Background: In the recent decade, there has been substantial progress in the technologies and philosophies associated with diagnosing and treating anterior cruciate ligament (ACL) injuries in China. The therapeutic efficacy of ACL reconstruction in re-establishing the stability of the knee joint has garnered widespread acknowledgment. However, the path toward standardizing diagnostic and treatment protocols remains to be further developed and refined. Objective: In this context, the Chinese Association of Orthopaedic Surgeons (CAOS) and the Chinese Society of Sports Medicine (CSSM) collaboratively developed an expert consensus on diagnosing and treating ACL injury, aiming to enhance medical quality through refining professional standards. Methods: The consensus drafting team invited experts across the Greater China region, including the mainland, Hong Kong, Macau, and Taiwan, to formulate and review the consensus using a modified Delphi method as a standardization approach. As members of the CSSM Lower Limb Study Group and the CAOS Arthroscopy and Sports Medicine Study Group, invited experts concentrated on two pivotal issues: “Graft Selection” and “Clinical Outcome Evaluation” during the second part of the consensus development. Results: This focused discussion ultimately led to a strong consensus on nine specific consensus terms. Conclusion: The consensus clearly states that ACL reconstruction has no definitive “gold standard” graft choice. Autografts have advantages in healing capability but are limited in availability and have potential donor site morbidities; allografts reduce surgical trauma but incur additional costs, and there are concerns about slow healing, quality control issues, and a higher failure rate in young athletes; synthetic ligaments allow for early rehabilitation and fast return to sport, but the surgery is technically demanding and incurs additional costs. When choosing a graft, one should comprehensively consider the graft's characteristics, the doctor's technical ability, and the patient's needs. When evaluating clinical outcomes, it is essential to ensure an adequate sample size and follow-up rate, and the research should include patient subjective scoring, joint function and stability, complications, surgical failure, and the return to sport results. Medium and long-term follow-ups should not overlook the assessment of knee osteoarthritis.

Bone morphogenetic protein-2 (BMP-2) has been commercially approved by the Food and Drug Administration for use in bone defects and diseases. BMP-2 promotes osteogenic differentiation of mesenchymal stem cells. In bone tissue engineering, BMP-2 incorporated into scaffolds can be used for stimulating bone regeneration in organoid construction, drug testing platforms, and bone transplants. However, the high dosage and uncontrollable release rate of BMP-2 challenge its clinical application, mainly due to the short circulation half-life of BMP-2, microbial contamination in bone extracellular matrix hydrogel, and the delivery method. Moreover, in clinical translation, the requirement of high doses of BMP-2 for efficacy poses challenges in cost and safety. Based on these, novel strategies should ensure that BMP-2 is delivered precisely to the desired location within the body, regulating the timing of BMP-2 release to coincide with the bone healing process, as well as release BMP-2 in a controlled manner to optimize its therapeutic effect and minimize side effects. This review highlights improvements in bone tissue engineering applying spatiotemporal and controlled BMP-2 delivery, including molecular engineering, biomaterial modification, and synergistic therapy, aiming to provide references for future research and clinical trials.

With the development of artificial intelligence technology and Unmanned Aerial Vehicle (UAV) technology, the traditional UAV target detection methods are difficult to achieve high detection accuracy in complex and changeable scenes. The paper selects Faster Region Convolutional Neural Network (Faster R-CNN) as the basic algorithm due to its high scalability and excellent classification performance. The convolution mode of the convolution layer in the network structure is adjusted to deformable convolution. Additionally, a hybrid attention mechanism module is added to combine channel attention and spatial attention modules behind the output layer of the Faster R-CNN network. Finally, the Soft Non-Maximum Suppression (Soft-NMS) method is selected to combine the improved Faster R-CNN algorithm of multiple individuals into the final target detection model. The performance of the improved Faster R-CNN algorithm and the original Faster R-CNN algorithm was verified through the VisDrone 2018-DET dataset and the full class average accuracy Mean Average Precision (mAP), accuracy and precision. The accuracy and logarithm of loss values of the improved Faster R-CNN algorithm and the original Faster R-CNN algorithm's Region Proposal Network (RPN) were 0.985, 0.981, 0.018, and 0.052, respectively. The target detection model based on the hybrid attention mechanism of ResNet-50 network fusion Spartial Attention Module (SAM) and Channel Attention Module (CAM) had the best classification performance, with the accuracy, precision and overall average accuracy of 25.8 %, 24.6 % and 22.7 %, respectively. The results are helpful to improve the target detection ability of UAV in complex environment, and contribute to the development of target detection technology in the future.

