24. Impact of COVID-19 on Engineering Students in the Middle Phases of the Pandemic: Academic Motivation, Valued College Experiences, and Stress.

Abstract

When COVID-19 struck, engineering schools responded to unique issues, including interrupted capstone projects, cooperative education, and study abroad. Students became a focus. This led to a funded study to investigate the pandemic's impact on engineering students' academic motivation, educational valuation, learning, and perceived stress, which were connected through a conceptual model. Approximately seven months after the onset, a large sample of undergraduate engineering students at a public U.S. university (n = 1,140) responded to a survey (41.6% response), followed by focus groups. Jones' MUSIC Model of Academic Motivation and the Perceived Stress Scale (PSS-10) were key components of the conceptual model. Seventy-eight percent (78%) said their motivation was less versus before remote instruction. Two dimensions of the M USIC Model were only at the middle point of the measurement scale --· interest and empowerment. Students scored higher on the PSS-10 (M = 22.2) seven months into the pandemic compared to other groups beforehand. Medium negative correlations were found between the MUSIC dimensions and the PSS-10 score, suggesting decreased motivation accompanied by increased stress. Remote coursework was the most - frequent de - motivator, and the valued co [lege experience cited most was Campus-based instruction. The most-frequent stressor was Academic. In all focus groups. low or decreased motivation was mentioned. This research informs Higher Education about undergraduates' motivation and stress, in particular during COVID-19 and contributes to use of the MUSIC Model and PSS-10 with engineering students. Abstract

Dynamic remodeling of the actin cytoskeleton is an essential feature for virtually all actin-dependent cellular processes, including cell migration, cell cycle progression, chromatin remodeling and gene expression, and even the DNA damage response. An altered actin cytoskeleton is a structural hallmark associated with numerous pathologies ranging from cardiovascular diseases to immune disorders, neurological diseases and cancer. The actin cytoskeleton in cells is regulated through the orchestrated actions of a myriad of actin-binding proteins. In thisReview, we provide a brief overview of the structure and functions of the actin-monomer-binding protein profilin-1 (Pfn1) and then discuss how dysregulated expression of Pfn1 contributes to diseases associated with the cardiovascular system. 27. Breast cancer cells promote osteoclast differentiation in an MRTF-dependent paracrine manner

Abstract

Bone is a frequent site for breast cancer metastasis. The vast majority of breast cancer–associated metastasis is osteolytic in nature, and RANKL (receptor activator for nuclear factor κB)-induced differentiation of bone marrow–derived macrophages to osteoclasts (OCLs) is a key requirement for osteolytic metastatic growth of cancer cells. In this study, we demonstrate that Myocardin-related transcription factor (MRTF) in breast cancer cells plays an important role in paracrine modulation of RANKL-induced OCL differentiation. This is partly attributed to MRTFs’ critical role in maintaining the basal cellular expression of connective tissue growth factor (CTGF), findings that align with a strong positive correlation between CTGF expression and MRTF-A gene signature in the human disease context. Luminex analyses reveal that MRTF depletion in breast cancer cells has a broad impact on OCL-regulatory cell-secreted factors that extend beyond CTGF. Experimental metastasis studies demonstrate that MRTF depletion diminishes OCL abundance and bone colonization of breast cancer cells in vivo, suggesting that MRTF inhibition could be an effective strategy to diminish OCL formation and skeletal involvement in breast cancer. In summary, this study highlights a novel tumor-extrinsic function of MRTF relevant to breast cancer metastasis.

30. mDia2 is an important mediator of MRTF-A-dependent regulation of breast cancer cell migration

Abstract

Dysregulated actin cytoskeleton gives rise to aberrant cell motility and metastatic spread of tumor cells. This study evaluates the effect of overexpression of wild-type versus functional mutants of MRTF-A on migration and invasion of breast cancer (BC) cells. Our studies indicate that SRF's interaction is critical for MRTF-A-induced promotion of both two-dimensional and three-dimensional cell migration, while the SAP-domain function is important selectively for three-dimensional cell migration. Increased MRTF-A activity is associated with more effective membrane protrusion, a phenotype that is attributed predominantly to SRF's interaction with MRTF. We demonstrate formin-family protein mDia2 as an important mediator of MRTF-stimulated actin polymerization at the leading edge and cell migration. Multiplexed quantitative immunohistochemistry and transcriptome analyses of clinical BC specimens further demonstrate a positive correlation between nuclear localization of MRTF with malignant traits of cancer cells and enrichment of MRTF–SRF gene signature in pair-matched distant metastases versus primary tumors. In conclusion, this study establishes a novel mechanism of MRTF-dependent regulation of cell migration and provides evidence for the association between MRTF activity and increased malignancy in human BC, justifying future development of specific small molecule inhibitors of the MRTF-SRF transcriptional complex as potential therapeutic agents in BC.

41. Novel Approaches to Modulate Endothelial Cell Migration and Angiogenesis

Abstract

Angiogenesis, the process of neovascularization from pre-existing vasculature, is fundamental to development and tissue repair. However, aberrant angiogenesis exacerbates progression of a number of diseases including but not limited to cancer and diabetic retinopathy. Endothelial cell (EC) migration, a key event for angiogenesis, is a highly regulated process that critically relies on the dynamic remodeling of the actin cytoskeleton via orchestrated actions of various actin-binding proteins. An important aspect of the dynamic remodeling of the actin cytoskeleton, cell migration, and angiogenesis is stimulus-induced regulation of de novo synthesis of many important molecular components of the actin cytoskeleton by MKL (Megakaryoblastic Leukemia)–SRF (Serum-Response Factor) transcriptional machineries. Given the previous genetic evidence for the importance of MKL/SRF signaling in EC migration and angiogenesis, we first set out to explore whether angiogenesis is susceptible to small molecule-mediated inhibition of the MKL/SRF pathway. Our studies showed that CCG-1423, a small molecule inhibitor of MKL, causes a prominent defect in EC migration and angiogenesis in vitro, ex vivo, and in vivo. Inhibition of MKL led to prominent depletion of actin-binding protein Profilin-1 (Pfn1), an important molecular player of membrane protrusion and EC migration/angiogenesis, but through an unconventional indirect mechanism that involves a cellular externalization control [Angiogenesis (2017) 20(4):663-672; J. Next, guided by computational-based rational design, we identified the first-generation small molecule inhibitors of the Pfn1:actin interaction, and further demonstrated the ability of these small molecules to inhibit EC migration, proliferation, and angiogenesis in vitro and ex vivo [J Biol Chem (2017) Dec 27, epub]. A simple and one-step highly diastereoselective synthesis method was developed to obtain bulky, configurational and air-stable P-chiral tricyclic phosphines 3 exhibiting an exceptionally strong nucleophilic character. The key steps in the method involve the reaction of the stable phosphonium sila-ylide with aryl- and alkyl-substituted acetylene derivatives to obtain enantiomerically pure phosphines.

Abstract

A stable phosphonium sila-ylide is prepared and its X-ray structure has disclosed a long Si-P bond and a pyramidalized silicon center, indicating a very weak P-Si [pi] reaction. An [alpha],[beta]-ambiphilic character with a nucleophilic silicon center is displayed, thus showing that the phosphonium sila-ylide can be used as a sila-Wittig reagent with carbonyl derivatives.
