Tofu prepared by conventional methods often has a bitter taste and poor water-holding capacity (WHC). To improve the quality of the product, alternative processes must be developed. Herein, the effect of ultrasound pretreatment on the properties of soymilk and tofu gel derived thereof were investigated. Treatment of soymilk with ultrasound gave rise to a reduction in the particle size and an enhancement in the surface hydrophobicity, whereby optimum values were obtained after 15 min treatment. Subsequently, microbial transglutaminase (MTG) was added to ultrasound-treated soymilk to promote the soy protein crosslinking. The gel strength, WHC, and nonfreezable water content of MTG-catalyzed tofu gel obtained from treated soymilk increased with the extension of the ultrasound pretreatment time, whereas the free sulfhydryl content decreased because of the formation of disulfide bonds. Fourier transform infrared spectroscopy demonstrated variations in the secondary structure of MTG-catalyzed tofu gel. Furthermore, soymilk's exposure to high-intensity ultrasound pretreatment led to a tofu gel with a dense, homogenous, and stable network structure, as evidenced by scanning electron microscopy. Therefore, this study answers for the theoretical support of the industrial production of MTG-catalyzed tofu gel from ultrasound-treated soymilk. In this study, effects of different concentrations of riboflavin (0, 0.02, and 0.1 μmol/g protein) on myofibrillar protein (MP, Scomberomorus niphonius) gel were characterized. The gel structure and properties were studied with or without Ultraviolet A (UVA) irradiation. Electron spin resonance results showed that riboflavin produced ·OH under UVA irradiation, which subsequently oxidized the MP. Compared with the control group, the addition of riboflavin with UVA irradiation increased the strength of the MP gel. The rheological results showed that under UVA irradiation, addition of riboflavin facilitated the sol-gel transition between 45 and 52°C, indicating that oxidation led to significant structural changes which in turn resulted in a more compact and uniform gel network. The presence of riboflavin led to increased carbonyl content and decreased sulfhydryl and free amino groups, which decreased the protein solubility and promoted alpha-helical conformational loss in the secondary structure of the MP. These results all indicated that the MP has been oxidized. Electrophoresis revealed that myosin heavy chains were aggregated in the UVA-treated riboflavin-added MP gel, indicating that protein cross-linking has been induced. The effect of phlorotannin extracts (PTE) (from sporophyll of Undaria pinnatifida) added at different levels (0, 25, 125, 625 µmol/g protein) on the gel properties of mackerel (Scomberomorus niphonius) myofibrillar protein (MP) was studied with and without ultraviolet A (UVA) irradiation. The results showed that the gel strength and cooking yield increased in a PTE dose-dependent manner, and at the level of 625 µmol/g protein PTE, the highest gel strength of 308.43 ± 8.12 (mN·cm) and cooking yield of 76.16 ± 1.40% were obtained in the samples treated with UVA irradiation. The same samples also showed increased carbonyl content, decreased total sulfhydryl, unwinding of α-helix, and quenching of fluorescence intensity of endogenous tryptophan, all of which indicated that elevated protein oxidation in these samples led to enhanced protein cross-linking. Results of sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) indicated aggregation of myosin heavy chains (MHCs) in the UVA-treated gels with PTE, also evidenced by the dense three-dimensional network structure in these samples visualized by scanning electron microscopy (SEM). Electron spin resonance (ESR) and spin trapping results indicated that free radicals were produced during the gelation process, possibly originated from UVA-treated PTE, which played a critical role of oxidizing fish MPs, and eventually led to the improvement of the textural properties of the mackerel MP gel. PRACTICAL APPLICATION: Brown algae are a family of high-yield marine algae. Phlorotannin extracts are highly active natural substances extracted from brown algae that can have many applications. Ultraviolet A (UVA) as a green and environmentally friendly physical processing method has been widely used in food processing in recent years. We cultured mouse brain capillary endothelial cell line bEnd.3 on the UV-irradiated Type I collagen gel. Morphology of bEnd.3 cells on the Type I collagen gel was drastically changed if the gel was crosslinked by UV irradiation. The interconnecting network of bEnd.3 cells which have cord-like morphology on the soft collagen gels was converted to the monolayer of the flat cells, tightly-bound each other covering the gel surface, in a confluent state. The collagen gels were mechanically stiffened by UV irradiation for 15 min with UV light at 254 nm showing approximately two times higher value of Young's modulus E (1.51 ± 0.58 kPa) than the control gel (3.17 ± 1.17 kPa). AFM images of the collagen fibrils were not severely changed after irradiation. Collagen subunit proteins were crosslinked and degraded simultaneously under UV irradiation proved by results of SDS-PAGE and separation by centrifugation. Expression of Integrin gene was measured by quantitative real-time PCR. Expression of the integrin α2 gene, tight junction protein 1 gene, and claudin 5 gene were down-regulated in cells on the UV irradiated collagen gel in comparison with the unirradiated one while expression of the integrin β1 gene and Integrin α1 gene did not significantly change. Thick actin filaments were more clearly observed in the cells on the UV-irradiated collagen gel than the unirradiated one by fluorescent microscopy. We conclude that UV irradiation made the collagen gel stiffened and changed the physiological state of bEnd.3 cells including their adhesion, extension, and proliferation., Competing Interests: Declaration of competing interest The author (H. A PEG-Tyr block copolymer forms a kinetically stable colloidal solution in water at room temperature which undergoes an irreversible conversion to a gel phase upon heating. A micellar solution and a gel can therefore be studied under identical experimental conditions. This made it possible to compare physical properties and chemical reactivity of micelles and gels in identical chemical environments and under identical conditions. EPR spectra of the spin-labelled copolymer showed that tyrosine mobility in gels was slightly reduced compared to micelles. Chemical reactivity was studied using photochemical degradation of tyrosine and tyrosine dimerization, in the absence and in the presence of an Fe(iii) salt. The reactivity trends were explained by reduced tyrosine mobility in the gel environment. The largest reactivity difference in gels and micelles was observed for bimolecular dityrosine formation which was also attributed to the reduction in molecular mobility.

Background: Canine otitis externa is a painful condition which can be challenging to treat due to difficulties in the administration of otic medication. This can be due to lack of owner compliance in the application of ear drops or due to the resentment that some dogs demonstrate when attempts are made to administer topical medication into a sensitive ear canal. The aim of the study was to assess the efficacy of a topical LED-illuminated gel (LIG) in canine otitis externa in comparison to standard of care therapy. Dogs with spontaneous otitis externa were randomly allocated in three groups: groups QW received LIG once weekly; BW received LIG twice weekly; group C received enrofloxacin and silver sulfadiazine twice daily. LIG consists of a topical application of a gel containing chromophores that, when illuminated by a LED lamp, re-emit fluorescent light which can stimulate physiological responses, promoting healing and controlling bacteria. The evaluation protocol (T 0 to T 5 ) considered clinical assessment (OTIS-3-index-scoring-system; pruritus-severity-scale; pain-severity-score; aural temperature), cytological scoring system, quali-quantitative bacteriologic assessment., Results: All groups (QW, n = 21; BW, n = 23; C, n = 20) showed improvement during the study (QW: P < 0.02 for cytological and pain scores, P < 0.003 for bacteriologic assessment, P < 10 - 4 for pruritus, total OTIS-3 and temperature assessments; BW: P < 10 - 4 for all clinical, cytological and bacteriologic assessments; C: P < 0.02 for all clinical and cytological assessments, P < 10 - 4 for bacteriologic assessment). The highest clinical score reduction occurred in Group BW (P < 0.014 in T 3 ; P < 0.001 in T 4 and P < 10 - 4 in T 5 ). BW reached the clinically relevant effect level at T 3 (- 3.26 ± 1.21 levels), QW reached it at T 4 (- 3.24 ± 0.99), C did not reach it. No differences between groups were seen in the reduction of CFU/mL (T 0 -T 5 )., Conclusions: All treatment groups showed a positive clinical effect. LIG administered twice-a-week was the most favourable protocol of the study. LIG may be considered beneficial in the management of canine otitis externa; it seems to be effective in controlling the clinical condition, including the signs of inflammation and local pain, the bacterial growth, and it may help increasing treatment compliance.