Frequent burst events in water distribution systems cause severe water loss and other environmental issues such as contamination and carbon emissions. The availability of massive monitored data has facilitated the development of data-driven burst detection methods. This paper proposes the flow subsequences clustering–reconstruction analysis method for burst detection in district metering areas (DMAs). The sliding window is used to create flow subsequence libraries for all time points of a day using a historical data set and thereafter the clustering–reconstruction analysis is conducted to obtain flow pattern libraries and reconstruction error subsequences. The threshold vector is determined by the detection matrix extracted from the reconstruction error subsequences at each time point. At the detection stage, the new flow subsequence is created and its reconstruction version is obtained based on the flow pattern library at the same time point. The new detection vector is extracted and compared with the threshold vector to identify bursts. The proposed method is applied to two real-world DMAs and its detection performance is demonstrated and compared with two previous methods. The proposed method is proven to be effective in detecting burst events with fewer false alarms. HIGHLIGHTS The flow subsequences clustering–reconstruction analysis method is proposed for burst detection.; The impact of parameterization on the detection performance is discussed.; The selection of the length of the historical data set directly affects the detection performance.; The proposed method performs better than the cumulative sum and shape similarity-based methods on two real-world DMA cases.;

Abstract Vision transformers (ViTs) have revolutionized computer vision by employing self-attention instead of convolutional neural networks and demonstrated success due to their ability to capture global dependencies and remove spatial biases of locality. In medical imaging, where input data may differ in size and resolution, existing architectures require resampling or resizing during pre-processing, leading to potential spatial resolution loss and information degradation. This study proposes a co-ordinate-based embedding that encodes the geometry of medical images, capturing physical co-ordinate and resolution information without the need for resampling or resizing. The effectiveness of the proposed embedding is demonstrated through experiments with UNETR and SwinUNETR models for infarct segmentation on MRI dataset with AxTrace and AxADC contrasts. The dataset consists of 1142 training, 133 validation and 143 test subjects. Both models with the addition of co-ordinate based positional embedding achieved substantial improvements in mean Dice score by 6.5% and 7.6%. The proposed embedding showcased a statistically significant advantage p-value< 0.0001 over alternative approaches. In conclusion, the proposed co-ordinate-based pixel-wise positional embedding method offers a promising solution for Transformer-based models in medical image analysis. It effectively leverages physical co-ordinate information to enhance performance without compromising spatial resolution and provides a foundation for future advancements in positional embedding techniques for medical applications.

Objective Spinal endoscopy radiofrequency is a minimally invasive technique for lumbar disc herniation (LDH) and low back pain (LBP). However, recurring LDH/LBP following spinal endoscopy radiofrequency is a significant problem. Paravertebral musculature plays a crucial role in spine stability and motor function, and the purpose of the present study was to identify whether patients’ baseline lumbar muscular electrophysiological function could be a predictor of recurring LDH/LBP. Methods This was a prospective follow‐up and case‐control study focusing on elderly patients with LDH who were treated in our department between January 1, 2018, and October 31, 2021. The end of follow‐up was recurring LBP, recurring LDH, death, missing to follow‐up or 2 years postoperation. The surface electromyography test was performed before the endoscopy C‐arm radiofrequency (ECRF) operation to detect the flexion–relaxation ratio (FRR) of the lumbar multifidus (FRRLM) and the longissimus erector spinae (FRRES), and the other baseline parameters included the general characteristics, the visual analogue scale, the Japanese Orthopaedic Association score, and the Oswestry Disability Index. Intergroup comparisons were performed by independent t‐test and χ2‐test, and further binary logistic regression analysis was performed. Results Fifty‐four patients completed the 2‐year follow‐up and were retrospectively divided into a recurring LDH/LBP group (Group R) (n = 21) and a no recurring group (Group N) (n = 33) according to their clinical outcomes. Highly sensitive and specific biomarker detection is of outstanding importance for the diagnosis and treatment of cancers. Herein, we developed robust photoelectrochemical (PEC) biosensors with low background noise and high sensitivity based on a heterojunction, which can improve semiconductor photoelectric properties by limiting the recombination of photogenerated electron–hole pairs and successfully widening the range of light absorption. Alpha-fetoprotein (AFP) was used as a target model to examine the analytical performances of the designed PEC biosensors. ZnO/Cs3MnBr5 heterogeneous film with a uniform porous structure and large surface area enhanced electron transfer and biomolecule immobilization, and significantly increased the photocurrent response. Under the optimal conditions, the designed PEC biosensor exhibited a linear detection range of 0.01–500 ng/mL and a detection limit of 12 pg/mL. In addition, this PEC biosensor performed well when testing human serum samples and exhibited good repeatability, stability over time, and specificity, showing enormous potential for the detection of cancer markers in future biological and clinical research.

