Abstract

Physalis peruviana is one of the most promising tropical fruit plants because of its rapid growth, high yield, and nutritional quality. This study was designed to investigate plant development under heavy metal contamination (Cd, Pb) and responsiveness to arbuscular mycorrhizal fungi (AMF) colonization by Rhizophagus clarum and Claroideoglomus claroideum. The antioxidant capacity, total lipid content and fatty acid profile in fruits, accumulation of Cd and Pb in different plant parts, plant dry biomass, and mycorrhizal colonization were determined. As a result of inoculation, a considerable reduction in Cd and Pb in the fruits was observed, compared with non-inoculated plants. The fruit number and dry weight increased in plants associated with C. claroideum. These plants also showed higher acid phosphatase activity, root protein accumulation and glomalin production. The type of antioxidant defense was AMF strain-dependent. Antioxidant activity and H2O2 neutralization were enzymatic rather than non-enzymatic processes in the fruits of C. claroideum plants compared with those forming an association with R. clarum. Mycorrhizal establishment changed the composition and concentration of fruits’ fatty acids. The ratio of unsaturated fatty acids was increased. With respect to the accumulation of bioactive compounds in golden berry the present findings are important for obtaining the optimum benefits of mycorrhizal association under unfavorable conditions.

23. Improving Yield and Quality-Related Physiological Characteristics of Lettuce by Integrated Inorganic and Organic Fertilizers Management

Abstract

The research aims to obtain more information on the nitrogen metabolism and nutraceutical properties of two types Lactuca sativa L. var. crispa L. type Batavia cv. 'Maritima' and type Lollo Rosso cv. 'Tuska' and one type Lactuca sativa L. var. capitata L. cv. 'Winter butterhead' under the implementation of different organic fertilizers. In this respect, the main goal of our study is to define a proper macro- and micronutrients source which has the most beneficial effect on yield as well as on various quality parameters of lettuce. The experiment was conducted in an unheated greenhouse in two consecutive seasons (2018/2019 and 2019/2020) under all the requirements for organic production. Four organic fertilizers (Italpollina”, “Arcobaleno”, “Lumbrical” and “EKOprop” were studied. From obtained results, it could be concluded that the impact of fertilizers on lettuce growth and quality has possessed cultivar dependence. The fertilizing with natural fertilizers, so that the nutrients they possess are strictly comprised of animal-based materials (dry bird manure “Italpolina”, mix from bovine, horse and chicken manure “Arkobaleno”) or compost (lumbricompost made by Red Californian worms “Lumbrical”) of the three lettuce cultivars resulted in lower plant FW and yield, compared with NPK fertilizing. Only fertilizing with a mix of different beneficial microorganisms leads to the same plant FW and yield as fertilizing with inorganic fertilizer. Based on the higher plant FW and yield of all three lettuce cultivars the treatment with the fertilizer composed with beneficial microorganisms can be substituted with the treatment with inorganic NPC fertilizer. “EKOprop” fertilizing showed the lowest leaf nitrate concentration which correlated with higher activity of nitrogen assimilating enzyme nitrate reductase.

24. Enzyme and Non-Enzyme Antioxidant Activity of Micro Propagated Cape Gooseberry (Physalis peruviana L.)

Abstract

Several studies have represented data on in vitro clonal multiplication of Physalis peruviana L. using different nutrient media supplemented with diverse growth regulators, but there is still scarce information on the changes in the antioxidant potential of the plants subjected to micropropagation. The present research investigates the effect of various plant growth regula-tors on the antioxidant activity of micropropagated P. peruviana L. The seedling explants of P. peruviana (L.) were cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of the studied citokines, 6-benzylaminopurine (BAP), zeatin and thidiazuron (TDZ).The micro-shoots produced normal roots within two weeks of cultivation on half-strength MS medium supplemented with the auxins indole-3-butyric acid (IBA) or indole-3-acetic acid (IAA) at concentrations 0.1 mg L-1 and 0.5 mg L-1, respectively. The most efficient plant rooting was achieved with plants grown on half-strength MS medium with 0.1 mg L-1 IBA (number of shoots per explant 12.1 with a shoot height 5.2 cm) as well as 0.1 mg L-1 IAA (number of shoots per explant 10.4 with a shoot height 7.3 cm) after four weeks of cultivation. The reported micropropagation protocol here is characterized by a rapid proliferation and facilitated rooting of the microshoots. The multiple plants were successfully adapted showing 100% survival rate after two months of ex vitro growth. The changes of en-zyme and non-enzyme antioxidant activity were found to depend on the concentration of the used cytokinins. At the lower concentration of 0.5 mg L-1 of studied cytikinins the antioxidant capacity was characterized by a higher concentration of metabolites with antioxidant poten-tial. The higher cytokinin concentration of 0.1 mg L-1 used in the micropropagation procedure resulted in higher antioxidant enzyme activity. Abstract

