Histopathological findings depend on the distinction between normal and subtle changes in mucosal tissues. These distinctions are made based on the nature of the cell lining, nuclear hyperchromasia and cyto-architecture of glands. Diagnosis is mostly based on the experience of the pathologists, which is a subjective procedure. This research focuses on automating the process through 3D reconstruction of gastric histopathology images enhancing quantitative diagnosis. It emphasizes the advantages of 3D reconstruction from light microscopic serial section gastric images over whole slide imaging for clinical decision-making. Comparative analysis shows that processing and analyzing WSI scans are more expensive than light microscopy. While 2D imaging with light microscopy faces challenges due to limited focal depth, especially for thin sections, the objective of this research is to highlight the importance of transforming 2D gastric images into 3D by layering sequential sections. It will offer insights into glandular texture and volume, aiding in distinguishing between malignant glands and benign mimickers. The paper presents a summary and comparative examination of the registration techniques necessary for reconstructing 3D histopathology images. Additionally, it discusses future directions in the field of 3D histopathology reconstruction and acknowledges its potential benefits for research scholars. This will hold potential interest for pathologists when grading gastric carcinoma.

This study investigated the progressive morphological alterations and digestive tract development in larval and juvenile red spotted grouper, Epinephelus akaara across growth stages. External shape observations were made using an optical microscope, and the development of the digestive tract was investigated using histological methods. At 1 day after hatching (DAH), the digestive tract appeared as a straight tube extending between the ventral side and yolk-sac. At 3 DAH, the yolk was nearly absorbed, liver and pancreatic tissue began to develop. At 5 DAH, the opening of the mouth and anus allowed for the ingestion of external food, and the expansion of the intestinal lumen was observed. Gastric lumen differentiation became evident between the anterior intestine and esophagus. At 10 DAH, mucosal folds had formed in the rectum, and goblet cells were observed in the esophagus. At 20 DAH, mucosal folds in the stomach developed, and an increase in goblet cells was observed throughout the digestive organs. At 30 DAH, pyloric caeca and further gastric development were observed. By 40-50 DAH, sphincters between the esophagus, stomach, and intestine were clearly defined, resembling the adult digestive system. These findings suggest transitioning larvae to commercial pellets around 30 DAH, coinciding with the development of gastric glands and pyloric caeca. Objective: Incisura angularis is one of the important parts for evaluating mucosal atrophy and cancer risk. We determined the type of mucosa at incisura angularis in Helicobacter pylori‐naïve normal stomach. Methods: Subjects aged 40 years or older who underwent esophagogastroduodenoscopy for dyspepsia or a routine health checkup were recruited in 24 facilities between March 2008 and February 2009. Serum antibody to H. pylori was measured. Endoscopic atrophy was evaluated according to Updated Kimura‐Takemoto classification. Five biopsy specimens were taken from the incisura angularis and greater and lesser curvatures of the antrum and corpus. These specimens were histologically classified as fundic, pyloric or transitional. H. pylori‐naïve normal stomach was defined with the strictest criterion among various combinations of histological, endoscopic and serum findings. We determined histological type of mucosa at incisura angularis in H. pylori‐naïve normal stomach. Results: A total of 270 subjects (122 men, mean 64.6 yo) were analyzed. The strictest criterion consists of serum antibody ≤ 3.0 U/mL, endoscopic atrophy C‐1 and histological grade 0 in all of the five items in Updated Sydney System. The numbers having fundic, transitional and pyloric mucosa at incisura angularis under the strictest criterion were 13 (50%), 13 (50%) and 0, respectively. The probability that the type of mucosa at incisura angularis would be pyloric was almost zero (97.5% confidence interval 0–0.132). A 55-year-old man visited our hospital for a detailed examination of a gastric submucosal tumor that was first detected 10 years prior. The tumor continued to grow and had developed a depressed area in its center. A histopathological examination of biopsy specimens revealed gastric adenocarcinoma of the fundic gland type (GA-FG). It was diagnosed as T2 based on the invasion depth as determined by white-light endoscopy and endoscopic ultrasonography. A total gastrectomy with lymphadenectomy was performed and a GA-FG in the mucosa and submucosa was confirmed histopathologically. However, there was a gradual transition to an infiltrative tubular adenocarcinoma with poorly differentiated components in the muscular and subserosal layers. Metastasis was identified in a dissected