Radiation therapy in the presence of a strong magnetic field is known to cause regions of enhanced and reduced dose at interfaces of materials with varying densities, in a phenomenon known as the electron return effect (ERE). In this study, a novel low-density gel dosimeter was developed to simulate lung tissue and was used to measure the ERE at the lung-soft tissue interface. Low-density gel dosimeters were developed with Fricke xylenol orange gelatin (FXG) and ferrous oxide xylenol orange (FOX) gels mixed with polystyrene foam beads of various sizes. The gels were characterized based on CT number, MR signal intensity, and uniformity. All low-density gels had CT numbers roughly equivalent to lung tissue. The optimal lung-equivalent gel formulation was determined to be FXG with  <1 mm polystyrene beads due to the higher signal intensity of FXG compared to FOX and the higher uniformity with the small beads. Dose response curves were generated for the optimal low-density gel and conventional FXG. The change in spin-lattice relaxation rate (R1) before and after irradiation was linear with dose for both gels. Next, phantoms consisting of concentric cylinders with low-density and conventional FXG were created to simulate the lung-soft tissue interface. The phantoms were irradiated in a conventional linear accelerator (linac) and in a linac combined with a 1.5 T magnetic resonance imaging (MRI) unit (MR-linac) to measure the effects of the magnetic field on the dose distribution. Hot and cold spots were observed in the dose distribution at the boundaries between the gels for the phantom irradiated in the MR-linac but not the conventional linac, consistent with the ERE.

A gel dosimeter has been developed utilising a recently reported system for reducing Fe 3+ diffusion in a Fricke gel dosimeter which chelates xylenol orange to the gelling agent poly(vinyl alcohol) (PVA). Formulations were investigated using both gelatin and PVA as the gelling agent, along with the inclusion of glyoxal. The resulting gel had an optical density dose response of 0.0031 Gy -1 , an auto-oxidation rate of 0.000 23 h -1 , and a diffusion rate of 0.132 mm 2 h -1 which is a significant improvement over previously reported gelatin based Fricke gel dosimeters. The gel was also shown to be energy and dose-rate independent and could be reused after irradiation. Thus, this gel dosimeter has the potential to provide a safe and practical solution to three dimensional radiation dosimetry in the medical environment.

The effects of pH (3.5, 4.5, and 5.5) and UV-C irradiation dose (12.8, 24.2, 35.8, and 54.6 mJ/cm 2 ) on the physicochemical properties changes in 10% Aloe vera gel blends; in addition, the acemannan concentration and structural changes in the precipitated polysaccharides were evaluated. A thermal treatment (TT; 45 s at 90 °C) was used for comparison. In contrast to TT, a dose of 24.2 mJ/cm 2 did not induce significant changes of free sugar content. Moreover, TT and UV-C irradiation did not significantly affect the content of mannose but increased those of galactose, fructose, and glucose. 1 H NMR analysis revealed minimal changes in the isolated fractions of acemannan, indicating that compared to the unprocessed control sample, the acemannan deacetylation was more pronounced by TT (27%) than by UV-C irradiation (11% at 54.6 mJ/cm 2 ), without any significant difference between the two. UV-C irradiation of Aloe vera gel blends at pH 3.5 and 24.2 mJ/cm 2 was an alternative to TT and efficiently preserve the characteristics of acemannan., (Copyright © 2019 Elsevier Ltd. Purpose: In low-density (LD) gel dosimeter, diffusive spin-spin relaxation rate (R2)-dispersion caused by susceptibility-induced internal gradient leads to a significant deviation in the measured R2 from the real value. In this study, the effect of induced internal gradient on R2 was visualized and quantified algebraically as an important cause of inaccuracy in LD gel dosimeters., Materials and Methods: In this method, two sets of LD and unit-density (UD) gel dosimeters were prepared. The LD gel was made by mixing the UD gel with expanded polystyrene spheres. The R2 was used to determine the spatially resolved decay rates due to diffusion in internal magnetic field. The internal gradient was calculated for a multiple spin-echo sequence., Results: It is shown that in a LD gel, the internal gradient leads to overestimation of mean R2 value (R2 mean ). Pixel-by-pixel R2 measurements inside a LD gel showed significant deviation from R2 mapping in UD gel., Conclusion: It appears that significant differences between R2 mean in a selected region of interest and pixel-by-pixel R2 values are the main source of inaccuracy in dose mapping of a LD gel., Competing Interests: None

Background: The human body consists of a variety of tissues and cavities with different physical and radiological properties. The most important among these are tissues and cavities including lungs, oral cavities, teeth, nasal passages, sinuses, and bones. Thus, the dose distribution in these tissues cannot be verified with soft tissue equivalent polymer gel dosimeters. This study is conducted to evaluate the effect of homogeneity on dose response of an anoxic low-density polymer gel dosimeter for radiation therapy., Materials and Methods: The low-density polymer gel dosimeter is composed of 12% gelatin, 5% methacrylic acid, 0.15% sodium dodecyl sulfate, 10 mm tetrakis (hydroxymethyl) phosphonium chloride, and 83% ultrapure deionized water. The time between irradiation and scanning for all gels experiments was 18 h. The gel dosimeters were imaged using a 1.5 T clinical magnetic resonance imaging scanner in a transmitter/receiver head coil., Results: The low-density gels had a mass density between 0.35 g/cm 3 and 0.45 g/cm 3 and the computed tomography (CT) values varied from approximately -650 to -750 Hounsfield units. It is clear that the CT numbers of the fabricated gel are close to those of the normal human lung tissue, which ranges from -770 to -875 Hounsfield units., Conclusion: Findings of the present study showed that if the density and homogeneity of the low-density polymer gel dosimeters are equal to those of the normal lung tissue, it may be used to measure the three dimensional dose distributions in lung tissue during radiation therapy., Competing Interests: There are no conflicts of interest

