flux and transport rates above the melting layer. Over the DCC lifetime, the height of the maximum downward mass flux decreased, whereas the height of the maximum net mass flux increased. During the dry season, the tracked DCCs had higher Z, propagation speed, and DCC area, and were more isolated spatially compared with the wet season. Dry season DCCs exhibit higher Z, mass flux, and mass transport rate while developing, whereas wet season DCCs exhibit higher Z, mass flux, and mass transport rates at later stages.

Dall’inizio del periodo pandemico, il personale sanitario è stato sottoposto a gravi difficoltà ed a continue emergenze, ed è sempre più esplicito il bisogno di avere un sostegno psicologico per gli operatori che affrontano tali disagi. In un'ottica di prevenzione nel campo della salute mentale per coloro che dovrebbero erogare ‘salute’, sarebbe auspicabile sviluppare una politica sanitaria che preveda l'inserimento strutturato della psicologia del lavoro e delle organizzazioni in contesti sanitari. In linea con gli obiettivi del "Nuovo sistema di garanzia per il monitoraggio dell'assistenza sanitaria" (del DM 12 marzo 2019 pubblicato in G.U. il 14 giugno 2019, operativo dal 1° gennaio 2020) che, nella prevenzione collettiva, descrive come indicatore di efficacia dei sistemi sanitari, il benessere organizzativo e luoghi di lavoro che erogano salute, si vuole avviare un’analisi del bisogno e delle criticità in merito. In questo studio ci si propone una prima review della letteratura sull’attuale presenza o assenza di interventi strutturati di psicologia del lavoro e delle organizzazioni nei sistemi sanitari, per analizzare le effettive politiche delle strutture sanitarie in merito, partendo dalla considerazione che non è semplice concretizzare tale presenza poiché aprirebbe le porte a quesiti su una verifica della reale efficacia della programmazione sanitaria, della modalità di reclutamento e di allocazione delle risorse umane e della leadership del management. Le suddette verifiche si rendono estremamente necessarie in quanto è dimostrato che l’inefficacia delle politiche sanitarie è una delle concause di malessere organizzativo, del burnout degli operatori ed una delle conseguenze della cosiddetta malasanità. Con l'obiettivo a lungo termine di uno sviluppo proattivo di una linea di attività specifica, distinta dalla psicologia clinica, nei contesti sanitari, qui si vuole esprimere una posizione innovativa utile agli operatori ed alle stesse aziende sanitarie che potrebbero beneficiare di un ruolo ed una funzione specifica che orienti, con la sua presenza e funzione, la cultura e la coscienza dell’organizzazione alla corretta gestione delle risorse umane e fornisca in modo sistematico quel necessario supporto psicologico agli operatori al fine di erogare benessere e non malessere agli utenti

Atmospheric aerosols affect the global energy budget by scattering and absorbing sunlight (direct effects) and by changing the microphysical structure, lifetime, and coverage of clouds (indirect effects). Both aerosol direct and indirect effects are affected by the vertical distribution of aerosols in the atmosphere, which is further influenced by a range of processes, such as aerosol dynamics, long-range transport, and entrainment. However, many observations of these processes are based on ground measurements, limiting our ability to understand the vertical distribution of aerosols and simulate their impact on clouds and climate. In this work, we examined the vertical heterogeneity of aerosols over the Atmospheric Radiation Measurement (ARM) Southern Great Plains (SGP) using data collected from the Holistic Interactions of Shallow Clouds, Aerosols and Land Ecosystems (HI-SCALE) campaign. The vertical profiles of meteorological and aerosol physiochemical properties up to 2500 m above are examined based on the 38 flights conducted during the HI-SCALE campaign. The aerosol properties over the SGP show strong vertical heterogeneity and seasonal variabilities. The aerosol concentrations at the surface are the highest due to strong emissions at ground level. In general, the mode diameter of aerosols during summer (∼ 100 nm) is larger than that during spring (∼ 30 nm), a result of enhanced condensational growth due to enriched volatile organic compounds in summer. The concentration of aerosols below 30 nm in the boundary layer (BL) (e.g., below 1000 m) during spring is higher than that during summer, a result of the stronger new particle formation (NPF) events due to the reduced condensation sink in spring. In the BL, the size of the aerosols gradually increases with altitude due to condensational growth and cloud processing. However, the chemical composition of the aerosols remained similar, with organics and sulfates representing 59.8 ± 2.2 % and 22.7 ± 2.1 %, respectively, of the total mass in the BL. Through the vertical profiles of aerosol properties, we observed NPF events in the upper BL during 7 out of 38 research flights, where the newly formed particles continue to grow as they are mixed down to the surface. There is also an indication that deep convection brings aerosols from the free troposphere (FT) to the surface, where they grow to contribute to the cloud condensation nuclei (CCN). Overall, the vertical heterogeneity of aerosols over the SGP is influenced by aerosol dynamics (new particle formation, growth, and cloud processing) and transport processes (mixing in the BL, long-range transport, entrainment, and convective downward transport). Case studies showing the influence of these factors are discussed.

