Abstract

The emergence of new SARS-CoV-2 variants poses challenges to global surveillance efforts, necessitating swift actions in their detection, evaluation, and management. Among the most recent variants, Omicron BA.2.86 and its sub-lineages have gained attention due to their potential immune evasion properties. This study describes the development of a digital PCR assay for the rapid detection of BA.2.86 and its descendant lineages, in wastewater samples. By using this assay, we analyzed wastewater samples collected in Italy from September 2023 to January 2024. Our analysis revealed the presence of BA.2.86 lineages already in October 2023 with a minimal detection rate of 2% which then rapidly increased, becoming dominant by January 2024, accounting for a prevalence of 62%. The findings emphasize the significance of wastewater-based surveillance in tracking emerging variants and underscore the efficacy of targeted digital PCR assays for environmental monitoring.

Abstract

The SARS-CoV-2 Omicron variant emerged in South Africa in November 2021, and has later been identified worldwide, raising serious concerns. A real-time RT-PCR assay was designed for the rapid screening of the Omicron variant, targeting characteristic mutations of the spike gene. The assay was used to test 737 sewage samples collected throughout Italy (19/21 Regions) between 11 November and 25 December 2021, with the aim of assessing the spread of the Omicron variant in the country. Positive samples were also tested with a real-time RT-PCR developed by the European Commission, Joint Research Centre (JRC), and through nested RT-PCR followed by Sanger sequencing. Overall, 115 samples tested positive for Omicron SARS-CoV-2 variant. The first occurrence was detected on 7 December, in Veneto, North Italy. Later on, the variant spread extremely fast in three weeks, with prevalence of positive wastewater samples rising from 1.0% (1/104 samples) in the week 5–11 December, to 17.5% (25/143 samples) in the week 12–18, to 65.9% (89/135 samples) in the week 19–25, in line with the increase in cases of infection with the Omicron variant observed during December in Italy. Similarly, the number of Regions/Autonomous Provinces in which the variant was detected increased from one in the first week, to 11 in the second, and to 17 in the last one. The presence of the Omicron variant was confirmed by the JRC real-time RT-PCR in 79.1% (91/115) of the positive samples, and by Sanger sequencing in 66% (64/97) of PCR amplicons. In conclusion, we designed an RT-qPCR assay capable to detect the Omicron variant, which can be successfully used for the purpose of wastewater-based epidemiology. We also described the history of the introduction and diffusion of the Omicron variant in the Italian population and territory, confirming the effectiveness of sewage monitoring as a powerful surveillance tool.

Abstract

The Erice 2018 Charter was unanimously approved at the conclusion of the 53rd Residential Course of the International School of Epidemiology and Preventive Medicine “Adapted Physical Activity in Sport, Wellness and Fitness; the role of the Departments of Prevention and of the National Health Service in doping prevention and health promotion”, held on 15-19 May 2018 in Erice, Italy, at the “Ettore Majorana” Foundation and Centre for Scientific Culture, and promoted by the Study Group on “Movement Sciences for Health” of the Italian Society of Hygiene, Preventive Medicine and Public Health. The event was part of a larger project supported by the Ministry of Health aimed at preventing doping in the general population involved in sport and physical activities. After an intense discussion the participants focused on ten statements involving the following critical issues: responsibility, priority, message, alphabetization, networks and alliances, school promoting health, player and opportunities, competences, know-how, programming and acting. These statements provide hints to approach doping within a public health frame and summarize the role of the Departments of Prevention and NHS in promoting and coordinating preventive actions with other institutions and stakeholders. Doping represents a complex phenomenon related to cultural, social, economic and legal issues. In addition to regulatory or repressive actions, education to health and legality is proposed as the fundamental strategy to contrast doping by promoting healthy lifestyles, based on scientific knowledge and respect for legality.

