The Thermodynamics of Advanced Fuels – International Database (TAF-ID) was developed using the Calphad method to provide a computational tool to perform thermodynamic calculations on nuclear fuel materials under normal and off-normal conditions. Different kinds of fuels are considered: oxide, metallic, carbide and nitride fuels. Many fission products are introduced as well as structural materials (e.g., zirconium, steel, concrete, SiC) and absorbers (e.g., B4C), in order to investigate the thermochemistry of irradiated fuels and to predict their chemical interaction with the surrounding materials. The approach to develop the database and the models implemented in the database are described. Examples of models for key chemical systems are presented. Finally, a few examples of application calculations on severe accidents with UO2 fuels, irradiated fuel chemistry of MOX and metallic fuels and metallic fuel/cladding interaction show how this tool can be used. To validate the database, the calculations are compared to the available experimental data. A good agreement is obtained which gives confidence in the maturity degree and quality of the TAF-ID database. The working version is only accessible to the participants of the TAF-ID project (Canada, France, Japan, the Netherlands, Republic of Korea, United Kingdom, USA). A public version is accessible by all the NEA countries. The current version contains models on the Am–Fe, Am–Np, Am-O-Pu, Am–U, Am–Zr, C–O–U-Pu, Cr–U, Np–U, Np–Zr, O–U–Zr, Re–U, Ru–U, Si–U, Ti–U, U-Pu-Zr, U–W systems. Im Rahmen der Arbeit wurden die Einflüsse proteolytischer Enzyme, welche von der Hefe ausgehen, auf den Bierschaum untersucht. Es konnte gezeigt werden, dass die Hefe großen Einfluss auf die Schaumhaltbarkeit des Bieres hat. Obwohl die Bestimmung des Bierschaums ein sehr schwierig zu kontrollierendes Qualitätsmerkmal darstellt, konnten Beziehungen von Hefevitalität und Bierschaum dargestellt werden. Weiterhin wurde eine neue angepasste Methode zur Bestimmung der Proteinase-A aktivität entwickelt und der Einfluss der Proteinase-A auf schaumaktive Proteine mittels SDS-PAGE untersucht., At first, the influence of proteolytic enzymes from brewers yeast on beerfoam was elucidated. The yeast shows a large influence on foamstability. Although the measurement of foamstability is a difficult quality characteristic, relations on yeastvitality and beerfoam could be shown. Der Blattkeim von Gerstenmalz ist reich an Lipiden und lipidabbauenden Enzymen. Da Lipide und deren Abbauprodukte einen negativen Einfluss auf die Geschmacksstabilität und die Schaumhaltbarkeit des Bieres haben, hat sich die Arbeit die Aufgabe gestellt, den vermuteten negativen Einfluss des Blattkeims auf die Bierqualität zu klären und technologische Maßnahmen aufzuzeigen, mit denen die Nachteile des Blattkeims minimiert werden können. In einer Reihe von Brauversuchen mit steigendem Blattkeimgehalt zur Schüttung konnte zweifelsfrei ein nachteiliger Einfluss des Blattkeims auf den Geschmack und die Geschmacksstabilität des Bieres herausgestellt werden. Technologische Möglichkeiten, mit denen die Nachteile eines forcierten Lipidabbaus minimiert werden können, wie Rohstoffauswahl, Malzlagerung, Schrotung, Schrotlagerung, (Ein-)Maischen, werden beschrieben und bewertet., The acrospire of malted barley is rich in lipids and lipid degrading enzymes. Since lipids and lipid-oxidation products are harmful in regard to flavour stability and foam stability of beer this work aimed to clarify the negative influence of the acrospire on the beer quality and to point out technological possibilities to minimise the detriments of the acrospire. In a series of brewing trials the addition of acrospire to the throw showed to be doubtless negative for the beer flavour and the flavour stability. Technological possibilities that enables a minimization of lipid-oxidation reactions in the brewing process, like the choice of raw materials, malt storage, milling, grist storage, different mashing(-in) systems, are described and evaluated.

