Only 11% of community college (associate's-degree) students transfer vertically and obtain a bachelor's degree within six years, despite over 80% originally intending to do so. These leaks in the transfer pipeline disproportionately affect students from underrepresented groups, who are more likely to attend community colleges and to leak out of the pipeline. To obtain insights about how to decrease these leaks, a survey was distributed to all City University of New York undergraduates; 31,511 responded. The survey concerned students' life and academic circumstances, as well as their information about and views on transfer. Analyses particularly compared responses of never-transferred associate's and bachelor's students and vertical transfer students. The results suggest multiple actions that higher education can take to decrease the pipeline leaks and increase higher education equity, including by increasing transfer students' belongingness, course availability, credit transfer, financial support, information, and time efficiency.

Abstract The Knight-Alzheimer Disease Research Center (Knight-ADRC) at Washington University in St. Louis has pioneered and led worldwide seminal studies that have expanded our clinical, social, pathological, and molecular understanding of Alzheimer Disease. Over more than 40 years, research volunteers have been recruited to participate in cognitive, neuropsychologic, imaging, fluid biomarkers, genomic and multi-omic studies. Tissue and longitudinal data collected to foster, facilitate, and support research on dementia and aging. The Genetics and high throughput -omics core (GHTO) have collected of more than 26,000 biological samples from 6,625 Knight-ADRC participants. Samples available include longitudinal DNA, RNA, non-fasted plasma, cerebrospinal fluid pellets, and peripheral blood mononuclear cells. The GHTO has performed deep molecular profiling (genomic, transcriptomic, epigenomic, proteomic, and metabolomic) from large number of brain (n = 2,117), CSF (n = 2,012) and blood/plasma (n = 8,265) samples with the goal of identifying novel risk and protective variants, identify novel molecular biomarkers and causal and druggable targets. Overall, the resources available at GHTO support the increase of our understanding of Alzheimer Disease.

IntroductionCover crop services depend on biomass production and species composition of the cover crop stand. In this study, we investigated the interactions in shoot biomass of dual cover crop mixtures and their competitiveness to suppress weeds before winter under different growing conditions.MethodsA field experiment was performed on sandy loam soil in Triesdorf, Germany, for two consecutive years. The cover crop species white mustard (Sinapis alba L.), oil radish (Raphanus sativus var. oleiformis L.), phacelia (Phacelia tanacetifolia J.), Egyptian clover (Trifolium alexandrinum L.), common vetch (Vicia sativa L.), field pea (Pisum sativum L.), and field bean (Vicia faba L.) were grown in pure stands or dual mixtures. Cover crops were sown in August, and shoot biomass was harvested at the end of the vegetation period. Cover crop shoot biomass was dried, and the share of each species and the sown test weed (Brassica napus L.) were determined.ResultsThe share of shoot biomass of a cover crop species in dual-species mixtures was closely related to its shoot biomass in its pure stand. In both years, mustard and phacelia showed similar interactions in shoot biomass production when growing with a second species. Regarding competitiveness against weeds, cruciferous species outperformed other cover crop species and could suppress weeds even if they were mixed with a less competitive partner. Weed suppression of mixtures with phacelia depended on the second component in the mixture and was more effective in a dry season. Legume species, especially Egyptian clover and common vetch could not suppress weeds in a pure stand.DiscussionOur results show that species with high weed suppression potential as single stands retain this ability in dual cover crop mixtures, regardless of the suppression potential of the second species that completes the mixture.

