Antimicrobial resistance in Escherichia coli presents a global challenge associated with nosocomial infections and increased mortality rates. Understanding resistance profiles is crucial for guiding treatment strategies and ensuring effective antibiotic use. This study aimed to investigate the prevalence and in vitro resistance of E. coli to community-acquired and nosocomial infections. Various clinical samples from 700 patients were cultured on MacConkey's medium and blood agar. The disk diffusion method was used to determine the antibiotic susceptibility profile of the E. coli isolates following the guidelines of the Clinical and Laboratory Standards Institute (CLSI). Urine, pus, seminal fluid, vaginal swabs, and other body fluids were among the clinical samples analyzed. Of the 112 E. coli isolates, 48.2% were from inpatients and 51.8% were from outpatients, with the majority (66%) isolated from urine samples. Higher resistance levels were observed in the urinary isolates than that in the previously recorded data from the same institutions. Notably, isolates exhibited high resistance to penicillin (98.2%), ampicillin (97.3%), first-generation cephalosporins (90.2%), erythromycin (72.2%), and roxithromycin (95.4%), whereas lower resistance was noted against piperacillin-tazobactam (25.0%), nitrofurantoin (12.5%), and imipenem (9.8%). The overall multidrug resistance rate was 62.5%, with higher rates observed in nosocomial infections (70%) compared to community-acquired isolates (55.6%). However, this difference was not statistically significant (p>0.05). This study underscores the prevalence of E. coli isolates (27.0%) and highlights the concerning level of resistance, particularly to older antibiotics. Background: Cryptosporidiosis is an opportunistic parasitic disease widely distributed worldwide. Although Cryptosporidium sp. causes asymptomatic infection in healthy people, it may lead to severe illness in immunocompromised individuals. Limited effective therapeutic alternatives are available against cryptosporidiosis in this category of patients. So, there is an urgent need for therapeutic alternatives for cryptosporidiosis. Recently, the potential uses of Eugenol (EUG) have been considered a promising novel treatment for bacterial and parasitic infections. Consequently, it is suggested to investigate the effect of EUG as an option for the treatment of cryptosporidiosis. Materials and methods: The in silico bioinformatics analysis was used to predict and determine the binding affinities and intermolecular interactions of EUG and Nitazoxanide (NTZ) toward several Cryptosporidium parvum (C. parvum) lowa II target proteins. For animal study, five groups of immunosuppressed Swiss albino mice (10 mice each) were used. Group I was left uninfected (control), and four groups were infected with 1,000 oocysts of Cryptosporidium sp. The first infected group was left untreated. The remaining three infected groups received NTZ, EUG, and EUG + NTZ, respectively, on the 6th day post-infection (dpi). All mice were sacrificed 30 dpi. The efficacy of the used formulas was assessed by counting the number of C. parvum oocysts excreted in stool of infected mice, histopathological examination of the ileum and liver tissues and determination of the expression of iNOS in the ileum of mice in different animal groups. Results: treatment with EUG resulted in a significant reduction in the number of oocysts secreted in stool when compared to infected untreated mice. In addition, oocyst excretion was significantly reduced in mice received a combination therapy of EUG and NTZ when compared with those received NTZ alone. EUG succeeded in reverting the histopathological alterations induced by Cryptosporidium infection either alone or in combination with NTZ. Moreover, mice received EUG showed marked reduction of the expression of iNOS in ileal tissues. Conclusion: Based on the results, the present study signified a basis for utilizing EUG as an affordable, safe, and alternative therapy combined with NTZ in the management of cryptosporidiosis. Borrelia burgdorferi sensu lato can infect horses. However, the extent to which Egyptian horses get infected and seroconvert owing to this pathogen is unclear. Thus, this study aimed to determine the seroprevalence of B. burgdorferi and estimate its associated risk factors. A total of 385 serum samples collected from asymptomatic horses reared in three governorates in Egypt were screened using a commercial IDEXX SNAP 4Dx Plus kit. The results revealed that 58 (15.1%) out of 385 horses had antibodies against B. burgdorferi with Giza governorate showing the highest prevalence rate (30/130; 23.1%). According to univariate logistic regression analysis, locality, age, breed of examined horses and application of hygienic measures in house were significantly associated with B. burgdorferi infection. Furthermore, the multivariate logistic regression study revealed that thoroughbred horses older than four years old and raised in Giza governorate under unsanitary conditions, were more likely to be infected with B. burgdorferi than others. The serological evidence of B. burgdorferi in Egyptian horses and assessment of the risk factors associated with infection, necessitate wide range screening of this disease in other areas as well for efficient diagnosis and control. Bovine viral diarrhea (BVD) is a disease that affects ruminants globally, including camels, and causing significant financial losses. The epidemiology of BVD in camels in Egypt are not well understood. Thus, this study aimed to determine the prevalence of anti-BVD virus (BVDV) antibodies in camels and identify the potential variables associated with the infection. A total of 400 camel sera from three Egyptian governorates were examined using commercial ELISA kit. The total seroprevalence was 4.8% in examined camels and the BVDV seropositivity were more prevalent in camels from Giza governorate. The highest seropositivity was found in aged camels more than 8 years (OR = 8.62, 95%CI: 1.03–71.87), camels from herd size less than 30 (OR = 4.20, 95%CI: 0.89–19.78), previously aborted animals (OR = 5.98, 95%CI: 2.12–16.92), and in animals kept in contact with sheep or goats (OR = 7.48, 95%CI: 2.56–21.86). Consequently, the camels may be a significant source of BVD infection for other ruminant animals in the same herd due to their susceptibility to the viral infection. [ABSTRACT FROM AUTHOR]

