Abstract

The interactions of meso-tetrakis(4N-(2-hydroxyethyl)pyridinium-4-yl)porphyrin (TEtOHPyP4), meso -tetrakis-(4N-allylpyridinium-4-yl)porphyrin (TAIPyP4) and meso-tetrakis(4N-metallylpyridinium-4-yl)porphyrin (TMet AIPyp4) with the poly(rA)poly(rU) and poly(rI)poly(rC) homopolymeric RNA duplexes at 18, 25. 35 and 45 degrees Celsius are studied. Combined circular dichroism (CD) and fluorescence results suggest that the first two porphyrins intercalate into the RNA duplexes, while TMetAIPyP4 forms outside-bound, self-stacked aggregates along the RNA helices.

22. THERMODYNAMIC PROFILE OF INTERACTION OF PORPHYRINS WITH NUCLEIC ACIDS.

Abstract

The binding of a number of novel water soluble porphyrins to nucleic acids (NA) were studied monitoring the changes in Soret region of absorbance spectra, fluorescence spectra and circular dichroism (CD) spectra of formed complexes. The binding modes for all complexes were dtermined. The binding isotherms were used to calculate the binding constant, Kb, and binding free energy, ⊿Gb = -RTlnKb. By performing these experiments as a function of temperature, we evaluated the van't Hoff binding enthalpies, ⊿Hb and the binding entropies, ⊿Sb. Generalization concerning the established correlation between the binding mode and thermodynamic profile was done. [ABSTRACT FROM AUTHOR]

24. Multiphoton spectral microscopy for imaging and quantification of tissue glycation

Abstract

Tissue glycation from diabetes and aging can result in complications such as renal failure, blindness, nerve damage and vascular diseases. In this work, we applied multiphoton microscopy for imaging and characterizing the extent of tissue glycation. The characteristic features of multiphoton autofluorescence (MPAF) and second harmonic generation (SHG) images as well as MPAF spectra of glycated bovine skin, cornea and aorta were acquired. The analysis of MPAF intensity change accompanying the glycation process shows that collagen is more responsive to the formation of autofluorescent advanced glycation endproducts (AGE) than elastic fibers. Changes in spectral features were also used to estimate the rate of glycation in tissues with intrinsic AF. Our study shows that multiphton imaging may be used for the in vitro investigation of the effects of tissue glycation and that this approach may be used for monitoring AGE formation in the clinical setting.