Il benessere digitale è soprattutto frutto del lavoro di chi ha il compito di rendere l’esperienza di fruizione del virtuale il più confortevole possibile. Il benessere nel digitale è un atto volitivo, conoscitivo ed euristico durante il quale, a partire dall’identificazione dei nostri bisogni e desideri, proviamo a trovare una loro soddisfazione nel digitale. Se da un lato l’applicazione dei principi dell’ergonomia alla costruzione degli spazi digitali risulta fondamentale per promuovere il benessere digitale, dall’altra esistono diversi tipi di App che propongono ai fruitori esperienze di benessere variamente declinate. Risulta, comunque, di fondamentale importanza coniugare lo sviluppo di competenze personali di natura affettiva e cognitiva con la costruzione di habitat digitali centrati sulla persona al fine di garantire il Ben-Essere delle persone che, in maniera parziale o totale, abitano il virtuale.

Electrical machines for transportation applications are subjected to harsh environmental conditions during their operations. Partial discharge (PD), which is one of the main reasons for insulation failure, is greatly affected by ambient conditions (i.e., temperature, pressure, and humidity). Countless efforts are made for a comprehensive understanding of the physics of PD under variable environmental factors. This paper aims to review recent works addressing temperature, pressure, and humidity impact on PD activity. The main content of the paper is organized into three sections dealing with each environmental factor. In every section, relevant publications are reviewed considering the type of samples tested, voltage waveform applied, mutual effects, and the most common PD modeling strategies used. The applicability of the PD measurements for PD risk assessment is also discussed. Based on the review, the current progress in understanding the environmental effects on the PD inception mechanism and PD characteristics is presented and discussed in detail, and future research trends in this field are outlined.

Despite improvements in B cell acute lymphoblastic leukemia (B-ALL) treatment, a significant number of patients experience relapse of the disease, resulting in poor prognosis and high mortality. One of the drawbacks of current B-ALL treatments is the high toxicity associated with the non-specificity of chemotherapeutic drugs. Targeted therapy is an appealing strategy to treat B-ALL to mitigate these toxic off-target effects. One such target is the B cell surface protein CD22. The restricted expression of CD22 on the B-cell lineage and its ligand-induced internalizing properties make it an attractive target in cases of B cell malignancies. To target B-ALL and the CD22 protein, we performed cell internalization SELEX (Systematic Evolution of Ligands by EXponential enrichment) followed by molecular docking to identify internalizing aptamers specific for B-ALL cells that bind the CD22 cell-surface receptor. We identified two RNA aptamers, B-ALL1 and B-ALL2, that target human malignant B cells, with B-ALL1 the first documented RNA aptamer interacting with the CD22 antigen. These B-ALL-specific aptamers represent an important first step toward developing novel targeted therapies for B cell malignancy treatments.