This study introduces the first 3D deformable dosimetry system based on x-ray computed tomography (CT) polymer gel dosimetry and establishes the setup reproducibility, deformation characteristics and dose response of the system. A N-isopropylacrylamide (NIPAM)-based gel formulation optimized for x-ray CT gel dosimetry was used, with a latex balloon serving as the deformable container and low-density polyethylene and polyvinyl alcohol providing additional oxygen barrier. Deformable gels were irradiated with a 6 MV calibration pattern to determine dosimetric response and a dosimetrically uniform plan to determine the spatial uniformity of the response. Wax beads were added to each gel as fiducial markers to track the deformation and setup of the gel dosimeters. From positions of the beads on CT images the setup reproducibility and the limits and reproducibility of gel deformation were determined. Comparison of gel measurements with Monte Carlo dose calculations found excellent dosimetric accuracy, comparable to that of an established non-deformable dosimetry system, with a mean dose discrepancy of 1.5% in the low-dose gradient region and a gamma pass rate of 97.9% using a 3%/3 mm criterion. The deformable dosimeter also showed good overall spatial dose uniformity throughout the dosimeter with some discrepancies within 20 mm of the edge of the container. Tracking of the beads within the dosimeter found that sub-millimetre setup accuracy is achievable with this system. The dosimeter was able to deform and relax when externally compressed by up to 30 mm without sustaining any permanent damage. Internal deformations in 3D produced average marker movements of up to 12 mm along the direction of compression. These deformations were also shown to be reproducible over 100 consecutive deformations. This work has established several important characteristics of a new deformable dosimetry system which shows promise for future clinical applications, including the validation of deformable dose accumulation algorithms.

Three dimensional dosimetry by optical CT readout of radiosensitive gels or solids has previously been indicated as a solution for measurement of radiotherapy 3D dose distributions. The clinical uptake of these dosimetry methods has been limited, partly due to impracticalities of the optical readout such as the expertise and labour required for refractive index fluid matching. In this work a fast laser beam optical CT scanner is described, featuring fluid-less and dual wavelength operation. A second laser with a different wavelength is used to provide an alternative reference scan to the commonly used pre-irradiation scan. Transmission data for both wavelengths is effectively acquired simultaneously, giving a single scan process. Together with the elimination of refractive index fluid matching issues, scanning practicality is substantially improved. Image quality and quantitative accuracy were assessed for both dual and single wavelength methods. The dual wavelength scan technique gave improvements in uniformity of reconstructed optical attenuation coefficients in the sample 3D volume. This was due to a reduction of artefacts caused by scan to scan changes. Optical attenuation measurement accuracy was similar for both dual and single wavelength modes of operation. These results established the basis for further work on dosimetric performance.

The aim of this study is to evaluate the application and accuracy of polymer gels for determining electron dose distributions in the presence of small heterogeneities made of bone and air. Different cylindrical phantoms containing MAGIC (Methacrylic and Ascorbic acid in Gelatin Initiated by Copper) normoxic polymer gel were used under the slab phantoms during irradiation. MR images of the irradiated gel phantoms were obtained to determine their R2 (spin-spin) relaxation maps for conversion to absorbed dose. One- and 2-dimensional lateral dose profiles were acquired at depths of 1 and 4 cm for 8 and 15 MeV electron beams. The results were compared with the doses measured by a diode detector at the same positions. In addition, the dose distribution in the axial orientation was measured by the gel dosimeter. The slope and intercept for the R2 versus dose curve were 0.509 ± 0.002 Gy s and 4.581 ± 0.005 s, respectively. No significant variation in dose-R2 response was seen for the two electron energies within the applied dose ranges. The mean dose difference between the measured gel dose profiles was smaller than 3% compared to those measured by the diode detector. These results provide further demonstration that electron dose distributions are significantly altered in the presence of tissue inhomogeneities such as bone and air cavity and that MAGIC gel is a useful tool for 3-dimensional dose visualization and qualitative assessment of tissue inhomogeneity effects in electron beam dosimetry.

This work presents an improvement of the VIPAR nd ('nd' stands for 'normoxic, double', or VIP) polymer gel dosimeter. The gel composition was altered by increasing the concentration of the monomeric components, N-vinylpyrrolidone (NVP) and N,N'-methylenebisacrylamide (MBA), in co-solvent solutions. The optimal composition (VIPAR CT , where 'CT' stands for computed tomography, or VIC) comprised: 17% NVP, 8% MBA, 12% t-BuOH, 7.5% gelatine, 0.007% ascorbic acid, 0.0008% CuSO 4   ×  5H 2 O and 0.02% hydroquinone. The following characteristics of VIC were achieved: (i) linear dose range of 0.9 &#95; 30 Gy, (ii) saturation for radiation doses of over 50 Gy, (iii) threshold dose of about 0.5 Gy, (iv) dose sensitivity of 0.171 Gy -1 s -1 , which is roughly 2.2 times higher than that of VIP (for nuclear magnetic resonance measurements). It was also found that VIC is dose- rate-independent, and its dose response does not alter if the radiation source is changed from electrons to photons for external beam radiotherapy. The gel responded similarly to irradiation with small changes in radiation energy but was sensitive to larger energy changes. The VIC gel retained temporal stability from 20 h until at least 10 d after irradiation, whereas spatial stability was retained from 20 h until at least 6 d after irradiation. The scheme adopted for VIC manufacturing yields repeatable gels in terms of radiation dose response. The VIC was also shown to perform better than VIP using x-ray computed tomography as a readout method; the dose sensitivity of VIC (0.397 HU Gy -1 ) was 1.5 times higher than that of VIP. Also, the dose resolution of VIC was better than that of VIP in the whole dose range examined.

Carboxyl group modified zinc phthalocyanines (ZnPc) are classic and widely used photosensitizers (PSs) in upconversion nanoparticles (UCNPs) mediated photodynamic therapy (PDT) for tumor treatment. To improve the PDT activity of the complex system of ZnPc and UCNPs, many UCNPs with high red emission intensity were design and prepared. ZnPc-(COOH) 4 tends to aggregate both in water and under physiological conditions, which can sharply decrease its PDT activity. Therefore, choosing monomeric COOH groups modified ZnPc as PSs is an alternative way to improve their activity. In this manuscript, three zinc(ii) phthalocyanines, substituted with 4 (ZnPc-(COOH) 4 ), 8 (ZnPc-(COOH) 8 ) and 16 (ZnPc-(COOH) 16 ) COOH groups, were synthesized. A comparison of the results indicated that ZnPc-(COOH) 16 existed in its monomeric form under physiological conditions because of its large substituents. Moreover, ZnPc-(COOH) 16 showed superior singlet oxygen ( 1 O 2 ) generation ability when compared to ZnPc-(COOH) 4 and ZnPc-(COOH) 8 . Therefore, we chose ZnPc-(COOH) 16 as PSs for absorption on the surface of the UCNPs. Then, they were encapsulated by crosslinked methacrylated hyaluronic acid (m-HA), which provides active tumor accumulation ability by binding its overexpressed receptors on the surface of cancer cells. The resulting nanoparticles can be effectively taken up by cancer cells and shows strong near-infrared (NIR) light triggered PDT in vitro.

