Background: Prostate cancer (PCa) is a malignant tumor of the male reproductive system, and its incidence has increased significantly in recent years. This study aimed to further identify candidate biomarkers with prognostic and diagnostic significance by integrating gene expression and DNA methylation data from PCa patients through association analysis., Material and Methods: To this end, this paper proposes a sparse partial least squares regression algorithm based on hypergraph regularization (HR-SPLS) by integrating and clustering two kinds of data. Next, module 2, with the most significant weight, was selected for further analysis according to the weight of each module related to DNA methylation and mRNAs. Based on the DNA methylation sites in module 2, this paper uses multiple machine learning methods to construct a PCa diagnosis-related model of 10-DNA methylation sites., Results: The results of Receiver Operating Characteristic (ROC) analysis showed that the DNA methylation-related diagnostic model we constructed could diagnose PCa patients with high accuracy. Subsequently, based on the mRNAs in module 2, we constructed a prognostic model for 7-mRNAs (MYH11, ACTG2, DDR2, CDC42EP3, MARCKSL1, LMOD1, and MYLK) using multivariate Cox regression analysis. The prognostic model could predict the disease free survival of PCa patients with moderate to high accuracy (area under the curve (AUC) =0.761). In addition, Gene Set EnrichmentAnalysis (GSEA) and immune analysis indicated that the prognosis of patients in the risk group might be related to immune cell infiltration., Conclusions: Our findings may provide new methods and insights for identifying disease-related biomarkers by integrating DNA methylation and gene expression data.

Clear cell renal cell carcinoma (ccRCC) is the most aggressive and deadly cancer of the urinary system and is regulated by multiple signaling pathways. However, the specific molecular mechanisms underlying ccRCC have not been fully studied or demonstrated. This study aimed to elucidate the function of lysosomal-associated transmembrane protein 5 (LAPTM5) in ccRCC cell lines and animal models and determine the potential underlying mechanisms. Our results demonstrated that LAPTM5 expression in patients with ccRCC was significantly higher in the tumor group than that in the adjacent nontumor group. Moreover, LAPTM5 promoted proliferation, migration, and invasion of ccRCC cells through the gain and loss of the function of LAPTM5 in 786-0 and Caki-1 cell lines. Similar results regarding LAPTM5 overexpression were obtained in BALB/c nude mice. In addition, LAPTM5 activated the Jun N-terminal kinase (JNK)/p38 signaling cascade by interacting with Ras-related C3 botulinum toxin substrate 1 (RAC1). Treatment with an RAC1 inhibitor eliminated the effects of LAPTM5 in ccRCC. Purpose: To evaluate the influence of removable thermoplastic appliances and fixed appliances on periodontal status., Methods: Cochrane library, PubMed, EMBASE, CBM, CNKI, VIP, Wanfang Database were searched from the date of inception to 2017.10.16，to find clinical trials about removable thermoplastic appliances and fixed appliances on periodontal status. The quality of the included studies was assessed by two reviewers independently, and meta analysis was performed using RevMan5.3., Results: Totally 11 studies including 543 patients were identified. The results of meta analysis showed there were significant difference in PLI and SPD at 3 months and PLI and GI at 6 months between removable thermoplastic appliances and fixed appliances, and no significant difference was observed in GI at 3 months and SPD at 6 months between the two groups., Conclusions: In the early orthodontic treatment, removable thermoplastic appliances has better effect than the fixed appliance on periodontal health.