AbstractMyxicola infundibulum Montagu, 1808 is the most reported species of its genus, showing an unusually wide distribution from the Mediterranean area to Australia, North Europe, and North America, a situation deriving from a wide synonymizing of numerous species with M. infundibulum. Recently, genetic analysis confirmed that the Australian form of this species is an introduced taxon from the Mediterranean area, while the examined North American specimens were genetically and morphologically different. In the present paper we travel through the history of M. infundibulum from the first descriptions, trying to trace both the origin of this taxon and the origin of its wide distribution, through an analysis of the descriptions of all valid and invalid taxa to date. We also examined material present in the collection of one of the authors previously identified as M. infundibulum, comparing Mediterranean material to some from the English Channel, and material from North America. This led to the erection of four taxa new to science from material recently collected along the Italian coasts, and the restoration of Myxicola pacifica Johnson, 1901. Positive symptoms of schizophrenia have been proposed to be an intrusion of dreaming in wakefulness; conversely, psychotic patients’ abnormal cognitive and behavioral features could overflow into sleep, so that their dreams would differ from those of healthy people. Here we assess this hypothesis by comparing dream features of 46 patients affected by schizophrenic spectrum disorders to those of 28 healthy controls. In patients, we also investigated correlations of dream variables with symptom severity and verbal fluency. Overall, patients reported fewer and shorter dreams, with a general impoverishment of content (including characters, settings, interactions) and higher spatiotemporal bizarreness. The number of emotions, mainly negative ones, was lower in patients’ reports and correlated inversely with symptom severity. Verbal fluency correlated positively with dream report length and negatively with perceptive bizarreness. In conclusion, our data show a significant impoverishment of dream reports in psychotic patients versus controls. Future research should investigate to what extent this profile of results depends on impaired verbal fluency or on impaired mechanisms of dream generation in this population. Moreover, in line with theories on the role of dreaming in emotion regulation, our data suggest that this function could be impaired in psychoses and related to symptom severity.

Microplastics (MPs) are insidious plastic particles with sizes ranging from 1 to 5000 µm. Their presence has been reported all over the world. Recently, bioremediation to remove MPs from water columns using filter feeders as biofilters has been proposed. In a previous lab experiment, the MP bioremediation potential of four fouling organisms from a mariculture facility (Mytilus galloprovincialis, Sabella spallanzanii, Phallusia mammillata, Paraleucilla magna) was separately assessed in single-species experiment. Herein, a follow-up of the work is presented using a multi-species approach. The four organisms were placed together in the same 5 L beaker and fed with a concentration of 250 p/L 6 µm red polystyrene discernible particles. After digesting the organisms and counting the MPs in both the water and the organisms, the results of the two experiments were compared. In the previous experiment, S. spallanzanii had the highest particle retention (PR) value (PR = 88.01%), while in this experiment, P. mammillata has the lowest PR value (PR = 31%). The multi-species approach resulted in a higher number of plastics being removed from the water (88%) compared to the single-species experiments. These fouling organisms naturally exist as a community, acting as an efficient filter with complex morphologies and hydrodynamic features. Here, this simple marine animal forest is re-evaluated by exploiting the ecosystem services provided by these organisms as a solution to MP pollution problem in a mariculture environment.

The low-voltage electric machine (EM) is a core technology for transportation electrification, and features like high power density and compact volume are essential prerequisites. However, these requirements are usually in conflict with the reliability property of EM, especially in the safety-critical industry such as aviation. Therefore, an appropriate balance between high-performance and reliability needs to be found. Often, the over-engineering method is applied to ensure safety, although it might have a detrimental effect on the EM volume. To address this issue, the EM reliability assessment is included at the EM design stage through the physics of failure (PoF) theory. In EMs, the windings play a key role in electromechanical energy conversion, but their insulation system is subject to frequent failure and represents a reliability bottleneck. Therefore, in-depth research on the root causes of insulation breakdown is beneficial for EM reliability improvement purposes. Indeed, increasing awareness and knowledge on the mechanism of the insulation degradation process and the related lifetime modeling enables the growth of appropriate tools for achieving reliability targets since the first EM design steps. In this work, the main aspects of the insulation system, in terms of materials and manufacturing, are first reviewed. Then, the principal stresses experienced by the winding insulation system are deeply discussed with the purpose of building a profound understanding of the PoF. Finally, an overview of the most common insulation lifetime prediction models is presented, and their use for accomplishing the reliability-oriented design (RoD) and the remaining useful life (RUL) estimation are examined.