In vivo dosimetry is a greatly underutilized tool for patient safety in clinical external beam radiotherapy treatments, despite being recommended by several national and international organizations (AAPM, ICRU, IAEA, NACP). The reasons for this underutilization mostly relate to the feasibility and cost of in vivo dosimetry methods. Due to the increase in the number of beam angles and dose per fraction in modern treatments, there is a compelling need for a novel dosimeter that is robust and affordable while able to operate properly in these complex conditions. This work presents a gel patch dosimeter as a novel method of in vivo dosimetry. DEFGEL, a 6% T normoxic polyacrylamide gel, was injected into 1 cm thick acrylic molds to create 1 cm thick small cylindrical patch dosimeters. To evaluate the change in optical density due to radiation induced polymerization, dosimeters were scanned before and after irradiation using an in-house developed laser densitometer. The dose-responses of three separate batches of gel were evaluated and compared to check for linearity and repeatability. The response development time was evaluated to ensure that the patch dosimeter could be high throughput. Additionally, the potential of this system to be used as an in vivo dosimeter was tested with a clinically relevant end-to-end in vivo phantom test. All irradiations were performed with a Varian Clinac 21EX at the University of Wisconsin Medical Radiation Research Center (UWMRRC). The dose-response of all three batches of gel was found to be linear within the range of 2-20 Gy. At doses below 0.5 Gy the statistical uncertainties were prohibitively large to make quantitative assessments of the results. The three batches demonstrated good repeatability in the range of 2 Gy to up to 10 Gy, with only slight variations in response at higher doses. For low doses the dosimeter fully developed within an hour while at higher doses they fully developed within four hours. During the in vivo phantom test the predicted patch absorbed dose was 4.23 Gy while the readout dose was evaluated to be 4.37 Gy, which corresponds to a 3.2% discrepancy. The dosimeter and densitometer pairing shows promise as an in vivo dosimetry system, especially for hypofractionated or MRI-guided radiotherapy treatments where higher doses are prescribed.

Gel dosimeters have acquired a unique status in radiotherapy, especially with the advent of the new techniques in which there is a need for three-dimensional dose measurement with high spatial resolution. One of the techniques in which the use of gel dosimeters has drawn the attention of the researchers is the boron neutron capture therapy. Background: Ion beam therapy represents a promising approach to treat prostate cancer, mainly due to its high conformity and radiobiological effectiveness. However, the presence of prostate motion, patient positioning and range uncertainties may deteriorate target dose and increase exposure of organs at risk. Spacer gel injected between prostate and rectum may increase the safety of prostate cancer (PC) radiation therapy by separating the rectum from the target dose field. The dosimetric impact of the application of spacer gel for scanned carbon ion therapy of PC has been analyzed at Heidelberg Ion-Beam Therapy Center (HIT)., Materials and Methods: The robustness of ion therapy treatment plans was investigated by comparison of two data sets of patients treated with and without spacer gel. A research treatment planning system for ion therapy was used for treatment plan optimization and calculation of daily dose distributions on 2 to 9 Computed Tomography (CT) studies available for each of the 19 patients. Planning and daily dose distributions were analyzed with respect to target coverage, maximal dose to the rectum (excluding 1 ml of the greatest dose; Dmax-1 ml) and the rectal volume receiving dose greater than 90% of prescribed target dose (V90Rectum), respectively., Results: The application of spacer gel did substantially diminish rectum dose. Dmax-1 ml on the treatment planning CT was on average reduced from 100.0 ± 1.0% to 90.2 ± 4.8%, when spacer gel was applied. The robustness analysis performed with daily CT studies demonstrated for all analyzed patient cases that application of spacer gel results in a decrease of the daily V90Rectum index, which calculated over all patient cases and CT studies was 10.2 ± 10.4 [ml] and 1.1 ± 2.1 [ml] for patients without and with spacer gel, respectively., Conclusions: The dosimetric benefit of increasing the distance between prostate and rectum using spacer gel for PC treatment with carbon ion beams has been quantified. Application of spacer gel substantially reduced rectal exposure to high treatment dose and, therefore, can reduce the hazard of rectal toxicity in ion beam therapy of PC. The results of this study enable modifications of the PC ion therapy protocol such as dose escalation or hypofractionation.

We aimed to investigate whether a cylindrical structure containing either air or fluid and with or without a metallic stent affects the volume and density of cavitation produced by high-intensity focused ultrasound via a gel phantom study. Sixteen tissue-mimicking phantoms based on a polyacrylamide gel mixed with bovine serum albumin with a cylindrical hole 1 cm in diameter and 7.5 cm in length were divided into four groups of four phantoms with air in the holes (group 1), four phantoms with fluid in the holes (group 2), four phantoms with air-containing metallic stents (group 3) and four phantoms with fluid-containing metallic stents (group 4). A pulsed high-intensity focused ultrasound beam (50% duty cycle, 40-Hz pulse repetition frequency) at 75 W of acoustic power was directed perpendicularly to the longitudinal axis of the hole, with its focus at the posterior wall of the hole. The size of the cavitation on the x-, y-, and z-axes was measured, and the volumes of cavitation and coagulation were calculated using the formula for the volume of an elliptical cone. The density of cavitation was measured in the tissue phantom anterior to the hole with a 1 × 1-cm square region of interest. For statistical analysis, the Kruskal-Wallis test and Mann-Whitney U-test were used. The phantoms with air-containing holes (groups 1 and 3) developed larger and denser cavitations anterior to the focus, without unnecessary coagulation posterior to the focus, compared with the phantoms with fluid-containing holes (groups 2 and 4), regardless of the presence of stents. All of the axes and volumes of the anterior cavitations were significantly larger than those of the posterior cavitations in groups 1 and 3 (all p-values <0.05). This study reports on the functional properties of 7% flaxseed oil/milk emulsion obtained by sonication (OM) using 20 kHz ultrasound (US) at 176 W for 1-8 min in two different delivery formulae, viz., ready-to-drink (RTD) and lactic acid gel. The RTD emulsions showed no change in viscosity after sonication for up to 8 min followed by storage up to a minimum of 9 days at 4±2 °C. Similarly, the oxidative stability of the RTD emulsion was studied by measuring the conjugated diene hydroperoxides (CD). The CD was unaffected after 8 min of ultrasonic processing. The safety aspect of US processing was evaluated by measuring the formation of CD at different power levels. The functional properties of OM gels were evaluated by small and large scale deformation studies. The sonication process improved the gelation characteristics, viz., decreased gelation time, increased elastic nature, decreased syneresis and increased gel strength. The presence of finer sono-emulsified oil globules, stabilized by partially denatured whey proteins, contributed to the improvements in the gel structure in comparison to sonicated and unsonicated pasteurized homogenized skim milk (PHSM) gels. Ultrasound has been previously investigated as an alternative readout method for irradiated polymer gel dosimeters, with authors reporting varying dose responses. We extend previous work utilizing a new computed tomography ultrasound scanner comprising of two identical 5 MHz, 128-element linear-array ultrasound transducers, co-axially aligned and submerged in water as a coupling agent, with rotational of the gel dosimeter between the transducers facilitated by a robotic arm. We have investigated the dose-dependence of both ultrasound bulk attenuation and broadband ultrasound attenuation (BUA) for the PAGAT gel dosimeter. The ultrasound bulk attenuation dose sensitivity was found to be 1.46  ±  0.04 dB m( -1) Gy( -1), being in agreement with previously published results for PAG and MAGIC gels. BUA was also found to be dose dependent and was measured to be 0.024  ±  0.003 dB MHz( -1) Gy( -1); the advantage of BUA being its insensitivity to frequency-independent attenuation mechanisms including reflection and refraction, thereby minimizing image reconstruction artefacts.