Background: Chronic muscle injury is characteristics of fatty infiltration and fibrosis. Recently, fibro/adipogenic progenitors (FAPs) were found to be indispensable for muscular regeneration while were also responsible for fibrosis and fatty infiltration in muscle injury. Many myokines have been proven to regulate the adipose or cell proliferation. Because the fate of FAPs is largely dependent on microenvironment and the regulation of myokines on FAPs is still unclear. We screened the potential myokines and found Interleukin-15 (IL-15) may regulate the fatty infiltration in muscle injury. In this study, we investigated how IL-15 regulated FAPs in muscle injury and the effect on muscle regeneration., Methods: Cell proliferation assay, western blots, qRT-PCR, immunohistochemistry, flow cytometric analysis were performed to investigate the effect of IL-15 on proliferation and adipogensis of FAPs. Acute muscle injury was induced by injection of glycerol or cardiotoxin to analyze how IL-15 effected on FAPs in vivo and its function on fatty infiltration or muscle regeneration., Results: We identified that the expression of IL-15 in injured muscle was negatively associated with fatty infiltration. IL-15 can stimulate the proliferation of FAPs and prevent the adipogenesis of FAPs in vitro and in vivo. The growth of FAPs caused by IL-15 was mediated through JAK-STAT pathway. In addition, desert hedgehog pathway may participate in IL-15 inhibiting adipogenesis of FAPs. Our study showed IL-15 can cause the fibrosis after muscle damage and promote the myofiber regeneration. Finally, the expression of IL-15 was positively associated with severity of fibrosis and number of FAPs in patients with chronic rotator cuff tear., Conclusions: These findings supported the potential role of IL-15 as a modulator on fate of FAPs in injured muscle and as a novel therapy for chronic muscle injury.

Objective: To evaluate the transverse displacement, stress distribution and tendency of change in tooth, alveolar bone and mid-palatal suture using three kinds of rapid maxillary expansion methods. Methods: Cone-beam CT image data was obtained by scanning skulls of a volunteer. Three-dimensional models of maxillary complex were re-established using Mimics and Geomagic Studio and models of Hyrax expander, Haas expander and miniscrew-assisted rapid palatal expander (MARPE) were established using ANSYS Workbench. Stress distribution, displacement and tendency of change in tooth, alveolar bone and mid-palatal suture were evaluated. Results: Hyrax expander brought 0.105 mm lateral displacement of crown, 0.022 mm mid-palatal suture width increase, wedge opening and clockwise rotation tendency of maxilla. Haas expander created uniform stress distribution, 0.216 mm lateral displacement of crown, and 0.031 mm mid-palatal suture width increase. In MARPE model, the lateral displacement of crown was 0.267 mm, and mid-palatal suture width increased 0.315 mm. The maximum of mid-palatal suture expansion and stress distribution appeared in the middle region, and maxilla had tendency of counterclockwise rotation. Conclusions: The lateral changes of teeth and bones brought by MARPE were the most significant. Haas expander had some advantages in comparison with Hyrax.

Extracellular matrix (ECM) is a system used to model the design of biomaterial matrices for tissue regeneration. Various biomaterial systems have been developed to mimic the composition or microstructure of the ECM. However, emulating multiple facets of the ECM in these systems remains a challenge. Here, a new strategy is reported which addresses this need by using silk fibroin and chitosan (CS) nanocomposite materials. Silk fibroin was first assembled into ECM-mimetic nanofibers in water and then blended with CS to introduce the nanostructural cues. Then the ratios of silk fibroin and CS were optimized to imitate the protein and glycosaminoglycan compositions. These biomaterial scaffolds had suitable compositions, hierarchical nano-to-micro structures, and appropriate mechanical properties to promote cell proliferation in vitro , and vascularization and tissue regeneration in vivo . Compared to previous silk-based scaffolds, these scaffolds achieved improvements in biocompatibility, suggesting promising applications in the future in tissue regeneration.

Objective: To investigate the stress distribution on the maxillary anterior teeth retracted with sliding mechanics and micro-implant anchorage using different retraction hook heights and positions., Methods: DICOM image data including maxilla and upper teeth were obtained with cone-beam CT. The three-dimensional finite element model was constructed using Mimics software. Brackets and archwire model were constructed using Creo software. The models were instantiated using Pro/Engineer software. Abaqus software was used to simulate the sliding mechanics by loading 2 N force on 0, 2, 4, 6, 8, 10 mm retraction hooks and three different positions, repectively. Rotation of the occlusal plane, the initial displacement and stress distribution of teeth were analyzed., Results: Lingual rotation of maxillary central incisor(0.021°), gingival movement of the maxillary first molar(0.005 mm), and clockwise rotation of the maxillary occlusal plane(0.012°) were observed when the force application point located at the archwire level (0 mm). In contrast, 0.235° labial rotation of the maxillary central incisor, 0.015 mm occlusal movement of the maxillary first molar, and 0.075° anti-clockwise rotation of the maxillary occlusal plane were observed when the force application point located at the higher level(10 mm retraction hook). The more the force application point was located posteriorly at the archwire level, the less lingual rotation of the maxillary central incisor and the more buccal displacement of maxillary first molar was observed., Conclusions: Maxillary anterior tooth rotation and retraction, vertical displacement of posterior segment, and rotation of the occlusal plane could be controlled by adjusting the height and position of the retraction hook in space closure using miniscrew and sliding mechanics.

