Abstract

Aging, often considered a result of random cellular damage, can be accurately estimated using DNA methylation profiles, the foundation of pan-tissue epigenetic clocks. Here, we demonstrate the development of universal pan-mammalian clocks, using 11,754 methylation arrays from our Mammalian Methylation Consortium, which encompass 59 tissue types across 185 mammalian species. These predictive models estimate mammalian tissue age with high accuracy (r > 0.96). Age deviations correlate with human mortality risk, mouse somatotropic axis mutations and caloric restriction. We identified specific cytosines with methylation levels that change with age across numerous species. These sites, highly enriched in polycomb repressive complex 2-binding locations, are near genes implicated in mammalian development, cancer, obesity and longevity. Our findings offer new evidence suggesting that aging is evolutionarily conserved and intertwined with developmental processes across all mammals.

Abstract

Homophilous behaviour plays a central role in the formation of human friendships. Individuals form social ties with others that show similar phenotypic traits, independently of relatedness. Evidence of such homophily can be found in bottlenose dolphins ( Tursiops aduncus) in Shark Bay, Western Australia, where females that use marine sponges as foraging tools often associate with other females that use sponges. 'Sponging' is a socially learned, time-consuming behaviour, transmitted from mother to calf. Previous research illustrated a strong female bias in adopting this technique. The lower propensity for males to engage in sponging may be due to its incompatibility with adult male-specific behaviours, particularly the formation of multi-level alliances. However, the link between sponging and male behaviour has never been formally tested. Here, we show that male spongers associated significantly more often with other male spongers irrespective of their level of relatedness. Male spongers spent significantly more time foraging, and less time resting and travelling, than did male non-spongers. Interestingly, we found no difference in time spent socializing. Our study provides novel insights into the relationship between tool use and activity budgets of male dolphins, and indicates social homophily in the second-order alliance composition of tool-using bottlenose dolphins.

4. Bothersome symptoms at midlife in relation to body fat percentage.

Abstract

Objectives: Increasing obesity has been associated with a higher frequency of symptoms at midlife. Bothersomeness represents an important measure of perceived symptom severity, but has received relatively little consideration, and relationships between symptom bothersomeness and obesity are not known. We evaluated the association between body fat percentage (%BF) and the bothersomeness of symptoms at midlife. Methods: This cross‐sectional study included women aged 40–60 in Qatar (n = 841). Participants reported frequency and bothersomeness of midlife symptoms hypothesized to be related to body composition. Initially, we characterized the relationship between continuous %BF and presence (yes/no) and bothersomeness (yes/no) for each symptom using restricted cubic spline (RCS) models to test nonlinearity. Subsequently, we used multinomial logistic regressions to evaluate associations between %BF and multilevel symptom outcomes, where categories were: (a) no symptoms or bother, (b) symptoms without bothersomeness, and (c) symptoms with bothersomeness. Results: The highest frequency of bothersomeness was reported for aches/stiffness in joints (51%), followed by trouble sleeping (34%), night sweats (21%), urinary incontinence (18%), hot flashes (16%), and shortness of breath (15%). In unadjusted multinomial logistic regressions, aches/stiffness in joints with and without bothersomeness had the same significant relationship with %BF. Bothersome night sweats, urinary incontinence, and hot flashes were significantly associated with %BF (p <.05), but those same symptoms without bothersomeness were not significantly associated with %BF. Conclusions: Our findings suggest that bothersomeness is an important variable that tracks with body fat and gives different information than report of the presence/absence of a symptom alone. Abstract

Background & aims: An estimated 38% of adults worldwide have non-alcoholic fatty liver disease (NAFLD). From individual impacts to widespread public health and economic consequences, the implications of this disease are profound. This study aimed to develop an aligned, prioritised fatty liver disease research agenda for the global health community. Methods: Nine co-chairs drafted initial research priorities, subsequently reviewed by 40 core authors and debated during a three-day in-person meeting. Following a Delphi methodology, over two rounds, a large panel (R1 n = 344, R2 n = 288) reviewed the priorities, via Qualtrics XM, indicating agreement using a four-point Likert-scale and providing written feedback. The core group revised the draft priorities between rounds. In R2, panellists also ranked the priorities within six domains: epidemiology, models of care, treatment and care, education and awareness, patient and community perspectives, and leadership and public health policy. Results: The consensus-built fatty liver disease research agenda encompasses 28 priorities. The mean percentage of ‘agree’ responses increased from 78.3 in R1 to 81.1 in R2. Five priorities received unanimous combined agreement (‘agree’ + ‘somewhat agree’); the remaining 23 priorities had >90% combined agreement. While all but one of the priorities exhibited at least a super-majority of agreement (>66.7% ‘agree’), 13 priorities had [removed]90% combined agreement. Conclusions: Adopting this multidisciplinary consensus-built research priorities agenda can deliver a step-change in addressing fatty liver disease, mitigating against its individual and societal harms and proactively altering its natural history through prevention, identification, treatment, and care. Abstract