The EphA2 receptor tyrosine kinase is overexpressed in most solid tumors and acts as the major driver of tumorigenesis. In this study, we developed a novel approach for targeting the EphA2 receptor using a 2′-fluoro-modified pyrimidine RNA aptamer termed ATOP. We identified the ATOP EphA2 aptamer using a novel bioinformatics strategy that compared aptamers enriched during a protein SELEX using recombinant human EphA2 and a cell-internalization SELEX using EphA2-expressing MDA231 tumor cells. When applied to EphA2-expressing tumor cell lines, the ATOP EphA2 aptamer attenuated tumor cell migration and clonogenicity. In a mouse model of spontaneous metastasis, the ATOP EphA2 aptamer slowed primary tumor growth and significantly reduced the number of lung metastases. The EphA2 ATOP aptamer represents a promising candidate for the development of next-generation targeted therapies that provide safer and more effective treatment of EphA2-overexpressing tumors.

This study uses surface disdrometer reflectivity factor estimates to calibrate the vertical and off-vertical pointing radar beams produced by an ultra high frequency (UHF) band radar wind profiler (RWP) deployed at the US Department of Energy (DOE) Atmospheric Radiation Measurement (ARM) program Southern Great Plains (SGP) Central Facility in northern Oklahoma from April 2011 through July 2019. The methodology consists of five steps. First, the recorded Doppler velocity power spectra are adjusted to account for Nyquist velocity aliasing and coherent integration filtering effects. Second, the spectrum moments are calculated. The third step increases the signal-to-noise ratio (SNR) due to inflated noise power estimates during convective rain events that cause SNR to be biased low. The fourth step determines the RWP calibration constant for one radar beam (called the “reference” beam) by comparing uncalibrated RWP reflectivity factors at 500 m above the ground to 1 min resolution surface disdrometer reflectivity factors. The last step uses the calibrated reference beam reflectivity factor to calibrate the other radar beams during precipitation. There are two key findings. The RWP sensitivity decreased by approximately 3 to 4 dB yr−1 as the hardware aged. This drift was slow enough that the reference calibration constant can be estimated over 3-month intervals using episodic rain events. The calibrated moments are available on the DOE ARM data archive, and the Python processing code is available on public repositories.

Isolated deep convective cloud life cycle and seasonal changes in storm properties are observed for daytime events during the US Department of Energy (DOE) Atmospheric Radiation Measurement (ARM) Green Ocean Amazon Experiment (GoAmazon2014/5) campaign to understand controls on storm behavior. Storm life cycles are documented using surveillance radar from initiation through maturity and dissipation. Vertical air velocity estimates are obtained from radar wind profiler overpasses, with the storm environment informed by radiosondes. Dry-season storm conditions favored reduced morning shallow cloud coverage and larger low-level convective available potential energy (CAPE) than wet-season counterparts. The typical dry-season storm reached its peak intensity and size earlier in its life cycle compared with wet-season cells. These cells exhibited updrafts in core precipitation regions (Z>35 dBZ) to above the melting level as well as persistent downdrafts aloft within precipitation adjacent to their cores. Moreover, dry-season cells recorded more intense updrafts to earlier life cycle stages as well as a higher incidence of strong updrafts (i.e., >5 m s−1) at low levels. In contrast, wet-season storms were longer-lived and featured a higher incidence of moderate (i.e., 2–5 m s−1) updrafts aloft. These storms also favored a shift in their most intense properties to later life cycle stages. Strong downdrafts were less frequent within wet-season cells aloft, indicating a potential systematic difference in draft behaviors, as linked to graupel loading and other factors between the seasons. Results from a stochastic parcel model suggest that dry-season cells may expect stronger updrafts at low levels because of larger low-level CAPE in the dry season. Wet-season cells anticipate strong updrafts aloft because of larger free-tropospheric relative humidity and reduced entrainment-driven dilution. Enhanced dry-season downdrafts are partially attributed to increased evaporation, dry-air entrainment mixing, and negative buoyancy in regions adjacent to sampled dry-season cores.

