Abstract

BackgroundSelf-management of Type 2 diabetes mellitus (T2D) is challenging. Regular self-monitoring of blood glucose and healthy lifestyles are required to improve glycometabolic control, thus delaying diabetes complications, and reducing hospitalizations. Digital technologies can empower patients in their disease management promoting self-management and motivation to change behaviors. We report the results of an exploratory trial aimed at evaluating the metabolic outcomes of using digital solutions for T2D self-management developed in the ProEmpower project, a European Commission funded Pre-Commercial Procurement.MethodsTwo digital solutions, DM4All and DiaWatch, which were codesigned with providers, patients, and caregivers, enabled the collection of clinical parameters by the patient using a smartphone integrated with the medical devices (glucometer, sphygmomanometer, scale, smart watch for heart rate monitoring and step counter). Data were automatically sent to the shared care plan allowing professionals to monitor adherence to treatment, set goals, and communicate more effectively with patients. At baseline and after an average follow-up of 8 months, glycosylated hemoglobin (HbA1c), body weight, blood pressure, and blood lipids were measured in 100 T2D patients using the ProEmpower solutions across different diabetes centers in Campania Region, age 45–79 years, both genders, and compared with a Control cohort of T2D patients (n = 100) with similar clinical characteristics and followed for a comparable period of observation in the same centers.ResultsAt baseline, the ProEmpower participants and the Control subjects were on average overweight, with a similar BMI in the two cohorts, and mean HbA1c was at acceptable levels (around 7.0%). Abstract

The purpose of this systematic review was to evaluate whether there are scientific evidence regarding the association between periodontitis and obstructive sleep apnea (OSA) in adults. An electronic search was performed on MEDLINE/PubMed for prospective and retrospective longitudinal studies, cohort studies, and case–control studies conducted in human adults affected by both OSA and periodontitis. Two reviewers extracted the data using a custom Excel spreadsheet. A methodological assessment of the quality of the studies was performed using the Newcastle–Ottawa Scale. Fourteen studies were included. All studies evaluated the association between periodontitis and OSA. None of the studies evaluated the cause–effect relationship. Eleven studies found a significant positive relationship between periodontitis and OSA, whereas three found no statistically significant association. Several study limitations were observed, such as lack of standardization of study groups, diagnosis of periodontitis and OSA, and differences in study design. Evidence of a plausible association between periodontitis and OSA was found. The possible relationship could be explained by systemic inflammation, oral breathing, and the comorbid relationship attributable to common risk factors. Observational and randomized controlled studies are needed to clarify the mechanism of interaction between the two conditions.

Abstract

Wide genome analysis of difference in gene expression between spermatogonial populations from 7-day-old mice and pachytene spermatocytes from 18-day-old mice was performed using Affymetrix gene chips representing similar to 12,500 mouse known genes or EST sequences. spanning approximately 1/3rd of the mouse genome. To delineate differences in the profile of gene expression between mitotic and meiotic stages of male germ cell differentiation, expressed genes were grouped in functional clusters. The analysis confirmed the previously described pre-meiotic or meiotic expression for several genes, in particular for those involved in the regulation of the mitotic and meiotic cell cycle. and for those whose transcripts are accumulated during the meiotic stages to be translated later in post-meiotic stages. Differential expression of several additional genes was discovered. In few cases (pro-apoptotic factors Bak, Bad and Bax), data were in conflict with the previously published stage-dependent expression of genes already known to be expressed in male germ cells. Northern blot analysis of selected genes confirmed the results obtained with the microarray chips. Six of these were novel genes specifically expressed in pachytene spermatocytes: a chromatin remodeling factor (chrac1/YCL1). a homeobox gene (hmx1), a novel G-coupled receptor for an unknown ligand (Gpr19), a glycoprotein of the intestinal epithelium (mucin 3), a novel RAS activator (Ranbp9), and the A630056B21Rik gene (predicted to encode a novel zinc finger protein). These studies will help to delineate the global patterns of gene expression characterizing male germ cell differentiation for a better understanding of regulation of spermatogenesis in mammals. Abstract

