Background: Dexrazoxane has been studied for its ability to prevent anthracycline-induced cardiac dysfunction (AICD) in several trials but its use in clinical practice remains limited. This is related to the low to moderate quality of the generated evidence, safety concerns and restricted prescribing indications. Additional randomized trials are needed before this drug can be routinely integrated into cardio-oncology clinical practice., Objectives: To describe the rationale and design of the HOVON 170 DLBCL - ANTICIPATE trial. This trial aims to establish the efficacy and safety of dexrazoxane for the primary prevention of AICD in patients diagnosed with Diffuse Large B-Cell Lymphoma (DLBCL) treated with six cycles R-CHOP 21 chemo-immunotherapy., Methods: This is a multicenter, parallel-group, open-label, phase III trial, randomizing 324 patients between either no cardioprotective treatment or dexrazoxane from the first R-CHOP cycle. The primary and co-primary endpoints are the incidence of AICD within 12 months of registration and the percentage of patients with complete metabolic remission at the end-of-treatment PET-CT respectively. The trial is registered at the EU Clinical Trials Register (EU-CT number 2023-505377-32) and ClinicalTrials.gov (NCT06220032)., Results: The medical research ethics committee approved the trial in May 2024. Recruitment has started in September 2024 and is expected to last for three years., Conclusions: This trial is poised to contribute crucial evidence concerning the efficacy and safety on the use of dexrazoxane in the primary prevention of AICD. Social media platform's lack of control over its content made way to the fundamental problem of misinformation. As users struggle with determining the truth, social media platforms should strive to empower users to make more accurate credibility judgements. A good starting point is a more accurate perception of the credibility of the message's source. Two pre-registered online experiments (N = 525;N = 590) were conducted to investigate how verified source information affects perceptions of Tweets (study 1) and generic social media posts (study 2). In both studies, participants reviewed posts by an unknown author and rated source and message credibility, as well as likelihood of sharing. Posts varied by the information provided about the account holder: (1) none, (2) the popular method of verified source identity, or (3) verified credential of the account holder (e.g., employer, role), a novel approach. The credential was either relevant to the content of the post or not. Study 1 presented the credential as a badge, whereas study 2 included the credential as both a badge and a signature. During an initial intuitive response, the effects of these cues were generally unpredictable. Yet, after explanation how to interpret the different source cues, two prevalent reasoning errors surfaced. First, participants conflated source authenticity and message credibility. Second, messages from sources with a verified credential were perceived as more credible, regardless of whether this credential was context relevant (i.e., virtual lab coat effect). These reasoning errors are particularly concerning in the context of misinformation. In sum, credential verification as tested in this paper seems ineffective in empowering users to make more accurate credibility judgements. Unlabelled: Recent studies suggest that the symptoms of chronic non-bacterial prostatitis (CP) or Chronic Pelvic Pain Syndrome (CPPS) may be due to or associated with pelvic floor muscle dysfunction. Therapies aimed to improve relaxation and proper use of the pelvic floor muscles such as biofeedback physical therapy and pelvic floor re-education are expected to give symptom improvement. The objective of this study was to evaluate the effect of biofeedback physical therapy on the symptoms of men with CPPS., Materials and Methods: Between March 2000 to March 2004, 33 consecutive men were diagnosed with CP/CPPS based on history including the NIH-CPSI questionnaire and physical examination including pelvic floor muscle tonus, urinalysis, uroflowmetry with residual urine measurement and transrectal ultrasonography of the prostate. All patients participated in a pelvic floor biofeedback re-educating program. A rectal EMG probe was used to measure resting tone of the pelvic floor muscles and was helpful for instruction pelvic floor muscles contraction and relaxation., Results: Two of the 33 men dropped out. In the remaining 31 men, mean age 43.9 years (range 23-70), the mean total Chronic Prostatitis Symptom Index (NIH-CPSI) changed from 23.6 (range 11-34) at baseline to 11.4 (range 1-25) after treatment (p<0.001). The mean value of the pelvic floor muscle tonus was 4.9 at diagnosis (range 2.0-10.0) and decreased to 1.7 (range 0.5-2.8) after treatment (p<0.001)., Conclusions: Our study clearly demonstrates a significant effect of biofeedback physical therapy and pelvic floor re-education for CP/CPPS patients, leading to a significant improvement of the symptom score. The correlation between the pelvic muscle tonus results with NIH-CPSI score is highly suggestive that the pelvic floor plays an important role in the pathophysiology of CP/CPPS.
