Lymph node metastasis (LNM) is one of the major prognostic factors in human gastrointestinal carcinomas (GICs). The lymph node-positive patients have poorer survival than node-negative patients. LNM is directly associated with the recurrence and poor survival of patients with GICs. The early detection of LNM in patients and designing effective therapies to suppress LNM may significantly impact the survival of these patients. The rapid progress made in proteomic technologies could be successfully applied to identify molecular targets for cancers at high-throughput levels. LC-MS/MS analysis enables the identification of proteins involved in LN metastasis, which can be utilized for diagnostic and therapeutic applications. Imbalanced and excessive use of chemical fertilizers has brought many soil fertility problems. Soil test crop response (STCR) approach aims to estimate precise fertilizer requirement for target yield under varying soil test values and gaining prominence for site-specific nutrient management. A field experiment to estimate STCR based fertilizer requirement for targeted yield of mungbean was conducted during 2019–2020 at G.B. Pant University of Agriculture and Technology, Pantnagar, India. Three fertility gradient plots of varying soil fertility levels were created by applying graded dose of N, P2O5 and K2O and growing wheat as an exhaust crop. Each of three soil fertility gradient plot was divided into 24 subplots (21 plots with different combinations of varying levels of N, P2O5, K2O and FYM, remaining 3 plots as controls) and mungbean was grown as main crop. Results revealed the requirement of 6.72, 0.883, and 3.52 kg of N, P and K, respectively for producing one quintal of mungbean grain. Soil, FYM, fertilizer and fertilizer along with FYM contributed 28.06%, 5.94%, 113.22%, and 135.01% for N; 24.73%, 3.18%, 15.1%, and 18.85% for P; 13.19%, 7.21%, 39.52%, and 55.84% for K, respectively. This information is used to develop STCR based fertilizer prescription equations along with ready reckoners of fertilizer doses at different soil fertility levels and yield targets of mungbean for providing balanced fertilization. Background: Cichorium intybus L. is cultivated in various temperate regions of the world for use of its different parts (mostly roots/seeds) as medicines or food. However, the anti-inflammatory, anti-proliferative and Epithelial Mesenchymal Transition (EMT)-inhibiting potential of C. intybus extracts have not been extensively studied at molecular level using suitable human cancer cell-line models. Objectives: Elucidation of the anti-inflammatory, anti-proliferative and EMT inhibiting potential of C. intybus extract using human cancer cell-line models. Materials and Methods: We prepared Methanolic Extract (MKE) of C. intybus leaves and studied its anti-inflammatory, anti-proliferative, and anti-EMT activities in THP-1 and U87MG cell lines through real time expression analysis and various functional assays including cell viability, cytotoxicity, colony forming and wound healing assays. Results: MKE significantly reduced the LPS-induced transcription of proinflammatory cytokines; TNF-α, and IL-6 from macrophage turned THP-1 cells. Also, expression of TGF-β gene was significantly downregulated by MKE in both THP-1 macrophages and U87MG cells. In U87MG glioma cells, MKE significantly downregulated the expressions of proliferation (Ki67, and PCNA), and EMT markers (LIX1, VIM, ZEB1, SNAIL, and SLUG), which are associated with aggressiveness, histological grade and poor prognosis in glioma. Conclusion: It should be emphasized that this is the first study done on examining the potential of C. intybus anti-inflammatory, anti-proliferative, and anti-EMT activities against the THP-1 macrophages and U87MG glioblastoma cells. Our findings strongly suggest C. intybus leaves as an efficient source of pharmaceutical formulations that can contribute to the treatment of inflammatory diseases and cancer and could add in improving human health significantly. An investigation was carried out to determine the impact of seed priming i.e. non-primed and primed seed with 2.0% KCl for 12 hr and twice foliar nutrition i.e. no spray, 0.5% ZnSO4, 0.5% CaCl2, 0.5% KNO3, and 2.0% urea on yield execution of normal sown (November 28) and late sown (Late sown) wheat. The results revealed that in both years of study, wheat planted on November 28 produced significantly higher grain yield with a comparable difference in number of spikes per square meter, spike length, number of fertile spikelets per spike, number of grains per spike, and 1000-grain