The introduction of beam intensity control concept in current radiotherapy techniques has increased treatment planning complexity. Thus, small-field dose measurement has become increasingly vital. Polymer gel dosimetry method is widely studied. It is the only dose measurement tool that provides 3D dose distribution. This study aims to use an N-isopropylacrylamide (NIPAM) gel dosimeter to conduct beam performance measurements of percentage depth dose (PDD), beam flatness, and symmetry for photon beams with field sizes of 3×3 and 4×4 cm(2). Computed tomography scans were used to readout the gel dosimeters. In the PDD measurement, the NIPAM gel dosimeter and Gafchromic™ EBT3 radiochromic film displayed high consistency in the region deeper than the build-up region. The gel dosimeter dose profile had 3% lower flatness and symmetry measurement at 5 cm depth for different fields compared with that of the Gafchromic™ EBT3 film. During gamma evaluation under 3%/3 mm dose difference/distance-to-agreement standard, the pass rates of the polymer gel dosimeter to the TPS and EBT3 film were both higher than 96%. Given that the gel is tissue equivalent, it did not exhibit the energy dependence problems of radiochromic films. Several tools are used for the dosimetric verification of intensity-modulated arc therapy (IMAT) treatment delivery. However, limited information is available for composite on-line evaluation of these tools. The purpose of this study was to evaluate the dosimetric verification of IMAT treatment plans using a 2D diode array detector (2D array), radiochromic film (RCF) and radiosensitive polymer gel dosimeter (RPGD). The specific verification plans were created for IMAT for two prostate cancer patients by use of the clinical treatment plans. Accordingly, the IMAT deliveries were performed with the 2D array on a gantry-mounting device, RCF in a cylindrical acrylic phantom, and the RPGD in two cylindrical phantoms. After the irradiation, the planar dose distributions from the 2D array and the RCFs, and the 3D dose distributions from the RPGD measurements were compared with the calculated dose distributions using the gamma analysis method (3% dose difference and 3-mm distance-to-agreement criterion), dose-dependent dose difference diagrams, dose difference histograms, and isodose distributions. The gamma passing rates of 2D array, RCFs and RPGD for one patient were 99.5%, 96.5% and 93.7%, respectively; the corresponding values for the second patient were 97.5%, 92.6% and 92.9%. Mean percentage differences between the RPGD measured and calculated doses in 3D volumes containing PTVs were -0.29 ± 7.1% and 0.97 ± 7.6% for the two patients, respectively. In conclusion, IMAT prostate plans can be delivered with high accuracy, although the 3D measurements indicated less satisfactory agreement with the treatment plans, mainly due to the dosimetric inaccuracy in low-dose regions of the RPGD measurements.

The use of small field sizes is increasingly becoming important in radiotherapy particularly since the introduction of stereotactic radiosurgery and intensity-modulated radiation therapy techniques. The reliable measurement of delivered dose from such fields with conventional dosimeters, such as ionization chambers, is a challenging task. In this work, methacrylic and ascorbic acid in gelatin initiated by copper polymer gel dosimeters are employed to measure dose in 3 dimensions. Field sizes of 5 × 5mm(2), 10 × 10mm(2), 20 × 20mm(2), and 30 × 30mm(2) are investigated for a 6-MV x-rays. The results show an agreement with Gafchromic film, with some variation in measured doses near the edge of the fields, where the film data decrease more rapidly than the other methods. Dose penumbra widths obtained with gel dosimeters and Gafchormic film were generally in agreement with each other. Purpose: The emergence of thermal modalities has promoted the use of heat-sensitive phantoms for calibration, measurement, and verification purposes. However, development of durable phantoms with high precision ability to represent the temperature distribution remains a challenge. This study aims to introduce a reusable phantom that provides an accurate assessment of the heated region in various thermal modalities., Materials and Methods: The phantom contains a thermochromic dye that is transparent blue at room temperature and becomes colourless after exceeding a threshold temperature. In order to determine the threshold temperature of the phantom, spectrophotometry analysis was performed. The various thermal (specific heat, thermal conductivity, melting point and latent heat of melting) and acoustic (sound speed, attenuation) properties of this phantom were measured and compared with those of the reference phantom without dye. The application of this phantom for radio-frequency and magnetic resonance guided focused ultrasound modalities was also examined., Results: The spectrophotometry analysis showed a threshold temperature of 50 ± 3 °C for this phantom. The results also demonstrated a 6 °C difference between the onset and ending temperatures of the discolouration process. Moreover, the starting temperature of colouration during cooling was found to be 4 °C lower than the ending temperature of discolouration. The sound speed, attenuation, specific heat, thermal conductivity and melting point of the heat-sensitive phantom were statistically equal to those of the reference phantom; however, the latent heat, and onset temperature of the melting of the heat-sensitive phantom were decreased by addition of the dye., Conclusions: The developed phantom is applicable for accurate evaluation of temperature variations in various thermal modalities.

Polymer gels are remarkable materials with physical structures that can adapt significantly and quite rapidly with changes in the local environment, such as temperature, light intensity, electrochemistry, and mechanical force. An interesting phenomenon observed in certain polymer gel systems is mechanochromism - a change in color due to a mechanical deformation. Mechanochromic photonic gels are periodically structured gels engineered with a photonic stopband that can be tuned by mechanical forces to reflect specific colors. These materials have potential as mechanochromic sensors because both the mechanical and optical properties are highly tailorable via incorporation of diluents, solvents, nanoparticles, or polymers, or the application of stimuli such as temperature, pH, or electric or strain fields. Recent advances in photonic gels that display strain-dependent optical properties are discussed. In particular, this discussion focuses primarily on polymer-based photonic gels that are directly or indirectly fabricated via self-assembly, as these materials are promising soft material platforms for scalable mechanochromic sensors., (Copyright © 2013 WILEY-VCH Verlag GmbH & Co. It is difficult to obtain images of thin slices from measurement of spin-spin relaxation (R2) with magnetic resonance imaging (MRI) using the traditional dose reading method of polymer gel dosimetry. In this study, the dose reading method was performed using X-ray computed tomography (CT) for proton beam measurements in order to enable collection of thin slices. In addition, three-dimensional (3D) images of polymer gels were constructed using volume rendering. As a result of acquisition of thin slices, more detailed 3D data consisting of smaller voxel sizes compared to R2 were acquired. However, it was found that with thin slice thicknesses and small voxels, the signal-to-noise ratio around the voxels deteriorated. Background: The application of spacer gel represents a promising approach to reliably spare the rectal frontal wall during particle therapy (IJROBP 76:1251-1258, 2010). In order to qualify the spacer gel for the clinical use in particle therapy, a variety of measurements were performed in order to ensure the biological compatibility of the gel, its physical stability during and after the irradiation, and a proper definition of the gel in terms of the Hounsfield Unit (HU) values for the treatment planning system. The potential for the use of the spacer gel for particle therapy monitoring with off-line Positron Emission Tomography (PET) was also investigated., Results: The spacer gel implanted to the prostate patient in direct neighbourhood to the clinical target volume does not interfere with the particle therapy treatment planning procedure applied at Heidelberg Ion Beam Therapy Centre (HIT). The performed measurements show that Bragg-peak position of the particles can be properly predicted on the basis of computed tomography imaging with the treatment planning system used at HIT (measured water equivalent path length of 1.011 ±0.011 (2σ), measured Hounsfield Unit of 28.9 ±6.1 (2σ)). The spacer gel samples remain physically unchanged after irradiation with a dose exceeding the therapeutic dose level. The independently measured Bragg-Peak position does not change within the time interval of 10 weeks., Conclusions: As a result of the presented experiments, the first clinical application of spacer gel implant during prostate cancer treatment with carbon ions and protons was possible at HIT in 2012. The reported pre-clinical investigations demonstrate that use of spacer gel is safe in particle therapy in presence of therapy target motion and patient positioning induced particle range variations. The spacer gel injected between prostate and rectum enlarge the distance between both organs, which is expected to clinically significantly decrease the undesirable exposure of the most critical organ at risk, i.e. rectal frontal wall. Further research on the composition of spacer gel material might lead to additional clinical benefits by validation of particle therapy of prostate via post-therapeutic PET-imaging or by patient positioning based on the gel as a radio-opaque marker.