Abstract Background Chronic lung disease affects nearly 37 million Americans and often results in significant quality of life impairment and healthcare burden. Despite guidelines calling for palliative care (PC) integration into pulmonary care as a vital part of chronic lung disease management, existing PC models have limited access and lack scalability. Use of telehealth to provide PC offers a potential solution to these barriers. This study explored perceptions of patients with chronic lung disease regarding a telehealth integrated palliative care (TIPC) model, with plans to use findings to inform development of an intervention protocol for future testing. Methods For this qualitative study, we conducted semi-structured interviews between June 2021- December 2021 with patients with advanced chronic lung disease. Interviews explored experiences with chronic lung disease, understanding of PC, and perceived acceptability of the proposed model along with anticipated facilitators and barriers of the TIPC model. We analyzed findings with a content analysis approach. Results We completed 20 interviews, with two that included both a patient and caregiver together due to patient preference. Perceptions were primarily related to three categories: burden of chronic lung disease, pre-conceived understanding of PC, and perspective on the proposed TIPC model. Analysis revealed a high level of disease burden related to chronic lung disease and its impact on day-to-day functioning. Although PC was not well understood, the TIPC model using a shared care planning approach via telehealth was seen by most as an acceptable addition to their chronic lung disease care. Conclusions These findings emphasize the need for a patient-centered, shared care planning approach in chronic lung disease. The TIPC model may be one option that may be acceptable to individuals with chronic lung disease. Future work includes using findings to refine our TIPC model and conducting pilot testing to assess acceptability and utility of the model.

Objective: There are many potential barriers to an individual's readiness and ability to use telehealth, including technology access, trust and knowledge. This qualitative study was a partnership between Jefferson and Esperanza Health Center (EHC), focused on addressing barriers to digital readiness among the Latino population served by EHC. Methods: We conducted semi-structured interviews with Latino patients to assess their experiences with technology and willingness to learn more about technology for healthcare. Interviews were transcribed, coded, and analyzed using a content analysis approach. Results: We completed 28 interviews. Most interviews (n = 23, 82 %) were conducted in Spanish. Barriers to using health technology included forgetting passwords, platforms not being available in multiple languages, and lacking digital skills. Conclusion: Participants identified a need and interest in receiving support to develop technological skills needed to access health information and engage in digital health services. Future work is needed to develop patient-centered interventions to support the digital health readiness needs of underserved populations. Innovation: Healthcare systems and community health organizations can partner to build the capacity of community members to identify barriers in digital health use. These partnerships have the potential to empower communities to create culturally sensitive interventions that aim to increase digital health literacy.

ObjectivesEfforts to promote COVID-19 vaccine acceptance must consider the critical role of the emergency department (ED) in providing health care to underserved patients. Focusing on patients who lacked primary care, we sought to elicit the perspectives of unvaccinated ED patients regarding COVID-19 vaccination concerns and potential approaches that might increase their vaccine acceptance.MethodsWe conducted this qualitative interview study from August to November 2021 at four urban EDs in San Francisco, California; Seattle, Washington; Durham, North Carolina; and Philadelphia, Pennsylvania. We included ED patients who were ≥18 years old, fluent in English or Spanish, had not received a COVID-19 vaccine, and did not have primary care physicians or clinics. We excluded patients who were unable to complete an interview, in police custody, under suspicion of active COVID-19 illness, or presented with a psychiatric chief complaint. We enrolled until we reached thematic saturation in relevant domains. We analyzed interview transcripts with a content analysis approach focused on identifying concerns about COVID-19 vaccines and ideas regarding the promotion of vaccine acceptance and potential trusted messengers.ResultsOf 65 patients enrolled, 28 (43%) identified as female, their median age was 36 years (interquartile range 29-49), and 12 (18%) interviews were conducted in Spanish. Primary concerns about COVID-19 vaccines included risk of complications, known and unknown side effects, and fear of contracting COVID-19 from vaccines. Trust played a major role for patients in deciding which sources to use for vaccine information and in engendering vaccine acceptance. Health care providers and family or friends were commonly cited as trusted messengers of information.ConclusionsWe characterized concerns about COVID-19 vaccines, uncovered themes that may promote vaccine acceptance, and identified trusted messengers-primarily health care professionals. These data may inform the development of nuanced COVID-19 vaccine messaging platforms to address COVID-19 vaccine hesitancy among underserved ED populations.

