In this thesis new non-viral gene delivery systems were synthesized and characterized. In the first part different questions concerning in vitro and in vivo gene delivery using HER2-targeted Polyethylenglycol (PEG)-modified Polyethylenimine (PEI) polyplexes were addressed. The second part deals with in vitro gene delivery using chitosan and its trimethyl- and PEG-trimethyl-derivatives. In the first part, different novel targeted gene delivery vectors consisting of PEI, PEG and the HER2 directed monoclonal antibody Trastuzumab are described. Conjugates differed in structure concerning PEG-PEI copolymers and linkers used for synthesis. The resulting PEG-PEI-Trastuzumab conjugates were characterized concerning hydrodynamic diameters and zetapotentials of polyplexes with DNA. All conjugate complexes were well below the size-threshold for endocytosis and the surface of the different conjugates was uncharged, decreasing binding to non-targeted cells. The ability of conjugate complexes to specifically bind to and be taken up into HER2-overexpressing cells was evaluated in vitro. Significant differences in the efficiency of binding and uptake were found depending on polyplex structure. These differences were reflected in transfection efficiency and specificity in HER2 overexpressing cells as well. The best results were obtained when Trastuzumab was coupled to PEI via a short PEG linker. The conjugate polyplexes were also characterized towards in vivo applicability. Blood compatibility was tested by incubation with erythrocytes determining the degree of hemolysis as well as erythrocyte morphology and aggregation. Conjugate polyplexes showed very low levels of hemolysis and erythrocyte morphology was virtually unchanged. Pharmacokinetics of conjugates revealed significant improvements over free plasmid DNA and PEI polyplexes. Furthermore, significantly decreased disposition of polyplexes in spleen and lung was observed in the case of conjugates. A HER2 overexpressing in vivo tumor model was established in SCID mice in an attempt to test conjugates in vivo. In the second part of the thesis, chitosan, trimethyl chitosan or polyethylenglycol-graft-trimethyl chitosan/DNA polyplexes were characterized concerning physicochemical properties such as hydrodynamic diameter, condensation efficiency and DNA release. The cytotoxicity of polymers and copolymers was studied in vitro. Furthermore, uptake- and transfection efficiency of polyplexes were evaluated in vitro. It was found that chitosan on the one hand showed very low cytotoxicity on the other hand, polyplexes tended to form large aggregates under physiological conditions, resulting in only 7% cellular uptake. Quaternization of chitosan improved the solubility, DNA condensation ability, and complex formation at physiological pH. Accordingly, cellular uptake was increased 8.5-fold compared to chitosan polyplexes and transfection efficiency was increased up to 678-fold. PEGylation of trimethyl chitosan not only decreased the cytotoxicity and further increased the solubility but also improved colloidal stability of polyplexes. These improvements resulted in a significant, up to 10-fold increase of transfection efficiency in NIH/3T3, L929 and MeWo cells compared to trimethyl chitosan. Cellular uptake pathways and intracellular processing of chitosan, trimethyl chitosan and polyethylenglycol-graft-trimethyl chitosan/DNA polyplexes were studied in two cell lines. The main uptake pathway in L929 cells was clathrin-mediated endocytosis accompanied by uptake via caveolae in the case of TMC and PTMC polyplexes. Transfection efficiency was strongly influenced by clathrin-mediated endocytosis. In A549 cells, uptake mainly occurred by clathrin-mediated endocytosis which was accompanied by macropinocytosis but transfection mainly depended on the former. However, it was found that besides the respective pathways other steps and barriers downstream polyplex uptake are at least of equal importance for efficient transfection with chitosan based vector systems., Die vorliegenden Arbeit befasst sich mit der Synthese und