1. Keel bone damage, such as deformations and fractures, is a severe problem regarding animal welfare in layers. To identify risk factors under commercial conditions, 33 layer flocks (22 barn, 11 free range) with white (n = 18), brown (n = 11) and mixed (n = 4) genotypes were examined. 2. Keel bone status was frequently scored by palpation throughout the laying period. Data on housing and management conditions were collected. Multiple regression and Generalized Estimating Equations procedure were used for analysis. 3. At 65–74 weeks of age, the prevalence of keel bone damage ranged between 26% and 74%. White genotypes and those kept in multi-tier systems developed significantly (p < 0.05) more keel bone damage than brown genotypes or those kept in single-tier systems. Wing feather condition was associated with keel bone damage (p < 0.05), while other investigated variables regarding health, housing and management were not associated. 4. In conclusion, housing and management should be adapted to meet the birds' specific needs in multi-tier systems, which may vary for brown and white genotypes. 1. Within a triennial project, 34 layer flocks with untrimmed beaks were examined regularly throughout the laying period to broaden knowledge on the occurrence and development of severe feather pecking and cannibalism, as well as on factors influencing this non-desirable behaviour. 2. Flocks involved 850 to 27,183 hens of seven different genetics, kept in a barn or on free-range systems. Damage to the plumage and skin was assessed in individual hens during each visit and their body weights recorded. Correlations smaller than 0.8 between different body areas for damage indicated the necessity to consider them separately. Accounting for the risk of bias due to unevenly distributed factors, regression functions were used to assess associations between pecking damage, losses and performance, housing and management conditions. In addition, temporal pattern of pecking damage for flocks with severe, medium and little damage was modelled using these functions. 3. As expected, plumage damage increased with age, whereas the development of skin lesions was less consistent. From 30 weeks of age, pecking damage on the back increased remarkably in flocks with the most severe scores compared to those with medium and little damage, especially during the later laying period. Associations were found between pecking damage on the back of hens with plumage quality of pullets when entering the layer house (point of lay). Damage to the vent/cloacal region was more pronounced in white compared to brown layers. However, this did not deny the importance of factors not significant in this study. 4. Metabolic monitoring is a tool that is helpful with the increasing requirements regarding feeding and health management in dairy herds. This study aimed at describing the trend and variability of different biochemical parameters in blood and urine in relation to the stages of lactation and parity, in a retrospective analysis of laboratory data from clinically healthy German Holstein cows. The results were derived from metabolic monitoring in Thuringia (Germany), during 2009-2019. A total of 361,584 measured values, of 13 different metabolic variables, were assigned to parity (primiparous and multiparous) and stage of lactation (10 classes from -30 to 300 days in milk). The Kruskal-Wallis test was applied for the evaluation of differences regarding parity or the stage of lactation. Non-esterified fatty acids, beta hydroxybutyrate, and the activity of aspartate aminotransferase in serum were clearly affected by parity and lactation. Serum concentrations of cholesterol, bilirubin, and phosphorus, as well as the serum activity of glutamate dehydrogenase, were affected by the stage of lactation, while parity impacted urea concentration. The serum activity of creatine kinase, serum concentrations of calcium, and urine concentrations of net acid base excretion, potassium, and sodium were not affected by parity or lactation. In conclusion, specific reference limits, with respect to parity and the stage of lactation, are necessary.

In the transition period from late gestation to early lactation, dairy cows undergo tremendous metabolic changes. Insulin is a relevant antilipolytic factor. Decreasing serum concentrations of insulin and glucose, increasing serum concentrations of nonesterified fatty acids (NEFA) and β-hydroxybutyrate (BHB), and changes in body condition score (BCS) reflect the negative energy balance around calving. This study investigated peripartum metabolic adaptation in 359 primiparous and 235 multiparous German Holstein cows from a commercial dairy herd under field conditions. Body condition score was recorded and blood samples were taken 10 to 1 d prepartum, 2 to 4 d postpartum, and 12 to 20 d postpartum. Generalized mixed models and generalized estimation equations were applied to assess associations between prepartum BCS; BCS changes during the transition period; insulin, glucose, NEFA, and BHB serum concentrations; and milk yield, which was taken from an electronic milk meter from d 6 of lactation. Serum insulin concentrations of multiparous postpartum cows were lower compared with prepartum, and compared with primiparous cows. In general, primiparous cows had lower postpartum NEFA and BHB concentrations than multiparous cows. In primiparous cows, we identified a positive association between prepartum BCS and prepartum serum insulin concentration. Prepartum obese multiparous cows, but not primiparous cows, were characterized by higher postpartum serum NEFA and BHB concentrations and lower milk yield than other cows in the same parity class. Primiparous cows with a smaller degree of BCS loss during the transition period had higher postpartum insulin and lower NEFA concentrations and lower milk yield than other primiparous cows. In conclusion, primiparous cows had less lipolysis and lower milk yield than multiparous cows, associated with higher insulin concentrations. Pregnancy-Associated Plasma Protein-A (PAPP-A) proteolyses insulin-like growth factor binding protein-4 (IGFBP-4), thereby regulating local IGF availability. Reduced PAPP-A mRNA expression has been reported in ovarian cancer specimens compared to normal ovarian surface epithelial cells (OSE). To characterize PAPP-A expression and proteolytic activity in OSE, we developed a lifespan-extended human cell model using a temperature-sensitive mutant of the SV40 large T antigen (SV40LT). These OSE(tsT) cells proliferate at 34 degrees C (i.e., when SV40LT-positive), but not at 39 degrees C, a temperature at which the SV40LT is unstable (SV40LT-negative). Proteolysis of radiolabeled IGFBP-4 in conditioned media from OSE(tsT) lines was IGF-dependent and blocked by anti-PAPP-A antisera. Temperature shifts that eliminated stable SV40LT induced a 7-fold increase in PAPP-A mRNA and a 4-fold increase in protein. The converse experiment (shifting to SV40LT-positive conditions) resulted in decreased levels of PAPP-A mRNA but little change in PAPP-A protein. Nevertheless, there was a marked reduction in IGF-BP-4 proteolytic activity in medium of SV40LT-positive OSE-(tsT) cells. This decreased PAPP-A activity coincided with a nearly 20-fold increase in mRNA encoding a physiological inhibitor of PAPP-A, the precursor form of eosinophil Major Basic Protein (proMBP), and 4- to 5-fold increases in proMBP protein. Primary cultures of unmodified OSE expressed high levels of PAPP-A and undetectable proMBP, and therefore produced abundant IGFBP-4 protease activity. Short-term ovarian tumor cell cultures expressed variable levels of PAPP-A and high levels of proMBP, and consequently secreted little or no IGFBP-4 protease activity. The concurrent regulation of PAPP-A and its inhibitor, proMBP, suggests that IGFBP-4 proteolysis and local regulation of IGF availability may be altered in malignant ovarian epithelial cells., (Copyright 2004 Wiley-Liss, Inc.)