BackgroundWe conducted in-depth interviews to characterize reasons for COVID-19 vaccine hesitancy in emergency department (ED) patients and developed messaging platforms that may address their concerns. In this trial, we seek to determine whether provision of these COVID-19 vaccine messaging platforms in EDs will be associated with greater COVID-19 vaccine acceptance and uptake in unvaccinated ED patients.MethodsThis is a cluster-randomized controlled trial (RCT) evaluating our COVID-19 vaccine messaging platforms in seven hospital EDs (mix of academic, community, and safety-net EDs) in four US cities. Within each study site, we randomized 30 1-week periods to the intervention and 30 1-week periods to the control. Adult patients who have not received a COVID-19 vaccine are eligible with these exclusions: (1) major trauma, intoxication, altered mental status, or critical illness; (2) incarceration; (3) psychiatric chief complaint; and (4) suspicion of acute COVID-19 illness. Participants receive an orally administered Intake survey. During intervention weeks, participants then receive three COVID-19 vaccine messaging platforms (4-min video, one-page informational flyer and a brief, scripted face-to-face message delivered by an ED physician or nurse); patients enrolled during non-intervention weeks do not receive these platforms. Approximately, an hour after intake surveys, participants receive a Vaccine Acceptance survey during which the primary outcome of acceptance of the COVID-19 vaccine in the ED is ascertained. The other primary outcome of receipt of a COVID-19 vaccine within 32 days is ascertained by electronic health record review and phone follow-up. To determine whether provision of vaccine messaging platforms is associated with a 7% increase in vaccine acceptance and uptake, we will need to enroll 1290 patients.DiscussionHighlighting the difficulties of trial implementation during the COVID-19 pandemic in acute care settings, our novel trial will lay the groundwork for delivery of public health interventions to vulnerable populations whose only health care access occurs in EDs.ConclusionsToward addressing vaccine hesitancy in vulnerable populations who seek care in EDs, our cluster-RCT will determine whether implementation of vaccine messaging platforms is associated with greater COVID-19 vaccine acceptance and uptake in unvaccinated ED patients.Trial statusWe began enrollment in December 2021 and expect to continue through 2022.Trial registrationClinicalTrials.gov NCT05142332 . Soil structure is sensitive to intensive soil management. It can be ameliorated by a reduction in soil cultivation and stimulation of plant and microbial mediators for aggregate formation, with the latter being a prerequisite and measure for soil quality. Cover crops (CCs) are part of an integrated approach to stabilize or improve soil quality. Thereby, the incorporation of diverse CC mixtures is hypothesized to increase the positive effects of CC applications. This study entailed an investigation of the legacy effect of CCs on soil aggregates after three crop rotations in the second main crop (winter wheat) after the last CC treatment. Four CCs (mustard, phacelia, clover, and oat) cultivated in pure stands and with a fallow treatment were compared to a mixture of the four CC species (Mix4) and a highly diverse 12-plant-species mixture (Mix12) in a long-term field experiment in Germany. The organic carbon (OC) distribution within macroaggregate fractions (16–8, 8–4, 4–2, 2–1, and mm) and their aggregate stability were measured by dry- and wet-sieving methods, and the mean weight diameter (MWD) was calculated from water-stable aggregates. The results showed that, compared to the fallow, all CCs increased the MWD between 10 % and 19 % in soil under the following main crop. The average MWD increase over the fallow was slightly higher for CC mixtures (16 %) than for single CCs (12 %). Most of the OC (67.9 % on average) was stored in the mm aggregate fraction, highest in the topsoil and decreasing with soil depth. The intermediate fractions (8–4 mm, 4–2 mm, 2–1 mm) stored 8.5 %, 10.5 %, and 11.0 % of the total OC, while 2.1 % was stored in the 16–8 mm fraction. Higher MWD improvement at the 20–30 cm depth also indicates additional benefits from a reduction in the cultivation depth. Structural equation modelling (SEM) suggests that single CCs were more likely to increase OC storage in small macroaggregates mm, while CC mixtures were more likely to increase OC in the largest fraction (8–16 mm). Different individual CC species or mixtures exhibited varying involvement in the formation of different aggregate fractions. We provide evidence that litter quality, root morphology, and rhizosphere input, which affect microbial mediators of aggregate formation, might be the main reasons for the observed differences between CC treatments. Cover crops are valuable multifunctional tools for sustainable soil management. Here, we showed that they contribute to structure amelioration in arable soils. Increasing the functional diversity of plant species in CC mixtures could be a strategy to further enhance the positive effects of CCs in agroecosystems.

