In this overview the genetic contributions to physical performance will be outlined with a special focus on strength, power and endurance characteristics. Two basic approaches have been used to study the genetic basis of performance phenotypes and related characteristics: the unmeasured genotype approach (top-down) and the measured genotype approach (bottom up). Assessment of heritability is based on the model that total variation (Vtot) in a phenotype is partitioned into genetic (VG), common environmental (VC) and individual-specific environmental (VE) components (Vtot=VG+VC+VE). Heritability (h 2 ) refers to the proportion of the total variation that can be attributed to genetic effects (VG/Vtot). Estimated heritabilities for strength vary widely between 0.27 and 0.58 in family studies and between 0.14 and 0.83 in twin studies. Heritabilities for dynamic strength of arm and leg muscle groups range from 0.29 to 0.87. For aerobic performance estimated heritabilities vary between 0.40 and 0.94. There is good evidence for genotype*training interaction for strength and aerobic performance. Association and linkage studies have indicated a number of potential interesting regions in the human genome. However few replications have been observed with the exception of associations between strength and ACE, ACTN3 and VDR and ACE for aerobic performance.

Short stature and later maturation of youth artistic gymnasts are often attributed to the effects of intensive training from a young age. Given limitations of available data, inadequate specification of training, failure to consider other factors affecting growth and maturation, and failure to address epidemiological criteria for causality, it has not been possible thus far to establish cause–effect relationships between training and the growth and maturation of young artistic gymnasts. In response to this ongoing debate, the Scientific Commission of the International Gymnastics Federation (FIG) convened a committee to review the current literature and address four questions: (1) Is there a negative effect of training on attained adult stature? (2) Is there a negative effect of training on growth of body segments? (3) Does training attenuate pubertal growth and maturation, specifically, the rate of growth and/or the timing and tempo of maturation? (4) Does training negatively influence the endocrine system, specifically hormones related to growth and pubertal maturation? The basic information for the review was derived from the active involvement of committee members in research on normal variation and clinical aspects of growth and maturation, and on the growth and maturation of artistic gymnasts and other youth athletes. The committee was thus thoroughly familiar with the literature on growth and maturation in general and of gymnasts and young athletes. Relevant data were more available for females than males. Youth who persisted in the sport were a highly select sample, who tended to be shorter for chronological age but who had appropriate weight-for-height. Data for secondary sex characteristics, skeletal age and age at peak height velocity indicated later maturation, but the maturity status of gymnasts overlapped the normal range of variability observed in the general population. Gymnasts as a group demonstrated a pattern of growth and maturation similar to that observed among short-, normal-, late-maturing individuals who were not athletes. Evidence for endocrine changes in gymnasts was inadequate for inferences relative to potential training effects. Allowing for noted limitations, the following conclusions were deemed acceptable: (1) Adult height or near adult height of female and male artistic gymnasts is not compromised by intensive gymnastics training. (2) Gymnastics training does not appear to attenuate growth of upper (sitting height) or lower (legs) body segment lengths. (3) Gymnastics training does not appear to attenuate pubertal growth and maturation, neither rate of growth nor the timing and tempo of the growth spurt. (4) Available data are inadequate to address the issue of intensive gymnastics training and alterations within the endocrine system. To construct reference values for height, body mass and BMI of children and adolescents from the Cariri region, Brazil; to compare the growth of Cariri children with those from CDC (Centers for Disease Control and Prevention) and with references from other Brazilian regions; to verify the associations between socioeconomic status and height, body mass and BMI in children and youth from both sexes.The sample comprised 3311 girls and 3280 boys aged 7-17 years, participating in the study 'Healthy Growth in Cariri'. Socioeconomic status was defined according to school attendance: private and public. Centile curves for height, body mass and BMI were constructed using the LMS method.Significant differences between children and adolescents from Cariri and those from other Brazilian regions and the CDC references were found for height and body mass. In girls from private schools, average differences in height compared to the CDC references ranged from 0.79-5.9 cm and in boys from 2.9-8.6 cm.Children from Cariri show a growth pattern in height, body mass and BMI that closely resembles the patterns observed in developed countries, but the absolute values in height and body mass are markedly lower than CDC references and growth references for other regions in Brazil.

Este estudo teve como objetivos: 1) apresentar cartas percentílicas e valores de referência para um conjunto variado de testes motores; e 2) comparar o desempenho das crianças e jovens cearenses com o de outros estudos desenvolvidos noutras regiões do país e do exterior. A amostra total é composta por 6.238 indivíduos (3.122 meninas e 3.116 meninos) com idades compreendidas entre os oito e os 17 anos. As cartas percentílicas foram construídas separadamente para cada sexo utilizando o método LMS implementado no "software" LMSchartmaker Pro versão 2.3. As cartas de referência produzidas para o Cariri apresentam um comportamento genérico semelhante ao verificado nos estudos considerados. Com exceção da prova do "trunk lift", constata-se uma nítida superioridade do desempenho dos meninos, enfatizando a presença de forte dimorfismo sexual. A comparação dos valores do P50 de crianças e jovens do Cariri relativamente às de Londrina (Brasil) e de Portugal mostraram performances consistentemente inferiores.

