Abstract

Scientific communication is facilitated by a data-driven, scientifically sound taxonomy that considers the end-user¿s needs and established successful practice. In 2013, the Fusarium community voiced near unanimous support for a concept of Fusarium that represented a clade comprising all agriculturally and clinically important Fusarium species, including the F. solani species complex (FSSC). Subsequently, this concept was challenged in 2015 by one research group who proposed dividing the genus Fusarium into seven genera, including the FSSC described as members of the genus Neocosmospora, with subsequent justification in 2018 based on claims that the 2013 concept of Fusarium is polyphyletic. Here, we test this claim and provide a phylogeny based on exonic nucleotide sequences of 19 orthologous protein-coding genes that strongly support the monophyly of Fusarium including the FSSC. We reassert the practical and scientific argument in support of a genus Fusarium that includes the FSSC and several other basal lineages, consistent with the longstanding use of this name among plant pathologists, medical mycologists, quarantine officials, regulatory agencies, students, and researchers with a stake in its taxonomy. In recognition of this monophyly, 40 species described as genus Neocosmospora were recombined in genus Fusarium, and nine others were renamed Fusarium. Here the global Fusarium community voices strong support for the inclusion of the FSSC in Fusarium, as it remains the best scientific, nomenclatural, and practical taxonomic option available

Abstract

Fusaria of the Liseola section were isolated from 1996 to 1998 corn survey samples (162, 104 and 111 samples respectively) and tested for mating. The incidence of isolates belonging to mating population A (F. verticillioides) ranged from 70.2 percent to 89.5 percent. In the 1996 to 1998 samples, from 63 percent to 91 percent, were contaminated with fumonisins, from 0.1 microg/g to 33.3 microg/g. Pearl millet samples were collected from 1996 to 1998. 119 isolates were paired with mating population A (F. verticillioides), D (F. proliferatum) and F (F. thapsinum) tester strains. No successful crosses were obtained with mating population F, 50.4 percent were fertile with mating population A and 10.1 percent crossed with mating population D. 39.5 percent did not form any perithecia. We isolated Fusarium pseudonygamai from Pearl millet for the first time in the United States, and believe that many of the isolates that were infertile belong to this species. This finding was confirmed by DNA sequence comparisons with known isolates from Nigeria. Fumonisins FB(1) and FB(2) were not detected in any of the 81 analyzed grain samples.

Abstract

SummaryTo assess interobserver variability of venography for screening for postoperative deep venous thrombosis (DVT), we used 185 bilateral ascending contrast venograms (366 lower limbs) which were performed using the long-leg film technique, in the frame of a trial of the efficacy of two low-molecular-weight-heparin fractions (initial evaluation). These venograms were submitted in a multicenter setting to three further readers who performed a serial scoring as DVT, no DVT or non-evaluable. DVTs were diagnosed in 78, 55 and 59/366 limbs (initial evaluation: 58). The proximal locations of DVTs were 16, 9 and 16 (initial evaluation: 15) and the non evaluable limbs 3, 5 and 18 (initial evaluation: 0). Apparent pairwise agreement between the three readers ranged from 87 to 90% (true coefficient of agreement Kappa 0.63-0.70). It ranged from 89 to 93% (Kappa 0.63-0.74) for the comparison between the readers and the initial evaluation. This considerable degree of disagreement among experienced readers should be taken into account in calculating sample sizes in prospective thromboprophylactic studies. Indeed, one of the two prophylactic regimens proved to be significantly (p = 0.012, p = 0.031, p = 0.049) or non-significantly (p = 0.073) superior to the other one depending upon the reading of venograms.

This study was designed to identify and compare the Fusariumspecies of the Gibberella fujikuroicomplex on pearl millet (Pennisetum glaucum(L.) R. Br) and corn (Zea maysL.) crops grown in southern Georgia, and to determine their influence on potential fumonisin production. Pearl millet and corn samples were collected in Georgia in 1996, 1997 and 1998. Three percent of the pearl millet seeds had fungi similar to the Fusariumspecies of the G. fujikuroispecies complex. One hundred and nineteen representative isolates visually similar to the G. fujikuroispecies complex from pearl millet were paired with mating population A (Fusarium verticillioides(Sacc.) Nirenberg), mating population D (F. proliferatum(Matsushima) Nirenberg) and mating population F (F. thapsinum(Klittich, Leslie, Nelson and Marasas) tester strains. Successful crosses were obtained with 50.4%, 10.1% and 0.0% of these isolates with the A, D and F tester strains, while 39.5 of the isolates did not form perithecia with any tester strains. Two of the typical infertile isolates were characterized by DNA sequence comparisons and were identified as Fusarium pseudonygamai(Nirenberg and O’Donnell), which is the first known isolation of this species in the United States. Based on the pattern of cross-compatibility, conidiogenesis, colony characteristics and media pigmentation, a majority of the infertile isolates belong to this species. Fumonisins FB1and FB2were not detected in any of the 81 pearl millet samples analyzed. The species of the G. fujikuroispecies complex were dominant in corn and were isolated from 84%, 74% and 65% of the seed in 1996, 1997 and 1998, respectively. Representative species of the G. fujikuroispecies complex were isolated from 1996 to 1998 Georgia corn survey (162, 104 and 111 isolates, respectively) and tested for mating compatibility.