Abstract Background Despite well-established guidelines to treat diabetes, many people with diabetes struggle to manage their disease. For many, this struggle is related to challenges achieving nutrition-related lifestyle changes. We examined how people with diabetes describe barriers to maintaining a healthy diet and considered the benefits of using a harm reduction approach to assist patients to achieve nutrition-related goals. Methods This is a secondary analysis of 89 interviews conducted with adults who had type 1 or type 2 diabetes. Interviews were analyzed using a content analysis approach. Themes regarding food or diet were initially captured in a “food” node. Data in the food node were then sub-coded for this analysis, again using a content analysis approach. Results Participants frequently used addiction language to talk about their relationship with food, at times referring to themselves as “an addict” and describing food as “their drug.” Participants perceived their unhealthy food choices either as a sign of weakness or as “cheating.” They also identified food’s ability to comfort them and an unwillingness to change as particular challenges to sustaining a healthier diet. Conclusion Participants often described their relationship with food through an addiction lens. A harm reduction approach has been associated with positive outcomes among those with substance abuse disorder. Patient-centered communication incorporating the harm reduction model may improve the patient-clinician relationship and thus improve patient outcomes and quality-of-life while reducing health-related stigma in diabetes care. Future work should explore the effectiveness of this approach in patients with diabetes. Trial registration Registered on ClinicalTrials.gov, NCT02792777. This longitudinal study of 17,455 students, the majority from underrepresented groups, investigated leaks in the vertical transfer pipeline from associate's-degree program entry to bachelor's-degree receipt. Investigated were both the size of the leaks and some associated variables. Pipeline progress examinations included quantification of early persistence, plus application to, enrollment and persistence in, and graduation from, bachelor's programs. The examined associated variables included student demographic characteristics and possible pipeline leak malleable factors: academic preparation and performance and financial need. Overall, only 23% of the original cohort received a bachelor's degree after eight years. In addition, this study is the first to identify and quantify the transfer melt leakage point (students accepted for vertical transfer who do not enroll in bachelor's programs), and the first to have quantified transfer shock (a transfer-related decrease in GPA) in a large general cohort (shown by 16 percentage points of the cohort). Variables involving quicker degree progress (e.g., being exempt from remediation and enrolling full-time) were negatively associated with pipeline leaks. Together the results provide guidance for policymakers and practitioners regarding how to increase vertical transfer pipeline output and increase bachelor's degrees by community college students, particularly those from underrepresented groups, thus increasing higher education equity.

Unbiased data-driven omic approaches are revealing the molecular heterogeneity of Alzheimer disease. Here, we used machine learning approaches to integrate high-throughput transcriptomic, proteomic, metabolomic, and lipidomic profiles with clinical and neuropathological data from multiple human AD cohorts. We discovered 4 unique multimodal molecular profiles, one of them showing signs of poor cognitive function, a faster pace of disease progression, shorter survival with the disease, severe neurodegeneration and astrogliosis, and reduced levels of metabolomic profiles. We found this molecular profile to be present in multiple affected cortical regions associated with higher Braak tau scores and significant dysregulation of synapse-related genes, endocytosis, phagosome, and mTOR signaling pathways altered in AD early and late stages. AD cross-omics data integration with transcriptomic data from an SNCA mouse model revealed an overlapping signature. Furthermore, we leveraged single-nuclei RNA-seq data to identify distinct cell-types that most likely mediate molecular profiles. Lastly, we identified that the multimodal clusters uncovered cerebrospinal fluid biomarkers poised to monitor AD progression and possibly cognition. Our cross-omics analyses provide novel critical molecular insights into AD.