Muscle strength is important in functional activities of daily living and the prevention of common pathologies. We describe the two-staged fine mapping of a previously identified linkage peak for knee strength on chr12q12-14. First, 209 tagSNPs in/around 74 prioritized genes were genotyped in 500 Caucasian brothers from the Leuven Genes for Muscular Strength study (LGfMS). Combined linkage and family-based association analyses identified activin receptor 1B (ACVR1B) and inhibin beta C (INHBC), part of the transforming growth factor beta pathway regulating myostatin - a negative regulator of muscle mass - signaling, for follow-up. Second, 33 SNPs, selected in these genes based on their likelihood to functionally affect gene expression/function, were genotyped in an extended sample of 536 LGfMS siblings. Strong associations between ACVR1B genotypes and knee muscle strength (P-values up to 0.00002) were present. Of particular interest was the association with rs2854464, located in a putative miR-24-binding site, as miR-24 was implicated in the inhibition of skeletal muscle differentiation. Rs2854464 AA individuals were similar to 2% stronger than G-allele carriers. The strength increasing effect of the A-allele was also observed in an independent replication sample (n=266) selected from the Baltimore Longitudinal Study of Aging and a Flemish Policy Research Centre Sport, Physical Activity and Health study. However, no genotype-related difference in ACVR1B mRNA expression in quadriceps muscle was observed. In conclusion, we applied a two-stage fine mapping approach, and are the first to identify and partially replicate genetic variants in the ACVR1B gene that account for genetic variation in human muscle strength. The aims of this study are twofold: (1) to present the latent growth model, its strategy and usefulness in modelling sport participation over a 6-year period in youngsters (2) and to study the impact of biological maturation in sport participation changes. A standardized sport participation questionnaire using h/week/year as the unit of analysis was applied to 588 Belgium boys 13-18 years of age followed longitudinally for 6 years. Skeletal age was used to estimate biological maturation. Growth curve modelling with robust estimation was used. Sport participation changes showed a curvilinear trend: baseline values (3.18+/-0.13 h/week/year), a linear trend that indicates the rate of change (0.70+/-0.11) and a quadratic trend indicating deceleration, i.e. a change in the rate of change (-0.07+/-0.02) were all statistically significant (P

Childhood overweight and obesity are increasing all over the world and have been associated with low levels of physical activity (PA).To determine the prevalence of overweight, obesity and PA levels in Azorean children according to age and sex; and to determine the association between levels of PA and prevalence of overweight and obesity.Weight, height and PA levels were measured in 3699 children aged 6-10 years, from the Azores Islands, Portugal. Overweight and obesity were classified according to the cut-offs of Cole et al. (BMJ 320:1240-1243, 2000) .In girls, prevalences of overweight and obesity were 22.8% and 13.2%, and in boys 17.6% and 12.3%, respectively. No age trends were found in the prevalence of overweight or obesity; however, girls had a higher risk of being overweight (OR = 1.4, 95% CI = 1.2-1.7) than boys. Levels of PA were higher in boys compared to girls (F(1) = 52.8, p0.001). A protective effect of PA practice (very active versus less active) was observed for obesity (OR = 0.7; 95% CI = 0.5-0.9).The results demonstrate the existence of high prevalence of overweight and obesity in children from the Azores Islands, which is associated with low levels of PA.

Objective: To model changes in body mass index (BMI), including its stability, and to investigate the association between physical activity, 1-mile run/walk and levels of gross motor coordination and BMI during 5 consecutive years. Design: A longitudinal study of children 6 years of age at baseline followed at annual intervals over 5 years. Subjects: A total of 285 children (143 boys and 142 girls) were enrolled in grade 1 (age 6 years) and followed through grade 5 (age 10 years). Measurements: BMI was recorded and physical activity was assessed by questionnaire, aerobic fitness was evaluated with the 1-mile run/walk and gross motor coordination was measured with the KTK test battery (Korperkoordination test fur Kinder). Multilevel modelling techniques were for the primary analysis. Results: Changes in BMI showed similar curvilinear trends in boys and girls, with ample inter-individual crossing trajectories that is, low tracking. Longitudinal changes in physical activity (PA) and aerobic fitness were not significantly associated with BMI-changes during the 5 years. Children who were more proficient in their motor coordination showed lower values of BMI during the 5 years. Conclusions: BMI trajectories of both boys and girls show low tracking of BMI-values. Considerable inter-individual variation exists both in baseline BMI-values and changes (velocity and acceleration) over time. PA and fitness were not associated with BMI-changes, but gross motor function was negatively associated with BMI-changes. No gender-specific associations were found. If confirmed in other populations these observations could be translated in the promotion of physical activities that improve gross motor function in children aged 6-10 years. This seems to be of major importance for the physical education curriculum of primary school children.

Repeated, intense use of muscles leads to a decline in performance known as muscle fatigue. Resistance to muscle fatigue depends on age, sex, muscle fiber type, activation by the nervous system and training. Heritability of muscle strength phenotypes ranges between 31% and 78%, although little is known about heritability of muscle fatigue. A first aim of this study was to estimate the heritability for fatigue resistance after a short bout of intense exercise of the knee musculature. The main purpose was to identify chromosomal regions linked to muscle fatigue applying genome-wide linkage analyses. A selection of 283 informative male siblings (17-36 years old), belonging to 105 families, was used to conduct a genome-wide SNP-based multipoint linkage analysis. Heritabilities for resistance to muscle fatigue ranged from 21% to 54%. The strongest linkage signal was found at 19q13.11 (LOD=2.158; P 1.5 were found (1p31.3, 3q29, 8p22, 11q25 and 19q12). When replicated in an independent sample, these results warrant further fine mapping studies aiming to detect genes that underlie variation in muscle fatigue.

Este estudo teve os seguintes objetivos: (1) apresentar procedimentos metodológicos utilizados na estimação da idade óssea; (2) descrever o método Tanner-Whitehouse (TW3), e (3) destacar os resultados da reprodutibilidade deste método em crianças e jovens. A metodologia adotada na preparação do processo de aprendizagem e reprodutibilidade do método TW3 foi estruturada em três fases, tendo sido realizadas em duas Instituições de ensino (Portugal e Bélgica), sob a orientação e supervisão de avaliadores experientes. Após várias etapas de treinamento, os resultados alcançados na avaliação inter e intraobservador situaram-se entre 81,3 e 87,9%. Os resultados finais do estudo possibilitaram comprovar a eficácia de um rigoroso processo de treino, como elemento essencial na preparação prévia do avaliador, para a utilização da avaliação da maturação esquelética com base no método TW3. The objectives of this study were: (1) to present methodological procedures used for the estimation of bone age; (2) describe the Tanner-Whitehouse method (TW3), and (3) to highlight the results of reproducibility of this method in children and adolescents. The method adopted for the preparation of the learning process and for evaluation of the reproducibility of the TW3 method was divided into three phases and was conducted at two teaching institutions (Portugal and Belgium) under the guidance and supervision of experienced raters. After several stages of training in the rating of radiographs, inter- and intraobserver reliability ranged from 81.3 to 87.9%. The final results of the study demonstrate the effectiveness of a rigorous training process as an essential element in the previous preparation of raters for the assessment of skeletal maturation by the TW3 method.