lymph node (LN). This is the first report of a GA-FG progressing to an aggressive cancer with LN metastasis. The H+,K+-ATPase was identified as the primary proton secretory pathway in the gastric parietal cell and is the pharmacological target of agents suppressing acid secretion. Recently, we identified a second acid secretory protein expressed in the parietal cell, the vacuolar H+-ATPase (V-type ATPase). The aim of the present study was to further characterize H+-ATPase activation by modulations in extracellular calcium via the calcium sensing receptor (CaSR). Isolated gastric glands were loaded with the pH indicator dye BCECF-AM [2',7'-bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein acetoxymethyl ester] to measure intracellular pH. Experiments were conducted in the absence of sodium and potassium to monitor H+-ATPase-specific transport activity. CaSR was activated with the calcimimetic R568 (400 nM) and/or by modulations in extracellular Ca2+. Elevation in calcium concentrations increased proton extrusion from the gastric parietal cell. Allosteric modification of the CaSR via R568 and calcium increased vacuolar H+--ATPase activity significantly (ΔpH/minlowCa2+(0.1mM) = 0.001 ± 0.001, ΔpH/minnormalCa2+(1.0mM) = 0.033 ± 0.004, ΔpH/minhighCa2+(5.0mM) = 0.051 ± 0.005). Carbachol significantly suppressed calcium-induced gastric acid secretion via the H -ATPase under sodium- and potassium-free conditions. We conclude that the V-type H+--ATPase is tightly linked to CaSR activation. We observed that proton pump inhibitor (PPI) exposure does not modulate H±-ATPase activity. This elevated blood calcium activation of the H±-ATPase could provide an explanation for recurrent reflux symptoms while taking a PPI therapy. We further demonstrate the modification of this proton excretion pathway by extracellular calcium and the activation of the calcium sensing receptor CaSR. The novelty of this paper is based on the modulation of the H+-ATPase via both extracellular Ca (activation) and the classical secretagogues histamine and carbachol (inactivation). Histological and histochemical features of the oesophagogastric segment of the alimentary canal as well as ultrastructure of gastric gland cells of freshwater tubenose goby Proterorhinus semilunaris were examined. The studies revealed that despite the lack of anatomical distinction, the oesophagogastric segment is histologically divided into the oesophagus, oesogaster and stomach, which provides evidence for the functional compartmentation of this organ. The oesophagus was characterised by the presence of numerous goblet cells secreting mainly a mixture of neutral and acid mucopolysaccharides. In the stomach, the apical zone of the surface epithelial cells contained neutral mucopolysaccharides. Numerous proliferating cells were scattered throughout the surface epithelium. In the lamina propria of the stomach, a well-developed layer of gastric glands was observed. The glands were of the alveolar type and occupied nearly the entire length of the stomach except the pyloric region. The gastric gland cells were varied into light and dark; however, their ultrastructure was identical. All cells had numerous mitochondria and a well-developed tubulovesicular system typical for the oxynticopeptic cells, but pepsinogen granules were not present in the cytoplasm of these cells. These findings contribute new evidence to literature reports that not all gobiid fish are stomachless. Moreover, they suggest higher adaptation of the species to utilise protein-rich food compared to stomachless fish, and its ability to adjust the alimentary canal quickly to changing diet. Abstract: The gastric glands of marine aquaculture fish species are known to start secretion of pepsinogens (Pep) during larval development, far after first feeding. Although characterization of gastric gland development is considered useful for managing larval feeding, the timing of initial Pep synthesis by the gastric gland has only been reported for a few species. Consequently, this study employed in situ hybridization (ISH) to characterize the onset of expression of genes encoding the gastric enzymes Pep and Chitinase (Chi), as well as the complete development of gastric glands in the larval stomach of Japanese flounder (Paralichthys olivaceus), spotted halibut (Verasper variegatus), seven-band grouper (Epinephelus septemfasciatus) and greater amberjack (Seriola dumerili). The expression of both Chi and Pep was observed to begin in the cardiac region of the larval stomach at 25days post-hatching (dph) in Japanese flounder, 36dph in spotted halibut, 38dph in seven-band grouper and 13dph in greater amberjack. The gastric glands of these species were fully developed in the stomach approximately 5–12days after the initiation of Pep expression. Development of gastric glands was observed to coincide with external manifestations of metamorphosis, such as the development of adult coloration in Japanese flounder and spotted halibut of the Pleuronectiformes, while it finished prior to adult-type coloration in seven-band grouper and greater amberjack of the Perciformes. Secretory