Abstract Background Osteoporosis, characterized by reduced bone mass and deterioration of bone quality, is a significant health concern for postmenopausal women. Considering that the specific role of circRNAs in osteoporosis and osteoclast differentiation remains poorly understood, this study aims to shed light on their involvement in these processes to enhance our understanding and potentially contribute to improved treatment strategies for osteoporosis. Methods An osteoporotic model was constructed in vivo in ovariectomized mouse. In vitro, we induced osteoclast formation in bone marrow-derived macrophages (BMDMs) using M-CSF + RANKL. To assess osteoporosis in mice, we conducted HE staining. We used MTT and TRAP staining to measure cell viability and osteoclast formation, respectively, and also evaluated their mRNA and protein expression levels. In addition, RNA pull-down, RIP and luciferase reporter assays were performed to investigate interactions, and ChIP assay was used to examine the impact of circZNF367 knockdown on the binding between FUS and CRY2. Results We observed increased expression of CircZNF367, FUS and CRY2 in osteoporotic mice and M-CSF + RANKL-induced BMDMs. Functionally, knocking down circZNF367 inhibited osteoporosis in vivo. Furthermore, interference with circZNF367 suppressed osteoclast proliferation and the expression of TRAP, NFATc1, and c-FOS. Mechanistically, circZNF367 interacted with FUS to maintain CRY2 mRNA stability. Additionally, knocking down CRY2 rescued M-CSF + RANKL-induced osteoclast differentiation in BMDMs promoted by circZNF367 and FUS. Conclusion This study reveals that the circZNF367/FUS axis may accelerate osteoclasts differentiation by upregulating CRY2 in osteoporosis and suggests that targeting circZNF367 may have potential therapeutic effects on osteoporosis.

60NiTi alloy is an important biomedical, structural and functional material. In this paper, the effects of heat treatment (temperature, time and cooling rate) on the microstructure and properties of 60NiTi alloy were studied, and the softening and hardening mechanisms were revealed. The results showed that the number of Ni4Ti3 phase enhanced with an increase of the cooling rate and the precipitation of Ni3Ti phase was inhibited after annealing at 950∼1075 °C followed by water-cooling. The orientation relationship between NiTi matrix and Ni4Ti3 phase obey the rule: [111]NiTi//[0001]Ni4Ti3 and (123¯)NiTi//(112¯0)Ni4Ti3. The faster the cooling rate was, the higher the hardness of the alloy was. The hardness of alloy under different cooling rate ranked from high to low was water-cooling, oil-cooling, liquid nitrogen-cooling, air-cooling and furnace-cooling. The hardness of the 60NiTi alloy was 316 HV after annealing at 1075 °C for 4 h followed by furnace-cooling, and the hardness sharply increased to 669 HV after annealing at 1075 °C for 4 h followed by water-cooling. The phase transformation of Ni4Ti3→Ni3Ti during cooling mainly contributed to the hardness decrease of the 60NiTi alloy.

Abstract We propose a general formalism beyond Weisskopf–Wigner approximation to efficiently calculate the coupling matrix element, evolution spectrum and population evolution of two quantum emitters in arbitrary metallic nanostructures. We demonstrate this formalism to investigate the radiative coupling and decay dynamics of two quantum emitters embedded in the two hot spots of three silver nano-spheroids. The vacuum Rabi oscillation in population evolution and the anti-crossing behavior in evolution spectrum show strong radiative coupling is realized in this metallic nanostructure despite its strong plasmon damping. Our formalism can serve as a flexible and efficient calculation tool to investigate the distant coherent interaction in a large variety of metallic nanostructures, and may be further developed to handle the cases for multiple quantum emitters and arbitrary dielectric–metallic hybrid nanostructures.