Cover crops have the potential to mitigate climate change by reducing negative impacts of agriculture on ecosystems. This study is first to quantify the net climate change mitigation impact of cover crops including land-use effects. A systematic literature and data review was conducted to identify major drivers for climate benefits and costs of cover crops in maize (Zea maize L.) production systems. The results indicate that cover crops lead to a net climate change mitigation impact (NCCMI) of 3.30 Mg CO2e ha-1 a-1. We created four scenarios with different impact weights of the drivers and all of them showing a positive NCCMI. Carbon land benefit, the carbon opportunity costs based on maize yield gains following cover crops, is the major contributor to the NCCMI (34.5% of all benefits). Carbon sequestration is the second largest contributor (33.8%). The climate costs of cover crops are mainly dominated by emissions from their seed production and foregone benefits due to land use for cover crops seeds. However, these two costs account for only 15.8% of the benefits. Extrapolating these results, planting cover crops before all maize acreage in the EU results in a climate change mitigation of 49.80 million Mg CO2e a-1, which is equivalent to 13.0% of the EU's agricultural emissions. This study highlights the importance of incorporating cover crops into sustainable cropping systems to minimize the agricultural impact to climate change.

Summary: There is a need for affordable, scalable, and specific blood-based biomarkers for Alzheimer’s disease that can be applied to a population level. We have developed and validated disease-specific cell-free transcriptomic blood-based biomarkers composed by a scalable number of transcripts that capture AD pathobiology even in the presymptomatic stages of the disease. Accuracies are in the range of the current CSF and plasma biomarkers, and specificities are high against other neurodegenerative diseases.

Parietal cortex RNA-sequencing (RNA-seq) data were generated from individuals with and without Alzheimer disease (AD; ncontrol = 13; nAD = 83) from the Knight Alzheimer Disease Research Center (Knight ADRC). Using this and an independent (Mount Sinai Brain Bank (MSBB)) AD RNA-seq dataset, cortical circular RNA (circRNA) expression was quantified in the context of AD. Significant associations were identified between circRNA expression and AD diagnosis, clinical dementia severity and neuropathological severity. It was demonstrated that most circRNA-AD associations are independent of changes in cognate linear messenger RNA expression or estimated brain cell-type proportions. Evidence was provided for circRNA expression changes occurring early in presymptomatic AD and in autosomal dominant AD. It was also observed that AD-associated circRNAs co-expressed with known AD genes. Finally, potential microRNA-binding sites were identified in AD-associated circRNAs for miRNAs predicted to target AD genes. Together, these results highlight the importance of analyzing non-linear RNAs and support future studies exploring the potential roles of circRNAs in AD pathogenesis.

Summary This paper presents the results of an advanced control strategy that employs active trailing edge flaps to reduce the fatigue loads of an experimental wind turbine. The strategy, called repetitive model predictive control, is a multiple‐input multiple‐output controller that aims at the alleviation of out‐of‐plane blade root bending moments. The strategy incorporates the control commands, output errors, and state deviation from the previous rotation. This way, the time lag in the strain sensor input due to the blade inertia is compensated. Additionally, a strategy to limit the computational costs is presented. The load alleviation performance is evaluated at different yaw cases and compared with different individual flap control strategies. The repetitive model predictive control is able to reduce the fatigue loads by up to 23% compared with the better performing individual flap control strategy. This improvement in load reduction is accompanied by an increase in flap travel of up to 7% compared with the individual flap control strategies.