compartments of neurons, endocrine cells, and exocrine glands are acidic and contain high levels of labile Zn2t Previously, we reported evidence that acidity is regulated, in part, by the content of Zn2+ in the secretory [i.e., tubulovesicle (TV)] compartment of the acid-secreting gastric parietal cell. Here we report studies focusing on the mechanisms of Zn2+ transport by the TV compartment in the mammalian (rabbit) gastric parietal cell. Uptake of Zn2+ by isolated TV structures was monitored with a novel application of the fluorescent Zn2+ reporter N-(6-methoxy-8-quinolyl)-para-toluenesulfonamide (TSQ). Uptake was suppressed by removal of external AlP or blockade of H+-K+-ATPase that mediates luminal acid secretion. Uptake was diminished with dissipation of the proton gradient across the TV membrane, suggesting Zn2+/H+ antiport as the connection between Zn2+ uptake and acidity in the TV lumen. In isolated gastric glands loaded with the reporter fluozin-3, inhibition of H+-K+-ATPase arrested the flow of Zn2+ from the cytoplasm to the TV compartment and secretory stimulation with forskolin enhanced vectorial movement of cytoplasmic Zn2+ into the tubulovesicle/lumen (TV/L) cornpartment. Our findings suggest that Zn2+ accumulation in the TV/L compartment is physiologically coupled to secretion of acid. These findings offer novel insight into mechanisms regulating Zn2+ homeostasis in the gastric parietal cell and potentially other cells in which acidic subcellular compartments serve signature functional roles. The gastric H+/K+-ATPase is located within an infolding (secretory canaliculus) of the apical plasma membrane of gastric parietal cells. Our aim was to measure the pH values in the cytosol and canaliculus of the acid-secreting parietal cell and the adjacent gland lumen in situ. We used ultrafine double-barreled tip-sealed microelectrodes at high acceleration rates for intracellular and canalicular measurements. Immunohistochemical staining of the parietal cells was used to identify the track of the electrode and to estimate the position of the electrode tip at the time of the last intracellular measurement. En route to the deepest regions of the mucosa, where the average gland lumen pH was approximately 3, and on advancing in steps of 2 μm, the electrode entered the cytosol of the parietal cells, where the pH value was 7.4. Advancing the electrode further resulted, in several instances, in a sharp decrease in pH to an average value of 1.7 ± 0.2, which we interpreted as the measurement within the canaliculus. When the electrode was advanced even further, the pH reading returned to the cytosolic value. From the difference in pH between the secreting canaliculus and the adjacent gland lumen, we concluded that the released acid was immediately buffered. Thus, the only cellular structure directly exposed to the highly acidic canalicular content is the apical membrane forming the canaliculus in the parietal cell. [ABSTRACT FROM AUTHOR]

The clonality of gastric glands remains a controversial topic. Chimeric mouse studies suggested that all gastric glands were monoclonal. However, using the X-linked transgenic mouse model, we have suggested that most glands are polyclonal during development and that the fraction of monoclonal glands increases after birth. Nevertheless, a fraction of glands is perpetually polyclonal even in the adult murine stomach. To examine the existence of gastric polyclonal glands in the adult mouse, we studied an X-linked intrinsic polymorphic gene, pgk-1, in 6 week-old female mice heterozygous for the X-linked pgk-1a and pgk-1b. The sequence containing the seventh HpaII site in the promoter region of the gene and the polymorphic sites was utilized. Twenty-four of 225 fundic glands (10.7%) and 3 of 167 pyloric glands (1.8%) were clonally heterotypic. Only 0.6% of colonic crypts were heterotypic. Striped bonito Sarda orientalis larvae and juveniles were reared in a laboratory from hatching to 14 days post hatching, and the development of their digestive system was investigated histologically. On day 2 (first feeding day), the larval-type digestive system was differentiated. On day 3, a blind sac with densely distributed gastric glands developed and pyloric ceca appeared, suggesting that the digestive system was attaining adult-type structure. The yolk was completely absorbed on day 4. Bonito larvae preyed on rotifers and/or Artemia nauplii on day 2, but almost the whole gut was occupied by fish larvae from day 4. In the striped bonito, the adult-type digestive system was established right after first feeding, and this precocious development was well synchronized with the appearance of piscivory. Gastric gland component cells were electron-microscopically and immunoelectronmicroscopically examined with high-pressure freezing followed by freeze substitution and a low-temperature embedding resin method and compared to that of the conventional chemical-fixation method. The rat gastric gland was high-pressure frozen, freeze-substituted with acetone-containing osmium or acrolein, and embedded in Epon 812 or Lowicryl K4M, respectively. Using