The local delivery of antiproliferative agents to inhibit neointimal growth is not specific to vascular smooth muscle cells (VSMC) and delays reendothelialization and vascular healing. This investigation was intended to evaluate the effect of luminal delivery of a VSMC-specific aptamer on endothelial healing. The impact of an RNA aptamer (Apt 14) was first examined on the migration and proliferation of primary cultured porcine aortic endothelial cells (ECs) in response to in vitro scratch wound injury. We further evaluated the impact of Apt 14 on reendothelialization when delivered locally in a swine iliofemoral injury model. Although Apt 14 did not affect EC migration and proliferation, in vitro results confirmed that paclitaxel significantly inhibited EC migration and proliferation. En face scanning electron microscopy demonstrated confluent endothelium with elongated EC morphology in Apt 14-treated arteries 14 and 28 days post-treatment. In contrast, vessels treated with paclitaxel-coated balloons displayed a cobblestone morphology and significant platelet and fibrin attachment at cell junctions. These results provide the first evidence of the efficacy of a cell-targeted RNA aptamer to facilitate endothelial healing in a clinically relevant large animal model.

This article examines the consequences of the Russian-Ukrainian war on the Italian economy, especially on inflation, wages and employment. The study shows that Italy, which in the pre-war period had not yet reabsorbed all the consequences of the 2008 crisis, with the structural weaknesses of its economy, came under renewed pressure due to the COVID-19 crisis and, afterwards, to inflation and the war. The paper indicates the need for an economic policy based on the ‘full and good employment’ strategy.

Hemocytes, the myeloid-like immune cells of Drosophila, fulfill a variety of functions that are not completely understood, ranging from phagocytosis to transduction of inflammatory signals. We here show that downregulating the hemocyte-specific Glial cell deficient/Glial cell missing (Glide/Gcm) transcription factor enhances the inflammatory response to the constitutive activation of the Toll pathway. This correlates with lower levels of glutathione S-transferase, suggesting an implication of Glide/Gcm in reactive oxygen species (ROS) signaling and calling for a widespread anti-inflammatory potential of Glide/Gcm. In addition, our data reveal the expression of acetylcholine receptors in hemocytes and that Toll activation affects their expressions, disclosing a novel aspect of the inflammatory response mediated by neurotransmitters. Finally, we provide evidence for acetylcholine receptor nicotinic acetylcholine receptor alpha 6 (nAchRalpha6) regulating hemocyte proliferation in a cell autonomous fashion and for non-cell autonomous cholinergic signaling regulating the number of hemocytes. Altogether, this study provides new insights on the molecular pathways involved in the inflammatory response.