BackgroundNumerous studies have investigated the risk factors of intravesical recurrence (IVR) after radical nephroureterectomy (RNU) in patients with upper urinary tract urothelial carcinoma (UTUC). However, few studies explore the predictors for unfavorable pathological types of IVR following RNU.MethodsWe retrospectively reviewed 155 patients diagnosed with bladder cancer (BC) following RNU. Binary logistic regression was used for the univariable and multivariable analyses. Nomograms were developed based on the multivariable analysis. The concordance index (C-index) was used to assess the performance of the nomograms. We performed internal validation by generating calibration plots.ResultsMuscle-invasive BC (MIBC) was significantly correlated with operation interval (p = 0.004) and UTUC T-stage (p = 0.016). Operation interval (p = 0.002) and UTUC T-stage (p = 0.028) were also risk factors for BC > 3 cm. UTUC grade (p = 0.015), operation interval (p = 0.003), and hydronephrosis (p = 0.049) were independent predictors for high-grade BC (HGBC). MIBC (p = 0.018) and surgical approach (p = 0.003) were associated with multifocal IVR. Besides, MIBC and HGBC were associated with UTUC grade (p = 0.009), operation interval (p = 0.001), and hydronephrosis (p = 0.023). Moreover, only operation interval (p = 0.036) was a predictor for BC with at least one unfavorable pathological type. We developed nomograms for MIBC, HGBC, BC > 3 cm, and MIBC and/or HGBC. The calibration curves of the nomograms showed good agreement between the observation and prediction cases. The C-indexes of the nomograms were 0.820 (95% CI, 0.747–0.894), 0.728 (95% CI, 0.649–0.809), 0.770 (95% CI, 0.679–0.861), and 0.749 (95% CI, 0.671–0.827), respectively.ConclusionsThe current study found that operation interval, UTUC T-stage, UTUC grade, surgical approach, and hydronephrosis are independent predictors for unfavorable pathological types of IVR following RNU. Nomograms based on these predictors were developed and internally validated to assess the risk of developing unfavorable pathological types of IVR. Furthermore, patients at high risk of developing unfavorable pathological types BC may benefit from more active follow-up 1 year after RNU by early detection of IVR.

ObjectivesTo summarize the clinicopathological diagnostic features and evolutionary trends of upper tract urothelial carcinoma (UTUC) in China over the past 20 years.MethodsAll patients diagnosed with upper tract urothelial carcinoma in the Peking University First Hospital from 2001 to 2020 were retrospectively collected. Data were divided into two groups (2001-2010 and 2011-2020) according to the date of diagnosis. Statistical analysis was done with the SPSS V22.0. Chi-square analysis and t-test were adopted to analyze depending on the data type. Subgroup analysis based on 5 years was used for visualization to present trends. Both Kaplan-Meier curve and Cox regression were used for univariate and multivariate survival analysis.ResultsThe study included 2561 cases diagnosed with upper tract urothelial carcinoma in total. Abstract Upper tract urothelial carcinomas (UTUCs) are rare entities that are usually diagnosed at advanced stages. Research on UTUC pathobiology and clinical management has been hampered by the lack of models accurately reflecting disease nature and diversity. In this study, a modified organoid culture system is used to generate a library of 25 patient‐derived UTUC organoid lines retaining the histological architectures, marker gene expressions, genomic landscapes, and gene expression profiles of their parental tumors. The study demonstrates that the responses of UTUC organoids to anticancer drugs can be identified and the model supports the exploration of novel treatment strategies. This work proposes a modified protocol for generating patient‐derived UTUC organoid lines that may help elucidate UTUC pathophysiology and assess the responses of these diseases to various drug therapies in personalized medicine.