The purpose of this study was to cross-validate the Beunen-Malina method for non-invasive prediction of adult height. Three hundred and eight boys aged 13, 14, 15 and 16 years from the Madeira Growth Study were observed at annual intervals in 1996, 1997 and 1998 and re-measured 7-8 years later. Height, sitting height and the triceps and subscapular skinfolds were measured; skeletal age was assessed using the Tanner-Whitehouse 2 method. Adult height was measured and predicted using the Beunen-Malina method. Maturity groups were classified using relative skeletal age (skeletal age minus chronological age). Pearson correlations, mean differences and standard errors of estimate (SEE) were calculated. Age-specific correlations between predicted and measured adult height vary between 0.70 and 0.85, while age-specific SEE varies between 3.3 and 4.7 cm. The correlations and SEE are similar to those obtained in the development of the original Beunen-Malina method. The Beunen-Malina method is a valid method to predict adult height in adolescent boys and can be used in European populations or populations from European ancestry. Percentage of predicted adult height is a non-invasive valid method to assess biological maturity.

Physical activity (PA) in children/adolescents of both genders from a rural community in Mozambique was estimated by accelerometry and by questionnaire and was compared with PA of Portuguese youth. Total PA, moderate (MPA), vigorous (VPA) and very vigorous (VVPA) were evaluated. Mozambican boys were more active than girls. Intensity of PA declined significantly with age. Survival activities, such as household tasks, were the predominant mode of PA. Compared with Portuguese children/adolescents, Mozambicans had significantly higher total PA; showed less decline of PA with age and engaged in fewer minutes at higher intensity PA. Environmental factors likely explain documented differences.

There have been few genetic studies of normal variation in body size and composition conducted in Africa. In particular, the genetic determinants of somatotype remain to be established for an African population. (1) To estimate the heritabilities of aspects of somatotype and (2) to compare the quantitative genetic effects in an African population to those that have been assessed in European and American populations. The sample composed of 329 subjects (173 males and 156 females) aged 7-17 years, belonging to 132 families. The sibships in the sample ranged in size from two to seven individuals. All sampled individuals were residents of the Calanga region, an area located to the north of Maputo in Mozambique. Somatotype was assessed using the Heath-Carter technique. Herit abilities were estimated using SAGE software. Moderate heritabilities were determined for each trait. Between 30 and 40% of the variation in each somatotype measure was attributable to genetic factors. The heritability of ectomorphy was 31%. Mesomorphy was similarly moderately heritable, with approximately 30% of the variationattributable to genetic factors. The heritability of endomorph was higher in the Calanga population (h(2) = 0.40). Quantitative genetic analyses of somatotype variation among siblings indicate that genetic factors significantly influence endomorphy, mesomorhpy, and ectomorphy. However, environmental factors also have significant effects on the variation in physique present in the population of Calanga. Lack of proper nutrition, housing, medical assistance, and primary health care, together with very demanding and sex-specific daily chores may contribute to the environmental effects on these traits.

Objective Evidence of a genetic basis of metabolic risk factors (MRFs) is growing. Studies examining the genetic and environmental basis of the clustering of MRFs, grouped together in the metabolic syndrome (MetS), are however sparse. The aim of this study therefore was to study the heritabilities of the MRFs and the genetic and environmental correlations between the MRFs. Methods Study participants were 768 Caucasian twins coming from 418 pairs (18–34 years). MRFs were those included in the National Cholesterol Education Program Adult Treatment Panel III definition of the MetS. Multivariate path analysis on the continuous MRFs of the MetS was implemented. Results Heritabilities ranged between 47.0% and 80.2% (men) and 58.5% and 77.9% (women) for the individual MRFs. Evidence was found for overarching genetic (A) and environmental (E) sources of variance, both however loading mainly on waist circumference. Furthermore, the model included a ‘lipids’ and a ‘blood pressure’-factor both in part attributable to A and E. The majority of the variance however was MRF-specific. Conclusion Based on our sample of young adults with a low prevalence of the MetS, it can be concluded that both genes and environment contribute significantly to the clustering of the MRFs although the majority of the variation is MRF-specific. Therefore, future QTL searches in young adults may want to focus on MRF-specific loci, rather than ‘cluster-phenotypes’ such as the MetS.

Children born small for gestational age are at increased risk of developing type 2 diabetes in adulthood. The satiety signal leptin that regulates food intake and energy expenditure might be a possible molecular link, as umbilical cord leptin levels are positively correlated with birth weight. In the present study, we examined whether common single nucleotide polymorphisms (SNPs) in the leptin (LEP; 19G>A) gene and its receptor (LEPR; Q223R and K109R) are associated with birth weight and adult metabolic risk factors for type 2 diabetes in twins.SNPs were genotyped in 396 monozygotic and 232 dizygotic twins (286 men and 342 women, mean age 25 years) recruited from the East Flanders Prospective Twin Survey. Data were analysed using linear mixed models.The LEPR K109R SNP was associated with birth weight (KK, KR and RR (95% confidence interval, CI): 2511 (2465-2557), 2575 (2516-2635) and 2726 (2606-2845) gram; P(additive)=0.001). Also the LEPR Q223R SNP showed a significant association with weight at birth (QQ, QR and RR (95% CI): 2492 (2431-2554), 2545 (2495-2595) and 2655 (2571-2740) gram; P(additive)=0.003). Furthermore, an interaction between the LEPR K109R and the Q223R SNP on birth weight was observed (P=0.014). G allele carriers of the LEP 19G>A SNP had higher high-density lipoprotein (HDL) cholesterol levels compared to 19A homozygotes (GX vs AA (95% CI): 1.62 (1.58-1.66) vs 1.49 (1.40-1.58) mmol l(-1); P(recessive)=0.013).This study indicates that leptin may act as a growth-promoting signal during fetal development, and suggests a possible role for the LEPR in explaining the inverse relationship between birth weight and the development of metabolic diseases in adulthood. Additionally, these results suggest that the LEP 19G>A SNP affect HDL cholesterol levels.

