Abstract

International audience; Members of the Streptomyces genus are among the most prolific microorganisms producing secondary metabolites with wide uses in medicine and in agriculture. Sequencing of the genome of the model Streptomyces, Streptomyces coelicolor, has highlighted an unexpected feature, i.e. that the potential of these organisms to synthesise secondary metabolites has been largely underestimated. They indeed possess many more gene clusters encoding natural product-like biosynthetic pathways than there are known natural products. Similar observations have since been made for other bacterial or fungal genomes. Thus, it became clear that microbial secondary metabolism had been seriously underestimated and that genome-based approaches were very promising for the search of new bioactive compounds. Here, we present an overview of the secondary metabolite biosynthetic potential of Streptomyces ambofaciens, a species known for decades as producer of the macrolide spiramycin and the pyrrolamide congocidine. Interestingly, genome analysis has revealed that despite of the close phylogenetic relatedness between S. coelicolor and S. ambofaciens, most of its secondary metabolite gene clusters are species-specific.

Abstract

Campylobacter jejuni is a microaerophilic and thermophilic pathogen genetically close to Helicobacter pylori. It represents one of the most of concern emerging food pathogens, as it is one of the primary sources of human enteritis, in developed as well as in developing countries. It is also a source of rare but serious neuropathies. Interest in this bacterium has constantly been growing during the last decade, since many antibiotic resistant strains are emerging. The sequencing of C. jejuni genome has revealed a small genome in which a lot of regulation systems are fully understood in well-studied bacteria were absent. To infect humans, a pathogen has to survive different environmental stresses and attacks of the immune system, and also be able to express its virulence factors. The Campylobacter conundrum is the latter: how does this microaerophilic (=sensitive to oxygen) bacterium, owning a small genome and lacking most of the well-known regulation systems, still constitute a hazard for humans? From what is known about C. jejuni genome, this book proposes to draw a parallel between on the one hand the different mechanisms and regulation pathways existing, and on the other hand stress resistance and virulence of this emerging food pathogen.

Abstract

Extraintestinal pathogenic Escherichia coli (ExPEC) are an important cause of urinary tract infections, neonatal meningitis and septicaemia in humans. Animals are recognized as a reservoir for human intestinal pathogenic E. coli, but whether animals are a source for human ExPEC is still a matter of debate. Pathologies caused by ExPEC are reported for many farm animals, especially for poultry, in which colibacillosis is responsible for huge losses within broiler chickens. Cases are also reported for companion animals. Commensal E. coli strains potentially carrying virulence factors involved in the development of human pathologies also colonize the intestinal tract of animals. This review focuses on the recent evidence of the zoonotic potential of ExPEC from animal origin and their potential direct or indirect transmission from animals to humans. As antimicrobials are commonly used for livestock production, infections due to antimicrobial-resistant ExPEC transferred from animals to humans could be even more difficult to treat. These findings, combined with the economic impact of ExPEC in the animal production industry, demonstrate the need for adapted measures to limit the prevalence of ExPEC in animal reservoirs while reducing the use of antimicrobials as much as possible.