Charakterisierung neuer nicht-viraler Genvektoren. Im ersten Teil werden verschiedene Fragestellungen hinsichtlich des Gentransfers mittels zielgerichteter Polyethylenglykol (PEG)-modifizierter Polyethylenimine behandelt. Der zweite Teil beschäftigt sich mit Chitosan und Chitosanderivaten und deren Eignung zum Gentransfer. Zunächst wurden basierend auf PEI, PEG und dem HER2 spezifischen monoklonalen Antikörper Trastuzumab verschiedene Konjugate hergestellt, wobei sowohl die verwendeten PEG-PEI Kopolymere als auch die Linker variiert wurden. Die DNA-Komplexe der hergestellten Konjugate wurden dann hinsichtlich ihrer hydrodynamischen Durchmesser und Zetapotentiale untersucht. Alle Konjugatkomplexe zeigten Größen, die für eine endozytotische Aufnahme geeignet waren. Weiterhin zeigten alle Polyplexe neutrale Komplexoberflächen, sodass die unspezifische Bindung an nicht-Zielzellen deutlich vermindert war. In der Zellkultur wurden die verschiedenen Polyplexe hinsichtlich ihrer Bindungs- und Aufnahmeeffizienz in HER2 überexprimierende Zellen untersucht. Hierbei wurden signifikante Unterschiede zwischen den verschiedenen Konjugaten festgestellt, welche sich auch in den Transfektionseffizienzen wieder spiegelten. Die vielversprechendsten Ergebnisse wurden mit einem Konjugat erhalten, bei dem Trastuzumab mittels eines kurzen PEG-Linkers and PEI gekoppelt wurde. Die hergestellten Konjugate wurden weiterhin hinsichtlich ihrer Anwendbarkeit in vivo untersucht. Die Blutkompatibilität wurde durch Inkubation der Polyplexe mit isolierten Erythrozyten und nachfolgender Bestimmung der Hämolyse und der Erythrozytenmorphologie untersucht. Sowohl die hämolytische Aktivität als auch der Einfluss auf die Erythrozytenmorphologie war bei allen Konjugatkomplexen äußerst gering. Untersuchungen zur Pharmakokinetik und Organverteilung ergaben eine gegenüber PEI und DNA signifikant erhöhte AUC und eine signifikant verminderte Deposition in der Lunge und Milz. Weiterhin wurde ein HER2 überexprimierendes Tumormodell in SCID-Mäusen etabliert, um in Zukunft weitere in vivo Untersuchungen solcher Konjugate zu ermöglichen. Im zweiten Teil der Arbeit wurden Chitosan, Trimethyl Chitosan bzw. Polyethylenglykol-graft-Trimethyl Chitosan/pDNA Komplexe hinsichtlich ihrer physikochemischen Eigenschaften charakterisiert. Weiterhin wurde die Zytotoxizität sowie die Aufnahme- und Transfektionseffizienz in der Zellkultur untersucht. In diesen Untersuchungen zeigte Chitosan zwar einerseits sehr niedrige Zytotoxizitäten, andererseits bildeten sich aber unter physiologischen Bedingungen große Aggregate und die zelluläre Aufnahme betrug lediglich 7%. Die Quarternisierung von Chitosan erhöhte die Löslichkeit, die DNA Kondensierung und die Komplexbildung bei physiologischem pH. Die zelluläre Aufnahme war 8.5-fach gegenüber Chitosan-Komplexen erhöht, die Transfektionseffizienz stieg um das 678-fache. Die PEGylierung von Trimethyl Chitosan führte nicht nur zu einer Verringerung der Zytotoxizität und zu einer weiter erhöhten Löslichkeit sondern erhöhte auch die kolloidale Stabilität der Polyplexe. Dies führte zu einer bis zu 10-fach erhöhten Transfektionseffizienz gegenüber Trimethyl Chitosan in verschiedenen Zelllinien. Die zellulären Aufnahmewege und die intrazelluläre Prozessierung der Komplexe wurde in zwei Zelllinien untersucht. In L929 Zellen wurden die Komplexe hauptsächlich durch Clathrin-vermittelte Endozytose aufgenommen, die im Fall von TMC und PEG-TMC von Aufnahme über Caveolae begleitet wurde. Die Transfektionseffizienz wurde wesentlich durch Inhibierung der Clathrin-vermittelten Endozytose beeinflusst. In A549 Zellen wurde die Komplexe ebenfalls im wesentlichen durch Clathrin-vermittelte Endozytose aufgenommen, die in diesem Fall von Macropinozytose begleitet wurde. Die Transfektionseffizienz wurde wiederum wesentlich von Clathrin-vermittelter Endozytose beeinflusst. In dieser Studie wurde festgestellt, dass neben der zellulären Aufnahme weitere Schritte, v.a. der intrazellulären Prozessierung der Komplexe, von mindestens ebenso großer Bedeutung für die Transfektionseffizienz sind.