Micropatterning is useful for regulating culture environments. We developed a highly efficient near-infrared-(NIR)-responsive gel and established a new technique that enables cell patterning by NIR irradiation. As a new culture substratum, we designed a tissue culture plate that was coated with a composite gel composed of agarose and carbon nanotubes (CNTs). A culture plate coated with agarose only showed no response to NIR irradiation. In contrast, NIR laser irradiation induced heat generation by CNTs; this permitted local solation of the CNT/agarose gel, and consequently, selective cell-adhesive regions were exposed on the tissue culture plate. The solation area was controlled by the NIR intensity, magnification of the object lens and CNT concentration in the gel. Furthermore, we formed circular patterns of HeLa cells and linear patterns of 3T3 cells on the same culture plate through selective and stepwise NIR irradiation of the CNT/agarose gel, and we also demonstrated that individual 3T3 cells migrated along a linear path formed on the CNT/agarose gel by NIR irradiation. These results indicate that our technique is useful for tailor-made cell patterning of stepwise and/or complex cell patterns, which has various biological applications such as stepwise co-culture and the study of cell migration.

The temperature dependence of an agar/gelatin phantom was evaluated. The purpose was to predict the material property response to high-intensity focused ultrasound (HIFU) for developing ultrasound guided dosing and targeting feedback. Changes in attenuation, sound speed, shear modulus and thermal properties with temperature were examined from 20°C to 70°C for 3 weeks post-manufacture. The attenuation decreased with temperature by a power factor of 0.15. Thermal conductivity, diffusivity and specific heat all increased linearly with temperature for a total change of approximately 16%, 10% and 6%, respectively. Sound speed had a parabolic dependence on temperature similar to that of water. Initially, the shear modulus irreversibly declined with even a slight increase in temperature. Over time, the gel maintained its room temperature shear modulus with moderate heating. Using computational modeling, we show that self-oscillating Belousov-Zhabotinsky (BZ) gels can both emit and sense a chemical signal and thus drive neighboring gel pieces to spontaneously self-aggregate, so that the system exhibits autochemotaxis. To the best of our knowledge, this is the closest system to the ultimate self-recombining material, which can be divided into separated parts and the parts move autonomously to assemble into a structure resembling the original, uncut sample. We also show that the gels' coordinated motion can be controlled by light, allowing us to achieve selective self-aggregation and control over the shape of the gel aggregates. By exposing the BZ gels to specific patterns of light and dark, we design a BZ gel "train" that leads the movement of its "cargo." Our findings pave the way for creating reconfigurable materials from self-propelled elements, which autonomously communicate with neighboring units and thereby actively participate in constructing the final structure.

A method is described for measuring absorbed electromagnetic energy radiated from cell phone antennae into ex vivo brain tissue. NMR images the 3D thermal dynamics inside ex vivo bovine brain tissue and equivalent gel under exposure to power and irradiation time-varying radio frequency (RF) fields. The absorbed RF energy in brain tissue converts into Joule heat and affects the nuclear magnetic shielding and the Larmor precession. The resultant temperature increase is measured by the resonance frequency shift of hydrogen protons in brain tissue. This proposed application of NMR thermometry offers sufficient spatial and temporal resolution to characterize the hot spots from absorbed cell phone radiation in aqueous media and biological tissues. Specific absorption rate measurements averaged over 1 mg and 10 s in the brain tissue cover the total absorption volume. Reference measurements with fiber optic temperature sensors confirm the accuracy of the NMR thermometry.

A relative humidity (RH) sensor based on long-period grating (LPG) with different responses is proposed by utilizing agarose gel as the sensitive cladding film. The spectral characteristic is discussed as the ambient humidity level ranges from 25% to 95% RH. Since increment of RH will result in volume expansion and refractive index increment of the agarose gel, the LPG is sensitive to applied strain and ambient refractive index; both the resonance wavelength and coupling intensity present particular responses to RH within two different RH ranges (25%-65% RH and 65%-96% RH). The coupling intensity decreases within a lower RH range while it increases throughout a higher RH range. The resonance wavelength is sensitive to the higher RH levels, and the highest sensitivity reaches 114.7 pm/% RH, and shares the same RH turning point with coupling intensity response. From a practical perspective, the proposed RH sensor would find its potential applications in high humidity level, temperature-independent RH sensing and multiparameter sensing based on wavelength/power hybrid demodulation and even static RH alarm for automatic monitoring of a particular RH value owing to the nonmonotonic RH dependence of the transmission power within the whole tested RH range.

One-dimensionally periodic block copolymer photonic lamellar gels with full-color tunability as a result of a direct exchange of counteranions were fabricated via a two-step procedure comprising the self-assembly of a hydrophobic block-hydrophilic polyelectrolyte block copolymer, polystyrene-b-poly(2-vinyl pyridine) (PS-b-P2VP), followed by sequential quaternization of the P2VP layers in 1-bromoethane solution. Depending on the hydration characteristics of each counteranion, the selective swelling of the block copolymer lamellar structures leads to large tunability of the photonic stop band from blue to red wavelengths. More extensive quaternization of the P2VP block allows the photonic lamellar gels to swell more and red shift to longer wavelength. Here, we investigate the dynamic swelling behavior in the photonic gel films through time-resolved in situ measurement of UV-vis transmission. We model the swelling behavior using the transfer matrix method based on the experimentally observed reflectivity data with substitution of appropriate counterions. These tunable structural color materials may be attractive for numerous applications such as high-contrast displays without using a backlight, color filters, and optical mirrors for flexible lasing.

We report a study of crack patterns formed in laponite gel drying in an electric field. The sample dries in a circular petri dish and the field is radial, acting inward or outward. A system of radial cracks forms in the setup with the center terminal positive, while predominantly cross-radial cracks form when the center is at a negative potential. The laponite accumulates near the negative terminal making the layer thicker at this end. A spring model on a square lattice is used to simulate the desiccation crack formation, with an additional radial force acting due to the electric field. With the radial force acting outward, radial cracks form and for the reversed field cross-radial cracks form. This conforms to the observation that laponite platelets become effectively positive due to overcharging and are attracted towards the negative terminal.