the high-pressure freezing method, the vitreous freezing range reached the depth of 150 mm from the surface. The ultrathin sections from both procedures embedding in Epon 812 and Lowicryl K4M were doubly stained with uranyl acetate and lead acetate, and histochemically or immuno-histochemically stained, respectively. In comparison to the conventional chemical fixation method, excellent results were obtained with respect to ultrastructural preservation. The stainings performed in this experiment included periodic acid-thiocarbohydrazide-silver proteinate staining, cationic colloidal cold at pH 2.5 staining, Helix pomatia lectin-staining, anti-alpha or -beta subunit antibodies of H+K+-ATPase immunostaining and pepsinogen immunostaining. The staining intensity of those was stronger than that of the conventional immersion-chemical fixation method. In addition to these results, the labels also showed good specific localization. In this paper, we provide a description of the high-pressure freezing followed by freeze substitution and low-temperature embedding resin method compared to the conventional chemical-fixation method. Little is known about the mechanisms that establish and maintain the proliferation and differentiation programs of the gastric epithelium. This is largely due to the complexity of the gastric epithelial units and the presence of the different epithelial lineage progenitors among heterogeneous populations of various mature cell types. This study is undertaken to establish an in vitro system highly enriched for gastric epithelial lineage progenitors. By using adult male rabbits, a simple method of isolating gastric epithelial cell fractions enriched in lineage progenitors was applied. Cultured cells labeled with bromodeoxyuridine were characterized by using lectin and immunohistochemical studies at light- and electron-microscopical levels. Analysis of primary cultures derived from the progenitor cell region of the epithelial units revealed that this system can support the proliferation and some of the differentiation programs of the progenitor cells. Cultured cells can be maintained for up to 5 days, while retaining most of the morphological features, molecular markers, and dynamic behavior of gastric epithelial progenitors. Differential cell counts at 1-day culture revealed that, while the undifferentiated progenitors formed about 30% of all attached cells, prepit, pit, and preparietal cells represented about 30%, 10%, and 2%, respectively. By 3 days, the increase in the percentage of pit and preparietal cells up to 25% and 9%, respectively, reflected their production in vitro. In conclusion, we have established a culture system enriched for gastric epithelial lineage progenitors that would hopefully allow the identification of factors and mechanisms involved in controlling their proliferative activity and differentiation pathways. High-pressure freezing/freeze substitution followed by Lowicryl K4M embedding provided an excellent morphology and antigenicity of the gastric glands, as well as the intraluminal fluid contents. Taking advantage of this, we histochemically investigated the secretory dynamics of the zymogenic contents in rat gastric gland, with special references to phospholipase A2 (PLA2) and phospholipase Cγ1 (PLCγ1). The combination of immunogold labeling and KMnO4–uranyl acetate–lead citrate staining for zymogenic contents clearly demonstrated the rapid diffusion of PLA2 molecules from the exocytosed zymogenic contents into the mucinous contents in gastric glandular lumens. In contrast, the exocytosed PLCγ1 molecules remained within the zymogenic contents in the glandular lumens. These findings indicated the distinction between the exocytosed PLA2 and PLCγ1 in their diffusion rate. In addition, the mucinous contents surrounding the exocytosed zymogenic contents were intensely labeled with Griffonia simplicifolia II lectin which specifically recognizes the mucin of mucous neck cells. Interestingly, some of the PLA2 immunolabeling on the mucinous contents was associated with the apical membranes of gastric epithelial cells, especially that of parietal cells. The distribution of lysozyme in normal gastric and duodenal mucosa was studied by light- and electronmicroscopic immunocytochemical techniques (direct enzyme-labeled antibody method). In the duodenal mucosa, lysozyme was found in the Paneth cells and the epithelial cells of Brunner's glands. Electron-microscopically, lysozyme was found in rough endoplasmic reticulum and perinuclear spaces, which were assumed to be protein-synthesizing organelles, and also in the secretory granules of Paneth cells. Additionally, lysozyme was detected in the stomach in mucinous granules and in some parts of the rough endoplasmic reticulum within the epithelial cells of the pyloric glands, the mucous neck cells of the fundic glands, and in several surface epithelial cells of the plyoric and fundic regions. This suggests that some quantity of lysozyme in gastrointestinal secretion originates from the gastric and duodenal glands, and that it acts as a defense mechanism in the gastrointestinal tract.