IntroductionPlants release a large variety of metabolites via their roots to shape physico-chemical soil properties and biological processes in the rhizosphere. While hydroponic growth conditions facilitate accessibility of the root system and recovery of root exudates, the natural soil environment can alter root metabolism and exudate secretion, raising the question to what extent the quantity and composition of root exudates released in hydroponic growth systems reflect those recovered from soil-grown roots.MethodsUsing a root washing method, we sampled root exudates from four field-grown cover crop species with wide taxonomic distance, namely white mustard, lacy phacelia, bristle oat, and Egyptian clover. A set of primary metabolites and secondary metabolites were analysed in a targeted and untargeted LC-MS-based approach, respectively, for comparison with exudates obtained from hydroponically cultured plants.Results and discussionWe found that hydroponically cultivated plants released a larger amount of total carbon, but that the recovery of total carbon was not indicative for the diversity of metabolites in root exudates. In the field, root exudates from phacelia and clover contained 2.4 to 3.8 times more secondary metabolites, whereas carbon exudation in hydroponics was 5- to 4-fold higher. The composition of the set of metabolites identified using the untargeted approach was much more distinct among all species and growth conditions than that of quantified primary metabolites. Among secondary metabolite classes, the presence of lipids and lipid-like molecules was highly indicative for field samples, while the release of a large amount of phenylpropanoids, organoheterocyclic compounds or benzenoids was characteristic for clover, mustard or oat, respectively, irrespective of the cultivation condition. However, at the compound level the bulk of released metabolites was specific for cultivation conditions in every species, which implies that hydroponically sampled root exudates poorly reflect the metabolic complexity of root exudates recovered from field-grown plants.

Improving N cycling in agroecosystems is one of the key challenges in reducing the environmental footprint of agriculture. Further, uncertainty in precipitation makes crop water management relevant in regions where it has not been necessary thus far. Here, we focus on the potential of winter-killed catch crops (CCs) to reduce N leaching losses from N mineralization over the winter and from soil water management. We compared four single CCs (white mustard, phacelia, Egyptian clover and bristle oat) and two CC mixtures with 4 and 12 plant species (Mix4 and Mix12) with a fallow treatment. High-resolution soil mineral N (Nmin) monitoring in combination with the modelling of spatiotemporal dynamics served to assess N cycling under winter-killed CCs, while soil water was continuously monitored in the rooting zone. Catch crops depleted the residual Nmin pools by between 40 % and 72 % compared to the fallow. The amount of residual N uptake was lowest for clover and not significantly different among the other CCs. Catch crops that produce high N litter materials, such as clover and mustard leaves, showed an early N mineralization flush immediately after their termination and the highest leaching losses from litter mineralization over the winter. Except for clover, all CCs showed Nmin values between 18 % and 92 % higher on the sowing date of the following maize crop. However, only Mix12 was statistically significant. Catch crops depleted the soil water storage in the rooting zone during their growth in autumn and early winter, but preserved water later on when their residues covered the ground. The shallow incorporation of CC residues increased water storage capacity during the cropping season of the main crop even under reduced soil water availability. Hence, catch cropping is not just a simple plant cover for the winter but improves the growth conditions for the following crop with decreased N losses. Mixtures have been shown to compensate for the weaknesses of individual CC species in terms of nutrient capture, mineralization and transfer to the following main crop as well as for soil water management. Detailed knowledge about plant performance during growth and litter mineralization patterns is necessary to make optimal use of their potential.