Middle East Respiratory Syndrome (MERS) is a viral respiratory disease caused b a special type of coronavirus called MERS-CoV. In the search for effective substances against the MERS-CoV main protease, we looked into compounds from brown algae, known for their medicinal benefits. From a set of 1212 such compounds, our computer-based screening highlighted four—CMNPD27819, CMNPD1843, CMNPD4184, and CMNPD3156. These showed good potential in how they might attach to the MERS-CoV protease, comparable to a known inhibitor. We confirmed these results with multiple computer tests. Studies on the dynamics and steadiness of these compounds with the MERS-CoV protease were performed using molecular dynamics (MD) simulations. Metrics like RMSD and RMSF showed their stability. We also studied how these compounds and the protease interact in detail. An analysis technique, PCA, showed changes in atomic positions over time. Overall, our computer studies suggest brown algae compounds could be valuable in fighting MERS. However, experimental validation is needed to prove their real-world effectiveness. Head lice (Pediculus humanus capitis) are a major global concern, and there is growing evidence of an increase in head lice prevalence among Saudi schoolchildren. The purpose of this study is to investigate the prevalence of an insecticidal resistance mutation in head lice collected from schoolchildren. A polymerase chain reaction (PCR) was used to amplify a segment of the voltage-gated sodium channel gene subunit to assess the prevalence and distribution of the kdr T917I mutation in head lice. Subsequently, the restriction fragment length polymorphism (RFLP) patterns revealed two genotypic forms: homozygous-susceptible (SS) and homozygous-resistant (RR). The results showed that 17 (37.80%) of the 45 samples were SS, whereas 28 (62.2%) were RR and T917I and L920F point mutations were found in the nucleotide and amino acid sequences of RR. Compared to other nations, the frequency of permethrin resistance mutation in the head louse population in Saudi Arabia was low. This study provides the first evidence of permethrin resistance mutation in human head lice in Saudi Arabia. Background: Neglected tropical diseases (NTDs) are chronic illnesses that affect people worldwide, especially the poor. Infection prevention and control is cost-effective and long-term by raising NTD awareness. Few studies have examined healthcare providers' knowledge of these infections and parasitic origin diseases in developing and developed countries, as well as in Saudi Arabia. Objectives: To assess medical and health sciences students' awareness of neglected tropical diseases (NTDs) with parasitic origins at King Abdulaziz University in Jeddah. Methods: From February to June 2022, King Abdulaziz University conducted a cross-sectional descriptive survey of 201 medicine and health sciences students. Questions covered NTD awareness, control, and participation. Categorical factors were presented as percentages and frequencies in statistical analysis. Results: The study had roughly equal male and female student participation. Over half of the students didn't know what NTDS was or that Saudi Arabia had NTDs. NTDs were a public health issue in Saudi Arabia, but only 6% of students had seen them. Most Saudi students didn't know about entity control actions. Medical and health students don't understand NTDs or their treatment. Saudi Arabia's health system may be strained by imported diseases due to this gap. Objective: To green synthesize and characterize copper nanoparticles (CuNPs) using Astragalus sinicus, as well as evaluate the acaricidal, larvacidal, and repellent activities of CuNPs against Hyalomma anatolicum (H. anatolicum), one of the most prevalent ticks infesting cattle in Saudi Arabia. Methods: CuNPs were green synthesized by adding the Astragalus sinicus extract to a copper sulfate solution. The acaricidal, larvicidal, and repellent activities of CuNPs against H. anatolicum were assessed via the adult immersion test, the larval packet test, and the vertical movement behavior of tick larvae, respectively. The effects of CuNPs on acetylcholinesterase as well as oxidative enzyme activities were examined. Results: The green synthesized CuNPs displayed a spherical form with a size range of 15-75 nm. After exposure of adult H. anatolicum to different concentrations of CuNPs, the viability rate of adult H. anatolicum and the mean number, weight, and hatchability of eggs were noticeably reduced, in comparison to the control group (P<0.001). In addition, the viability rate of larvae considerably declined (P<0.001) with the LC50 and LC90 values of 11.30 and 20.34 µg/mL, respectively. The maximum repellent activity of CuNPs was observed at 50, 100, and 200 µg/mL with complete repellent activity after 60, 120, and 180 min of exposure, respectively. CuNPs, mainly at ½LC50 and LC50 concentrations, markedly suppressed the acetylcholinesterase activity of the larval stage of H. anatolicum (P<0.001). Moreover, CuNPs, mainly at LC50 dose, significantly elevated malondialdehyde level while declining glutathione-Stransferase level in H. anatolicum larvae (P<0.001). Conclusions: CuNPs show potent acaricidal, larvicidal, and repellent activities against adults and larvae of H. anatolicum. However, further studies must be performed to clarify the precise mechanisms and the efficacy of CuNPs in practical use. [ABSTRACT FROM AUTHOR]