Abstract

The aim of the following study is to identify and quantify microorganisms in the saliva of black stain patients using species-specific probes for Actinomyces israelii, Actinomyces naeslundii, Streptococcus mutans and Lactobacillus spp, as well as a universal probe for bacterial 16S rDNA, to obtain the figure of the total bacterial counts (for test and control group). This study represents a first phase of the research line on extrinsic tooth pigmentation on developmental age patients. Materials and methods. The selected sample is composed of 20 subjects aged between 2 and 15 years and in good general and oral health among the patients included in a dental examination trial at the Department of Pediatric Dentistry of the University Hospital of Palermo, after obteining informed consent from parents. Subjects with a positive anamnesis for systemic diseases or with enamel defects were excluded from the study. Exclusion criteria were also a diet including regular use of tea, coffee, iron supplements, or antibiotic therapies in the 3 months prior to enrollment and the use of mouthwashes/toothpastes containing antiseptics. Parents/tutors were asked to complete a questionnaire on their occupational status and level of education, as well as their eating habits, the frequency of brushing and the patient's dental records. A sample of 20 patients without BS was used as control group. From all subjects participating to the study a sample of 5ml of unstimulated saliva was recovered from the right and left buccal area through a sterile syringe, after 2 minutes waiting for the accumulation of an adequate amount of saliva. Statistical analysis was carried out using descriptive and inferential statistics (mean and standard deviation; chi-square test, t test for non-paired data). StatView 5.0.1 statistical software (SAS Institute, Cary, NC, USA) was used for this purpose, and the tests were considered significant at p values less than or equal to 0.05. The study protocol will be presented for approval to the Palermo 1 ethics committee.

Abstract

The Erice 50 Charter titled "Strategies for Diseases Prevention and Health Promotion in Urban Areas" was unanimously approved at the conclusion of the 50th Residential Course "Urban Health. Instruments for promoting health and for assessing hygienic and sanitary conditions in urban areas", held from 29th March to 2nd April 2017 in Erice, at the "Ettore Majorana" Foundation and Centre for Scientific Culture and promoted by the International School of Epidemiology and Preventive Medicine "G. D'Alessandro" and the Study Group "Building Hygiene" of the Italian Society of Hygiene, Preventive Medicine and Public Health (SItI). At the conclusion of the intense learning experience during the Course, with more than 20 lectures, workshops and long-lasting discussions between Professors and Students, the participants identified the major points connecting urban features and Public Health, claiming the pivotal role of urban planning strategies for the management of Diseases Prevention and Health Promotion activities. The Erice 50 Charter is configured as a Decalogue for Healthy Cities and as a Think Tank for designing effective strategic actions and best practices to develop urban regeneration interventions and improve the urban quality of contemporary cities. The Decalogue is structured into the following key strategic objectives: 1. Promoting urban planning interventions that address citizens towards healthy behaviours; 2. Improving living conditions in the urban context; 3. Building an accessible and inclusive city, with a special focus on the frail population; 4. Encouraging the foundation of resilient urban areas; 5. Supporting the development of new economies and employment through urban renewal interventions; 6. Tackling social inequalities; 7. Improving stakeholders' awareness of the factors affecting Public Health in the cities; 8. Ensuring a participated urban governance; 9. Introducing qualitative and quantitative performance tools, capable of measuring the city's attitude to promote healthy lifestyles and to monitor the population's health status; 10. Encouraging sharing of knowledge and accessibility to informations. Finally, all the participants underlined that a multidisciplinary team, composed of Physicians specialized in Hygiene, Preventive Medicine, Public Health and Technicians as Architects, Urban planners and Engineers, is needed to deepen the research topic of Urban Health.