Abstract Background This study was conducted with the aim of exploring the effect of the BMP signaling pathway on osteoblastic differentiation in rat bone marrow mesenchymal stem cells (rBMSCs) in rats with osteoporosis (OP). Methods The bilateral ovaries of female SD rats were resected for the establishment of a rat OP model. The osteoblastic differentiation of isolated rBMSCs was identified through osteogenic induction. Adipogenetic induction and flow cytometry (FCM) were used to detect adipogenic differentiation and the expression of rBMSC surface markers. The rBMSCs were grouped into the blank group, NC group, si-BMP2 group, and oe-BMP2 group. The expression levels of key factors and osteogenesis-related factors were determined by Western blot and quantitative real-time polymerase chain reaction (qRT-PCR). The formation of calcified nodules was observed by alizarin red staining. ALP activity was measured by alkaline phosphatase staining. This work describes the fabrication of AZ91D/A5052 clad strips by direct cladding from molten metals using a horizontal twin roll caster. Subsequently, the effects of roll speed, pouring sequence, and solidification length on the AZ91D/A5052 clad strips were investigated. The AZ91D/A5052 clad strips with a thickness of 4.9 mm were successfully cast at a roll speed of 9 m/min and with a 5 mm roll gap. The cladding ratio of AZ91D/A5052 was about 1:1. The single-roll casting results showed that the experimental solidification constants of AZ91D and A5052 were 62 mm/min0.5 and 34 mm/min0.5, respectively. The twin-roll casting results showed that the effect of rolling speed on the surface condition of A5052 was greater than that of AZ91D. In addition, the high melting point A5052 alloy poured into the lower nozzle could solve the remelting problem of the low melting point AZ91D. Moreover, extending the upper solidification distance could reduce the generation of intermetallic compounds. The EDS analysis results showed no voids at the bonding interface, while three intermetallic compound layers were also found at the bonding interface of AZ91D/A5052 strips, namely α-Mg + Mg17Al12, Mg17Al12, and Al3Mg2. This study could be instructive for dissimilar sheet metal bonding.

A significant gain in optical Kerr susceptibility of semiconductor quantum systems is of great importance for enabling nonlinear optical devices for the applications such as optical switches and high-density on-chip integration. Here, we investigate the optical Kerr susceptibility of the semiconductor quantum dot in the hybrid structure consisting of a semiconductor quantum dot and a metal nanoparticle. Taking into account the exciton-plasmon coupling, the modified expression for the optical Kerr susceptibility of the semiconductor quantum dot in the hybrid structure is given using iterative method and density matrix theory. One finds that the optical Kerr susceptibility of the semiconductor quantum dot in the hybrid structure is one to two orders of magnitude larger than that of an isolated semiconductor quantum dot. The magnitude of such enhancement is strongly dependent on the surrounding dielectric environment. Furthermore, the magnitude of the optical Kerr susceptibility of the semiconductor quantum dot in the hybrid structure exhibits a decrease followed by an increase with the increase of the radius of the semiconductor quantum dot, while that produced by the isolated semiconductor quantum dot demonstrates an increase. Such non-monotonic dependence makes the hybrid structure a promising candidate for developing nonlinear optical nano-devices with good performances.

Osteosarcoma is the most common primary malignant tumor, especially in children and adolescents. Circular RNAs (circRNAs) are found to play roles in the progression of osteosarcoma. However, the exact functions of circRNAs in osteosarcoma development still need to be clarified. We obtained differentially expressed circRNAs and miRNAs from a GSE99671 data set (GEO database). The gene co-expression network of ceRNAs and osteosarcoma-related genes was analyzed using the STRING database. qRT-PCR was used to detect the expression of circ-03955 and miR-3662. Transwell assays and flow cytometry were performed to detect phenotypic changes in cell function. A xenograft tumor model was established using BALB/c nude mice. Dual luciferase activity and RNA immunoprecipitation assays were performed to assess the relationship between circ-03955, miR-3662, and metadherin (MTDH). Immunohistochemistry, immunofluorescence, and Western blotting were used to assess protein expression levels. Circ-03955 was significantly upregulated, and miR-3662 was downregulated in osteosarcoma. Circ-03955 silencing inhibited the growth and metastasis of osteosarcoma. Mechanism analysis revealed that circ-03955 could bind to miR-3662, and the latter could target MTDH, leading to its suppressed expression and facilitating epithelial-mesenchymal transition (EMT). All these findings demonstrate that the presence of circ-03955 promotes EMT in osteosarcoma by acting as miR-3662 sponge-mediated MTDH expression.