Creative approaches at the interface of ecology, statistics, mathematics, informatics, and computational science are essential for improving our understanding of complex ecological systems. For example, new information technologies, including powerful computers, spatially embedded sensor networks, and Semantic Web tools, are emerging as potentially revolutionary tools for studying ecological phenomena. These technologies can play an important role in developing and testing detailed models that describe real-world systems at multiple scales. Key challenges include choosing the appropriate level of model complexity necessary for understanding biological patterns across space and time, and applying this understanding to solve problems in conservation biology and resource management. Meeting these challenges requires novel statistical and mathematical techniques for distinguishing among alternative ecological theories and hypotheses. Examples from a wide array of research areas in population biology and community ecology highlight the importance of fostering synergistic ties across disciplines for current and future research and application. © 2005 American Institute of Biological Sciences.

10. Synthetic satellite data generation to inform hydrological models in data scarce areas

Abstract

The creation of synthetic images of precipitation, temperature, evapotranspiration, and terrestrial water storage is proposed to address the gaps in satellite data availability prior to the year 2000 and extend the data to pre-satellite periods. This is necessary to model and manage water resources and evaluate the impact of climate change on hydrological processes in regions with limited data. The synthetic images should closely resemble real satellite images.The approach is based on the relationship between meteorological factors and existing satellite images and the idea that, under similar weather conditions, patterns of specific processes may repeat over time. The ERA5 reanalysis data is used as the meteorological predictor, and a K-Nearest Neighbor algorithm with a process-specific similarity metric is applied to generate the synthetic images.The method is tested in the Volta River Basin in West Africa where water resources are critically impacted by climate change. The synthetic images are input into a spatially-distributed hydrological model for calibration and validation, and their quality is assessed by their ability to reproduce historical streamflow time series. The goal of this testing phase is to improve the generation technique and produce synthetic images that closely approximate unobserved processes and improve the accuracy of the modeling.

13. Impact of Changing Distance-Learning Formats in Physical Therapy Professional Education.

Abstract

AIMS: In early 2020, the COVID-19 pandemic caused a majority of higher education to shift content delivery formats to allow for social distancing to decrease spread of the virus. The purpose of this investigation was to discover physical therapy students' perceived impact from changing from a synchronous videoconferencing format to a more hybrid program format. METHODS: A qualitative case study design bounded by the time of switching formats due to the pandemic was chosen (March 2020 through September 2020). Physical therapy students participated in an agreement survey (n=38) and semi-structured interviews (n=12). Survey and interview data were coded and analyzed to form categories and themes of discovered perspectives. RESULTS: Students' perceived proficiency of hands-on skill was negatively affected. Cross-campus student and faculty interactions improved. Students felt changing formats would not result in detrimental effects on their overall learning nor on their career potential as physical therapists. DISCUSSION: Educators in entry-level professional physical therapy programs utilizing distance-education models should consider and adjust timing of hands-on skill instruction to match didactic content to encourage better connection and clinical application. Distance-learning educators should foster more interaction with students who may feel isolated. Interaction between distance-separated cohorts can reduce feelings of competition and inequality between campus locations and create improved learning communities. [ABSTRACT FROM AUTHOR]

Abstract

This article investigates the viability of using forced allocation as a mechanism to alleviate capacity challenges through scenarios of varying allowable citrus throughput at the Port of Durban. Rule-based resource allocation techniques are used to divide the allowable citrus throughput at the constrained port between production regions by setting quotas. The allocation model framework minimises the impact of the forced allocation on the citrus export cold chain by setting the quotas as constraints in an integer programming (IP) formulation of a minimum cost transportation problem (TP) for the system. The results show that forced allocation is feasible under at least one resource allocation technique for four of the scenarios tested. [ABSTRACT FROM AUTHOR]

Abstract

Non-alcoholic fatty liver disease (NAFLD) is a potentially serious liver disease that affects approximately one-quarter of the global adult population, causing a substantial burden of ill health with wide-ranging social and economic implications. It is a multisystem disease and is considered the hepatic component of metabolic syndrome. Unlike other highly prevalent conditions, NAFLD has received little attention from the global public health community. Health system and public health responses to NAFLD have been weak and fragmented, and, despite its pervasiveness, NAFLD is largely unknown outside hepatology and gastroenterology. There is only a nascent global public health movement addressing NAFLD, and the disease is absent from nearly all national and international strategies and policies for non-communicable diseases, including obesity. In this global Delphi study, a multidisciplinary group of experts developed consensus statements and recommendations, which a larger group of collaborators reviewed over three rounds until consensus was achieved. The resulting consensus statements and recommendations address a broad range of topics — from epidemiology, awareness, care and treatment to public health policies and leadership — that have general relevance for policy-makers, health-care practitioners, civil society groups, research institutions and affected populations. These recommendations should provide a strong foundation for a comprehensive public health response to NAFLD. © 2021, Springer Nature Limited.