This study presents a method to identify and distinguish insects, clouds, and precipitation in 35GHz (Ka-band) vertically pointing polarimetric radar Doppler velocity power spectra and then produce masks indicating the occurrence of hydrometeors (i.e., clouds or precipitation) and insects at each range gate. The polarimetric radar used in this study transmits a linear polarized wave and receives signals in collinear (CoPol) and cross-linear (XPol) polarized channels. The measured CoPol and XPol Doppler velocity spectra are used to calculate linear depolarization ratio (LDR) spectra. The insect-hydrometeor discrimination method uses CoPol and XPol spectral information in two separate algorithms with their spectral results merged and then filtered into single value products at each range gate. The first algorithm discriminates between insects and clouds in the CoPol Doppler velocity power spectra based on the spectra texture, or spectra roughness, which varies due to the scattering characteristics of insects vs. cloud particles. The second algorithm distinguishes insects from raindrops and ice particles by exploiting the larger Doppler velocity spectra LDR produced by asymmetric insects. Since XPol power return is always less than CoPol power return for the same target (i.e., insect or hydrometeor), fewer insects and hydrometeors are detected in the LDR algorithm than the CoPol algorithm, which drives the need for a CoPol based algorithm. After performing both CoPol and LDR detection algorithms, regions of insect and hydrometeor scattering from both algorithms are combined in the Doppler velocity spectra domain and then filtered to produce a binary hydrometeor mask indicating the occurrence of cloud, raindrops, or ice particles at each range gate. Forty-seven summertime days were processed with the insect-hydrometeor discrimination method using US Department of Energy (DOE) Atmospheric Radiation Measurement (ARM) program Ka-band zenith pointing radar observations in northern Oklahoma, USA. For these 47d, over 70% of the hydrometeor mask column bottoms were within ±100m of simultaneous ceilometer cloud base heights. All datasets and images are available to the public on the DOE ARM repository.

This study investigated the systematic rise in cognitive ability scores over generations, known as the "Flynn Effect," across middle childhood and early adolescence (7-15 years; 291 monozygotic pairs, 298 dizygotic pairs; 89% White). Leveraging the unique structure of the Louisville Twin Study (longitudinal data collected continuously from 1957 to 1999 using the Wechsler Intelligence Scale for Children [WISC], WISC-R, and WISC-III ed.), multilevel analyses revealed between-subjects Flynn Effects--as both decrease in mean scores upon test re-standardization and increase in mean scores across cohorts--as well as within-child Flynn Effects on cognitive growth across age. Overall gains equaled approximately three IQ points per decade. Novel genetically informed analyses suggested that individual sensitivity to the Flynn Effect was moderated by an interplay of genetic and environmental factors.

The current analysis investigates genetic and environmental influences on the bidirectional relationships between temperament and general cognitive ability (GCA). Measures of GCA and three temperament factors (persistence, approach, and reactivity) were collected from 486 children ages 4-9 years (80% white, 50% female) from the Louisville Twin Study from 1976 to 1998. The results indicated a bidirectional dynamic model of temperament influencing subsequent GCA and GCA influencing subsequent temperament. The dynamic relationship between temperament and GCA arose primarily from shared genetic variance, particularly in families with higher socioeconomic status, where input from temperament contributed on average 20% to genetic variance in GCA versus 0% in lower SES families.

Acute lung injury (ALI) is a syndrome of acute inflammation, barrier disruption, and hypoxemic respiratory failure associated with high morbidity and mortality. Diverse conditions lead to ALI, including inhalation of toxic substances, aspiration of gastric contents, infection, and trauma. A shared mechanism of acute lung injury is cellular toxicity from damage-associated molecular patterns (DAMPs), including extracellular histones. We recently described the selection and efficacy of a histone-binding RNA aptamer (HBA7). The current study aimed to identify the effects of extracellular histones in the lung and determine if HBA7 protected mice from ALI. Histone proteins decreased metabolic activity, induced apoptosis, promoted proinflammatory cytokine production, and caused endothelial dysfunction and platelet activation in vitro. HBA7 prevented these effects. The oropharyngeal aspiration of histone proteins increased neutrophil and albumin levels in bronchoalveolar lavage fluid (BALF) and precipitated neutrophil infiltration, interstitial edema, and barrier disruption in alveoli in mice. Similarly, inhaling wood smoke particulate matter, as a clinically relevant model, increased lung inflammation and alveolar permeability. Treatment by HBA7 alleviated lung injury in both models of ALI. These findings demonstrate the pulmonary delivery of HBA7 as a nucleic acid-based therapeutic for ALI.