Only 11% of community college (associate's-degree) students transfer vertically and obtain a bachelor's degree within six years, despite over 80% originally intending to do so. These leaks in the transfer pipeline disproportionately affect students from underrepresented groups, who are more likely to attend community colleges and to leak out of the pipeline. To obtain insights about how to decrease these leaks, a survey was distributed to all City University of New York undergraduates; 31,511 responded. The survey concerned students' life and academic circumstances, as well as their information about and views on transfer. Analyses particularly compared responses of never-transferred associate's and bachelor's students and vertical transfer students. The results suggest multiple actions that higher education can take to decrease the pipeline leaks and increase higher education equity, including by increasing transfer students' belongingness, course availability, credit transfer, financial support, information, and time efficiency. Introduction: Cover crop services depend on biomass production and species composition of the cover crop stand. In this study, we investigated the interactions in shoot biomass of dual cover crop mixtures and their competitiveness to suppress weeds before winter under different growing conditions. Methods: A field experiment was performed on sandy loam soil in Triesdorf, Germany, for two consecutive years. The cover crop species white mustard (Sinapis alba L.), oil radish (Raphanus sativus var. oleiformis L.), phacelia (Phacelia tanacetifolia J.), Egyptian clover (Trifolium alexandrinum L.), common vetch (Vicia sativa L.), field pea (Pisumsativum L.), and field bean (Vicia faba L.) were grown in pure stands or dual mixtures. Cover crops were sown in August, and shoot biomass was harvested at the end of the vegetation period. Cover crop shoot biomass was dried, and the share of each species and the sown test weed (Brassica napus L.) were determined. Results: The share of shoot biomass of a cover crop species in dual-species mixtures was closely related to its shoot biomass in its pure stand. In both years, mustard and phacelia showed similar interactions in shoot biomass production when growing with a second species. Regarding competitiveness against weeds, cruciferous species outperformed other cover crop species and could suppress weeds even if they were mixed with a less competitive partner. Weed suppression of mixtures with phacelia depended on the second component in the mixture and was more effective in a dry season. Legume species, especially Egyptian clover and common vetch could not suppress weeds in a pure stand. Discussion: Our results show that species with high weed suppression potential as single stands retain this ability in dual cover crop mixtures, regardless of the suppression potential of the second species that completes the mixture. Rotavirus is the most common pathogen causing pediatric diarrhea and an important cause of morbidity and mortality in low- and middle-income countries. Previous evidence suggests that the introduction of rotavirus vaccines in national immunization schedules resulted in dramatic declines in disease burden but may also be changing the rotavirus genetic landscape and driving the emergence of new genotypes. We report genotype data of more than 16,000 rotavirus isolates from 40 countries participating in the Global Rotavirus Surveillance Network. Data from a convenience sample of children under five years of age hospitalized with acute watery diarrhea who tested positive for rotavirus were included. Country results were weighted by their estimated rotavirus disease burden to estimate regional genotype distributions. Globally, the most frequent genotypes identified after weighting were G1P[8] (31%), G1P[6] (8%) and G3P[8] (8%). Genotypes varied across WHO Regions and between countries that had and had not introduced rotavirus vaccine. G1P[8] was less frequent among African (36 vs 20%) and European (33 vs 8%) countries that had introduced rotavirus vaccines as compared to countries that had not introduced. Our results describe differences in the distribution of the most common rotavirus genotypes in children with diarrhea in low- and middle-income countries. G1P[8] was less frequent in countries that had introduced the rotavirus vaccine while different strains are emerging or re-emerging in different regions.

In most terrestrial ecosystems, plant growth is limited by nitrogen and phosphorus. Adding either nutrient to soil usually affects primary production, but their effects can be positive or negative. Here we provide a general stoichiometric framework for interpreting these contrasting effects. First, we identify nitrogen and phosphorus limitations on plants and soil microorganisms using their respective nitrogen to phosphorus critical ratios. Second, we use these ratios to show how soil microorganisms mediate the response of primary production to limiting and non-limiting nutrient addition along a wide gradient of soil nutrient availability. Using a meta-analysis of 51 factorial nitrogen-phosphorus fertilization experiments conducted across multiple ecosystems, we demonstrate that the response of primary production to nitrogen and phosphorus additions is accurately predicted by our stoichiometric framework. The only pattern that could not be predicted by our original framework suggests that nitrogen has not only a structural function in growing organisms, but also a key role in promoting plant and microbial nutrient acquisition. We conclude that this stoichiometric framework offers the most parsimonious way to interpret contrasting and, until now, unresolved responses of primary production to nutrient addition in terrestrial ecosystems.