Background: Military operation personnel often suffer from sleep difficulty because of their work requirements. In this study, we investigated the efficacy of zaleplon at two doses to induce afternoon-sleep under noise interference and its effects on psychomotor performance and vestibular function; we subsequently established the optimal dosage regimen for military operation personnel., Methods: Twenty-two healthy young male volunteers were recruited for the study. Eight subjects took 10 mg or 15 mg of zaleplon and placebo alternately and then were exposed to noise. Changes in polysomnography (PSG) indices, including sleep latency (SL), sleep efficiency (SE) and sleep structure, were recorded after drug administration. After awakening, the volunteers' subjective judgments of sleep quality and sleepiness were measured. Eight volunteers underwent 3 psychomotor performance tests at a one-week interval, and the psychomotor performance tests were conducted before and after taking zaleplon and placebo. Six volunteers participated in the vestibular function test session, and parameters, including optokinetic nystagmus (OKN), vestibular ocular reflex (VOR), visual-vestibular ocular reflex (VVOR) and vestibular ocular reflex fixation suppression (VOR-Fix), were detected by the same experimental design as described above. The data of sleep observations were subjected to one-way variance analysis., Results: Compared with the placebo group, SL was shortened significantly, and the scores of subjective sleep quality and sleep depth were clearly increased in the zaleplon 10 mg group (P < 0.05). Moreover, the SE and the percent of REM (rapid eye movement) sleep were increased remarkably in the zaleplon 15 mg group (P < 0.01). Furthermore, the SE, percent of REM sleep and scores of subjective sleep depth in the zaleplon 15 mg group were significantly higher than in the zaleplon 10 mg group (P < 0.05). The psychomotor performance did not change significantly after ingestion of 10 mg or 15 mg of zaleplon, whereas the OKN and VOR gains were lower in the two dose groups of zaleplon (P < 0.05) and restored to normal 3 h after drug ingestion., Conclusion: Zaleplon is an ideal hypnotic for military personnel, and its hypnotic efficiency is dose-related under noise interference; a 15 mg dose of zaleplon could provide significantly better sleep than a 10 mg dose of zaleplon.