Insulin resistance and obesity are underlying causes of type 2 diabetes and therefore much interest is focused on the potential genes involved. A series of anthropometric and metabolic characteristic were measured in 240 MZ and 112 DZ twin pairs recruited from the East Flanders Prospective Twin Survey. Microsatellite markers located close to ABCC8, ADIPOQ, GCK, IGF1, IGFBP1, INSR, LEP, LEPR, PPARγ and the RETN gene were genotyped. Univariate single point variance components linkage analyses were performed using two methods: (1) the standard method, only comprising the phenotypic and genotypic data of the DZ twin pairs and (2) the extended method, also incorporating the phenotypic data of the MZ twin pairs. Suggestive linkages (LOD > 1) were observed between the ABCC8 marker and waist-to-hip ratio and HDL-cholesterol levels. Both markers flanking ADIPOQ showed suggestive linkage with triglycerides levels, the upstream marker also with body mass and HDL-cholesterol levels. The IGFBP1 marker showed suggestive linkage with fat mass, fasting insulin and leptin levels and the LEP marker showed suggestive linkage with birth weight. This study suggests that DNA variants in ABCC8, ADIPOQ, IGFBP1 and LEP gene region may predispose to type 2 diabetes. In addition, the two methods used to perform linkage analyses yielded similar results. This was however not the case for birth weight where chorionicity seems to be an important confounder.

Alongitudinal study of growth and physical fitness of twins and their parents was designed in 1985. The major aims of this Leuven Longitudinal Twin Study were to quantify the genetic and environmental determination of (1) somatic characteristics, biological maturation and physical performance characteristics during the growth process, (2) the growth and developmental patterns, and (3) the covariation in somatic and performance characteristics.

Fine mapping of linkage peaks is one of the great challenges facing researchers who try to identify genes and genetic variants responsible for the variation in a certain trait or complex disease. Once the trait is linked to a certain chromosomal region, most studies use a candidate gene approach followed by a selection of polymorphisms within these genes, either based on their possibility to be functional, or based on the linkage disequilibrium between adjacent markers. For both candidate gene selection and SNP selection, several approaches have been described, and different software tools are available. However, mastering all these information sources and choosing between the different approaches can be difficult and time-consuming. Therefore, this article lists several of these in silico procedures, and the authors describe an empirical two-step fine mapping approach, in which candidate genes are prioritized using a bioinformatics approach (ENDEAVOUR), and the top genes are chosen for further SNP selection with a linkage disequilibrium based method (Tagger). The authors present the different actions that were applied within this approach on two previously identified linkage regions for muscle strength. This resulted in the selection of 331 polymorphisms located in 112 different candidate genes out of an initial set of 23,300 SNPs.

To investigate the association of muscular strength and aerobic fitness with a continuous metabolic syndrome risk score in male and female adults.This cross-sectional study included 1019 (571 men) Flemish adults, aged 18-75. Muscular strength was evaluated by measuring isometric knee extension and flexion peak torque, using a Biodex System Pro 3 dynamometer. Aerobic fitness was quantified as VO2peak and was determined during a maximal cycle ergometer exercise test. Both strength and aerobic fitness were scaled for differences in FFM, using allometric analyses. A validated metabolic syndrome risk score that was based on waist circumference, triglycerides, blood pressure, fasting plasma glucose, and HDL cholesterol was used. Metabolic syndrome risk score, strength, and aerobic fitness were analyzed as continuous variables using multiple linear regression.Metabolic syndrome risk was inversely associated with strength, independently of aerobic fitness, and after adjustment for age, height, education level, smoking status, and dietary intake in women (beta = -0.172, P0.001). In men, however, adjustment for aerobic fitness attenuated the inverse association between strength and metabolic syndrome risk (beta = -0.044, P0.05). Independently of strength, aerobic fitness was inversely associated with metabolic syndrome risk (men: beta = -0.309, P0.001; women: beta = -0.208, P0.001). Furthermore, independent associations were found for strength and aerobic fitness with several individual metabolic syndrome risk factors in women, and most of these associations were only partially mediated by central and general adiposity indicators.Although cross-sectional, the present results support inclusion of strength training in addition to aerobic exercise in physical activity recommendations for women, because both types of activity might show additional effects in reducing the risk of the metabolic syndrome.

Aims/hypothesis We determined the genetic contribution of 18 anthropometric and metabolic risk factors of type 2 diabetes using a young healthy twin population. Methods Traits were measured in 240 monozygotic (MZ) and 138 dizygotic (DZ) twin pairs aged 18 to 34 years. Twins were recruited from the Belgian population-based East Flanders Prospective Twin Survey, which is characterised by its accurate zygosity determination and extensive collection of perinatal and placental data, including information on chorionicity. Heritability was estimated using structural equation modelling implemented in the Mx software package. Results Intra-pair correlations of the anthropometric and metabolic characteristics did not differ between MZ monochorionic and MZ dichorionic pairs; consequently heritabilities were estimated using the classical twin approach. For body mass, BMI and fat mass, quantitative sex differences were observed; genetic variance explained 84, 85 and 81% of the total variation in men and 74, 75 and 70% in women, respectively. Heritability estimates of the waist-to-hip ratio, sum of four skinfold thicknesses and lean body mass were 70, 74 and 81%, respectively. The heritability estimates of fasting glucose, fasting insulin, homeostasis model assessment of insulin resistance and beta cell function, as well as insulin-like growth factor binding protein-1 levels were 67, 49, 48, 62 and 47%, in that order. Finally, for total cholesterol, LDL-cholesterol, HDL-cholesterol, total cholesterol:HDL-cholesterol ratio, triacylglycerol, NEFA and leptin levels, genetic factors explained 75, 78, 76, 79, 58, 37 and 53% of the total variation, respectively. Conclusions/interpretation Genetic factors explain the greater part of the variation in traits related to obesity, glucose intolerance/insulin resistance and dyslipidaemia. It is widely believed that active participation in sports during youth is an important prerequisite for adult involvement in sports. However, data from reliable longitudinal studies tracking patterns of sports participation from youth into adulthood are scarce. This study addresses the leisure-time sports participation of adult women, 32—41 years of age, from a lifetime sports socialization perspective. Some 20 years after they participated in 1979 in the Leuven Growth Study on Flemish Girls, 257 female adults participated again in a comprehensive questionnaire and face-to-face interview. Inter-age correlations for sports participation are calculated from adolescence into adulthood. Logistic regression modeling and structural equation modeling are used to explain individual differences in adult sports participation. Outcomes indicate that tracking of sports involvement between late adolescence and adulthood is moderately high ( r = .41; beta .42). The results from the multivariate analysis show that sport participation during adolescence is a better predictor of adults' involvement in sports than educational level or parental socioeconomic status. The variances accounted for are rather small, indicating that sport experiences and social background characteristics only partially explain the sport participation behavior of adults. In the sports socialization process, late adolescent sports experience, along with the school program in which an adolescent is involved, appear to play a crucial role in sport involvement in later life. We recommend that youth sports programs need to be examined critically with regard to their contribution to lifetime sports participation.