Purpose: Interfraction and intrafraction variation in anatomic structures is a significant challenge in contemporary radiotherapy. The objective of this work is to develop a novel tool for deformable structure dosimetry, using a tissue-equivalent deformable gel dosimeter that can reproducibly simulate targets subject to deformation. This will enable direct measurement of integrated doses delivered in different deformation states, and the verification of dose deforming algorithms., Methods: A modified version of the nPAG polymer gel has been used as a deformable 3D dosimeter and phantom to investigate doses delivered to deforming tissue-equivalent geometry. The deformable gel (DEFGEL) dosimeter/phantom is comprised of polymer gel in a latex membrane, moulded (in this case) into a cylindrical geometry, and deformed with an acrylic compressor. Fifteen aluminium fiducial markers (FM) were implanted into DEFGEL phantoms and the reproducibility of deformation was determined via multiple computed tomography (CT) scans in deformed and nondeformed states before and after multiple (up to 150) deformations. Dose was delivered to the DEFGEL phantom in three arrangements: (i) without deformation, (ii) with deformation, and (iii) cumulative exposures with and without deformation, i.e., dose integration. Irradiations included both square field and a stereotactic multiple dynamic arc treatment adapted from a patient plan. Doses delivered to the DEFGEL phantom were read out using cone beam optical CT., Results: Reproducibility was verified by observation of interscan shifts of FM locations (as determined via CT), measured from an absolute reference point and in terms of inter-FM distance. The majority (76%) of points exhibited zero shift, with others shifting by one pixel size consistent with setup error as confirmed with a control sample. Comparison of dose profiles and 2D isodose distributions from the three arrangements illustrated complex spatial redistribution of dose in all three dimensions occurring as a result of the change in shape of the target between irradiations, even for a relatively simple deformation. Discrepancies of up to 30% of the maximum dose were evident from dose difference maps for three orthogonal planes taken through the isocenter of a stereotactic field., Conclusions: This paper describes the first use of a tissue-equivalent, 3D dose-integrating deformable phantom that yields integrated or redistributed dosimetric information. The proposed methodology readily yields three-dimensional (3D) dosimetric data from radiation delivery to the DEFGEL phantom in deformed and undeformed states. The impacts of deformation on dose distributions were readily seen in the isodose contours and line profiles from the three arrangements. It is demonstrated that the system is potentially capable of reproducibly emulating the physical deformation of an organ, and therefore can be used to evaluate absorbed doses to deformable targets and organs at risk in three dimensions and to validate deformation algorithms applied to dose distributions.

This study evaluates the dosimetric performance of the polymer gel dosimeter 'Methacrylic and Ascorbic acid in Gelatin, initiated by Copper' and its suitability for quality assurance and analysis of I-131-targeted radionuclide therapy dosimetry. Four batches of gel were manufactured in-house and sets of calibration vials and phantoms were created containing different concentrations of I-131-doped gel. Multiple dose measurements were made up to 700 h post preparation and compared to equivalent Monte Carlo simulations. In addition to uniformly filled phantoms the cross-dose distribution from a hot insert to a surrounding phantom was measured. In this example comparisons were made with both Monte Carlo and a clinical scintigraphic dosimetry method. Dose-response curves generated from the calibration data followed a sigmoid function. The gels appeared to be stable over many weeks of internal irradiation with a delay in gel response observed at 29 h post preparation. This was attributed to chemical inhibitors and slow reaction rates of long-chain radical species. For this reason, phantom measurements were only made after 190 h of irradiation. For uniformly filled phantoms of I-131 the accuracy of dose measurements agreed to within 10% when compared to Monte Carlo simulations. A radial cross-dose distribution measured using the gel dosimeter compared well to that calculated with Monte Carlo. Small inhomogeneities were observed in the dosimeter attributed to non-uniform mixing of monomer during preparation. However, they were not detrimental to this study where the quantitative accuracy and spatial resolution of polymer gel dosimetry were far superior to that calculated using scintigraphy. The difference between Monte Carlo and gel measurements was of the order of a few cGy, whilst with the scintigraphic method differences of up to 8 Gy were observed. A manipulation technique is also presented which allows 3D scintigraphic dosimetry measurements to be compared to polymer gel dosimetry measurements without generating misleading errors due to the limited spatial resolution.

In this feasibility study an organic plastic scintillator is calibrated against ionisation chamber measurements and then embedded in a polymer gel dosimeter to obtain a quasi-4D radiation detector. This hybrid dosimeter was irradiated with megavoltage x-rays from a linear accelerator, with temporal measurements of the dose rate being acquired by the scintillator and spatial measurements acquired with the gel dosimeter. The detectors employed in this study are radiologically equivalent; and we show that neither detector perturbs the intensity of the radiation field of the other. By employing these detectors in concert, spatial and temporal variations in the radiation intensity can now be detected and gel dosimeters can be calibrated for absolute dose from a single irradiation.

It was more than 50 years ago that an appearance of birefringence in alginate gels prepared under cation flow was reported for the first time, however, the anisotropic structure of the alginate gel has not been studied in detail. In the present study, anisotropic Ca-alginate gels were prepared within dialysis tubing in a high Ca(2+)-concentration external bath, and optical and small-angle X-ray scattering (SAXS) measurements were performed to characterize the structure of the gel. The observations of the gel with crossed polarizers and with circular polarizers revealed the molecular orientation perpendicular to the direction of Ca(2+) flow. Analyses of the SAXS intensity profiles indicated the formation of rod-like fibrils consisting of a few tens of alginate molecules and that the anisotropy of the gel was caused by the circumferential orientation of the large fibrils. From the observed asymmetric SAXS pattern, it was found that the axis of rotational symmetry of the anisotropic structure was parallel to the direction of Ca(2+) flow. The alignment factor (A(f)) calculated from the SAXS intensity data confirmed that the orientation of the fibrils was perpendicular to the direction of Ca(2+) flow.

Purpose: Three dimensional dose polymer gel dosimetry measurements provide unique information on sophisticated dose distributions. In this study, the authors propose a novel method to improve the accuracy of polymer gel dosimeters by inserting a plastic scintillation detector (PSD) to provide a dose reference., Methods: PSD dosimeters were calibrated using chromatic deconvolution and then inserted into polyacrylanide gel (PAG) dosimeters. The gel and the PSDs were immersed into water and irradiated with 6 MV wedge filtered beams to obtain a wide range of dose variation. Calibration vials containing the same gel were also irradiated to generate a standard calibration curve. The distribution of magnetic nuclear transverse relaxation rate (R2) values of the gel was determined with a multislice multiecho MRI sequence at 1.5 T. Another calibration curve was obtained by assigning the R2 values in the gel surrounding the scintillators to the dose determined by the PSDs. A reference calibration point from a PSD located in a low dose gradient area served to correct the standard calibration method yielding three novel calibration methods. The results were compared with EBT2 GAFCHROMIC film measurements acquired in the same condition and with the Pinnacle3 treatment planning dose calculations,, Results: The mean absolute error of the standard calibration method ranged from 6.1 to 12.4%. The corresponding gamma index (3%/3 mm distance to agreement) criterion was satisfied for only 56% of the pixels in the middle slice of the gel compared to Pinnacle3 dose calculations and to EBT2 film measurements in the center part of the field. Calibration methods using a PSD reduced the mean absolute error to less than 4%; this value was under 2.6% for one of the three methods. In that case, 98% of the pixels satisfied the gamma index criterion., Conclusions: The accuracy of PAG dosimeters may be highly improved using one reference dose point measurement using a plastic scintillation detector. The best calibration procedure corrected the slope of the calibration curve derived from the calibration vials to match the R2 value around a PSD calibration, while keeping the R2 value at 0 Gy constant.