In this paper the strong electrochemiluminescence (ECL) nanoparticles have been prepared based on the anionic polyelectrolyte sodium polyacrylate (PAA)-ECL enhancement for Ru(bpy)3(2+), which were loaded by the carrier of SiO2 nanoparticle. There were two kinds of Ru(bpy)3(2+) for the as-prepared nanoparticles, the doped one and the exchanged one. The former was loaded inside the ECL nanoparticles by doping, in a form of ion-pair macromolecules PAA-Ru(bpy)3(2+). The corresponding ECL was enhanced about 2 times owing to the doping increase of Ru(bpy)3(2+). The latter was loaded on the PAA-doped Nafion membrane by ion exchange. The corresponding ECL was enhanced about 3 times owing to the ion-exchanging increase of Ru(bpy)3(2+). At the same time, ECL intensity of the doped-inside Ru(bpy)3(2+) was further enhanced 13 times because polyelectrolyte PAA in the doped membrane could obviously enhance electron transfer between the doped Ru(bpy)3(2+) and the working electrode. Furthermore, based on hydrophobic regions of the doped membrane antibody labeling could be easily realized by the as-prepared nanoparticles and then a high sensitive ECL immunoassay for HBsAg was developed. The linear range was between 1.0 and 100 pg mL(-1) (R(2)=0.9912). The detection limit could be as low as 0.11 pg mL(-1) (signal-to-noise ratio=3)., (Copyright © 2015 Elsevier B.V. Research on the association between maternal periodontal disease and the risk of preeclampsia has generated inconsistent results. This meta-analysis was conducted to evaluate the association between maternal periodontal disease and the risk of preeclampsia. A literature search of PubMed and Embase was performed to identify relevant papers published before March 2013. Only observational studies that assessed maternal periodontal disease and the risk of preeclampsia were selected. Patients' periodontal status was examined at different time points during pregnancy or after delivery (at 14-32 weeks of gestation, within 48 h prior to or within 5 days after delivery). Pooled odds ratios (ORs) and corresponding 95% confidence intervals (CIs) were calculated for cases and controls. Cases were defined as women with concurrent hypertension and proteinuria after 20 weeks of gestation. Eleven studies involving 1118 women with preeclampsia and 2798 women without preeclampsia were identified and analyzed. Women with periodontal disease before 32 weeks of gestation had a 3.69-fold higher risk of developing preeclampsia than their counterparts without periodontal disease (OR=3.69; 95% CI=2.58-5.27). Periodontal disease within 48 h prior to delivery was associated with a 2.68-fold higher risk of preeclampsia (OR=2.68; 95% CI=1.39-5.18). Pregnant women with periodontal disease within 5 days after delivery had a 2.22-fold higher risk of preeclampsia than women without periodontal disease (OR=2.22; 95% CI=1.16-4.27). In conclusion, this meta-analysis suggests that maternal periodontal disease is an independent predictor of preeclampsia.

The aim of the present study was to assess the atypical imaging manifestations of branchial cleft cysts (BCCs) confirmed by pathology. Computerized tomography (CT) or magnetic resonance imaging (MRI) of 17 BCC cases were reviewed. The imaging features, including laterality, location, border, attenuation and internal architecture, were evaluated. All 17 cases were second BCCs, including 5 cases of Bailey type I classification cysts and 12 cases of type II classification cysts. The atypical imaging features included signal and morphological abnormalities. The abnormal signal intensities were caused by intracapsular bleeding (n=2) or solidification of cystic fluid (n=2). Intracystic hemorrhaging revealed homogeneous hyperintensity on T1-weighted image (T1WI) and T2-weighted image (T2WI). Solidification of cystic fluid revealed slightly homogeneous hyperintensity compared with muscle on T1WI and homogeneous hypointensity on T2WI without enhancement. The aberrant morphology mainly presented as thickening of the cystic wall (n=13). Thickened walls of BCCs with ill- (n=5) or well- (n=8) defined borders were observed in 13 patients. In 3 patients, significant enhancement was identified following intravenous gadolinium administration (n=4). When with atypical CT or MRI features are presented, the typical location of BCCs can help in the diagnosis, as it is located at the lateral portion of the neck adjacent to the anterior border of the mandibular angle or sternocleidomastoid muscle. The atypical observations, including variable signals, imply that the cystic content has changed. Thickened walls indicate inflammation or cancerous tendency and patients with ill-defined margins, vascular involvement or lymphadenopathy atelectasis indicate malignant conversion.

Objective: To evaluate the accuracy of cone-bean CT (CBCT) on the measurement of the tooth length., Methods: One hundred and sixty single root premolars extracted for orthodontic purposes were selected. The tooth length was measured with vernier caliper. The premolars were divided into 10 groups randomly, 16 premolars in each group.In each group, the teeth were lined in molar area, premolar area, canine area and anterior tooth area in maxilla and mandible. Then CBCT was taken, and the tooth length was measured using the software supplied by the manufacturer in coronal and sagittal plane. Accuracy was compared between vernier caliper measurement and CBCT measurement., Results: The tooth length measured by CBCT was smaller than that measured with the vernier caliper. Significant differences were found in six areas, except the maxillary and mandibular premolar areas. With the increase of mesial-distal inclination or buccolingual torque angle, the difference between tooth length measured by CBCT and that measured with vernier caliper increased. The correlation between buccolingual torque angle (∠X1) and the coronal view of CBCT radiology tooth length (Spearmen index was 7.00, P < 0.001), and the correlation between mesial-distal inclination and the coronal view of CBCT radiology tooth length (Spearmen index was 0.624, P < 0.001) were found., Conclusions: The tooth length measured by CBCT was smaller than that measured with vernier caliper.

