Southern African savanna rangelands are facing a widespread degradation pattern called bush encroachment. This is associated with implications for various aspects of the water cycle and in particular canopy transpiration. At the individual‐tree scale, it is estimated by scaling sap‐flux density by sapwood area. However, the direct measurement of sapwood area is impracticable at landscape scale and general allometric equations of the West‐Brown‐Enquist (WBE) model relating sapwood area to primary size measures seem to fail for some species and climates. Therefore, we conducted intensive field measurements to establish species‐specific allometric relationships between sapwood area and sizes (stem diameter, crown area) in six dominant shrub species involved in bush encroachment in Namibia (Colophospermum mopane, Senegalia mellifera, Vachellia reficiens, Dichrostachys cinerea, Vachellia nebrownii, Catophractes alexandri). We found strong allometric relationships between sapwood area and stem diameter as well as between sapwood area and crown area for all six species. These relations are largely in line with the WBE theory but still provide estimates that are more accurate. Only in D. cinerea, the sapwood area was significantly smaller than predicted by the WBE theory, which might be caused by a larger need for stabilizing heartwood. Our results are useful to estimate water loss via transpiration at a large scale using remote sensing techniques and can promote our understanding of the ecohydrological conditions that drive species‐specific bush encroachment in savannas. Aims Plant–plant interactions, being positive or negative, are recognized to be key factors in structuring plant communities. However, it is thought that root competition may be less important than shoot competition due to greater size symmetry belowground. Because direct experimental tests on the importance of root competition are scarce, we aim at elucidating whether root competition may have direct or indirect effects on community structure. Indirect effects may occur by altering the overall size asymmetry of competition through root–shoot competitive interactions. Methods We used a phytometer approach to examine the effects of root, shoot and total competition intensity and importance on evenness of experimental plant communities. Thereby two different phytometer species, Festuca brevipila and Dianthus carthusianorum, were grown in small communities of six grassland species over three levels of light and water availability, interacting with neighbouring shoots, roots, both or not at all. Important Findings We found variation in community evenness to be best explained if root and shoot (but not total) competition were considered. However, the effects were species specific: in Dianthus communities increasing root competition increased plant community evenness, while in Festuca communities shoot competition was the driving force of this evenness response. Competition intensities were influenced by environmental conditions in Dianthus, but not in Festuca phytometer plants. While we found no evidence for root–shoot interactions for neither phytometer species root competition in Dianthus communities led to increased allocation to shoots, thereby increasing the potential ability to perform in size-asymmetric competition for light. Fire and grazing shape biodiversity in savannah landscapes. In land use management, knowing the effects of fire and grazing on biodiversity are important in order to ensure environmental sustainability. Beetles specifically are indicators of the biodiversity response to fire and grazing.A grazing exclusion and burning experiment in a split‐plot design was used in order to investigate the interacting effects of fire and wildlife grazing on biomass, diversity, and species composition of darkling beetles (Coleoptera, Tenebrionidae) over time after fire.Darkling beetle species richness and diversity were responding in a three‐way‐interaction to fire, grazing, and time after fire, whereby biomass of darkling beetles remained unaffected and species compositional changes were attributed to seasonal changes of time only. Fire on ungrazed plots had a negative effect on species diversity and richness 2 weeks and 6 months post fire, whereas fire on grazed plots had no impact on species diversity and richness. Grazing only lowered species diversity and richness 6 months after fire treatments.Results suggest that grazing overrides the effects of fire and that the similar effects caused by fire and grazing are due to niche and assemblage simplification of the habitat. Shrub encroachment in semi‐arid savannas is induced by interacting effects of climate, fire suppression, and unsustainable livestock farming; it carries a severe risk of land degradation and strongly influences natural communities that provide key ecosystem functions. However, species‐specific effects of shrub cover on many animal groups that act as indicators of degradation remain largely unknown. We analysed the consequences of shrub encroachment for ground‐dwelling beetles in a semi‐arid Namibian savanna rangeland, where beetles and vegetation were recorded along a shrub cover gradient (<1% to >30%). Focusing on species niche breadths and optima, we identified two crucial shrub cover thresholds (2.9% and 10.0%), corresponding to major changes in the beetle communities with implications for savanna ecosystem functioning. Niche optima of most species were between the first and second thresholds; beyond the second threshold, saprophagous, coprophagous, and rare predatory beetles declined in numbers and diversity. This is problematic because beetles provide important ecosystem functions, such as decomposition and nutrient cycling. However, we also found that certain species were adapted to high shrub cover, thus providing examples of niche differentiation. Despite the predominantly negative effects of heavy shrub encroachment on beetle communities, shrubs in their early life stages apparently provide essential structures, which enhance habitat quality for ground‐dwelling beetles. Our results demonstrate that shrub encroachment can have mixed effects on ground‐dwelling beetle communities and hence on savanna ecosystem functioning. We, therefore, conclude that rangeland management and restoration should consider the complex trade‐offs between species‐specific effects and the level of encroachment for sustainable land use. Topics of discussion included the presentation of digitized archival materials on the Internet, the selection of archival materials for digitization and preservation, and the debate whether archival processing and the creation of online finding aids should take precedence over mass digitization.

