5. CuI/Cu(OSO2CF3)2 catalysed convenient approach to dichromenopyridines and triazole-thiazole appended chromone derivatives

Abstract

A variety of new pentacyclic dichromenopyridines have been synthesized from 2-amino chromone and O-propargyloxy salicilaldehydes through intramolecular Povarov reaction using CuI/Cu(OSO2CF3)2 as a Lewis acid catalyst. The dichromenopyridines synthesis involve two sequential steps, insitu N-heteryl aldimine formation and an intramolecular Aza Diels-Alder[4 + 2] cycloaddition. Furthermore the key intermediate, terminal alkyne tethered Schiff bases subjected to Cu(I) mediated click [3 + 2] cycloaddition with thiazole appended azide to afford imine linked chromone-1,2,3-triazole -thiazole tri heterocyclic molecular hybrids.

Abstract

Abstract

An alternate synthesis of an endothelin receptor antagonist bosentan monohydrate is reported. This new synthetic route involves the coupling of p - tert -butyl- N -[6-chloro-5-(2-methoxy-phenoxy)(2,2′-bipyrimidin)-4-yl]benzene sulfonamide with commercially available raw material (2,2-dimethyl-1,3-dioxolan-4-yl)methanol as the key step. Attractive features of this approach are its versatileness, commercial availability of raw materials, usage of eco-friendly reagents, and it efficiently provides the desired bosentan monohydrate free from reported impurities such as dimer, N-alkylated, and pyrimidinone impurities.

10. A New and Competitive Synthetic Approach for an Antihistamine Agent, Bilastine

Abstract

Efforts towards the novel synthesis of second generation non-sedating antihistamine drug, Bilastine was described in this manuscript. This competitive synthetic approach involves the convergent synthesis of Bilastine via simple Friedel-Crafts acylation as an alternate for earlier reported Stille and Suzuki couplings. The selectivity in Friedel-Crafts acylation reaction with chloro acetyl chloride on different substituted arenes was studied and employed the best conditions for the synthesis of Bilastine. Further synthetic approach involves the deoxygenation of aryl ketone to corresponding alkane in single step and finally provides Bilastine with 39% of improved overall yields, utilizing simple and cost-effective reagents, suitable for kilogram scale synthesis.

Abstract

A new scalable synthesis of the common thiazole fragment (C12-C21 unit) of epothilone family of molecules (epothilone A-D) has been developed via palladium-catalyzed Wacker oxidation as a key step using (S)-2,2,3,3,9,9,10,10-octamethyl-5-vinyl-4,8-dioxa-3,9-disilaundecane, which has been prepared from commercially available chiral synthon (R)-ethyl-4-cyano-3-hydroxybutanoate. Then further chemical modifications using Horner-Wadsworth-Emmons reaction and Wittig reaction result in the common thiazole fragment (C12-C21 unit) in good yields. [ABSTRACT FROM AUTHOR]