Commonly used indicators of biological maturation are discussed, including sexual, skeletal, morphological, and dental maturity, and the hypothalamus–pituitary–end organ axes that regulate the growth and maturation processes. Interrelationships among indicators and the tempo, timing, and sequence of maturational events are also considered. Environmental factors that influence the level of maturity at a given point in time and the process of maturation are also discussed: undernutrition, obesity, ethnic/racial background, social class, familial characteristics, climate, and altitude. Recommendations for the design of studies of maturational events are made, and an overview of secular changes before and after 1970 is provided. The review concludes with specific recommendations for the inclusion of a maturity indicator or maturity indicators in the construction of an international growth standard for preadolescent and adolescent children

Purpose: To verify the applicability of the prediction equation for maturity offset in a sample of female gymnasts followed longitudinally through adolescence. Methods: Fifteen gymnasts were followed longitudinally for 6-7 yr across adolescence. Weight, height, and sitting height were measured at annual intervals. The Preece-Baines Model I was fitted to longitudinal height data for individual gymnasts to derive age at peak height velocity (PHV). The curve-fitting protocol was successfully fit to the height records of 13 of the 15 gymnasts with standard errors of estimate between 0.02 and 0.28 cm. Maturity offset was calculated from measurements taken at each observation for the 13 gymnasts and also added to chronological age at each observation point to provide an estimated age at PHV. Age at PHV derived with the Preece-Baines model was used as the criterion. Differences between the criterion age at PHV and predicted age at PHV were calculated. Results: Maturity offset overestimates age at PHV in gymnasts. Mean predicted ages at PHV deviate linearly from the criterion age at PHV, but the difference is significant only at 9 yr. Correlations between maturity offset predicted ages at PHV and criterion age at PHV range from -0.13 to +0.76. The Bland-Altman plot of criterion and predicted ages at PHV suggest a systematic bias in the predictions. Conclusion: Maturity offset appears to have limitations when applied to female gymnasts. Care is warranted in utilizing maturity offset per se and predicted age at PHV based on maturity offset as an indicator of maturity timing in female gymnasts and perhaps other short females.

Tracking of Physical Fitness and Physical Activity from Youth to Adulthood in Females. Med. Sci. Sports Exerc., Vol. 38, No. 6, pp. 1114 – 1120, 2006. Purpose: To evaluate stability of physical fitness and physical activity from adolescence into middle adulthood in Flemish females. Methods: Within the scope of the Leuven Longitudinal Study on Lifestyle, Fitness and Health, 138 females (mean age = 16.6 T 1.1 yr) from the Leuven Growth Study of Flemish Girls were seen in adulthood (mean age = 40.5 T 1.1 yr). Several body dimensions and motor fitness tests were taken. Physical activity was assessed by means of a sports participation inventory. Interage correlations were calculated between adolescent and adult values. Cross-tabulation was used to identify the percentage of subjects remaining in the same BMI and physical activity group or shifting from one group to another from adolescence to adulthood. Odds ratios for less activity and overweight in adulthood according to adolescent activity or weight status were calculated. Results: Except for flamingo balance, plate tapping, leg lifts, and arm pull, all anthropometric and physical fitness characteristics were stable from adolescence to adulthood (r ranging from 0.49 to 0.96). Sports participation was not a stable characteristic (r = 0.13). From adolescence to adulthood, 84.5 and 63.6%, respectively, remained in the normal-weight and overweight group, whereas 62.5 and 54.4%, respectively, remained in the less active and active group. The odds of being overweight in adulthood was 9.53 (95% CI: 3.1–29.8) times greater in overweight compared with normal-weight adolescent girls. Conclusion: In Flemish females, anthropometric and fitness characteristics demonstrate higher levels of stability from adolescence to middle adulthood than physical activity. Weight status during adolescence is indicative of adult weight status, and a pattern of less activity rather than activity tends to continue

The purpose of this study was to determine whether the observed phenotypic stability in static strength during adolescence, as measured by interage correlations in arm pull, is mainly caused by genetic and/or environmental factors. Subjects were from the Leuven Longitudinal Twin Study ( n = 105 pairs, equally divided over 5 zygosity groups). Arm-pull data were aligned on age at peak height velocity to attenuate the temporal fluctuations in interage correlations caused by differences in timing of the adolescent growth spurt. Developmental genetic models were fitted using structural equation modeling. After the data were aligned on age at peak height velocity, the annual interage correlations conformed to a quasi-simplex structure over a 4-yr interval. The best-fitting models included additive genetic and unique environmental sources of variation. Additive genetic factors that already explained a significant amount of variation at previous measurement occasions explained 44.3 and 22.5% of the total variation at the last measurement occasion in boys and girls, respectively. Corresponding values for unique environmental sources of variance are 31.2 and 44.5%, respectively. In conclusion, the observed stability of static strength during adolescence is caused by both stable genetic influences and stable unique environmental influences in boys and girls. Additive genetic factors seem to be the most important source of stability in boys, whereas unique environmental factors appear to be more predominant in girls.