Abstract

Investigations into cooperative partner choice should consider both potential and realised partners, allowing for the comparison of traits across all those available. Male bottlenose dolphins form persisting multi-level alliances. Second-order alliances of 4–14 males are the core social unit, within which 2–3 males form first-order alliances to sequester females during consortships. We compared social bond strength, relatedness and age similarity of potential and realised partners of individual males in two age periods: (i) adolescence, when second-order alliances are formed from all available associates, and (ii) adulthood, when first-order allies are selected from within second-order alliances. Social bond strength during adolescence predicted second-order alliance membership in adulthood. Moreover, males preferred same-aged or older males as second-order allies. Within second-order alliances, non-mating season social bond strength predicted first-order partner preferences during mating season consortships. Relatedness did not influence partner choice on either alliance level. There is thus a striking resemblance between male dolphins, chimpanzees and humans, where closely bonded non-relatives engage in higher-level, polyadic cooperative acts. To that end, our study extends the scope of taxa in which social bonds rather than kinship explain cooperation, providing the first evidence that such traits might have evolved independently in marine and terrestrial realms. © The Authors

Abstract

Aedes mosquitoes (Diptera: Culicidae), principle vectors of several arboviruses, typically lay eggs in man‐made water‐filled containers located near human dwellings. Given the widespread emergence of insecticide resistance, stable and biofriendly alternatives for mosquito larviciding are needed. Laboratory studies have demonstrated that inactivated yeast interfering RNA tablets targeting key larval developmental genes can be used to facilitate effective larvicidal activity while also promoting selective gravid female oviposition behaviour. Here we examined the efficacy of transferring this technology toward development of lure‐and‐kill ovitraps targeting Aedes aegypti (L.) and Aedes albopictus (Skuse) female mosquitoes. Insectary, simulated field and semi‐field experiments demonstrated that two mosquito‐specific yeast interfering RNA pesticides induce high levels of mortality among larvae of both species in treated large volume containers. Small‐scale field trials conducted in Trinidad, West Indies demonstrated that large volume ovitrap containers baited with inactivated yeast tablets lure significantly more gravid females than traps containing only water and were highly attractive to both A. aegypti and A. albopictus females. These studies indicate that development of biorational yeast interfering RNA‐baited ovitraps may represent a new tool for control of Aedes mosquitoes, including deployment in existing lure‐and‐kill ovitrap technologies or traditional container larviciding programs. 50. Molecular epidemiology of methicillin-resistant staphylococci amongst veterinary personnel, personnel-owned pets, patients and the hospital environment of two companion animal veterinary hospitals

Abstract

This study investigated the transmission cycle of methicillin-resistant Staphylococcus aureus (MRSA) and Staphylococcus pseudintermedius (MRSP) in small companion animal veterinary practice. Sampling was undertaken at two small animal veterinary hospitals in Sydney, Australia. Samples were collected from 46 veterinary personnel, 79 personnel-owned dogs and cats, 151 clinically normal canine hospital admissions and 25 environmental sites. Nasal swabs were collected from veterinary personnel. Nasal, oral and perineal swabs were collected from animals. Methicillin resistance was detected by growth on BrillianceTM MRSA 2 Agar and confirmed by cefoxitin and oxacillin broth microdilution for S. aureus and S. pseudintermedius, respectively. MRSA and MRSP isolates were characterised using whole genome sequencing including mecA gene screening and multilocus sequence typing. MRSA was isolated from four (8%) veterinary personnel but no animals. MRSP was isolated from 11/151 (7%) of canine hospital admissions and 4/53 (8%) of personnel-owned dogs but no veterinary personnel or cats. No MRSA or MRSP was isolated from the environment. MRSP isolates were resistant to significantly more antimicrobial classes than MRSA. The main MRSP clone carried by canine patients (ST496) was distinct to that carried by personnel-owned dogs (ST64). One veterinary nurse, who carried Panton Valentine leucocidin-positive ST338 MRSA, also owned a ST749 MRSP-positive dog. Besides MRSP-positive dogs from the same household sharing the same clone of MRSP, MRSA and MRSP were not shared between humans, animals or environment. Therefore, in the non-outbreak setting of this study, there was limited MRS transmission between veterinary personnel, their pets, patients or the veterinary environment.