Abstract The humidity role in the partial discharge (PD) inception mechanism is quite challenging, especially when considering the environmental temperature. Indeed, there is no general rule to explain the humidity effect on the PD phenomenon. In this paper, the PD activity in inter‐turn insulation is experimentally investigated for different relative humidity (RH) conditions at three different ambient temperatures, that is, 30°C, 60°C, and 90°C. Partial discharge inception voltage (PDIV) is directly measured through a photomultiplier tube (PMT), whereas the tip‐up tests are performed aiming at monitoring both dissipation factor (tanδ) and insulation capacitance (IC). These extra measurements (diagnostic dielectric markers) allow better assessing the insulation status. The adoption of the tip‐up test enables the insulation properties measurement. Based on the tip‐up tests’ findings, the interfacial polarization process starts at 75% RH under 60°C, while the high conductivity area is already formed at 75% RH when the ambient temperature is 90°C. The water film formation deduced from the tip‐up test is then used to explain the trend of PDIV, and the validity is further proved by finite element analysis (FEA).

Abstract Analysis of cell-cell interactions, cell function and cell lineages greatly benefits selective destruction techniques, which, at present, rely on dedicated, high energy, pulsed lasers and are limited to cells that are detectable by conventional microscopy. We present here a high resolution/sensitivity technique based on confocal microscopy and relying on commonly used UV lasers. Coupling this technique with time-lapse enables the destruction and following of any cell(s) in any pattern(s) in living animals as well as in cell culture systems.

Abstract Background The WAVE/SCAR complex, consisting of CYFIP (PIR121 or Sra1), Kette (Nap1), Abi, SCAR (WAVE) and HSPC300, is known to regulate the actin nucleating Arp2/3 complex in a Rac1-dependent manner. While in vitro and in vivo studies have demonstrated that CYFIP, Kette, Abi and SCAR work as subunits of the complex, the role of the small protein HSPC300 remains unclear. Results In the present study, we identify the HSPC300 gene and characterize its interaction with the WAVE/SCAR complex in the Drosophila animal model. On the basis of several lines of evidence, we demonstrate that HSPC300 is an indispensable component of the complex controlling axonal and neuromuscular junction (NMJ) growth. First, the Drosophila HSPC300 expression profile resembles that of other members of the WAVE/SCAR complex. Second, HSPC300 mutation, as well as mutations in the other complex subunits, results in identical axonal and NMJ growth defects. Third, like with other complex subunits, defects in NMJ architecture are rescued by presynaptic expression of the respective wild-type gene. Fourth, HSPC300 genetically interacts with another subunit of the WAVE/SCAR complex. Fifth, HSPC300 physically associates with CYFIP and SCAR. Conclusion Present data provide the first evidence for HSPC300 playing a role in nervous system development and demonstrate in vivo that this small protein works in the context of the WAVE/SCAR complex.

Abstract Fluid biomarkers are currently measured in cerebrospinal fluid and blood for Alzheimer's disease diagnosis and are promising targets for drug development and for patients’ follow‐up in clinical trials. These biomarkers have been grouped in an unbiased research framework, the amyloid (Aβ), tau, and neurodegeneration (AT[N]) biomarker system to aid patients’ early diagnosis and stratification. Metrological approaches relying on mass spectrometry have been used for the development of reference materials and reference measurement procedures. Despite their excellent performances as clinical tools, fluid biomarkers often present an important between‐laboratory variation. Standardization efforts were carried out on the biomarkers currently included in the AT(N) classification system, involving the collaboration of national metrology institutes, clinicians, researchers, and in vitro diagnostic providers. This article provides an overview of current activities towards standardization. These reference methods and reference materials may be used for recalibration of immunoassays and the establishment of standardized cutoff values allowing a better stratification of Alzheimer's disease patients. Highlights The AT(N) biomarker system allows stratifying AD patients on the basis of biomarker profiles. Fluid biomarker measurements often present an important between‐laboratory variation preventing the establishment of standardized cutoff values. Overview on the standardization initiatives involving the fluid biomarkers currently included in the AT(N) framework.