This study reports the results of a multipoint linkage study that aims to unravel the genetic basis of muscle strength and muscle mass in humans. Myostatin ( GDF8) is known to be a strong inhibitor of muscle growth in animals. However, studies examining human myostatin polymorphisms are rare and are limited to the GDF8 gene itself. Here, the contribution to isometric and concentric knee strength of nine key proteins involved in the myostatin pathway is studied in a nonparametric multipoint linkage analysis by means of a variance components and regression method. A sample of 367 healthy young male siblings was phenotyped on an isokinetic dynamometer and genotyped for markers of the myostatin pathway genes. Three of the loci were found significantly linked with a quantitative trait locus (QTL) for knee muscle strength. First, D13S1303 showed replication of an explorative single-point linkage study with a maximum LOD score of 2.7 ( P = 0.0002). Second, maximum LOD scores of 3.4 ( P = 0.00004) and 3.3 ( P = 0.00005) were observed for markers D12S1042 and D12S85, respectively, at 12q12–14. Finally, marker D12S78 showed an LOD score of 2.7 at 12q22–23. We conclude that several genes involved in the myostatin pathway, but not the myostatin gene itself, are important QTLs for human muscle strength. An additional set of valuable candidate genes that were not part of the myostatin pathway was found in the chromosome 12 and 13 genomic regions.

Objectives Three questions were addressed: (1) Do female gymnasts have adolescent growth spurts in height, sitting height, and leg length? (2) Are the sequence and magnitude of spurts comparable with female adolescent non-athletes? (3) How do the data compare with other female gymnasts and with short girls? Study design Height and sitting height were measured annually on 15 Belgian gymnasts from 8.7 ± 1.5 to 15.5 ± 1.5 years. The gymnasts trained, on average, approximately 15 h/wk. Leg length was estimated as height minus sitting height. The Preece-Baines Model I was fitted to individual growth records to estimate ages at peak velocity and peak velocities for the three dimensions. Age at menarche and skeletal age were also assessed. Results Gymnasts have clearly defined adolescent spurts in height, estimated leg length, and sitting height that occur approximately 1 year later and are slightly less intense than in nonathletic adolescent girls. Age at menarche and skeletal age are consistent with later somatic maturation. The pattern of adolescent growth and maturation is similar to that of other gymnasts, short normal late-maturing girls, and late-maturing girls with short parents. Conclusions The results emphasize a primary role for constitutional factors in the selection process of female gymnasts at relatively young ages.

Previous studies have shown that low birth weight (LBW) is a risk factor for renal impairment in adult life. The effects of LBW and renal function were studied by using twins, which allows distinguishing among fetoplacental, maternal, and genetic influences. Perinatal data were obtained at birth, and absolute creatinine clearance (not corrected for body surface area) was measured at a mean age of 25.6 yr in 653 individuals. Twins were considered both as individuals and as members of twin pairs. Statistical analyses were performed with and without adjusting for gestational age, zygosity, gender, age, body mass index, glucose level, BP, and smoking status. However, the intra-uterine environment may differ according to the chorionicity of the monozygotic twins, which may have biased previous heritability estimates of blood pressure. OBJECTIVE: The aim of the present study was to assess whether the heritability of blood pressure, derived from measurements in monozygotic and dizygotic twins, differs according to the chorionicity of the monozygotic twins. METHODS: Conventional and 24-h ambulatory blood pressures were measured in 125 dizygotic twin pairs and in 97 dichorionic and 128 monochorionic monozygotic twin pairs at the age of 18-34 years. The twin sample was drawn from the East Flanders Prospective Twin Survey, in which perinatal data were collected at birth. Intra-pair correlation coefficients were calculated and compared between both types of monozygotic twin pairs. Heritability was estimated from model-fitting and path analysis, based on the dizygotic twins and, respectively, all monozygotic twins and the two subtypes. RESULTS: Intra-pair correlation coefficients for the various blood pressures, after adjustment for body mass index, ranged from 0.45 to 0.71 in the monozygotic twin pairs and did not differ significantly according to chorionicity. Heritability estimates of blood pressure were between 52 and 64%, and were similar when calculated from dizygotic twins and, respectively, dichorionic and monochorionic monozygotic twins. CONCLUSIONS: Heritability estimates of conventional and ambulatory blood pressure do not differ significantly according to the chorionicity of the monozygotic twins.

The purpose of this study was to investigate stability and change in different expressions of strength development in adolescent boys using structural equation modeling. Three models were used: Markov simplex to study stability or tracking, Wiener or random walk to investigate fanning-out or spread effects in change, and latent growth to study differences in individual pathways of change as well as group changes. In the Leuven Growth Study, 588 male subjects were followed for 6 years with a mean age of 12.7 years at the onset of the study. Vertical jump, arm pull, and bent arm hang were used to mark the following strength factors: explosive strength, static strength, and functional strength. All models were tested with robust estimation procedures based on the software EQS 6.0. Main results and conclusions are as follows: 1) all strength factors showed moderate to high tracking, with low values of instability in relative position of the subjects in their developmental channels; 2) the fanning-out effect is not obvious, although some evidence showed a spread effect in functional and explosive strength; 3) there are marked interindividual differences in developmental pathways of strength manifestations; 4) strength development is linear and also has some curvilinearity, something akin to a breaking effect; 5) linear trend is negatively correlated with initial status and the leveling-off effect is also negatively correlated with the linear change.

The aim of this study is to quantify the genetic and environmental variation in isometric and explosive strength (power) in children and adolescents, using structural equation models. Arm pull (static strength) and vertical jump (explosive strength, power) were measured in 105 twin pairs from the Leuven Longitudinal Twin Study. Boys and girls were tested at annual intervals between 10 and 16 years and at 18 years. Path models were fitted to the observed strength characteristics and a gender heterogeneity analysis was performed at each age level. A model including additive genetic and specific environmental factors (AE-model) allowing for a difference in total phenotypic variance or in genetic/environmental variance components in boys and girls best explains both strength characteristics at most age levels. The additive genetic contribution for isometric strength varies between a2 = .44 and a2 = .83, and for explosive strength between a2 = .47 and a2 = .92, except at 16 years in males. In conclusion there is good evidence that during the growth period both static and explosive strength are under moderate to moderately strong genetic influence.