The effects of microbial transglutaminase (MTGase, 0.5%, w/w), dietary fiber (konjac flour, Kjc, 1. 0%, w/w) and ultraviolet irradiation, UV (254 nm, 3300 µW/cm(2) and 40 min), on heat-induced gels from gilthead seabream mince (Sparus aurata) were studied. MTGase addition improved texture, force at rupture increased from 44.3±18.1 to 131.9 ± 56.7 N, and increased pH and water-holding capacity. Moreover, MTGase reduced the elastic modulus E(1) and darkened the gels; protein solubility declined, meaning greater protein aggregation, according to electropherograms. Evidence was found that disulfide bonding has a role in textural improvement by MTGase. Kjc increased the hardness from 15.1±3.1 to 20.6 ± 4.7 N, the elastic modulus and WHC. Kjc itself and not its effect upon proteins may explain the hardening effect. However, gels containing Kjc were less deformable and Kjc reduced the extractable protein in the various selective bond media. UV did not present advantages as a gelation-promoting technology. MTGase and Kjc were texturally advantageous, although without synergies for gel strength: it fell from 56.9±7.1 to 24.6±5.9 N mm as a result of adding Kjc to gels containing MTGase. Nevertheless, the hardening effect of Kjc and MTGase combined surpassed the sum of the individual effects, thereby indicating the existence of a mutual reinforcement of the hardness through MTGase and konjac. Additionally, this study showed that gilthead seabream may be used to produce good quality (concerning texture, color and WHC) heat-induced gels.

Objective: This study focuses on the diverse effects of MRI parameters on image quality using widely available imaging pulse sequences., Methods: A tissue equivalent gel system has been used for Magnetic Resonance Imaging by using Xylenol orange dye with Fricke-Benzoic solution which was formulated by Kelly R.G (1998). The gel system was radiated while using 6MV photons from Varian Clinic Linear Accelerator., Results: The qualitative analysis include the effect of TR and TE on signal nonuniformity according to which the escalating repetition time gives a uniform signal having the average values calculated are 0.76%, 0.83%, 1.43% and 1.89% for the conventional spin echo (CSE), fast spin echo (FSE), gradient recalled echo (GRE) and fluid attenuated inversion recovery (FLAIR) respectively. FLAIR showed contradictory results due to longitudinal relaxation of the signal. This is because, at the longer value of inversion time (TI), the signal from simple fluids is nulled, thus reducing signal intensity., Conclusion: The evaluation of signal uniformity for different pulse sequences demonstrates that the repetition time (TR) affects the signal homogeneity as it maintains image quality for CSE and FSE. However, a careful selection is required for FLAIR due to its sensitive behaviour for image uniformity.

This study compares the dose response of irradiated polymer gel with acrylic and styrofoam housing while applying multi-detector CT (MDCT) and cone-beam CT (CBCT). The dose response for MDCT and CBCT, while using an acrylic phantom is 1.34 and 0.67 DeltaHU Gy(-1), respectively, and becomes 1.54 and 0.84 DeltaHU Gy(-1) while using styrofoam, suggesting styrofoam is the better housing material. While the dose response of MDCT is better than that of CBCT, CBCT is yet a promising 3D dosimetry technique, given its potentially better spatial resolution and sensitive dose interpretation capability., (Copyright 2009 Elsevier Ltd. The aim of this study was to evaluate the reliability of Fricke gel-layer dosimeters for the measurement of in-phantom dose distributions produced by a ((192))Ir brachytherapy source. The doses obtained were compared to measurements performed with thermoluminescent dosimeters and treatment planning calculations. Gel dosimetry has been studied mainly for medical applications. The radiation induced ferric ions concentration can be measured by different techniques to be related with the absorbed dose. Aiming to assess gamma/thermal neutrons dose from research reactors, Fricke gel and alanine gel solutions produced at IPEN using 300 bloom gelatin were mixed with Na(2)B(4)O(7) salt, and the mixtures were irradiated at the beam hole #3 of the IEA-R1 research reactor, (BH#3) adapted to BNCT studies, and the dose-response was evaluated using spectrophotometry technique., (Copyright 2010 Elsevier Ltd. Normoxic polymer gel dosimeters are studied, with the aim of achieving a valid and advantageous method for in-phantom 3D dose determinations. Developments were carried out in the application of such dosimetric material to the method based on dosimeter gel layers that has shown good reliability for absorbed dose imaging in radiotherapy. The technique has been improved, in particular taking care of minimizing the oxygen infiltration into the gel matrix in order to suitably avoid its effect of inhibiting the polymerization process after exposure. A suitable choice of the material of dosimeter walls has brought to achieve good steadiness in time of dosimeter sensitivity and satisfactory results in dose imaging and depth-dose profiling., (Copyright 2009 Elsevier Ltd. We have developed a novel phantom material: a solution of polyvinyl alcohol (PVAL) in ethanol and water, freeze-thawed to produce a solid yet elastically compressible gel. The x-ray attenuation and mechanical properties of these gels are compared with published measurements of breast tissue. Gels with PVAL concentrations from 5 to 20% w/v were produced. The linear x-ray attenuation coefficients of these gels range from 0.76 to 0.86 cm(-1) at 17.5 keV, increasing with PVAL concentration. These values are very similar to the published values of breast tissue at this energy, 0.8-0.9 cm(-1). Under compression cancerous breast tissue is approximately ten times stiffer than healthy breast tissue. The Young's moduli of the gels increase with PVAL concentration. Varying the PVAL concentration from 7.5 to 20% w/v produces gels with Young's moduli from 20 to 220 kPa at 15% strain. These values are characteristic of normal and cancerous breast tissue, respectively.

In this study a new branched methacrylated poly(propylene glycol-co-lactic acid) (PPG-PLA-IEM) and methacrylated cellulose acetate butyrate resin (CAB-IEM) were synthesized. Hydrogels with various amounts of PPG-PLA-IEM and CAB-IEM (25, 50 and 75 wt% IEM modified) were prepared by photopolymerization. Collagen tethered PEG-monoacrylate (PEGMA-collagen) was prepared and introduced as a bioactive moiety to modify the hydrogel in order to enhance cell affinity. In vitro attachment and growth of 3T3 mouse fibroblasts and human umbilical vein endothelial cells (HUVEC) on the hydrogels with and without collagen were also investigated. It was observed that, the collagen improves the cell adhesion onto the hydrogel surface. With the increasing amount of collagen, cell viability increased by 28% for ECV304 (P < 0.05) and 30% for 3T3 (P < 0.05).