Translation of stored mRNAs accounts for protein synthesis during the transcriptionally inactive stages of spermatogenesis. A key step in mRNA translation is the assembly of the initiation complex EIF4F, which is regulated by the MTOR (mammalian target of rapamycin) and MNK1/2 (MAP kinase-interacting kinase 1 and 2) pathways. We investigated the expression and activity of regulatory proteins of these pathways in male germ cells at different stages of differentiation. All translation factors analyzed were expressed in germ cells throughout spermatogenesis. However, while EIF4G and PABP1 (poly[A]-binding protein 1) were more abundant in postmeiotic cells, MTOR and its target EIF4EBP1 (4E-BP1) decreased steadily during spermatogenesis. In vivo labeling showed that pachytene spermatocytes display higher rates of protein synthesis, which are partially dependent on MTOR and MNK activity. By contrast, haploid spermatids are characterized by lower levels of protein synthesis, which are independent of the activity of these pathways. Accordingly, MTOR and MNK activity enhanced formation of the EIF4F complex in pachytene spermatocytes but not in round spermatids. Moreover, external cues differentially modulated the activity of these pathways in meiotic and haploid cells. Heat shock decreased MTOR and MNK activity in pachytene spermatocytes, whereas round spermatids were much less sensitive. On the other hand, treatment with the phosphatase inhibitor okadaic acid activated MTOR and MNK in both cell types. These results indicate that translational regulation is differentially dependent on the MTOR and MNK pathways in mouse spermatocytes and spermatids and suggest that the late stages of germ cell differentiation display constitutive assembly of the translation initiation complex.

Abstract

Palaeolithic rock art belongs to the most ancient artistic representations of human beings. Famous examples are the Altamira and Lascaux paintings. These paintings are conserved under very exceptional conditions. Basically, the pigments are composed of iron oxides or ochre for red and yellow paints, and charcoal and manganese oxides for black colors. Many studies have already been devoted to the characterization of the employed pigments, their treatment prior to application and the artistic techniques (Baffier et al. 1999, Pomiès et al. 1999, Chalmin et al. 2003 and 2004). However, rock art is today threatened by numerous alteration phenomena, among them the formation and interaction with calcium carbonate concretions. The cave “Grande Grotte” of Arcy-sur-Cure (24500-28000 BP, Yonne, France) with prehistoric paintings is particularly concerned by this problem. This cave is generally known for its impressive calcite speleothems as stalactites and stalagmites. Important calcite concretions also cover the walls. Basically, two types of calcite have been observed on the walls: translucent yellowish layers and opaque white or grey layers. Other calcite types are present in the lakes of the cave (floating calcite rafts at the surface of the lake and soft calcite at the bottom of the lake). Prehistoric figures were discovered in 1990 after a cleaning of the dark walls, fouled during several centuries of visits, of this karst cave with an acidic solution,. In the past, the presence of the calcite layers on the walls had a protective effect of the prehistoric paints. Today, prehistorians try to identify the ensemble of prehistoric representations in the cave. So, identification and restoration of a part of the cave paintings has been done by removing the opaque white calcite layer by mechanical abrasion while keeping another underlying translucent layer, in order to evidence and to study the paintings (Girard et al., 2001).

Abstract

International audience; Palaeolithic rock art belongs to the most ancient artisitc representations of human beings. Conserved under very exceptional conditions, it is today threatened by numerous alteration phenomena, among them the formation and interaction with calium carbonate concretions. The cave “Grande Grotte” of Arcy-sur-Cure (Yonne, France) with prehistoric paintings shows important calcite concretions. Two types of calcite have been observed on the walls: translucent yellowish layers and opaque white or grey layers that obscure the paintings. Other calcite types are present in the lakes of the cave (floating calcite rafts at the surface of the lake and soft calcite at the bottom of the lake). The morphology of the different calcites was observed at different scales by optical microscopy with normal and polarized light, SEM and TEM. The elemental composition was measured by micro-PIXE and the structure by XRD, IR and Raman spectroscopy. The bacterial diversity and its role in calcite formation were assessed by culture and 16S-SSCP in order to distinguish and to assess various abiotic and biotic formation mechanisms. The investigation of calcite characteristics enables conclusions on the formation mechanism and on a biotic or abiotic origin of the calcites. The change of calcite types on the walls reveals changes of the environmental cave parameters. In addition, interactions of calcites with the prehistoric paint layer could be evaluated.