A substantial portion of soil organic matter (SOM) is of microbial origin. The efficiency with which soil microorganisms can convert their substrate carbon (C) into biomass, compared to how much is lost as respiration, thus co-determines the carbon storage potential of soils. Despite increasing insight into soil microbial C cycling, empirical measurements of microbial C processing across biomes and across soil horizons remain sparse. The theory of ecological stoichiometry predicts that microbial carbon use efficiency (CUE), i.e. growth over uptake of organic C, strongly depends on the relative availability of C and nutrients, particularly N, as microorganisms will either respire excess C or conserve C while mineralising excess nutrients. Microbial CUE is thus expected to increase from high to low latitudes and from topsoil to subsoil as the soil C:N and the stoichiometric imbalance between SOM and the microbial biomass decrease. To test these hypotheses, we collected soil samples from the organic topsoil, mineral topsoil, and mineral subsoil of seven sites along a 1500-km latitudinal transect in Western Siberia. As a proxy for CUE, we measured the microbial substrate use efficiency (SUE) of added substrates by incubating soil samples with a mixture of 13C labelled sugars, amino sugars, amino acids, and organic acids and tracing 13C into microbial biomass and released CO2. In addition to soil and microbial C:N stoichiometry, we also determined the potential extracellular enzyme activities of cellobiohydrolase (CBH) and phenol oxidase (POX) and used the CBH:POX ratio as an indicator of SOM substrate quality. We found an overall decrease of SUE with latitude, corresponding to a decrease in mean annual temperature, in mineral soil horizons. SUE decreased with decreasing stoichiometric imbalance in the organic and mineral topsoil, while a relationship of SUE with soil C:N was only found in the mineral topsoil. However, contrary to our hypothesis, SUE did not increase with soil depth and mineral subsoils displayed lower average SUE than mineral topsoils. Both within individual horizons and across all horizons SUE was strongly correlated with CBH:POX ratio as well as with climate variables. Since enzyme activities likely reflect the chemical properties of SOM, our results indicate that SOM quality exerts a stronger control on SUE than SOM stoichiometry, particularly in subsoils were SOM has been turned over repeatedly and there is little variation in SOM elemental ratios.

The occurrence of dark fixation of CO2 by heterotrophic microorganisms in soil is generally accepted, but its importance for microbial metabolism and soil organic carbon (C) sequestration is unknown, especially under C-limiting conditions. To fill this knowledge gap, we measured dark 13CO2 incorporation into soil organic matter and conducted a 13C-labelling experiment to follow the 13C incorporation into phospholipid fatty acids as microbial biomass markers across soil profiles of four tundra ecosystems in the northern circumpolar region, where net primary productivity and thus soil C inputs are low. We further determined the abundance of various carboxylase genes and identified their microbial origin with metagenomics. The microbial capacity for heterotrophic CO2 fixation was determined by measuring the abundance of carboxylase genes and the incorporation of 13C into soil C following the augmentation of bioavailable C sources. We demonstrate that dark CO2 fixation occurred ubiquitously in arctic tundra soils, with increasing importance in deeper soil horizons, presumably due to increasing C limitation with soil depth. Dark CO2 fixation accounted on average for 0.4, 1.0, 1.1, and 16% of net respiration in the organic, cryoturbated organic, mineral and permafrost horizons, respectively. Genes encoding anaplerotic enzymes of heterotrophic microorganisms comprised the majority of identified carboxylase genes. The genetic potential for dark CO2 fixation was spread over a broad taxonomic range. The results suggest important regulatory function of CO2 fixation in C limited conditions. The measurements were corroborated by modeling the long-term impact of dark CO2 fixation on soil organic matter. Our results suggest that increasing relative CO2 fixation rates in deeper soil horizons play an important role for soil internal C cycling and can, at least in part, explain the isotopic enrichment with soil depth.
