Abstract

A sensitive and specific immunoassay for the simultaneous detection of Clostridium botulinum type C (BoNT/C) and type and type D neurotoxin is developed. Streptavidin-poly-horseradish peroxidase is used as a conjugate and a precipitating substrate allowed the direct semiquantitative readout of the assay and for a more accurate quantitative detection, the substrate can be eluted and measured in a photometer.

Abstract

A multiplexed quantitative real-time PCR assay was developed for the simultaneous detection of the human pathogenic Clostridium botulinum serotypes A, B, E and F. The pentaplexed assay was found to show high sensitivity and specificity and facilitated for the simultaneous screening and differentiation of specimens for C. botulinum A, B, E and F.

Abstract

The thermal inactivation of BoNT/A, BoNT/A complex, BoNT/B, and BoNT/B complex in cow's milk in a pasteurization process similar to the industry standard is described. The high-temperature short-time (HTST) pasteurization process in the dairy industry is found to remarkably reduce the risk of consumer harm even if larger amounts of BoNT were deliberately released into the milk supply chain.

30. Production and purification of Clostridium botulinumtype C and D neurotoxin

Abstract

Neurotoxins of Clostridium botulinumare needed in basic neurologic research, but as therapeutic agent for certain neuro‐muscular disorders like strabism as well. A method for the production and purification of botulinum neurotoxins C and D is reported using a two‐step hollow‐fiber cross flow filtration and a newly developed chromatographic purification procedure. Hollow‐fiber filtration proved to be a rapid and safe concentration and pre‐purification step, which can easily be scaled up. The chromatographic purification included hydrophobic interaction, anion exchange and size exclusion chromatography runs. Botulinum neurotoxins C and D could be recovered with an overall yield of 12.6% and 10.6%, respectively. A specific toxicity of 1.86×107minimal lethal dose mg−1(type C) and 5.26×107minimal lethal dose mg−1(type D) was determined in the mouse bioassay.

Abstract

Since 1970 our laboratory is specialized in diagnosis of clostridial diseases, including Clostridium botulinum and botulism. Since 1995, samples from more than 900 suspected botulinal cases were received, mainly in cattle, horses and men. 524 outbreaks were diagnosed as clearly positive by toxin neutralisation; 83 cases remained inconclusive with the toxin neutralisation. The geographical distribution of the positive cases in Germany is demonstrated for cattle and horses. Dispatch and treatment of specimens and interpretation of results are discussed.

32. [Development of an in vitro assay for the detection of Botulinum Neurotoxins]

Abstract

Botulism is a severe, often lethal intoxication, which affects man and animal. It is caused by the neurotoxins (BoNT) of Clostridium botulinum, an anaerobic spore-former. At present the toxin types A-G have been identified. The extreme toxicity of these substances still make the animal testing inevitable. No alternative method could reach the sensitivity of the mouse bioassay. Based on the investigations presented, a serological test shall be developed, which concentrates the sample and detects the toxin in one step. Preliminary results of an in vitro test detecting BoNT/D are discussed.