Anti-proliferative agents have been the primary therapeutic drug of choice to inhibit restenosis after endovascular treatment. However, recent safety and efficacy concerns for patients who underwent peripheral artery disease revascularization have demonstrated the need for alternative therapeutics. The aim of this investigation was to investigate the efficacy of a cell-specific RNA aptamer inhibiting vascular smooth muscle cell proliferation and migration. First, the impact of the RNA aptamer (Apt 14) on the wound healing of primary cultured porcine vascular smooth muscle cells (VSMCs) was examined in response to a scratch wound injury. We then evaluated the effect of local luminal delivery of Apt 14 on neointimal formation in a clinically relevant swine iliofemoral injury model. In contrast with a non-selected control aptamer (NSC) that had no impact on VSMC migration, Apt 14 attenuated the wound healing of primary cultured porcine VSMCs to platelet-derived growth factor-BB. Histological analysis of the Apt 14-treated arteries demonstrated a significant reduction in neointimal area percent diameter stenosis compared with arteries treated with saline and NSC controls. The findings of this study suggest that aptamers can function as selective inhibitors and thus provide more fine-tuning to inhibit selective pathways responsible for neointimal hyperplasia.

This paper provides an account of the evolution of the notion of comparatio in the main dialectical works of Valla, Agricola, and Vives. It highlights the elements of continuity and discontinuity and sheds light on the original contributions of Vives’s treatment. In Valla, Agricola and Vives, the notion of comparatio characterizes: a) the locus of the relation to another in the inventio method; b) the cognitive act through which one can grasp the relationships of similarity and difference between concepts; c) the epistemic method for weighing the degree of plausibility of probable arguments. The paper also shows how Vives enhances the role of comparatio within dialectical art. Firstly, he attributes a pre-eminent position to the locus of the comparatio by virtue of the transversality of its application to all the other loci. Secondly, he identifies the explanation of the key concept of syllogism in the act of compering two sentences with a third. Finally, he finds the essence of the disputatio in the method of comparing equally probable contradictory arguments. This can rightly be considered an innovative element of the Vivesian dialectical treatment with respect to the most advanced European humanist movement of the first decades of the sixteenth century.

Introduction: Breast cancer (BC) diagnostics lack noninvasive methods and procedures for screening and monitoring disease dynamics. Admitted CellSearch® is used for fluid biopsy and capture of circulating tumor cells of only epithelial origin. Here we describe an RNA aptamer (MDA231) for detecting BC cells in clinical samples, including blood. The MDA231 aptamer was originally selected against triple-negative breast cancer cell line MDA-MB-231 using cell-SELEX.Methods: The aptamer structure in solution was predicted using mFold program and molecular dynamic simulations. The affinity and specificity of the evolved aptamers were evaluated by flow cytometry and laser scanning microscopy on clinical tissues from breast cancer patients. CTCs were isolated form the patients’ blood using the developed method of aptamer-based magnetic separation. Breast cancer origin of CTCs was confirmed by cytological, RT-qPCR and Immunocytochemical analyses.Results: MDA231 can specifically recognize breast cancer cells in surgically resected tissues from patients with different molecular subtypes: triple-negative, Luminal A, and Luminal B, but not in benign tumors, lung cancer, glial tumor and healthy epithelial from lungs and breast. This RNA aptamer can identify cancer cells in complex cellular environments, including tumor biopsies (e.g., tumor tissues vs. margins) and clinical blood samples (e.g., circulating tumor cells). Breast cancer origin of the aptamer-based magnetically separated CTCs has been proved by immunocytochemistry and mammaglobin mRNA expression.Discussion: We suggest a simple, minimally-invasive breast cancer diagnostic method based on non-epithelial MDA231 aptamer-specific magnetic isolation of circulating tumor cells. Isolated cells are intact and can be utilized for molecular diagnostics purposes.