Maintaining the stability of therapeutic proteins is a hallmark of successful drug product (DP) development. The stability of DPs is closely monitored during various stages such as development, manufacturing, and in clinical preparation. However, the stability of a DP can change considerably after administration (in vivo stability). Assessing protein stability under physiologic conditions can improve the safety and efficacy profile of commercial DPs. The first part of the thesis focuses on protein aggregation and fragmentation of monoclonal antibodies (mAbs), after simulated intravenous administration. The molecules showed markedly different stability in human serum with some mAbs being subjected to substantial fragmentation, while others remained stable. Moreover, substantial sub-visible particle formation in serum was detected with developed methods such as fluorescence microscopy and flow cytometry. In the second part of this thesis, we simulated the subcutaneous administration of mAbs without fluorescence labeling. In line with our previous observations, mAb candidates had substantial differences in their stability under physiologic conditions. Furthermore, we showed that phosphate-buffered saline does not stress mAbs to the same extent as bicarbonate-buffers and human serum. The in vivo stability of therapeutic proteins should be an integral part of holistic stability assessments during early stages of development.

Even though tumor treatment has achieved remarkable successes in recent years, cancer is still one of the severest diseases there is. According to statistics published by the WHO, cancer is the second leading cause of death worldwide. Chemotherapy is one of the most widely applied cancer treatment methods since it emerged in the 1940s. Despite of research and development efforts made meanwhile, many compounds used in chemotherapy present major drawbacks. Nanotechnology, as an addon to the anti-cancer drug development, is a promising strategy to cope with certain shortcomings of otherwise encouraging compounds. In this PhD thesis introduction, the reader will be given the understanding, basics and vocabulary to successfully navigate through the publications. At first, the basics of cancer are discussed and points of attack for treatments are described. Then, a short overview will present the development of chemotherapeutics from the beginnings up to nanomedicine. In our studies, we explored different approaches to developing enzyme-triggered nanoformulations. Our enzyme-responsive formulations were tested in vitro and, to some extent, in vivo. The conclusion is focused on putting the nanoformulation in context with current progress and presuming opportunities and risks.

Im Rahmen dieser Promotionsarbeit wurden β-Faltblatt-bildende SSP-Systeme auf ihre Hydrogeleigenschaften, ihren Einfluss auf die Zellreaktion, ihre intrinsische antimikrobielle Aktivität und ihr Antibiotika-Freisetzungspotential untersucht. Insgesamt ergab diese Promotionsarbeit, dass P11-SSP-Hydrogele eine Reihe von Eigenschaften besitzen, welche erforderlich sind, um sie zu potentiellen Kandidaten im Rahmen neuartiger therapeutischer Strategien für die Behandlung der Parodontitis weiterzuentwickeln., Within this PhD project, β-sheet forming SAP systems were investigated for their hydrogel properties, their influence on cell response, their intrinsic antimicrobial activity and their antibiotic delivery potential. Overall, this thesis showed that P11-SAP hydrogels possess a number of properties that are necessary to develop them into potential candidates in novel therapeutic strategies for the treatment of periodontitis.