Asymmetric coupled quantum wires provide the possibility for realizing obvious nonlinear optical rectification due to breaking the inversion symmetry. Here, we investigated nonlinear optical rectification (NOR) and electronic states in asymmetric coupled quantum wires (ACQWs). We find that the NOR max value of the ACQWs exhibits double peak structure with the increase of the geometrical asymmetry (i.e. the left wire radius deviating from the right wire radius), thus offering a direction for achieving remarkable NORS. In the double peak structure, the max value is one to two orders of magnitude larger than the min value. Furthermore, we find that the tunneling effect with respect to the ground state is reduced with the increase of the geometrical asymmetry, while the tunneling effect with respect to the first-excited state is firstly reduced, subsequently enhanced and then reduced with the increase of the geometrical asymmetry. Such coupling pictures elucidate the double peak structure in the NOR max value of the ACQWs. Our work enriches our understanding for the NOR and quantum states of the ACQWs, thereby laying the groundwork for the design and optimization of nonlinear nano-optical devices based on asymmetric coupled quantum wires such as THz radiation sources and infrared photo-detectors.

Recent advances in deep learning have improved the segmentation accuracy of subcortical brain structures, which would be useful in neuroimaging studies of many neurological disorders. However, most existing deep learning based approaches in neuroimaging do not investigate the specific difficulties that exist in segmenting extremely small but important brain regions such as the subnuclei of the amygdala. To tackle this challenging task, we developed a dual-branch dilated residual 3D fully convolutional network with parallel convolutions to extract more global context and alleviate the class imbalance issue by maintaining a small receptive field that is just the size of the regions of interest (ROIs). We also conduct multi-scale feature fusion in both parallel and series to compensate the potential information loss during convolutions, which has been shown to be important for small objects. The serial feature fusion enabled by residual connections is further enhanced by a proposed top-down attention-guided refinement unit, where the high-resolution low-level spatial details are selectively integrated to complement the high-level but coarse semantic information, enriching the final feature representations. As a result, the segmentations resulting from our method are more accurate both volumetrically and morphologically, compared with other deep learning based approaches. To the best of our knowledge, this work is the first deep learning-based approach that targets the subregions of the amygdala. We also demonstrated the feasibility of using a cycle-consistent generative adversarial network (CycleGAN) to harmonize multi-site MRI data, and show that our method generalizes well to challenging traumatic brain injury (TBI) datasets collected from multiple centers. This appears to be a promising strategy for image segmentation for multiple site studies and increased morphological variability from significant brain pathology.

The association of epilepsy with structural brain changes and cognitive abnormalities in midlife has raised concern regarding the possibility of future accelerated brain and cognitive aging and increased risk of later life neurocognitive disorders. To address this issue we examined age-related processes in both structural and functional neuroimaging among individuals with temporal lobe epilepsy (TLE, N = 104) who were participants in the Epilepsy Connectome Project (ECP). Support vector regression (SVR) models were trained from 151 healthy controls and used to predict TLE patients’ brain ages. It was found that TLE patients on average have both older structural (+6.6 years) and functional (+8.3 years) brain ages compared to healthy controls. Accelerated functional brain age (functional – chronological age) was mildly correlated (corrected P = 0.07) with complex partial seizure frequency and the number of anti-epileptic drug intake. Functional brain age was a significant correlate of declining cognition (fluid abilities) and partially mediated chronological age-fluid cognition relationships. Chronological age was the only positive predictor of crystallized cognition. Accelerated aging is evident not only in the structural brains of patients with TLE, but also in their functional brains. Understanding the causes of accelerated brain aging in TLE will be clinically important in order to potentially prevent or mitigate their cognitive deficits. Abstract Background To develop and evaluate the feasibility of a data-driven deep learning approach (deepAC) for positron-emission tomography (PET) image attenuation correction without anatomical imaging. A PET attenuation correction pipeline was developed utilizing deep learning to generate continuously valued pseudo-computed tomography (CT) images from uncorrected 18F-fluorodeoxyglucose (18F-FDG) PET images. A deep convolutional encoder-decoder network was trained to identify tissue contrast in volumetric uncorrected PET images co-registered to CT data. A set of 100 retrospective 3D FDG PET head images was used to train the model. The model was evaluated in another 28 patients by comparing the generated pseudo-CT to the acquired CT using Dice coefficient and mean absolute error (MAE) and finally by comparing reconstructed PET images using the pseudo-CT and acquired CT for attenuation correction. Paired-sample t tests were used for statistical analysis to compare PET reconstruction error using deepAC with CT-based attenuation correction. Results deepAC produced pseudo-CTs with Dice coefficients of 0.80 ± 0.02 for air, 0.94 ± 0.01 for soft tissue, and 0.75 ± 0.03 for bone and MAE of 111 ± 16 HU relative to the PET/CT dataset. deepAC provides quantitatively accurate 18F-FDG PET results with average errors of less than 1% in most brain regions. Conclusions We have developed an automated approach (deepAC) that allows generation of a continuously valued pseudo-CT from a single 18F-FDG non-attenuation-corrected (NAC) PET image and evaluated it in PET/CT brain imaging.