The aim of this study was to determine whether the use of third-generation OC has an impact on markers of CV disease in genetically identical women. We performed an intrapair comparison in 27 monozygotic twin pairs, one of whom was taking third-generation OC, whereas the other was not using OC. Biometric parameters were ascertained and conventional and 24-h ambulatory blood pressure (BP) was recorded. A fasting blood sample was taken for the measurement of glucose, insulin, proinsulin, lipids, and insulin-like growth factor binding protein-1 (IGFBP-1). Insulin resistance and beta-cell function were calculated by homeostasis model assessment (HOMA). A 24-h urine sample for cortisol was obtained. Third-generation OC use increased 24-h ambulatory systolic and diastolic BP by 5.2 and 3.9 mmHg, respectively (both P=0.0003). There was no effect on glucose, insulin and proinsulin levels, and on HOMA parameters, but the IGFBP-1 levels were markedly raised (P=0.0009). The lipid profile showed a 34% increase in triglyceride levels (P < 0.0001), but also a 7% increase in HDL-cholesterol levels (P=0.037). Use of third-generation OC impacts on CV disease markers in young-adult genetically identical women. Some changes are beneficial (increased HDL-cholesterol), whereas others may be deleterious (increased BP and triglyceride levels) or have unknown effects at this time (increased IGFBP-1 levels).

Within the context of the effects of training for sports on growth and maturation, there is very little menarcheal data for elite rowing athletes. Knowledge of the relationship of the maturational status with training level, different boat categories, and somatic features of the athletes will clarify the assumed impact of rowing training on the growth and maturational process of youngsters.The aim of this study was to determine the age at menarche in world top junior rowing athletes and to investigate its relationship with anthropometric characteristics, and competition level, rowing style and boat category.The sample consisted of 212 female junior rowers, with a mean chronological age of 17.6 +/- 0.8 years, all participants at the 1997 FISA World Junior Rowing Championships. Anthropometric dimensions, somatotype and body composition characteristics were measured, and age at menarche and training data were retrospectively obtained by questionnaires.Results revealed that the mean age at menarche of the total group of rowers was 12.8 +/- 1.2 years and did not differ from a non-athletic reference population. Rowers who started their rowing training before menarche (n = 78) showed a significant (p/= 0.01) later age at menarche compared with rowers who started their training after menarche (n = 134), with mean ages of 13.4 and 12.4 years, respectively. No significant relationship between the age at menarche and physical and body composition characteristics could be demonstrated, with r varying between -0.11 and 0.11. Furthermore, no significant differences in ages at menarche between competition levels (finalists versus non-finalists/medallists versus non-medallists) and between the different boat categories could be observed.Based on the results of this study, there is no direct evidence to state that intensive rowing training has a negative influence on the maturation status of junior female athletes.

Skeletal maturity and physical fitness of children and youth from the Autonomous Region of Madeira. The general aim of this mixed longitudinal study is to demonstrate dif- ferences in fitness levels between contrasting skeletal maturity groups for children and youth from the Autonomous Region of Madeira. A representative sample of 507 students of both sexes aged 8, 10, 12, 14 and 16 years has been observed at annual intervals for a period of three years. Several fitness components were observed by means of the Eurofit test battery. Skeletal maturity was assessed using the Tanner-Whitehouse Mark II system. ANOVA and the post-hoc Tukey tests were used to verify the statistical significance of the observed differences. The results of this study show that (1) boys and girls who mature ear- lier are stronger than their average and late-maturing peers. This is probably partly explained by the bigger body size and muscular mass of the early-maturing youngsters; (2) no differences between maturity groups were observed for plate tapping and shuttle run tests; (3) for girls the absence of statistical significant differences also applies for sit and reach, standing broad jump, sit ups and 12-minute run/walk; (4) boys (10-11 years of age) and girls (7-9 and 10-11 years of age) who mature later are more proficient than early- and average- maturing youngsters for bent arm hang. Again, the higher body mass of early matures, exerting a negative influence in the performance of this test, can partly explain this negative association. At the age of 14-15 the direction of the relationship changes, and early-maturing boys show better results than average-and late-maturing boys. At these age levels, the larger increase in strength that accompanies male adolescence com- pensates the higher body mass; (5) boys and girls who mature late achieved better results than early ones for flamingo balance.

Heritability of somatotype components from early adolescence into young adulthood: a multivariate analysis on a longitudinal twin study.Peeters MW, Thomis MA, Claessens AL, Loos RJ, Maes HH, Lysens R, Vanden Eynde B, Vlietinck R, Beunen G.Faculty of Physical Education and Physiotherapy, Katholieke Universiteit Leuven, Leuven, Belgium.BACKGROUND: Several studies with different designs have attempted to estimate the heritability of somatotype components. However they often ignore the covariation between the three components as well as possible sex and age effects. Shared environmental factors are not always controlled for. AIM: This study explores the pattern of genetic and environmental determination of the variation in Heath-Carter somatotype components from early adolescence into young adulthood. SUBJECTS AND METHODS: Data from the Leuven Longitudinal Twin Study, a longitudinal sample of Belgian same-aged twins followed from 10 to 18 years (n = 105 pairs, equally divided over five zygosity groups), is entered into a multivariate path analysis. Thus the covariation between the somatotype components is taken into account, gender heterogeneity can be tested, common environmental influences can be distinguished from genetic effects and age effects are controlled for. RESULTS: Heritability estimates from 10 to 18 years range from 0.21 to 0.88, 0.46 to 0.76 and 0.16 to 0.73 for endomorphy, mesomorphy and ectomorphy in boys. In girls, heritability estimates range from 0.76 to 0.89, 0.36 to 0.57 and 0.57 to 0.76 for the respective somatotype components. Sex differences are significant from 14 years onwards. More than half of the variance in all somatotype components for both sexes at all time points is explained by factors the three components have in common. CONCLUSIONS: The finding of substantial genetic influence on the variability of somatotype components is further supported. The need to consider somatotype as a whole is stressed as well as the need for sex- and perhaps age-specific analyses. Further multivariate analyses are needed to confirm the present findings.

Relationships between skeletal maturation and fundamental motor skills and gross motor coordination were evaluated in 429 children (213 boys and 216 girls) 7-10 years. Skeletal age was assessed (Tanner-Whitehouse 2 method), and stature, body mass, motor coordination (Körperkoordinations Test für Kinder, KTK) and fundamental motor skills (Test of Gross Motor Development, TGMD-2) were measured. Relationships among chronological age, skeletal age (expressed as the standardised residual of skeletal age on chronological age) and body size and fundamental motor skills and motor coordination were analysed with hierarchical multiple regression. Standardised residual of skeletal age on chronological age interacting with stature and body mass explained a maximum of 7.0% of the variance in fundamental motor skills and motor coordination over that attributed to body size per se. Standardised residual of skeletal age on chronological age alone accounted for a maximum of 9.0% of variance in fundamental motor skills, and motor coordination over that attributed to body size per se and interactions between standardised residual of skeletal age on chronological age and body size. In conclusion, skeletal age alone or interacting with body size has a negligible influence on fundamental motor skills and motor coordination in children 7-10 years.