Stevia rebaudiana Bertoni is a valuable plant whose products are increasingly used in medicine, pharmacy and the food industry. This necessitates the use of biotechnological approaches for its mass propagation. Establishing optimal conditions for in vitro cultivation is essential for obtaining high biomass and secondary metabolites production. A large number of articles considering the role of plant growth regulators and other additives in the culture medium in the growth and development of Stevia are available in the literature. However, there are no summarized data about the use of nanoparticles in Stevia tissue cultures. Therefore, this review also includes the research conducted so far on the effect of nanoparticles on Stevia micropropagation. Furthermore, the influence of different elicitors on secondary metabolite production and antioxidant activity of in vitro-cultivated Stevia plants have been discussed. By referring to the collected literature, we concluded that biotechnological approaches applied to S. rebaudiana cultivation might improve the agronomic traits of plants and steviol glycosides production. 35. ASSESSMENT OF THE ANTIOXIDANT POWER OF IN VITRO OBTAINED COLEUS FORSKOHLII BRIQ

Abstract

In thе prеsent studу, a simрle protосоl for in vitro rooting induction of the important medicinal plant Coleus forskohlii Briq with a high content of secondary metabolites with antioxidant activity was described. For the in vitro germination seeds of C. forskohlii were placed on MS medium suррlemented with 0.4 mg L-1 gibberellic acid and 1.0 mg L-1 CaCl2. The оbtаinеd shoots were cultured on a half-strength ½ MS medium containing indole-3-butyric acid (IBA) or indole-3-acetic acid (IAA) at concentrations of 0.1 and 0.5 mg L-1. The maximum rooting of plants (95%) with mean values of 9.2 roots per explant, root length (5.1 cm) and root fresh weight (0.085 g) was obtained on half-strength MS medium with 0.1 mg L-1 IAA after three weeks of culture. The plants were successfully ex vitro adapted with 93.3% survival on the mixture substrate of soil, peat and sand (2:1:1 v/v/v). The antioxidant enzymes асtivitiеs (superoxide dismutase, catalase, ascorbate peroxidase and guaiacol peroxidase) showed the highest values when IBA was used for C. forskohlii in vitro cultivation. Applied auxins influenced the level of secondary metabolites synthesis. Half strength MS culture medium supplemented with 0.5 mg L-1 IAA caused the highest total phenol and flavonoid content as well as the highest antioxidant activity level (radical scavenging activity and ferric reducing antioxidant power). The results indiсаte thаt orgаnоgenеsis is а very соmplicated biologiсаl prосеss involving up and downregulation of plant enzyme and non-enzyme antioxidant protection systems, which plау an imроrtant role during the organogenesis of plants.

Abstract

The study was performed to evaluate the nutritional value and antioxidant properties of cowpea grain, where plants were grown under conditions of tripartite symbiosis (cowpea plants, nitrogen-fixing bacteria (Bradyrhizobium japonicum) and arbuscular mycorrhizal fungi, (Glomus intraradices) without fertilization. Cowpea plants (Vigna unguiculata (L.) Walp) cv. Hrisi were grown in a glasshouse until the full maturity of grain on the Alluvial meadow soil (Eutric Fluvisol) at three levels of water-holding capacity (40, 60 and 80%). Bradyrhizobium japonicum solely and in combination with arbuscular mycorrhizal fungi increased the content of proteins, soluble sugars and total phenols in the cowpea seeds. When dual inoculation has applied the levels of proteins and soluble sugars in the cowpea grain did not reduce at 40%, nor at 80% in comparison with the optimal water-holding capacity level (60%). Antioxidant capacity at 40% water-holding capacity was determined by the increased content of water soluble antioxidants, flavonoid content and increased ferric reducing power activity and increased antioxidant levels was expressed more distinguished in the treatments with dual inoculation. Slight change of antioxidants in the grain of cowpea was established at an elevated level of water supply. Nutritional value of a grain of cowpea after dual inoculation with Bradyrhizobium strains and Glomus intraradices was not determined by the water-holding capacity levels.

Abstract

Helianthus annuus is one of the most important oil species, with a comparatively narrow genetic base. The development of new sunflower cultivars is the focus of many research and breeding programs. Intergeneric hybridization between H. annuus and Echinacea purpurea, a valuable medicinal plant, has not yet been utilized in cultivar development. This paper describes 2 advanced hybrid lines produced from an intergeneric cross between the cultivated sunflower inbred line 6650 and an accession of E. purpurea. The hybrid plants have successfully been grown in the field. The hybrids, showing expression of traits from both parental species, were intermediate between both parents for plant height, leaf shape, leaf color, floral characteristics, tocopherol content, and fatty acid profile. It could be postulated that the hybridization Helianthus × Echinacea offers opportunities for combining desirable traits that increase the variability of cultivated sunflower.