Abstract Background 5-Hydroxymethylcytosine (5hmC) is converted from 5-methylcytosine (5mC) by a group of enzymes termed ten-eleven translocation (TET) family dioxygenases. The loss of 5hmC has been identified as a hallmark of most types of cancer and is related to tumorigenesis and progression. However, the role of 5hmC in bladder cancer is seldom investigated. Vitamin C was recently reported to induce the generation of 5hmC by acting as a cofactor for TET dioxygenases. In this study, we explored the role of 5hmC in bladder cancer and the therapeutic efficacy of vitamin C in increasing the 5hmC pattern. Results 5hmC was decreased in bladder cancer samples and was related to patient overall survival. Genome-wide mapping of 5hmC in tumor tissues and vitamin C-treated bladder cancer cells revealed that 5hmC loss was enriched in cancer-related genes and that vitamin C treatment increased 5hmC levels correspondingly. Vitamin C treatment shifted the transcriptome and inhibited the malignant phenotypes associated with bladder cancer cells in both in vitro cell lines and in vivo xenografts. Conclusions This study provided mechanistic insights regarding the 5hmC loss in bladder cancer and a rationale for exploring the therapeutic use of vitamin C as a potential epigenetic treatment for bladder cancer.

Abstract Background Lymph node dissection (LND) is not routinely performed during radical nephroureterectomy (RNU) in upper tract urothelial carcinomas (UTUC) and the role of LND has been controversial. We aim to investigate whether patients with LND had improved survival in UTUC patients. Methods We performed a systematic literature search of PubMed, Embase, and Cochrane library for citations published prior to January 2016, describing LND performed among UTUC patients and conducted a standard meta-analysis of survival outcomes. Results Eleven eligible studies containing 7516 patients satisfied the inclusion criteria. Pooled HRs for cancer-specific survival (CSS) and recurrence-free survival (RFS) were 1.17 (P = 0.18) and 1.33 (P = 0.19) respectively. Abstract Background There's no consensus about the difference between renal pelvic and ureteral tumors in terms of clinical features, pathological outcomes, epigenetic biomarkers and prognosis. Methods The data of 341 patients with renal pelvic tumors and 271 patients with ureteral tumors who underwent radical nephroureterectomy between 1999 and 2011 were retrospectively reviewed. The clinicopathologic features, gene promoters methylation status and oncologic outcomes were compared. Regression analysis was performed to identify oncologic prognosticators. Results Patients with ureteral tumors were relatively older (p = 0.002), and had higher likelihood of pre-operative renal insufficiency (p 0.05), while gene methylation status was demonstrated to be an independent prognostic factor. Conclusion Renal pelvic tumors and ureteral tumors exhibited significant differences in clinicopathologic characteristics and epigenetic biomarkers. Gene promoter methylation might be an important mechanism in explaining distinct tumor patterns and behaviors in UTUC.

Abstract A chitosan-based microsphere delivery system has been fabricated for controlled release of alendronate (AL). The present study aimed to incorporate the chitosan/hydroxyapatite microspheres-loaded with AL (CH/nHA-AL) into poly(L-lactic acid)/nanohydroxyapatite (PLLA/nHA) matrix to prepare a novel microspheres-scaffold hybrid system (CM-ALs) for drug delivery and bone tissue engineering application. The characteristics of CM-ALs scaffolds containing 10% and 20% CH/nHA-AL were evaluated in vitro, including surface morphology and porosity, mechanical properties, drug release, degradation, and osteogenic differentiation. The in vivo bone repair for large segmental radius defects (1.5 cm) in a rabbit model was evaluated by radiography and histology. In vitro study showed more sustained drug release of CM-AL-containing scaffolds than these of CM/nHA-AL and PLLA/nHA/AL scaffolds, and the mechanical and degradation properties of CM-ALs (10%) scaffolds were comparable to that of PLLA/nHA control. The osteogenic differentiation of adipose-derived stem cells (ASCs) was significantly enhanced as indicated by increased alkaline phosphates (ALP) activity and calcium deposition. In vivo study further showed better performance of CM-ALs (10%) scaffolds with complete repair of large-sized bone defects within 8 weeks. A microspheres-scaffold-based release system containing AL-encapsulated chitosan microspheres was successfully fabricated in this study. Our results suggested the promising application of CM-ALs (10%) scaffolds for drug delivery and bone tissue engineering.