The neurofibromatosis type 2 gene product merlin is known to provoke gliogenic tumors as a result of its mutagenic loss. Merlin's physiological anti-mitogenic function makes it unique among its ezrin-radixin-moesin (ERM) family members. Although ERM proteins and merlin are known to be expressed in glial cells of the peripheral nervous system and CNS, the neuronal expression pattern and function of merlin have been less well investigated. We report here expression of merlin in developing and mature neurons of the murine CNS. Within cerebellar Purkinje cells (PCs), merlin was localized in the soma, sprouting dendrites and axons. Merlin expression in PCs was high during the period of initial dendrite regression and declined during later phases of dendrite elongation. Consistently, merlin expression in vivo was increased in Engrailed-2-overexpressing PCs, which are characterized by a reduced dendritic extension. Furthermore, overexpression of merlin in dissociated cerebellar cultures and in neurogenic P19 cells caused a significant decline in neurite outgrowth, while, conversely, inhibition of merlin expression increased process formation. This effect was dependent on phosphorylation of serine 518 and involved the inactivation of the growth-promoting GTPase Rac. We thus provide evidence that merlin plays a pivotal role in controlling the neuronal wiring in the developing CNS. [ABSTRACT FROM AUTHOR]

Receptor tyrosine kinases participate in several signaling pathways through small G proteins such as Ras (rat sarcoma). An important component in the activation of these G proteins is Son of sevenless (SOS), which catalyzes the nucleotide exchange on Ras. For optimal activity, a second Ras molecule acts as an allosteric activator by binding to a second Ras-binding site within SOS. This allosteric Ras-binding site is blocked by autoinhibitory domains of SOS. We have reported recently that Ras activation also requires the actin-binding proteins ezrin, radixin, and moesin. Here we report the mechanism by which ezrin modulates SOS activity and thereby Ras activation. Active ezrin enhances Ras/MAPK signaling and interacts with both SOS and Ras in vivo and in vitro. Moreover, in vitro kinetic assays with recombinant proteins show that ezrin also is important for the activity of SOS itself. Ezrin interacts with GDP-Ras and with the Dbl homology (DH)/pleckstrin homology (PH) domains of SOS, bringing GDP-Ras to the proximity of the allosteric site of SOS. These actions of ezrin are antagonized by the neurofibromatosis type 2 tumor-suppressor protein merlin. We propose an additional essential step in SOS/Ras control that is relevant for human cancer as well as all physiological processes involving Ras.

Spinal cord injury (SCI) results in a pathophysiology characterized by multiple locomotor and sensory deficits, resulting in altered nociception and hyperalgesia. SCI triggers an early and prolonged inflammatory response, with increased interleukin-1beta levels. Transient changes are observed in subunit populations of the transcription factor nuclear factor-kappaB (NF-kappaB). There were decreases in neuronal c-Rel levels and inverse increases in p65 and p50 levels. There were no changes in neuronal p52 or RelB subunits after SCI at any time point tested. Similarly, SCI had no effect on oligodendroglial levels of any NF-kappaB subunit. There were significant early increases in COX-2 and inducible nitric oxide synthase mRNA and protein levels after SCI. We used synthetic double-stranded "decoy" deoxyoligonucleotides containing selective NF-kappaB protein dimer binding consensus sequences. Decoys targeting the p65/p50 binding site on the COX-2 promoter decreased SCI-induced cell losses, NF-kappaB p65/p50 DNA-binding activity, and COX-2 and iNOS protein levels. NF-kappaB p65/p50 targeted decoys improved early locomotor recovery after moderate but not severe SCI, yet ameliorated SCI-induced hypersensitization after both moderate and severe SCI. To determine whether changes in GABA activity played a role in decreased hypersensitivity after SCI and p65/p50 targeted decoy, we counted gamma-aminobutyric acid (GABA)-containing neurons in laminae 1-3.