Purpose: To establish an animal model for maxillary incisors intrusion with mini-implant as the anchorage and study the effect on the surface morphology of cementum after the force had been given., Methods: 9 dogs were selected and randomly divided into 5 groups, group I was the control group(1 dog) with no force given; groups II,III,IV,V were experimental groups (2 dogs each). Mini-implants were inserted into the labial alveolar bone between the roots of second and third maxillary incisors on both sides, 100 gram force was imposed on the first and the second incisors on both sides.The animals were sacrificed at the end of 1, 2, 4,12 weeks (activation for 4 weeks,then retention for 8 weeks after force removal) respectively after activation.Scanning electron microscopy was used to examine the surface of the cementum., Results: In group II, the surfaces of root tip had a little scratch, which was small,confined. In group III, near the apical 1/3 of root, lacunas were fused, but no dentin exposed. In group IV, at the junction of the apical 1/3 and the middle 1/3 of the root, lacunas went deep into the dentin,which were wide and serious. In group V, the surfaces of lacunas were low ,smooth and even., Conclusions: As the intrusion goes on, the area of absorption expands from the root tip to neck. Root resorption increases with the duration of the applied force prolonged and repair of the cementum occurs at the resorbed area after force removal.

This paper introduces the present situation of medical modernization construction in Zhejiang Province, including provincial health E-government affairs, the public health system, digital hospitals, community health services, and the new rural cooperative medical system, and analyses the problems exiting, and points out the future construction tasks.

Based on IHE technical framework, this paper proposes the HIS and RIS/PACS integration strategy in combination with our hospital's practice. And it has been proved to be efficient in our practice.

AIM:To investigate the changes of gastric mucosal ascorbic acid secretion in patients with nonulcer dyspepsia and the effect of gastrin on it, and to relate any observed changes to H.pylori infection and mucosal histology.METHODS:Ascorbic acid secretions in patients were examined by collecting continuously gastric juice for one hour after having aspirated and discarded fasting gastric juice. Using the clearance rate (mL/min) of ascorbic acid from blood to gastric juice represented ascorbic acid secretion in the gastric mucosa.Ascorbic acid concentrations in plasma and juice were measured by ferric reduced method.RESULTS:Gastric ascorbic acid secretions in H.pylori-positive patients (1.46mL/min,range 0.27-3.78) did not significantly differ from those in H.pylori-negative patients(1.25mL/min, 0.47-3.14)(P>0.05). There were no significant differences in ascorbic acid secretions between patients with mild (1.56mL/min, 0.50-3.30), moderate (1.34mL/min, 0.27-2.93) and severe (1.36mL/min, 0.47-3.78) inflammation (P >0.05). There were no significant differences in ascorbic acid secretions between patients without activity (1.45mL/min, 0.27-3.14) and with mild (1.32mL/min, 0.61-2.93), moderate (1.49mL/min, 0.50-3.78) and severe (1.43mL/min, 0.51-3.26) activity of chronic gastritis either (P>0.05). Ascorbic acid secretions in patients with severe atrophy (0.56mL/min, 0.27-1.20) were markedly lower than those in patients without atrophy (1.51mL/min, 0.59-3.30) and with mild (1.43mL/min, 0.53-3.78) and moderate (1.31mL/min,0.47-3.16)atrophy(P<0.005). There was a significant negative correlation between ascorbic acid secretion and severity of atrophy (correlation coefficient =-0.43, P<0.005). After administration of pentagastrin, ascorbic acid secretions were markedly elevated (from 1.39mL/min, 0.36-2.96 to 3.53mL/min, 0.84-5.91) (P <0.001).CONCLUSION:Ascorbic acid secretion in gastric mucosa is not affected by H.pylori infection. Gastric ascorbic acid secretion is markedly related to the severity of atrophy, whereas not related to the severity of inflammation and activity. Gastrin may stimulate gastric ascorbic acid secretion. A decreased ascorbic acid secretion may be an important factor in the link between atrophic gastritis and gastric carcinogenesis.