Quantum Information: quantum interference suppresses dephasing of qubits Scientists have realized a novel quantum device to improve the robustness of quantum information encoded in a qubit against noise. It is a well known difficulty in quantum information processing that the noise in the environment disturbs the quantum state of a qubit and degrades the qubit's performance. Andrew Houck and colleagues at Princeton University have designed and implemented a superconducting qubit that utilizes the quantum interference effect to decouple itself from microwaves in a nearby cavity. The researchers demonstrate the robust preparation, manipulation and measurement of the qubit in the presence of noise. This approach provides a promising element for the storage and processing of quantum information, mitigating effects of cross-talk when trying to control or measure multiple qubits.

Docetaxel (Doc) is the gold standard of care for castration-resistant prostate cancer (CRPC) patients, although the therapeutic effects are modest. Fuzheng Yiliu decoction (FZYL) comprises multiple herbs, and has been used for >10 years to treat various cancers, including hepatocellular tumors, malignant gastrointestinal tumors, and prostate cancer. In the study reported, we evaluated the anticancer effects of FZYL and of a combination of Doc and FZYL in CRPC tumor-bearing mice, and explored the underlying mechanisms. PC-3 tumor-bearing mice were treated with FZYL, Doc, Doc + FZYL or vehicle solution. Tumor volume was monitored, and tumor weight, proliferation and apoptosis of tumor tissues were measured. Deep sequencing was used to profile the miRNA expression patterns in tumor tissues. Our results suggested that FZYL alone could depress tumor growth, and the combination of Doc and FZYL treatment exhibited enhanced anticancer effects. Doc + FZYL regulated the expression of 10 miRNAs: miR-34b-5p, miR-674-3p, miR-140-3p, miR-342-3p, miR-214-3p, miR-149-5p, miR-378c, miR-29b-3p, miR-218-5p, and miR-378a-3p, involving in the PI3K-Akt pathway in the treatment of CRPC.

There is relatively little literature on prevalence of chronic kidney disease (CKD) prior to surgery in patients with upper urinary tract urothelial carcinoma (UTUC). We evaluated the prevalence and clinical associated factors of baseline CKD in patients with UTUC. Methods: There were 785 patients with a pathologic diagnosis of UTUC from January 2002 to December 2011 who were analyzed in this study. Estimated glomerular filtration rate (eGFR) was calculated by re-expressed Modification of Diet in Renal Disease (MDRD) formulas for the Chinese population. A multivariate logistic regression was performed to evaluate the odds ratios (ORs) for CKD stage 3 or higher in UTUCs after data differences were tested. Results: The prevalence of CKD in UTUCs presenting at our hospital was 58.6% and 70.8% in the group age 70 years and older. Older age [per year increased; OR = 1.050; 95% confidence interval (CI): 1.034–1.067], lower tumor stage (T stage; per stage increased; OR = 0.666; 95% CI: 0.544–0.816), higher tumor grade (per grade increased; OR = 1.392; 95% CI: 1.004–1.930) and the main tumor locating in the pelvis (ureter as reference; OR = 0.648; 95% CI: 0.475–0.885) were independently associated with decreased kidney function in the multivariate logistic regression. The use of serum creatinine (Scr) only to evaluate the renal function would ignore a large proportion of patients suffering from CKD stage 3 in UTUCs, especially in those older than 70 years (39.3% vs. 54.1%, p = 0.022). Conclusion: We demonstrated a high prevalence (58.6%) of CKD in patients with UTUC, particularly in the group older than 70 years (70.8%). Older age, lower T stage, higher tumor grade, and the main tumor locating in pelvis (ureter as reference) were independently associated with CKD in UTUCs.