weight. There was 23.0% and 24.0% decrease in grain yield for the late sowing on Late sown when compared with normal sowing on November 28 in first year and second year, respectively. Primed seeds were not significantly impacted in comparison to the non-primed seed. In contrast with no spray, spraying of various nutrients twice at maximum tillering and 80% flowering recorded significant improvement in yield of wheat crop. Highest yields were recorded with a foliar spray of 2.0% urea followed by 0.5% ZnSO4, 0.5% KNO3, and 0.5% CaCl2. The significant interaction between sowing dates and foliar spraying demonstrates that foliar spray of urea at the rate of 2.0% and ZnSO4 at the rate of 0.5% at maximum tillering and 80% flowering stage were financially more reasonable choices for both the planting dates and can increase grain yield. Protected cultivation is a resource-efficient method of crop production, however, at the same time, it is resource intensive. An optimum rate and time of fertilizer application is required in order to maximize crop yield vis-à-vis resource use efficiency. However, these aspects are scarcely studied for tomato under low- and medium-tech greenhouses in Indian humid sub-tropics. In this regard, a two-year study was conducted to assess the effect of four NPK fertilization rates (i.e., 120, 100, 80, and 60% of the recommended dose of fertilizers, RDF) and three fertigation scheduling approaches—fertigation at different stages in different proportions of NPK, along with an additional treatment, i.e., farmers' practice (soil-based application of recommended NPK) for tomatoes under a naturally ventilated polyhouse. The plant growth attributes, the tomato yield- and quality-related traits, the nutrient (NPK) accumulation by the plants, the water use efficiency (WUE), and economics were studied in response to different fertigation rates and scheduling approaches. These parameters were affected by both the rates of NPK fertilization and their time of application (scheduling) over the different growth stages. Among the different rates and time of fertigation, the recommended dose of fertilizer (100% RDF) (i.e., 300 kg N, 150 kg P2O5, and 150 kg K2O per ha and their scheduling as 15% N, 10% P2O5, and 10% K2O of RDF during 15–45 days after transplanting (P1); 40% N, 40% P2O5, and 40% K2O of RDF during 47–76 DAT (P2); 30% N, 40% P2O5, and 40% K2O of RDF during 77–107 DAT (P3); and 15% N, 10% P2O5, and 10% K2O of RDF during 108–138 DAT (P4)) was found to be the optimum for fruit yield, WUE, and economics of tomato under protected condition. Gallbladder cancer (GBC) is the sixth most common gastrointestinal tract cancer with a very low overall survival and poor prognosis. Profiling of cancer-derived extracellular vesicles (EVs) is an emerging strategy for identification of candidate biomarkers for the detection and prognosis of the disease. The aim of the study was to analyse the protein content from GBC cell line- derived EVs with emphasis on proteins which could be used as candidate biomarkers for the detection of GBC. NOZ and OCUG-1 cell lines were cultured and EVs were isolated from conditioned media. LC-MS/MS analysis of total EV proteins led to the identification of a total of 268 proteins in both the cell lines. Of these, 110 proteins were identified with ≥2 unique peptides with ≥2 PSMs in at least two experimental and technical replicate runs. STRING (Search Tool for the Retrieval of Interacting Genes/Proteins) database was used to perform bioinformatics analysis of 110 proteins which showed 'cell adhesion molecule binding', 'integrin binding', 'cadherin binding' among the top molecular functions and 'focal adhesion' to be among the top pathways associated with the EV proteins. A total of 42 proteins including haptoglobin (HP), pyruvate kinase (PKM), annexin A2 (ANXA2), thrombospondin 1 (THBS1), were reported to be differentially abundant in GBC tissue. Of these, 16 proteins were reported to be differentially abundant in plasma and plasma-derived EVs. We infer these proteins to be highly important to be considered as potential circulatory biomarkers for the detection of GBC. To check the validity of this hypothesis, one of the proteins, haptoglobin (HP) as a representative case, was analysed in plasma by quantitative Enzyme linked immunosorbent assay (ELISA) and we observed its increased levels in GBC in comparison to controls (p value= 0.0063). Receiver operating character (ROC) curve analysis for GBC vs controls showed an Area under the ROC Curve (AUC) of 0.8264 for HP with 22% sensitivity against 100% specificity.