To determine the efficacy of pyridoxine in treating seizures, 90 infants and children with recurrent convulsions primarily due to acute infectious diseases were enrolled in the present study. Forty patients were treated with high-dose pyridoxine (30 or 50 mg/kg/day) by intravenous infusion, and 50 subjects served as controls. Antiepileptic drugs and other therapies were similar in the two groups except for pyridoxine. Clinical efficacy criteria were based on the frequency of convulsions per day and on the duration of individual seizures after therapy was initiated. The results indicated that total response rates in the pyridoxine group and control group were 92.5% and 64%, respectively (chi-square = 14.68, P < .001). After initiation of therapy, seizures resolved after 2.4 +/- 1.4 days in the pyridoxine group and after 3.7 +/- 2.0 days in the control group (t = 3.67, P < .001). No adverse effects of pyridoxine were apparent during the observation period. We conclude that pyridoxine is an effective, safe, well-tolerated, and relatively inexpensive adjunct to routine antiepileptic drugs for treatment of recurrent seizures in children.

Radiation-induced apoptosis in chinese hamster ovary (CHO) cell lines is characterized by endonucleolytic cleavage of cellular DNA and changes of cell morphology within hours after radiation exposure. We investigated the capacity of ebselen [2-phenyl-1,2-benzisoselenazol-3(2H)-one], a seleno-organic compound with selenium-dependent glutathione peroxidase (GPx) activity, to protect cells from radiation-induced apoptosis. This phenomenon was studied by the quantitation of apoptotic cells and DNA gel electrophoresis after 6 Gy X-ray exposure. We also measured the activity of GPx and membrane lipid peroxidation. It was observed that 20 µM ebselen efficiently blocked apoptotic cell formation and DNA fragmentation 48 h post irradiation. Furthermore the data demonstrated that lipid peroxides increased significantly in irradiated cells and ebselen inhibited this process by elevating the cellular GPx activity. The results presented here indicate the requirement of free radicals for radiation-induced apoptosis and ultimately may yield insight necessary for designing protocols to modulate the process of radiation-induced apoptosis with antioxidant agents that scavenge radiation-induced free radicals.

A fusion gene was constructed in a plasmid in which the coding regions of human GM&#95;CSF and LIF cDNAs were connected by a synthetic linker sequence encoding a short peptide G-S-G-G-S through DNA recombinant techniques. It was then subcloned into the pBV220 expression vector, and expressed in E. coli after transformation and temperature induction. The expressed protein named as rhGM-LIF was confirmed by Western blot. After purification, the determination of activities showed that rhGM-LIF exhibited both GM-CSF and LIF activities.

A granulocytic leukemic cell line, called L833 has been established through culturing in vitro from mouse bone marrow of transplantable granulocytic leukemia. More than 280 passages have been performed in 3 years. Cell growth has been rapid and stable. Incidence of tumour formation was 100% with various routes of inoculation. The nature of granulocytic leukemia was confirmed by examination of cytology, cytochemistry, pathology and ultrastructural changes. Analysis proved chromosomes to be hypodiploid with model number of 39, loss of chromosomes, and presence of a marker. Besides the chromosomal change as mentioned above, both L883-A and L833-B derived from colonies formed from the L833 cells cultured in semi-solid agar medium, have their own marker. The cell line was sensitive to various types of antitumour agents in varying degrees. It was also sensitive to ionizing radiation. D0 values of L833 and L883-A cells were 98.8 and 104.9 rad, respectively. The results were similar to that of L801. Establishment of this cell line is of important significance for studying leukemia and screening anti-tumour agents, as well as provides a useful direct, economic and precisely quantitative tool for other relative studies.