Abstract

The Erice Charter was unanimously approved at the conclusion of the 47th Residential Course "Adapted Physical Activity in Sport, Wellness and Fitness: New Challenges for Prevention and Health Promotion", held on 20-24 April 2015 in Erice, Italy, at the "Ettore Majorana" Foundation and Centre for Scientific Culture, and promoted by the International School of Epidemiology and Preventive Medicine "G. D'Alessandro" and the Study Group on Movement Sciences for Health of the Italian Society of Hygiene, Preventive Medicine and Public Health. After an intense discussion the participants identified the main points associated with the relevance of physical activity for Public Health, claiming the pivotal role of the Department of Prevention in coordinating and managing preventive actions. The participants underlined the importance of the physicians specialized in Hygiene, Preventive Medicine and Public Health. The contribution of other operators such as physicians specialized in Sport Medicine was stressed. Further, the holders of the new degree in Human Movement and Sport Sciences were considered fundamental contributors for the performance of physical activity and their presence was seen as a promising opportunity for the Departments of Prevention. Primary prevention based on recreational physical activities should become easily accessible for the population, avoiding obstacles such as certification steps or complex bureaucracy. The Sport Doctor is recognized as the principal referent for preliminary physical evaluation and clinical monitoring in secondary and tertiary prevention actions based on adapted physical activities. Developing research in the field is essential as well as implementing higher education on physical activity management in Schools of Public Health.

Abstract

La termoporazione dinamica irreversibile (DIT) consiste in un nuovo tipo di tecnologia di pastorizzazione ideato da Koulik e colleghi basato su un processo termico dinamico, caratterizzato da riscaldamenti molto rapidi (shock termici) a temperature finali non superiori a 70°C. L’obiettivo del nostro studio è stato la valutazione dell’influenza di parametri di processo quali la temperatura di partenza (T1 ) del campione da trattare, la temperatura finale in corso di trattamento (T 2), la differenza tra temperatura iniziale e temperatura finale (ΔT), la velocità di riscaldamento (θ) e la durata del trattamento sull’abbattimento della carica batterica nella matrice alimentare

Abstract

Avalanching plays a crucial role in granular materials dynamics, in particular in the evolution of the shape of the leeward side of sand dunes. This paper presents a physically-based mathematical model capable of reproducing the kinematic evolution of the surface of sand piles and to obtain eventually the stationary configurations, in the presence of external sources as well. Simulation results with different boundary conditions and geometries are reported in order to show the high flexibility of the model. Abstract

Living organisms respond to chemicals at all the levels of biological organization, from molecules to ecosystems. In general, biochemical and physiological responses (molecules, cells, tissues) are triggered at low concentrations and after short exposure times, while ecological responses (life cycle traits, populations, communities, ecosystems) can be evidenced only at higher concentrations and after longer exposures. On the other hand, responses at the higher levels of organization are considered more relevant from an environmental point of view, as they often result in irreversible severe damage to ecological systems. It is important to clarify the relationship between low-level and high-level responses to different toxicants, particularly considering the increasing application of biomarkers as early warning systems to assess environmental pollution. In accordance with this view, effects of exposure to hexavalent chromium, Cr(VI), on life cycle endpoints were compared to effects on a battery of eight biomarkers in the earthworm Eisenia andrei. In spite of the importance of Cr(VI) as an environmental contaminant, its toxicity to earthworms, a major component of soil fauna, has received little attention; in particular almost no data are available on biochemical and physiological responses. A natural clayey soil was spiked at six Cr(VI) concentrations, from 6.25 to 200 mg/kg. Adult worms were exposed in laboratory to the spiked soils and to a control treatment (unspiked soil). Two sets of experimental containers were prepared: the first to measure lysosomal membrane stability, lysosomal lipofuscins and neutral lipids accumulation, metallothionein content, Ca2+ ATPase, catalase, glutathione S-transferase and acyl CoA oxydase activities, after 10 d of exposure; the second to asses survival, growth and chromium bioaccumulation after 28 d, and reproduction after 56 d of exposure. All the biological effects were significantly affected by Cr(VI) exposure and showed a clear concentration-effect relationship, except for the content of metallothioneins. Biomarkers showed different trends at increasing Cr(VI) concentrations, that could be classified as typical “increasing responses” (neutral lipids), “decreasing responses” (lysosomal membrane stability, Ca2+-ATPase and acyl Co-A oxydase activities) and “bell-shaped responses” (catalase and glutathione S-transferase activities). Biomarkers were more sensitive to Cr(VI) than life-cycle endpoints; among these, reproduction was the most responsive.