Obwohl die Hefe Saccharomyces cerevisiae sich in den letzten 50 Jahren als Modell für alle Genom- und Proteomanalysen durchgesetzt hat und der einzige vollständig „entschlüsselte“ Organismus ist, so gibt es bis jetzt keine Methode, die eine aussagekräftige und exakte Beurteilung des physiologischen Zustandes einer Bierhefe in allen Einsatzstadien der Bierherstellung ermöglicht. Die Vitalität einer Hefe wird heutzutage meist anhand von Globalbestimmungsmethoden wie Zellvermehrungsrate, Gärgeschwindigkeit, pH-Sturz u.a. beurteilt, die allerdings keine Aussagen über die Vorgänge innerhalb einer Zelle machen. Das Ansetzen von Probegärungen liefert die Ergebnisse frühestens nach 3 Tagen und bis dahin findet sich die Hefe bereits im Einsatz, so dass nachträgliche Korrekturen nicht mehr möglich sind und es wird in Kauf genommen, dass eine gärschwache Hefe zum Einsatz kommt. Weitere Nachteile der heutigen Analysen sind die hohen Kosten und der Bedarf an entsprechend geschultes Personal, um die Geräte zu bedienen oder um den zusätzlichen Arbeitsaufwand zu bewältigen. Mit Hilfe einfacher enzymkinetischer Methoden wurde versucht, einen Einblick in die Vorgänge innerhalb der Zelle zu bekommen. Das Hauptaugenmerk der Untersuchungen lag in den Aktivitäten der Enzyme Maltase, PDH, PDC und ADH, die alle eine Schlüsselfunktion im Metabolismus der Hefe besitzen. Da die bisherigen Studien dieser Enzyme sich auf Laborhefestämme beschränkte, mussten zunächst Erfahrungswerte mit den Aktivitäten der Hefe in allen Stadien der Bierherstellung gewonnen werden. Dazu wurden verschiedene Fermentationen im Labormaßstab am Institut mit der Messung von Enzymaktivitäten begleitet und mit Untersuchungen in einer Brauerei ergänzt. Mit Hilfe einer erstellten Formel zur Berechnung der Gärleistung war die Möglichkeit zum Vergleich der verschiedenen Fermentationen gegeben. With the help of uncomplicated methods for measuring enzyme kinetics, we attempted to gain insight into the inner workings of the cell. The primary focus of this analysis was the enzyme activity of maltase (MAL), pyruvate dehydrogenase (PDH), pyruvate decarboxylase (PDC) and alcohol dehydrogenase (ADH), all of which have a key function in yeast metabolism. Due to the fact that past studies of these enzymes were conducted with laboratory strains, our first task was to gather data on yeast enzyme activity during all stages of beer production. For this purpose, a number of laboratory-scale fermentations were performed at the institute and the enzyme activity was measured and further supplemented with analysis results from a brewery. In dieser Arbeit wurden eine bestimmte Betriebshefe und zahlreiche Vergleichshefen hinsichtlich ihrer physiologischen und genetischen Eigenschaften untersucht. Zur physiologischen Charakterisierung wurden Wachstumsparameter, das Bruchbildungsverhalten, die Verwertung von Kohlehydraten sowie die Bildung von Gärungsnebenprodukten in Modellgärungen eingesetzt. Bei den Gärungsnebenprodukten zeigten sich zum Teil große Abweichungen, die für einen Hefestamm charakteristisch waren. Die genetische Typisierung umfaßte die Methoden der Pulsfeld-Gelelektrophorese (PFGE), Restriktionsfragment-Längenpolymorphismen (RFLP), Sequenzierungen, Mikrosatelliten-Längenpolymorphismen (MLP) und Amplified Fragment Length Polymorphism (AFLP). Für die Stammdifferenzierung der Brauhefen war die AFLP die am besten geeignete Methode, da hiermit praktisch alle untersuchten Hefestämme unterschieden werden konnten., In this work a particular brewery yeast strain and a number of strains for comparison were analyzed for their physiological and genetical properties. For a physiological characterization several growth parameters, flocculation, utilization of carbohydrates and the production of fermentation byproducts were analyzed in model fermentations. The fermentation byproduct analysis showed great differences between some of the yeast strains. The genetical typing was performed by the methods of Pulsed Field Gel Electrophoresis (PFGE), Restriction Fragment Length Polymorphisms (RFLP), sequencing, Microsatellite Length Polymorphisms (MLP) and Amplified Fragment Length Polymorphisms (AFLP), respectively. The AFLP method was found to be the best suitable for strain differentiation and was able to distinguish practically all of the examined yeast strains.