The association between bone mass, body structure, and body composition was explored in 156 men, 40 years of AGE. Bone mineral density (total body, lumbar spine, left arm, and left leg) was obtained by dual-energy X-ray absorptiometry (DXA; Hologic QDR 4500A). Body structure was determined from a battery of anthropometric dimensions with a principal components analysis. Body composition was estimated with DXA. From the 24 anthropometric dimensions, three components were extracted and identified as muscle, fat, and skeletal length. Significant correlations between the muscle component and all BMD measurements (r = 0.28–0.44) were obtained. Except for BMD of the left arm, which correlated significantly, but negatively, with the fat component (r = −0.16), no significant relations were found between the fat component and BMD. There were significant correlations between lean mass, fat mass, and BMD measurements. The correlations were higher between lean mass and BMD (r = 0.22–0.44) than between fat mass and BMD (r = 0.08–0.24). The multiple regression analysis revealed that except for BMD of the left arm only lean mass or the muscle component, and not fat mass or the fat component, were independent predictors of BMD. Background: Substantial interindividual variation is observed in sports participation and physical activity levels in youth. This study aimed to (1) estimate the relative contribution of genes, along with shared and nonshared environmental factors, to variation in sports participation index (SPI) and leisure-time physical activity (LTPA); and (2) test differences in those factors in males and females. Methods: The sample was comprised of 411 Portuguese twin pairs of different zygosity aged 12 to 25 years. The SPI and LTPA were assessed with the Baecke questionnaire. Quantitative genetic modeling was used to test alternative models for the presence of additive gene effects (a2), common or shared environment within the family (c2), and unique environmental factors (e2). Results: The best-fitting models showed sex-specific effects for the two phenotypes. Variance components for SPI in males were a2=68.4%, c2=20%, and e2=11.6%; and in females, a2=39.8%, c2=28.4%, and e2=31.8%. For variation in LTPA, genetic factors in males explained 63%, common environment was not significant, and unique environment explained 37%. In females, contributing factors were a2=32%, c2=38%, and e2=30%. Conclusions: Genetic effects explained a considerable amount of variation in SPI and LTPA, which were greater in males than in females. The relevance of shared environmental factors (family and peers) and nonshared environmental factors in SPI and LTPA is particularly evident in females.

This study investigated the relationship between sports participation/physical activity during youth (13 - 18 years of age) and adulthood (30 - 40 years of age), and cardiovascular risk factors (body fat and fat distribution, blood pressure, lipoprotein levels and cardiorespiratory fitness) at 40 years of age. Subjects were 166 Flemish males from "The Leuven Longitudinal Study on Lifestyle, Fitness and Health". Physical activity was assessed by means of a sports participation inventory and the Tecumseh community Health Study Questionnaire. In addition to correlation and multiple stepwise regression analyses, different groups (at risk, not at risk) were contrasted on sports participation/physical activity parameters using ANOVA. Long-term exposure during adulthood to daily physical activity was slightly related to a low/high risk profile for waist circumference, percent body fatness, triglycerides and peak VO(2). Sports participation during adolescence was not related to levels of cardiovascular risk factors at 40 years of age.

Purpose: The range of variability between individuals of the same chronological age (CA) in somatic and biological maturity is large and especially accentuated around the adolescent growth spurt. Maturity assessment is an important consideration when dealing with adolescents, from both a research perspective and youth sports stratification. A noninvasive, practical method predicting years from peak height velocity (a maturity offset value) by using anthropometric variables is developed in one sample and cross-validated in two different samples. Methods: Gender specific multiple regression equations were calculated on a sample of 152 Canadian children aged 8-16 yr (79 boys; 73 girls) who were followed through adolescence from 1991 to 1997, The equations included three somatic dimensions (height, sitting height, and leg length), CA, and their interactions. The equations were cross-validated on a Combined sample of Canadian (71 boys, 40 girls measured from 1964 through 1973) and Flemish children (50 boys, 48 girls measured from 1985 through 1999). Results: The coefficient of determination (R2) for the boys' model was 0.92 and for the girls' model 0.91 the SEEs were 0.49 and 0.50, respectively, Mean difference between actual and predicted maturity offset for the verification samples was 0.24 (SD 0.65) yr in boys and 0,001 (SD 0.68) yr in girls. Conclusion: Although the cross-validation meets statistical standards or acceptance, caution 1, warranted with regard to implementation. It is recommended that maturity offset be considered as a categorical rather than a continuous assessment. Nevertheless, the equations presented are a reliable, noninvasive and a practical solution for the measure of biological maturity for matching adolescent athletes.

The present study has the following purposes: (1) to present and situate the studies made in the Community of Portuguese Speaking Countries and (2) to present some recommendations for future investigations. Seventeen studies are considered and the following conclusions are drawn: (1) almost the totality of the studies present a cross-sectional design; (2) in the Community of Portuguese Speaking Countries context, the height and weight of Portuguese and Brazilian children are nearly coincident while the youngsters of Mozambique are shorter and less heavy. The children from Cabo Verde and Mozambique have less subcutaneous fat than the Portuguese and Brazilian children. On sit and reach, Mozambique presents the best performances but the opposite holds true for sit ups. On 12-minute run/walk, Portuguese perform better than Brazilians; (3) in European context, boys and girls from Brazil and Mozambique are slightly shorter than Belgians. Children and youth from Mozambique are less heavy and have lower subcutaneous fat levels than Belgian children. On sit and reach Mozambique boys and girls perform better than their Belgian peers. Future growth studies need to consider to use: (1) longitudinal or mixed-longitudinal designs; (2) multidisciplinary research teams; (3) well defined methodologies in training of test team; (4) rigorous quality control during data collection and data entry; and (5) inclusion of parameters related to health.
